Transcriptomic Analysis of Cellular Senescence: One Step Closer to Senescence Atlas

  • Senescent cells that gradually accumulate during aging are one of the leading causes of aging. While senolytics can improve aging in humans as well as mice by specifically eliminating senescent cells, the effect of the senolytics varies in different cell types, suggesting variations in senescence. Various factors can induce cellular senescence, and the rate of accumulation of senescent cells differ depending on the organ. In addition, since the heterogeneity is due to the spatiotemporal context of senescent cells, in vivo studies are needed to increase the understanding of senescent cells. Since current methods are often unable to distinguish senescent cells from other cells, efforts are being made to find markers commonly expressed in senescent cells using bulk RNA-sequencing. Moreover, single-cell RNA (scRNA) sequencing, which analyzes the transcripts of each cell, has been utilized to understand the in vivo characteristics of the rare senescent cells. Recently, transcriptomic cell atlases for each organ using this technology have been published in various species. Novel senescent cells that do not express previously established marker genes have been discovered in some organs. However, there is still insufficient information on senescent cells due to the limited throughput of the scRNA sequencing technology. Therefore, it is necessary to improve the throughput of the scRNA sequencing technology or develop a way to enrich the rare senescent cells. The in vivo senescent cell atlas that is established using rapidly developing single-cell technologies will contribute to the precise rejuvenation by specifically removing senescent cells in each tissue and individual.

A Study on the Oral Health Knowledge and Education Needs of Adult's Lifestage -Research on Basic Data for Developing Oral Health Education Plan for Well-aging- (성인의 생애주기별 구강보건 지식과 교육 요구도에 관한 연구 -웰에이징 구강보건교육안 개발 기초자료 조사-)

  • This study investigated the oral health knowledge and educational needs of adults by life cycle. We aimed to provide basic data for developing oral health education plans for well -aging. The survey was conducted online and through mobiles by Gallup Korea during February 2021. A total of 319 adults over the age of 19 were surveyed. The study aimed at investigating the general characteristics, the quality of life-related to oral health, knowledge of oral health, and the need for well-aging education. The PASW Statistics ver 18.0 statistical program was used for data analysis. The analysis of the data suggests that prevention was the highest priority among all age groups. Furthermore, there was a difference in the perceived educational need among different age groups. The cause of the youth, the treatment of the middle-aged and the elderly were significantly related to oral and systemic disease. The need for education related to oral diseases was ranked in the order of aches, bad breath, periodontal diseases, and tooth discoloration. For information, the subjects reached out to the internet, dental and medical institutions, and public health centers. The teaching methods experienced by those surveyed included lectures combined with experience, videos, and expert lectures. We believe that this study can be used to develop an education plan that reflects the needs of the subjects when conducting life cycle education for well-aging.

A study of well-dying and well-aging through consideration of factors related to the Korean social community (한국사회 공동체 관련 요소들의 고찰을 통한 웰다잉(well-dying)과 웰에이징(well-aging) 연구)

  • This study historically examined nine representative community-related elements such as Hyangyak, Doore, Pumassi, Dongje, Gwallye, Wedding, Funeral, Ancestral Rites, Saemaul Undong, etc. in our society. This study aimed to create a better Korean society by revealing the relationship between well-aging and well-dying. This study investigated and analyzed 45 recent papers published between 2016 and 2021. As a result of the analysis, out of 45 cases, 22 cases are connected to the Joseon Dynasty, and 15 cases are related to modern society. When divided into life and death, 6 out of 9 items were bound to life, and 3 items encompassed life and death. And based on the relationship between well-aging and well-dying, it was specified as Type A-D. In conclusion, this study allows our society to achieve social integration and share the interests of the community through the principle of reciprocity and solidarity. This study can contribute from the perspective of convergence to becoming a true community that can share the interests of the community.

A Qualitative Case Study on the old people's Perception of Dignifiable Aging (노인의 품위 있는 노화 인식에 대한 질적사례연구)

  • This study aims to examine how old people think about 'dignifiable aging'. For the study, the authors have tried to understand and analyze of old people's view on 'classy and gracious aging' through their life experiences. The study focuses on the questions of 'how the participants experience aging', 'how they understand well, classy and successful aging and 'how they explain their ideas of the aging, then analyzes their ideas as types of 'within cases' as well as 'inter cases'. Indepth interview has employed as main methodology of data collection with the analysis of concerned written documents such as on-line and off-line essays as secondary data. The collected data had been basically sorted and analyzed by sorting commonimity and differences. Then they have been classified into the thems that indicate meanings and forms of 'dignified aging': 'ageless mind', 'happiness determined by one's mind', 'responsibility on family', 'desire for learning and social activities'. These meaning-units are added up to the main meaning of '(fullfilled) satisfaction through life-values' mediated by 'acceptance and acknowledgement', 'efforts', 'others' gaze', 'dreams and hopes', '(fullfilled) Satisfaction through life-values' can be achieved by 'sharing with others'. An interesting fact is that the meaning-units can be devided into two groups: 'the real life is the ideal life' which explains the actualization of the ideal in the daily life and 'life, conflict, the ideal' in which the real and the ideal are parellel with constant conflicts. In the conclusion, some implications and suggestions derived from the analysis are described from the v iew of social welfare.

