응유효소 생산을 위한 Mucor mucedo C-7의 배양조건 (The Culture Conditions of Mucor mucedo C-7 for Producing the Milk-Clotting Enzyme)

  • 효과적인 응유효소 생산을 위해 응유효소 생산 균주로 선정된 Mucor mucedo C-7을 밀기울 고체배지에 배양하여 최적 배양 조건을 검토하였다. 그 결과 밀기울에 대한 첨수량은 밀기울량에 100% 첨수하였을때 효소 생산이 가장 양호하였으며 배양 은도와 시간은 각각 3$0^{\circ}C$, 72시간이 가장 적당함을 알 수 있었다. 또 밀기울 배지에 증류수 대신 Macllvaine 완충용액 (pH4.5)을 첨가 하였을 경우 효소 생산은 현저히 증가하였으며 응유효소의 활성도 중류수를 첨수한 경우 보다 안정함을 알 수 있었다.

Investigation of supplementation with a combination of fermented bean dregs and wheat bran for improving the growth performance of the sow

  • To investigate the effect of dietary supplementation with a fermented mixture of bean dregs and wheat bran (FBW) on sow performance. FBW was given to sows during late gestation and lactation; in total, 24 sows were randomly assigned to 4 groups (control diet; 3% FBW diet; 6% FBW diet; 9% FBW diet, n = 6). The weight ratio of bean dregs (wet) to wheat bran was 4:6. Sows were fed different diets from 85 d of gestation until weaning. The results showed that supplementation with FBW increased average daily feed intake (ADFI) during lactation (p < 0.05). FBW supplementation also increased litter weight and milk yield (p < 0.05). The contents of Escherichia coli in the feces of the treatment groups were significantly reduced by FBW supplementation (p < 0.01). FBW supplementation significantly improved the fecal morphology (p < 0.05), alleviating sows' constipation. In conclusion, FBW could increase the ADFI, improve lactation and piglet litter weight in sows and reduce the pathogenic bacterial content in sow feces and constipation.

Hot Water Extract of Wheat Bran Attenuates White Matter Injury in a Rat Model of Vascular Dementia

  • Vascular dementia is characterized by white matter lesions involving the demyelination and activation of astrocytes and microglia. In a previous study, we showed that the supernatant of a laboratory-scale, hot water extract of ground whole wheat (TALE) attenuated white matter injury and astrocytic activation in a rat model of bilateral common carotid artery occlusion (BCCAO). In the present study, we made several modifications to the hot water extraction process to remove starch and enable large-scale production. We used wheat bran (WB), which contains less starch, instead of ground whole wheat. In addition, we removed starch granules with a decanter before hot water extraction. The final product, wheat bran extract (WBE), contained 2.42% arabinose, a surrogate marker of arabinoxylan, which is an active constituent of WBE. Supplementation of the rat model of BCCAO with WBE (400 mg/kg/day) for 33 days attenuated white matter injury, which was assessed by Luxol Fast Blue staining, in the corpus callosum (cc) and optic tract (opt) regions. Attenuation of white matter injury in the opt region was accompanied by improvement of the pupillary light reflex. Immunochemical staining revealed that supplementation with WBE reduced astrocytic activation in the cc and opt regions and reduced microglial activation in the opt region. These findings indicate that supplementation with WBE is effective at attenuating white matter injury accompanied by the inhibition of astrocytic and microglial activation. Therefore, extracts from WB, a cheap by-product of wheat milling, can be developed as a nutraceutical to prevent vascular dementia, a disease for which there is no approved pharmaceutical treatment.

Chemoprevention of Azoxymethane Induced Colon Cancer in Rats by Feeding Grange Juice, Soy, Wheat Bran and Flaxseed

  • Epidemiologic studies consistently demonstrate an inverse relationship between risk for colon canter and consumption of fruits and vegetables. Wheat bran, flax and soy contain dietary fiber and phytochemicals, such as lignans and isoflavones, that may inhibit colon carcinogenesis. Orange juice contains hesperidin, a flavanone glucoside that protects against colon carcinogenesis. This study determined if feeding orange juice, wheat bran, soy and flaxseed (combined diet) would inhibit azoxymethane (AOM) induced colon cancer. Cancer was initiated in male Fisher 344 rats by injecting 15 mg AOM/kg of weight at 22 and 29 days of age. One week after the second AOM injection, rats (N = 30) in the combined diet group received dry diet containing wheat bran (4%), soy with ethanol soluble phytochemirals(13%) and flaxseed (8%) and orange juice replaced drinking water. The control group remained on the control diet and received distilled water to drink. The rats were killed 28 weeks later, and colon tissues and tumors were removed for histologic analysis. Feeding the combined diet significantly reduced tumor incidence (p < 0.05), however tumor multiplicity was not changed (p > 0.05, 0.9 tumors/rat fed the combined diet vs 1.2 for controls). Also, tumor burden was only marginally reduced in rats fed the combined diet vs control rats (65 vs 210 mg of tumor/rats, respectively). The reduction in tumor incidence was associated with a decreased labeling index and proliferation zone in normal appearing colon mucosa. Therefore, this study shows that phytochemicals in wheat bran, soy, flax and orange juice reduce colon carcinogenesis, presumably by decreasing cell proliferation and enhancing cell differentiation.

