흰쥐의 섬유아세포 L-929를 이용한 새로운 Soft Contact Lens 소재의 생물안전성 검증 (Biosafety of the New Soft Contact Lens Materials in the Fibroblast L-929 Cell Line)

  • In this study, we polymerized new materials for soft contact lens using HEMA (2-hydroxyethyl methacrylate) which is the based-monomer of soft contact lens, EGDMA (ethylene glycol dimethacrylate) as cross linkage agent, and the new additives of monoester or di-ester derived from itaconic acid commercially produced by the fermentation of Asp. itaconicus. New polymer materials for soft contact lens were synthesized with the mixture of HEMA and mono- or diester at different ratios and presented to a good water content and oxygen transmissibility (Dk/L) values. In case of polymerization with HEMA and mono-ester (15%), the water content and oxygen transmissibility of contact lens were found to be good values at 57.6% and 28.5 Dk respectively. The mixture of HEMA and mono-ester is more excellent than HEMA/di-ester in the water content and oxygen transmissibility. The toxicity of new contact lens materials were confirmed in the fibroblast L-929 cell line using a agar overlay test and a growth inhibition test with the extract solution of contact lens.

Improvement of Biomineralization of Sporosarcina pasteurii as Biocementing Material for Concrete Repair by Atmospheric and Room Temperature Plasma Mutagenesis and Response Surface Methodology

  • Microbially induced calcium carbonate precipitation (MICP) has recently become an intelligent and environmentally friendly method for repairing cracks in concrete. To improve on this ability of microbial materials concrete repair, we applied random mutagenesis and optimization of mineralization conditions to improve the quantity and crystal form of microbially precipitated calcium carbonate. Sporosarcina pasteurii ATCC 11859 was used as the starting strain to obtain the mutant with high urease activity by atmospheric and room temperature plasma (ARTP) mutagenesis. Next, we investigated the optimal biomineralization conditions and precipitation crystal form using Plackett-Burman experimental design and response surface methodology (RSM). Biomineralization with 0.73 mol/l calcium chloride, 45 g/l urea, reaction temperature of 45℃, and reaction time of 22 h, significantly increased the amount of precipitated calcium carbonate, which was deposited in the form of calcite crystals. Finally, the repair of concrete using the optimized biomineralization process was evaluated. A comparison of water absorption and adhesion of concrete specimens before and after repairs showed that concrete cracks and surface defects could be efficiently repaired. This study provides a new method to engineer biocementing material for concrete repair.

경기도내 수계시설에서 분리된 레지오넬라균의 분포현황 및 Legionella pneumophila serogroup 1의 유전학적 다양성 연구 (Distribution of Legionella species from water systems and genetic diversity of L. pneumophila serogroup 1 in Gyeonggi-do)

  • 레지오넬라증은 인공수계환경이 레지오넬라균에 오염되었을 때 에어로졸의 전파에 의하여 집단 발생되는 심각한 폐렴과 높은 치명률을 일으키는 호흡기질환이다. 본 연구에서는 경기도내 수계시설로부터 147주의 레지오넬라균을 검출하였고, 시설별, 수계환경별 분포현황을 조사하였다. 분리한 레지오넬라균 중에서 Legionella pneumophila의 검출률은 85.7%로 높게 나타났으며, 혈청군을 분석한 결과 L. pneumophila serogroup (sg) 1이 주로 검출되었다. Non-L. pneumophila 중에서는 L. wadsworthii이 가장 많이 검출되었다. L. pneumophila sg 1의 유전학적 다양성을 분석하기 위해 pulsed field gel electrophoresis (PFGE)와 sequence-based typing (SBT) 방법을 사용하여 비교 분석하였다. 32주에 대해 PFGE 분석 결과 22개의 PFGE pattern을 보였고, SBT 분석 결과 9개의 유형으로 나누어졌다. PFGE와 SBT 분석에서 모두 크게 3개 group으로 분류되어 매우 유사한 결과를 나타냈다. SBT 유형 중 ST1이 가장 우점하였고, 2개의 새로운 유형도 찾아낼 수 있었다. SBT는 데이터베이스가 잘 구축되어 있어 균주 간의 유전학적 유형분석을 쉽고, 간단하게 할 수 있기 때문에 레지오넬라증의 분자역학적 연구에 유용한 방법임을 확인할 수 있었다.

