Comparison of the Therapeutic Effects of $Curosurf^{(R)}$ and $Newfactan^{(R)}$ in Respiratory Distress Syndrome (신생아 호흡 곤란 증후군에서 $Curosurf^{(R)}$$Newfactan^{(R)}$의 치료 효과 비교)

  • Purpose : The aim of this study was to compare the clinical effects of modified porcine (Curosurf$^{(R)}$) and bovine (Newfactan$^{(R)}$) surfactants in the treatment of neonatal respiratory distress syndrome. Methods : Between April 2004 and December 2006, 65 neonates (birth weight ${\leq}$2,500 g and gestational age ${\leq}$35 weeks) with neonatal respiratory distress syndrome were treated in our neonatal intensive care unit with surfactant. Thirty-one neonates received Curosurf$^{(R)}$ and 34 neonates received Newfactan$^{(R)}$. The neonates were not enrolled if they had major congenital anomalies or meconium aspiration syndrome. We compared the changes in respiratory parameters after surfactant instillation, the incidences of acute and chronic complications, and the mortality between the two treatment groups. Results : Neonatal and maternal demographic characteristics were not different between the groups. The patterns of change in the respiratory parameters after surfactant instillation were not statistically different between the groups. The incidences of surfactant reinstillation and acute complications, such as pneumothorax, patent ductus arteriosus, pulmonary hemorrhage, and grade 3-4 intraventricular hemorrhage, were not different between the neonates who received Curosurf$^{(R)}$ and the neonates who received Newfactan$^{(R)}$. There were no statistically significant differences in the duration of mechanical ventilation, oxygen therapy, hospitalization, prevalence of bronchopulmonary dysplasia, periventricular leukomalacia, retinopathy of prematurity, necrotizing enterocolitis, and mortality between the groups. Conclusion : In the present comparative study, no significant differences in the clinical effects of Curosurf$^{(R)}$ and Newfactan$^{(R)}$ were observed.

Cell Growth in Suspension-Culture of Populus nigra var. italica and the Efficiency of Micro-Callus Formation according to Cell Plating Method (Populus nigra var. italica현탁배양(懸濁培養) 세포(細胞)의 생장(生長) 및 Cell Plating방법(方法)에 따른 Micro-Callus형성능력(形成能力))

  • In order to know the growth of suspended cells by explant sources, the change of nitrogen contents of cultured cells following the growth periods, capability of micro-callus formation according to cell plating methods, growth of suspended cells on various media, and efficiency of micro-callus formation by using growth regulators and different N strengths were investigated. 1. When suspension culture was tried by using the callus induced from internode and petiole, cell fresh weight and packed cell volume increased with similar way and the growth reached at stationary phase after 12 culture days. 2. N-contents of cultured cells increased upto 3 days and decreased around 6days. But the values increased again upto 9 days, after that they showed gradual decreases. 3. Of cell plating methods, embedding method was the best for micro-callus formation. 4. Growth of suspened cells showed the rest performanoes, when they were cultured on LM medium with 1/2N strengths and BAP 0.01.2.4-D 0.1, and NAA $1.0mg/{\ell}$, after 15 cultured days(upto 76.9 folds). LM medium was better than MS or GD. The combination of auxin and cytokinin was better for cell growing than auxin-treatment only. 5. Micro-callus from single cell and small cell aggregates was formed only on MS and LM media with 2,4-D $1.0mg/{\ell}$.

Effects of Elevated Spring Temperatures on the Growth and Fruit Quality of the Mandarin Hybrid 'Shiranuhi' (봄철 가온처리가 부지화의 생장과 과실품질에 미치는 영향)

  • The effects of elevated spring temperatures on the growth and fruit quality of the mandarin hybrid 'Shiranuhi' [(Citrus unshiu ${\times}$ C. sinensis) ${\times}$ C. reticulata] were investigated in plastic greenhouses, to develop a cropping system to improve the quality of the fruit and increase the income of growers on Jeju Island, South Korea. Under conditions of elevated temperature I ($25/15^{\circ}C$, day/night) and elevated temperature II ($28/18^{\circ}C$, day/night) during early spring, budburst was advanced by 11 and 15 d, and full bloom by 22 and 45 d, respectively, compared to those of the plants grown at ambient air temperature in a plastic greenhouse. Elevated temperatures decreased the number of spring shoots but increased mean spring shoot length and leaf area. Growing 'Shiranuhi' trees at elevated temperatures resulted in increases in mean fruit weight and fruit L/D ratio (> 1.0). In addition, fruit color development was significantly advanced in trees grown under elevated temperatures during early spring, which allowed the fruit to be harvested 1-2 months earlier than trees grown under ambient air temperature. Fruit soluble solids content (SSC) and titratable acidity (TA) at harvest were similar between elevated temperature I and ambient air temperature, but were significantly higher than at elevated temperature II. Considering fruit quality, harvest time, and yield, the elevated temperature treatment regime of $25/15^{\circ}C$ (day/night) during early spring could be useful for cultivation of the mandarin hybrid 'Shiranuhi' to increase the income of growers.