주정발효용(酒精醱酵用) 효소제(酵素劑) 배양제조(培養製造) 조건(條件)이 Amylase 활성(活性)에 미치는 영향(影響) (The Effect on the Culture Condition on the Activity of Amylase used for Alcohol Fermentation)

  • The culture used wheat bran as media for four kind of mold strains such as Aspergillus oryzae, Aspergillus kawachii, Aspergillus usamii and Rhizopus javanicus to examine which strain could higher the activity of amylase most which is used for alcohol fermentation. It also provided three different kind of wheat bran media containing starch of 47%, 51% and 55% respectively for each strain. For each media it also added three different kind of nitrogen sources; ammonium sulfate, casein, and ammonium sulfate and casein equally mixed. Each nitrogen source added was subordinately differentiated into three different percentages, 2%, 4% and 6% respectively, except the 2% for the ammonium sulfate. The results obtained were summarized as follows (1) The activity of ${\alpha}-amylase$ was highest in the media of starch value 47% of wheat bran with 6% of casein added. (2) The activity of ${\beta}-amylase$ was highest in the media of starch value 51% of wheat bran with 2% of the equal mixture of ammonium sulfate and casein added. (3) The activities of both ${\alpha}-amylase$ and ${\beta}-amylase$ of Aspergillus usamii were highest in the media of starch value 47% wheat bran with no addition of nitrogen source. (4) Of the four strains examined, the activities of ${\alpha}-amylase$ and ${\beta}-amylase$ cultured in Rhizopus javanicus were both relatively higher. (5) The activities of ${\alpha}-amylase$ and ${\beta}-amylase$ of the strains examined became lower as the percentage of starch contents increased except in Rhizopus javanicus.

열처리 밀기울의 수용성 분획의 특징 (Characteristics of Water Soluble Fractions of Wheat Bran Treated with Various Thermal Processes)

  • Autoclaving, microwaving, 압출성형 등의 열 및 기계적 에너지에 대한 밀기울 세포벽의 반응성을 구명하기 위하여 수용성 분획의 특정을 연구하였다. 수용성 분획의 화학적 분석과 겔여과 크로마토그래피 결과에 의하면 밀기울에 포함되어 있는 전분의 호화가 가장 중요한 수용화 기작으로 작용하며, 이 과정에서 비전분성 세포벽 구성물질의 수용화도 일어냐는 것으로 밝혀졌다. 수용성의 비전분 다당류의 GC 분석결과 적용된 열처리 과정에서 세포벽의 arabinoxylan 부위가 많이 수용화되는 현상을 보였다. 특히, 고온파 고전단력을 동시에 제공하는 압출성형은 밀기울의 수용화에 가장 효율적이었으며, 또한 상대적으로 고분자량의 비전분성 구성성분을 수용 화시켰다.

Effects of brewer's spent grain on the growth and nutrition of the giant mealworm beetle, Zophobas atratus

  • The giant mealworm beetle, Zophobas atratus (Coleoptera: Tenebrionidae), is a tropical beetle. As this beetle can be used as an ingredient in animal feed, the effects of brewer's spent grain (BSG) on the development and nutritional value of Z. atratus were investigated. As results, there were no significant differences on the larval survival rate, body weight, and duration at different content of BSG, mixed with conventional feed, wheat bran (WB). Based on these results, BSG can be mixed with wheat bran as a Z. atratus food ingredient without any problems. The nutritional value was compared between 100% wheat bran control and 50% BSG experimental groups. The moisture, crude protein, carbohydrates, and amino acids were 1.1-1.4 times higher in the 50% BSG group. In the BSG group, the linoleic acid content was 1.6 times higher than that in the control group. In the wheat bran group, the oleic acid content was 38.4%, which was 1.3 times higher than that in the BSG group. As minerals, the control and BSG groups showed high potassium and phosphorus contents. In terms of hazardous materials, four heavy metals (lead, mercury, arsenic, and cadmium) and two microorganisms (Escherichia coli and Salmonella spp.) were not detected. There were no significant differences in developmental characteristics between the wheat bran and BSG mixed groups, and the nutritional values were better in the BSG mixed group. Therefore, BSG can be used as alternative food source for rearing Z. atratus.

