Molecular Control of Gene Co-suppression in Transgenic Soybean via Particle Bombardment

  • Molecular co-suppression phenomena are important to consider in transgene experiments. Embryogenic cells were obtained from immature cotyledons and engineered with two different gene constructs (pHV and pHVS) through particle bombardment. Both constructs contain a gene conferring resistance to hygromycin (hpt) as a selective marker and a modified glycinin (11S globulin) gene (V3-1) as a target. sGFP(S65T) as a reporter gene was, however, inserted into the flanking region of the V3-1 gene (pHVS). Fluorescence microscopic screening after the selection of hygromycin, identified clearly the expression of sGFP(S65T) in the transformed soybean embryos bombarded with the pHVS construct. Stable integration of the transgenes was confirmed by polymerase chain reaction (PCR) and Southern blot analysis. Seeds of transgenic plants obtained from the pHV construct frequently lacked an accumulation of endogenous glycinin, which is encoded by homologous genes to the target gene V3-1. Most of the transgenic plants expressing sGFP(S65T) showed highly accumulation of glycinin. The expression of sGFP(S65T) and V3-1 inherits into the next generations. sGFP(S65T) as a reporter gene may be useful to increase the transformation efficiency of transgenic soybean with avoiding gene co-suppression.

T7 RNA Polymerase Is Expressed in Plants in a Nicked but Active Form (T7 RNA polymerase 유전자의 담배식물에서의 발현)

  • We have prepared several chimeric constructs containing the bacteriophage T7 RNA polymerase gene under control of the wound-inducible potato proteinase inhibitor II (pin2) promoter and have transformed Nicotiana tabacum plants with these constructs. Southern blot analyses indicate that either one or two copies of the gene constructs are present in the transgenic plants. Northern blot analyses indicate that mRNA encoding T7 RNA polymerase is expressed in a wound-inducible manner. We purified T7 RNA polymerase and prepared antiserum. This antiserum was used for Western blot analyses to demonstrate that a protein which is cross reactive with T7 RNA polymerase is produced. The molecular mass of this protein is 80 kDa, a size which is consistant with the nicked form of the polymerase as is often seen when expressed in E. coli. RNA polymerase assays were used to indicate that the nicked form of T7 RNA polymerase is active and capable of incorporating labeled nucleotides into transcripts in vitro. Analysis of transgenic plants did indeed show that wound-inducible activation of the T7 RNA polymerase permits the establishment of a genetic system to overexpress genes in plants using T7 RNA polymerase(Received March 20, 1997; accepted May 2, 1997)

Modification of Turnip yellow mosaic virus coat protein and its effect on virion assembly

  • Turnip yellow mosaic virus (TYMV) is a positive strand RNA virus. We have modified TYMV coat protein (CP) by inserting a c-Myc epitope peptide at the N- or C-terminus of the CP, and have examined its effect on assembly. We introduced the recombinant CP constructs into Nicotiana benthamiana leaves by agroinfiltration. Examination of the leaf extracts by agarose gel electrophoresis and Western blot analysis showed that the CP modified at the N-terminus produced a band co-migrating with wild-type virions. With C-terminal modification, however, the detected bands moved faster than the wild-type virions. To further examine the effect, TYMV constructs producing the modified CPs were prepared. With N-terminal modification, viral RNAs were protected from RNase A. In contrast, the viral RNAs were not protected with C-terminal modification. Overall, the results suggest that virion assembly and RNA packaging occur properly when the N-terminus of CP is modified, but not when the C-terminus is modified.

A Study on Conceptions of Giftedness in Arts (예술영재성의 의미와 특성: 음악, 전통예술, 발레 분과를 중심으로)

  • Exploring the conceptions of giftedness in arts is very important as the essential prerequisite to identification of giftedness and organizing the curriculum for the gifted in arts. So I have explored the conceptions of giftedness in western music, korean traditional arts(music, theatricals, dance) and ballet based on the results of various studies and cases. The three fields of performing arts mentioned above are operating in the present fields on gifted education in Korea. As the result of this study, I have converged three or four factors on main psychological constructs of giftedness in arts. The giftedness in western and korean traditional music is composed of three sub-factors: high ability in relevant domain, high creativity in general and specific areas, and high motivation including task commitment. However, the giftedness in traditional theatricals, dance, and ballet consists four sub-factors: the physical condition, high ability in relevant domain, high creativity in a specific area, and high motivation including task commitment. The internal characteristics of the gifted in arts mentioned above can be maximized through supporting external conditions like social environments including family and school, and crucial opportunity to meet the teacher who affects the development of giftedness.

  • This paper explores critically and historically some of the popular academic views concerning the development of the study of Southeast Asia through the lens of the contributions of particular scholars and institutions. Within the broad field of Southeast Asian Studies the focus is on the disciplines of geography, history and ethnology. There are certain views concerning the development of scholarship on Southeast Asia which continue to surface and have acquired, or are in the process of acquiring "mythical" status. Among the most enduring is the claim that the region is a post-Second World War construction primarily arising from Western politico-strategic and economic preoccupations. More specifically, it is said that Southeast Asian Studies for a considerable period of time has been subject to the American domination of this field of scholarship, located in programs of study in such institutions as Cornell, Yale and California, Berkeley, and, within those institutions, focused on particular scholars who have exerted considerable influence on the directions which research has taken. Another is that, based on the model or template of Southeast Asian Studies (and other area studies projects) developed primarily in the USA, it has distinctive characteristics as a scholarly enterprise in that it is multidisciplinary, requires command of the vernacular, and assigns special importance to what has been termed 'groundedness' and historical, geographical and cultural contextualization; in other words, a Southeast Asian Studies approach as distinct from disciplinarybased studies addresses local concerns, interests, perspectives and priorities through in-depth, on-the-ground, engaged scholarship. Finally, views have emerged that argue that a truly Southeast Asian Studies project can only be achieved if it is based on a set of locally-generated concepts, methods and approaches to replace Western ethnocentrism and intellectual hegemony.