Effects of Cyanobacterial Bloom on Zooplankton Community Dynamics in Several Eutrophic Lakes (부영양호수에서 남조류 bloom이 동물플랑크톤 군집변화에 미치는 영향)

  • Toxin production and low digestibility of cyanobacteria are known to cause low exploitability of cyanobacteria by zooplankton. In this study, we compared relative tolerance and compatibility of zooplankton taxa in eight eutrophic lakes, exposed to frequent cyanobacterial blooms, uring the summer season of 1999. Microcystis, Anabaena, Oscillatoria and Phormidium were common cyanobacteria in all lakes. with relatively lower $NO_3-N$ concentration (<0.2 mgN/l) and TN/TP ratio (<20), compared with other lakes where colonial cyanobacteria dominated. Rotifers were dominant zooplankton in most lakes, and among them, Keratella, Polyarthra and Hexathra were common. The laboratory feeding experiment showed that relative copepods that greatly decreased (90%) after 4 day when cyanobacteria were used as the food source of zooplankton, while rotifers gradually increased with the change of dominant taxa from Keratella through Pompholyx to Monostyla. These results suggest that rotifers may be capable of coexisting with cyanobacteria by exploiting them for the food source.

Host Dependent Oviposition and Development of Azuki Bean Weevil (Callosobruchus chinensis L.) in Different Leguminous Seeds (기주에 따른 팥바구미(Callosobruchus chinensis L.)의 산란 선호성 및 성장)

  • To investigate the oviposition preference and development of azuki bean weevil (Callosobruchus chinensis L.), the following six different leguminous seeds were used in this study: red bean (Vigna angularis (Willd.) Ohwi & Ohashi), black soybean (Glycine max (L.) Merr.), soybean (Glycine max (L.) Merr.), seoritae (Glycine max (L.) Merr.), small black bean (Rhynchosia nulubilis) and kidney bean (Phaseolus vulgaris var. humilis Alef.). In the study of oviposition preference, the numbers of eggs per leguminous seed on red bean, black soybean, soybean, seoritae and small black bean were 1.23, 0.61, 0.69, 1.05 and 1.13, respectively. The maximum daily number of eggs was observed at 48 hours and the minimum was at 96 hours. According to each host leguminous seed, developmental time for each host seed was different. The shortest adult emergence time was on red bean (25.27 days). The other five leguminous seeds increased or doubled the adult emergence time. Adult emergence rates feeding on red bean, seoritae, black soybean, soybean, small black bean were 83.33%, 28.23%, 27.87%, 20.44%, and 11.59%, respectively. Emergence rate on red bean was four times higher than the rate on other seeds. The longevity of emerged female adults was almost all longer than that of males. The male adults weighed the lowest of feeding on small black bean. Female adults weighed the lowest of feeding on soybean. Adult weights were the heaviest for both males and females feeding on red bean. As a result, hosts of azuki bean weevil could decrease oviposition rate, emergence rate, adult longevity, and adult weight but increase emergence time. Especially in kidney bean, adult was not completely emerged. No eggs were laid. These results suggest that there might be emergence inhibitors in kidney bean. These imformation might be used to control damages caused by azuki bean weevils.

Ultrastructural Changes Induced by Telluric Acid in the Rat Liver (Telluric Acid가 흰쥐 간조직의 미세구조에 미치는 영향)