The Thermal Performance of Building Insulation Materials According to Long-Term Aging (건축용 단열재의 장기 경시변화에 따른 열성능 특성)

  • This study is to draw thermal property data during long-term aging, by testing the thermal conductivity of building insulation materials in Korea. The thermal resistance of extruded insulation within 3 days from manufacture performed well over the KS Standard. After 50 to 110 days, however, the thermal performance had deteriorated to the level of the KS standard. Eventually, after 4,000 days, the insulation performance had deteriorated to about 25.4~41.8% of the initial performance. Therefore, this research will be utilized as a reference for thermal properties during long-term aging, in order to improve standards and regulations related to building insulation materials.

Chestnut Extracts as new Anti-aging agent

  • Inner nutshell of Castanea crenata has been used as an anti-aging and anti-wrinkle agent from the ancient time in east Asia. In order to develop new anti-aging and anti-wrinkle, ethanolic extract of inner nutshell of Castanea crenata was prepared and various biological activities were evaluated. Cor-285 possessed potent tree radical scavenging activity in vitro compared to gallic acid. Cor-285 showed the preventive effect against UV-induced cytotoxicity of fibroblast at concentration of 25-250mg/ml. When Cor-285 was evaluated for its anti-allergic activity, it effectively inhibited histamine release from mast cells induced by compound 48/80. The inhibitory activity was stronger than that of glycyrrhizinate. Cor-285 also showed on vivo inhibition against delayed hypersensitivity as well as croton-oil induced ear edema in mice when topically applied. These results strongly suggest that Cor-285 may reduce immunoregulatory/inflammatory skin trouble. From the attempts to isolate the scavenger, were isolated. In a clinical trial of twenty healthy volunteers with aged skin, 6 weeks application of Cor-285 decreased wrinkle about 26% and increased moisturizing 20% on the skin. All of these results indicate that Cor-285 may be an effective anti-aging and anti-wrinkle agent.

  • Face aging aims to synthesize future face images by reflecting the age factor on given faces. In recent years, deep learning-based approaches have made outstanding progress in simulating the aging process of the human face. However, generating accurate and high-quality aging faces is still intrinsically difficult. We propose a new method that incorporates gender information into the model, which achieves comparable and stable performance. Experimental results demonstrate that our method can preserve the identity well and generate diverse aged faces.

Berberine Inhibits the Production of Lysophosphatidylcholine-induced Reactive Oxygen Species and the ERK1/2 Pathway in Vascular Smooth Muscle Cells

  • Lysophosphatidylcholine (lysoPC) induces vascular smooth muscle cell (VSMC) proliferation and migration, which has been proposed to initiate the intimal thickening in coronary atherosclerotic lesions. Berberine is an alkaloid in Berberis aquifolium and many other plants. Recently, it has been shown to have beneficial effects on the cardiovascular system, such as anti-hyperglycemic and cholesterol-lowering activity. In this study, we investigated its effects on lysoPC-induced VSMC proliferation and migration. Berberine inhibited lysoPC-induced DNA synthesis and cell proliferation in VSMCs, as well as migration of the lysoPC-stimulated VSMCs. It also inhibited the activation of extracellular signal-regulated kinases (ERKs) and reduced transcription factor AP-1 activity and the lysoPC-induced increases in intracellular reactive oxygen species (ROS). These results indicate that the inhibitory effects of berberine on lysoPC-stimulated VSMC proliferation and migration are attributable to inhibition of ROS generation and hence of activation of the ERK1/2 pathway. This suggests that berberine has potential in the prevention of atherosclerosis and restenosis.

Research Trends of Psychology Studies Published in 『Journal of the Korean Gerontological Society』 (『한국노년학(韓國老年學)』의 심리학 분야 연구동향)

  • Academic research of the elderly psychology flourishes in an aging society. The basic concern of both psycho-gerontology dealing with psychological processes of aging and geronto-psychology dealing with psychology of the elderly is with the maintenance of high quality of life and well-being. Given this context, the present study examined research trends of 91 depression and suicide studies and 31 aging and successful aging studies, and compared the trends with those identified in studies published within the first 30 years. It was found that a majority of studies are concerned with the direct or mediating effects of depression on suicide and everyday life, risk factors influencing depression, perceptions of aging and factors influencing those perceptions, and successful aging and factors influencing successful aging. Further, the number of studies of these topics is greater than that of studies published within the first 30 years. The findings were discussed in the context of heightening the quality of life of the older adults.

Study on the Experimental Aging Estimation Technique for HTPB based Solid Propellant Considering Post Curing Effect (후경화를 고려한 HTPB 고체 추진제의 실험적 노화평가 기법 연구)

  • Post curing effects are estimated by specimen tests. Propellant specimen accelerated aging tests are performed when post curing is estimated to be complete and the coefficients of Arrhenius aging equations are acquired. Simulated motors with cylindrical grain are designed and fabricated to confirm the application. Accelerated aging tests are conducted, and aged properties are measured and estimated for the inner bore, center and bond parts of the grain. The measured aging ratios of the modulus are compared with the ones predicted by the equations. As the results, the accelerated aging equations predict well the propellant aging trends; however, some differences are observed at the bond part. Therefore, the specimen extraction part must be carefully chosen to suit the test purpose when a rocket motor grain is used for the aging test.