  • With the continued integration through scientification and the progress I the technology of construction around the world Korea's inability to stay within the mainstream of the movement has put it in danger of losing it's very identity. As such Korean cityscapes are filled with overwhelming prevalence of buildings and constructs void of national identity but rather reflective of unconditional application of the western standards of architecture which has so far been the compass of the movement. Accordingly the purpose of this research is the understanding of the current world view concerning of architecture the critical regionalism through the examination of the background of theory which reflect the architectural Kenneth Prampton and also through Ricardo Legorrata Charles Correa Antoine Predock's analysis. Furthermore this research hopes to explore and offer a methodology through which a unique Korean identity can be established in Korea's Modern architecture.

National Culture and Relational Selling: Antecedents, Outcomes and Boundary Conditions of ASB and Customer-Oriented Selling in Korea

  • Globalization and the emergence of new markets has placed increased emphasis on cross-cultural sales and marketing research. Despite considerable advances in the personal selling literature, little is known about the cross-cultural transferability of key constructs. Given the degree to which well-accepted relational sales behaviors such as ASB and customer oriented selling reflect Western values such as individualism and low uncertainty avoidance, the relative efficacy of such practices in alternative cultural context is less clear. Using a Korean sample, our results confirm the beneficial direct effects of these relational selling practices upon performance. However, we also find strong workgroup interdependence diminishes the effects of ASB and that performance fully mediates the relationship between both sales behaviors and job satisfaction. In contrast to prior results, we find female salespeople and those newer to their position demonstrate stronger ASB.

  • To investigate the region of ErbB2 for the $ErbB2-{\beta}-catenin$ interaction, a proteasome $resistant-{\beta}-catenin$ and various ErbB2 constructs were transfected in COS7 cells. ErbB2 proteins were immunoprecipitated, and coimmunoprecipitated ${\beta}-catenin$ was examined by Western blotting. ${\beta}-catenin$ coimmunoprecipitated with full length ErbB2. Of the truncated ErbB2 proteins DT (1-1123), DHC (1-1031) and DK (1-750), the ErbB2 constructs containing the kinase domain, DT and DHC, precipitated together with ${\beta}-catenin$ but DK containing no kinase domain did not. To further test the requirement of the kinase domain for ${\beta}-catenin-ErbB2$ interaction, the presence of ${\beta}-catenin$ in the immunocomplex was examined following transfection with an ErbB2 mutant (${\triangle}750-971$) whose kinase domain is internally deleted and subsequent immunoprecipitation of the ErbB2 mutant. ${\beta}-catenin$ was not detected in the immunocomplex. These results suggest that the ErbB2 kinase domain comprises a potential site for ${\beta}-catenin$ binding to the receptor tyrosine kinase.

A Consensus Technique for Tropical Cyclone Intensity Prediction over the Western North Pacific (북서태평양 태풍 강도 예측 컨센서스 기법)

  • In this study, a new consensus technique for predicting tropical cyclone (TC) intensity in the western North Pacific was developed. The most important feature of the present consensus model is to select and combine the guidance numerical models with the best performance in the previous years based on various evaluation criteria and averaging methods. Specifically, the performance of the guidance models was evaluated using both the mean absolute error and the correlation coefficient for each forecast lead time, and the number of the numerical models used for the consensus model was not fixed. In averaging multiple models, both simple and weighted methods are used. These approaches are important because that the performance of the available guidance models differs according to forecast lead time and is changing every year. In particular, this study develops both a multi-consensus model (M-CON), which constructs the best consensus models with the lowest error for each forecast lead time, and a single best consensus model (S-CON) having the lowest 72-hour cumulative mean error, through on training process. The evaluation results of the selected consensus models for the training and forecast periods reveal that the M-CON and S-CON outperform the individual best-performance guidance models. In particular, the M-CON showed the best overall performance, having advantages in the early stages of prediction. This study finally suggests that forecaster needs to use the latest evaluation results of the guidance models every year rather than rely on the well-known accuracy of models for a long time to reduce prediction error.

Analysis of C. elegans VIG-1 Expression

  • Double-stranded RNA (dsRNA) induces gene silencing in a sequence-specific manner by a process known as RNA interference (RNAi). The RNA-induced silencing complex (RISC) is a multi-subunit ribonucleoprotein complex that plays a key role in RNAi. VIG (Vasa intronic gene) has been identified as a component of Drosophila RISC; however, the role VIG plays in regulating RNAi is poorly understood. Here, we examined the spatial and temporal expression patterns of VIG-1, the C. elegans ortholog of Drosophila VIG, using a vig-1::gfp fusion construct. This construct contains the 908-bp region immediately upstream of vig-1 gene translation initiation site. Analysis by confocal microscopy demonstrated GFP-VIG-1 expression in a number of tissues including the pharynx, body wall muscle, hypodermis, intestine, reproductive system, and nervous system at the larval and adult stages. Furthermore, western blot analysis showed that VIG-1 is present in each developmental stage examined. To investigate regulatory sequences for vig-1 gene expression, we generated constructs containing deletions in the upstream region. It was determined that the GFP expression pattern of a deletion construct (${\Delta}-908$ to -597) was generally similar to that of the non-deletion construct. In contrast, removal of a larger segment (${\Delta}-908$ to -191) resulted in the loss of GFP expression in most cell types. Collectively, these results indicate that the 406-bp upstream region (-596 to -191) contains essential regulatory sequences required for VIG-1 expression.