  • This experiment was carried out to investigate the effects of telluric acid on the histological and fine structural changes in the rat liver. Fischer 344 rats($150{\sim}200gm$) were used in this study as control and experimental groups. Telluric acid(5 mg/100 gm of body weight) suspensed in olive oil was given intraperitoneally to the animals of the experimental group and only olive oil to those of the control group. At the intervals of 3, 6 and 12 hours, 1, 2, 3, 5, 10, 20, 30 and 60 days after administration, the animals were sacrificed, and livers were obtained from the rats. For light microscopic examination of the liver, sections($5{\mu}m$) were stained with hematoxylineosin(H-E). For electron microscopic examination of the liver, sections were stained with uranyl acetate and lead citrate, finally examined with Zeiss EM 109 electron microscopes. The results obtained were as follows. 1. In the control group, round nucleus. well developed mitochondria, Golgi apparatus, rough endoplasmic reticulum(RER) and numerous glycogen particles were observed in the cytoplasm of the hepatocyte. In the cytoplasmic membranes of the hepatocyte, sinusoidal surface had numerous microvilli and cellular surface is combinated adjacent hepatocyte with desmosomes. The RER cisterns were dilated and zymogen granules were fewer than those of the dark cells. Kupffer cells with irregular nuclear membrane were observed. Fat storing cell and collagenous fiber bundle were observed in the Disse space. 2. Kupffer cell, inflammatory cells in the connective tissue of hepatic triad and lysosome were increased in the 3, 6, and 12 hour experimental group comparing with that of the control group. 3. In the 1 day experimental group, infiltration of inflammatory cells in interlobular connective tissue, dilatation of sinusoidal capillary and increasing of Kupffer cell were observed. Atropic change of hepatocyte and aggregation of glycogen particles in the cytoplasm of hepatocyte were observed. In this group, desmosome near bile canaliculi and collagenous fiber bundle in the Disse space were increased comparing with that of the 12 hours experimental group. In the 2 days experimental group, desmosome, lysosome, peroxisome and collagenous fiber bundle were increased comparing with that of the 1 day experimental group. Furthermore, lamellated bodies were also seen in the cytoplasm of the hepatocyte. 4. In 3 and 5 days experimental groups, transformations of hepatic cell cord and degeneration of the hepatocyte were markedly inclosed comparing with the all experimental groups. And damaged RER and mitochondria. collagenous fiber bundle were also inclosed comparing with that of the 2 days experimental group. Autophagosome and fat storing cells with large lipid droplets were also observed comparing with that of the 2 days experimental group. Tight junction and desmosome between the hepatocytes were separated. These degenerating changes were severe through the all experimental groups. 5. In the 10 and 20 days experimental groups, arrangement of hepatic cell cords and cell organelles of hepatocytes were similar to those of the control group. However, aggregation of glycogen particles, dilatation of sinusoidal capillary and infiltration of inflammatory cells remained. 6. In the 30 days experimental group, the tissue findings were similar to those of the control grout. But lamellated bodies in some hepatocytes and lysosome were remained in the cytoplasms of the Kupffer cells. In the 60 days experimental group, these all changes were recovered as the control group. In conclusion, telluric acid would directly induce the degenerative and necrotic changes on the hepatic tissue. However, these changes were perfectly recoverd in the 60 days experimental group as the control group.

Effects of Dietary Supplementation of Coffee Meal on Intestinal Enzyme Activity, Biochemical Profiles and Microbial Population in Broiler Chicks (커피박 첨가가 육계의 소장 효소 활성도, 생화학 지표 및 장내 미생물 균총에 미치는 영향)

  • The current study was performed to investigate the effects of dietary supplementation of dried coffee meal (CM) on growth performance, intestinal and blood biochemical index, intestinal enzymes, and cecal microbial populations. A total of 162, 3-day-old male broiler chicks were randomly allocated into three dietary groups: control group (CON), basal diet added with 0.5% CM (CM I), and basal diet added with 1.0% CM (CM II). Dietary supplementation of CM did not change bird performance and the relative weight of intestinal mucosal tissues. The birds fed the diet supplemented with CM (0.5 and 1.0%) significantly decreased mucosal glucose concentration (P<0.05) without affecting blood glucose level compared with those fed control diet. The level of blood aspartate aminotransferase (AST) significantly increased in CM II group (P<0.05) without affecting ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GTP) compared with that in the CON group. The specific activity of intestinal maltase, leucine aminopeptidase (LAP) and alkaline phosphatase (ALP) were not affected by dietary supplementation of CM, whereas sucrase activity in birds fed the diet supplemented with CM was decreased (P<0.05) compared to that in the control birds. The colony forming units (CFU) of E. coli in the cecum of CM-fed birds was significantly decreased (P<0.05) compared with that of control birds without changing the CFU of Lactobacillus. In conclusion, dietary supplementation of lower level of CM (0.5%) can be used as a beneficial feed resource without liver toxicity in broiler chicks.

Weed Population Distribution and Change of Dominant Weed Species in Paddy Field of Southern Gyeonggi Region (경기 남부지역 논잡초 발생분포 및 군락변화)

  • Survey was carried out in paddy fields to investigate the dominance of weed species and weed occurrence on 200 fields of southern Gyeonggi region in Korea during the year 2013. We observed 3 species of grass, 5 species of sedge, and 11 species of broadleaf and other weeds. The most dominant weed species was Eleocharis kuroguwai, followed by Echinochloa crus-galli, Sagittaria trifolia, Monochoria vaginalis, Scirpus juncoides and Bidens tripartita. The dominance value of E. kuroguwai and B. tripartita decreased compared to the survey results performed in 2005 but that of M. vaginalis, S. juncoides, S. trifolia and E. crus-galli increased. Weed occurrence increased from 2.83 g in 2005 to 3.51 g in 2013 based on weed dry weight per $m^2$. Occurrence ratio of annual weeds to perennial weeds changed from 45.2% : 54.8% in 2005 to 45.8% : 54.2% in 2013. If we analyze dominance of weed species based on the rice transplanting timing, E. kuroguwai and E. crus-galli were the most dominant weed species at mid-May transplanting, and M. vaginalis and S. juncoides were the most dominant weed species at late- May transplanting, and M. vaginalis and E. kuroguwai were the most dominant weed species at early-June transplanting. Since paddy weeds resistant to sulfonylurea herbicides are expected to be continuously increased, the regular monitoring of their occurrence and efficient control methods should be considered in future.