Aspergillus niger 의 고체상태 발효 시스템에서의 β-Glucosidase 회수 (β-Glucosidase Recovery from a Solid-State Fermentation System by Aspergillus niger)

  • 밀기울 발효에서 효소회수의 모델로 Aspergillus niger를 고체상태로 발효시켜 조사하였다. 발효시킨 밀기울에서 증류수로 효소추출 효율은 초산 완충액, 구연산 완충액, 구연산-인산 완충액 및 5% 메탄올 처리보다 높았다. 따라서, 추출 용매로 증류수를 이용하여 최적 조건을 상세히 검토하였다. 최적 조건은 고체와 액체 용매를 1:5의 비율로 증류수로서 세 번 세척하였을 때에 최대 회수율을 0.025 U/g으로 확보하였다.

Aspergillus 속 곰팡이를 이용한 액체종국 제조 및 밀누룩의 품질특성 (Quality Characteristics of wheat Nuruk and Optimum Condition of Liquid Starters for Aspergillus sp.)

  • 고역가 액체종국 제조 및 배양기술을 개발하기 위해, Aspergillus 속의 곰팡이(A. oryzae, A. niger)를 이용한 밀기울 첨가율 별(0, 5, 10, 15 및 20%) 배지를 이용한 액체종국을 제조하여 이들의 품질 특성을 조사하였다. 액체종국의 일반성분 및 효소활성(glucoamylase, acidic protease)을 조사한 결과, 배지에 밀기울 첨가량이 많을수록 균사체량이 많아졌을 뿐만 아니라 산도, 아미노산도도 증가하는 것을 알 수 있었고 액체종국에 첨가하는 밀기울 농도가 달라짐에 따라 각 균주의 효소활성이 바뀌는 것을 알 수 있었다. 또한, 효소활성에 따른 최적 배양시간은 곰팡이 종류에 따라 차이가 있었고 A. oryzae와 A. niger 곰팡이의 액체종국 배양은 10~15% 밀기울을 첨가하여 48~72시간 배양하는 최적조건을 규명하였다. 기존에 사용되었던 고체종국과 비교하여 밀입국제조에 종국으로써 적합한지를 구명하고자 A. oryzae와 A. niger를 사용한 고체종국을 접종한 밀누룩을 대조구로 하여 밀기울 10% 액체배지로 만든 액체종국의 접종량 별(1, 3, 5, 10%) 밀누룩의 배양시간 별(0, 15, 20, 36, 38, 40 hrs) 및 이화학적 특성과 효소활성(${\alpha}$-amylase, glucoamylase, acidic protease)변화를 비교하였다. 결론적으로 시간과 인력등의 경제적 가치가 많이 투입되는 고체종국 보다는 누룩 제조시 액체종국을 사용하는 것이 여러 가지 측면에서도 유리 하다고 여겨진다.

Bacillus subtilis 분리균의 Mannanase와 Xylanase 생산성과 효소 특성 (Production and Properties of Mannanase and Xylanase by a Bacillus subtilis Isolate)

  • 새만금 갯벌로부터 xylan과 locust bean gum (LBG)을 분해하는 미생물을 분리하여 그 생화학적 특성과 16S rRNA 서열을 조사한 결과 Bacillus subtilis로 확인되었다. 분리균 YB-30은 konjac이나 LBG가 존재한 상태에서 배양할 경우 mannanase의 생산성이 급격하게 증가되며, 밀기울이 존재하에서는 xylanase의 생산성이 증가되였다. Konjac (3.5%)과 밀기울(1%)이 동시에 첨가된 배지에서 균의 성장이 정지기에 진입하였을 때 mannanase와 xylanase의 생산성이 최고 수준에 도달하였다. 배양상등액의 mannanase와 xylanase는 60℃와 pH 6.0, 55℃와 pH 5.5에서 각각 최대 활성을 보였다. 고온에서는 두 효소 모두 안정하지는 않았으며 xylanase가 mannanase보다 안정성이 낮았다. 밀기울은 쌀기울보다 많은 양의 arabinoxylan을 함유하고 있으므로 배양상등액으로 분해하였을 때 밀기울로부터 생성되는 환원당의 양이 더 많은 것으로 확인되었으며, 이로 보아 B. subtilis YB-30에 의해 생산되는 xylanase와 mannanase는 사료첨가용 효소로 활용 가능성이 높다고 여겨진다.