Changes of Furfural and Levulinic Acid Yield from Small-diameter Quercus mongolica Depending on Dilute Acid Pretreatment Conditions (약산 전처리 조건에 따른 소경 신갈나무 유래 푸르푸랄 및 레불린산의 함량 변화)

  • In this study, dilute acid pretreatment was operated using small-diameter Quercus mongolica for evaluating the yield change of furfural and levulinic acid depending on pretreatment factors. The dilute acid pretreatment was conducted depending on reaction temperature ($140-180^{\circ}C$), reaction time (10-30 min), and sulfuric acid concentration (0-2%, w/w). Then, glucose, XMG (xylose + mannose + galactose), furfural, and levulinic acid contents in the liquid hydrolyzate were measured and analyzed after pretreatment. Glucose content increased to 16.02% as reaction temperature, reaction time, and sulfuric acid concentration increased, but it decreased at the sulfuric acid concentration of 2% (reaction temperature: > $170^{\circ}C$, reaction time: > 20 min). On the other hand, reaction temperature had a strong influenced on XMG content, and XMG content decreased to 1.63% through increasing of reaction temperature and sulfuric acid concentration, but XMG content was less affected by changes of reaction time. Furfural content increased with the increase of reaction temperature, reaction time, and sulfuric acid concentration, and maximum furfural content was 7.61% (reaction temperature: $180^{\circ}C$, reaction time: 20 min, sulfuric acid concentration: 1%) based on a weight of raw material, while furfural content was dropped in more severe condition than in maximum furfural content condition. Levulinic acid content also increased with higher reaction temperature, reaction time, and sulfuric acid concentration. Especially, the sharp increase of levulinic acid content was observed above $170^{\circ}C$, and maximum levulinic acid content was 10.98% (reaction temperature: $180^{\circ}C$, reaction time: 30 min, sulfuric acid concentration: 2%). However, less than 1% of furfural and levulinic acid content was obtained in non-acidic catalyst condition that in whole conditions of reaction temperature and reaction time.

Rectal Temperature Maintenance Using a Heat Exchanger of Cardioplegic System in Cardiopulmonary Bypass Model for Rats (쥐 심폐바이패스 모델에서 심정지액 주입용 열교환기를 이용한 직장체온 유지)

  • Background: Small animal cardiopulmonary bypass (CPB) model would be a valuable tool for investigating path-ophysiological and therapeutic strategies on bypass. The main advantages of a small animal model include the reduced cost and time, and the fact that it does not require a full scale operating environment. However the rat CPB models have a number of technical limitations. Effective maintenance and control of core temperature by a heat exchanger is among them. The purpose of this study is to confirm the effect of rectal temperature maintenance using a heat exchanger of cardioplegia system in cardiopulmonary bypass model for rats. Material and Method: The miniature circuit consisted of a reservoir, heat exchanger, membrane oxygenator, roller pump, and static priming volume was 40 cc, Ten male Sprague-Dawley rats (mean weight 530 gram) were divided into two groups, and heat exchanger (HE) group was subjected to CPB with HE from a cardioplegia system, and control group was subjected to CPB with warm water circulating around the reservoir. Partial CPB was conducted at a flow rate of 40 mg/kg/min for 20 min after venous cannulation (via the internal juglar vein) and arterial cannulation (via the femoral artery). Rectal temperature were measured after anesthetic induction, a ter cannulation, 5, 10, 15, 20 min after CPB. Arterial blood gas with hematocrit was also analysed, 5 and 15 min after CPB. Result: Rectal temperature change differed between the two groups (p<0.01). The temperatures of HE group were well maintained during CPB, whereas control group was under progressive hypothermia, Rectal temperature 20 min after CPB was $36.16{\pm}0.32^{\circ}C$ in the HE group and $34.22{\pm}0.36^{\circ}C$ in the control group. Conclusion: We confirmed the effect of rectal temperature maintenance using a heat exchanger of cardioplegia system in cardiopulmonary bypass model for rats. This model would be a valuable tool for further use in hypothermic CPB experiment in rats.