A Laboratory-Scale Study of the Applicability of a Halophilic Sediment Bioelectrochemical System for in situ Reclamation of Water and Sediment in Brackish Aquaculture Ponds: Establishment, Bacterial Community and Performance Evaluation

  • In this study, we investigated the potential of using sediment bioelectrochemical systems (SBESs) for in situ treatment of the water and sediment in brackish aquaculture ponds polluted with uneaten feed. An SBES integrated into a laboratory-scale tank simulating a brackish aquaculture pond was established. This test tank and the control (not containing the SBES) were fed with shrimp feed in a scheme that mimics a situation where 50% of feed is uneaten. After the SBES was inoculated with microbial sources from actual shrimp pond sediments, electricity generation was well observed from the first experimental week, indicating successful enrichment of electrochemically active bacteria in the test tank sediment. The electricity generation became steady after 3 weeks of operation, with an average current density of $2.3mA/m^2$ anode surface and an average power density of $0.05mW/m^2$ anode surface. The SBES removed 20-30% more COD of the tank water, compared to the control. After 1 year, the SBES also reduced the amount of sediment in the tank by 40% and thus could remove approximately 40% more COD and approximately 52% more nitrogen from the sediment, compared to the control. Insignificant amounts of nitrite and nitrate were detected, suggesting complete removal of nitrogen by the system. PCR-DGGE-based analyses revealed the dominant presence of Methylophilus rhizosphaerae, Desulfatitalea tepidiphila and Thiothrix eikelboomii, which have not been found in bioelectrochemical systems before, in the bacterial community in the sediment of the SBES-containing tank. The results of this research demonstrate the potential application of SBESs in helping to reduce water pollution threats, fish and shrimp disease risks, and thus farmers' losses.

Bacterial Community Structure Shifted by Geosmin in Granular Activated Carbon System of Water Treatment Plants

  • We investigated the relation between the presence of geosmin in water and the bacterial community structure within the granular activated carbon (GAC) system of water treatment plants in South Korea. GAC samples were collected in May and August of 2014 at three water treatment plants (Sungnam, Koyang, and Yeoncho in Korea). Dissolved organic carbon and geosmin were analyzed before and after GAC treatment. Geosmin was found in raw water from Sungnam and Koyang water treatment plants but not in that from Yeoncho water treatment plant. Interestingly, but not surprisingly, the 16S rRNA clone library indicated that the bacterial communities from the Sungnam and Koyang GAC systems were closely related to geosmin-degrading bacteria. Based on the phylogenetic tree and multidimensional scaling plot, bacterial clones from GAC under the influence of geosmin were clustered with Variovorax paradoxus strain DB 9b and Comamonas sp. DB mg. In other words, the presence of geosmin in water might have inevitably contributed to the growth of geosmin degraders within the respective GAC system.

  • As the genetically modified microorganisms (GMMs) and their recombinant plasmid DNAs could be released into natural environments, their stabilities and impacts to indigenous microorganisls have become very importhant research subjects concerning with environmental and ecological aspects. In this study, the genetically modified E. coli CU103 and its recombinant pCU103 plasmid DNA, in which pcbCD genes involving in degradation of biphenyl and 4-chlorobiphenyl were cloned, were studied for their survival and stability in several different waters established under laboratory conditions. E. coli CU103 and its host E. coli XL1-Blue survived longer in sterile distilled water (SDW) and filtered autoclaved river water (FAW) than in filtered river water (FW). A lot of extracellular DNAs were released from E. coli CU103 by lytic action of phages in FW and the released DNAs were degraded by DNase dissolved in the water. Such effects of the factors in FW on stability of the recombinant pCU103 plasmid were also observed in the results of gel electrophoresis, quantitative analysis with bisbenzimide, and transformation assay. Therefore, the recombinant plasmids of pCU103 were found to be readily liberated from the genetically modified E. coli CU103 into waters by normal metabolic processes and lysis of cells. And the plasmid DNAs were quite stable in waters, but their stabilities could be affected by physicoKDICical and biological factors in non-sterile natural waters.

The Dynamics of Protein Decomposition in Lakes of Different Trophic Status - Reflections on the Assessment of the Real Proteolytic Activity In Situ

  • The aim of this paper is to discuss the methodology of our investigation of the dynamics of protein degradation and the total in situ protealytic activity in meso/eutrophic, eutrophic, and hypereutrophic freshwater environments. Analysis of the kinetics and rates of enzymatic release of amino acids in water samples preserved with sodium azide allows determination of the concentrations of labile proteins $(C_{LAB})$, and their half-life time $(T_{1/2})$. Moreover, it gives more realistic information on resultant activity in situ $(V_{T1/2})$ of ecto- and extracellular proteases that are responsible for the biological degradation of these compounds. Although the results provided by the proposed method are general y well correlated with those obtained by classical procedures, they better characterize the dynamics of protein degradation processes, especially in eutrophic or hypereutrophic lakes. In these environments, processes of protein decomposition occur mainly on the particles and depend primarily on a metabolic activity of seston-attached bacteria. The method was tested in three lakes. The different degree of eutrophication of these lakes was clearly demonstrated by the measured real proteolytic pattern and confirmed by conventional trophic state determinants.

Purification and Characterization of Biosurfactants Produced by Pseudomonas sp. SW1

  • Pseudomonas sp. SW1 grew and produced biosurfactants on 3% hexadecane as the energy and carbon source. As a result of biosurfactant synthesis, the surface tension of the medium was reduced from 72 dyne/cm to 30 dyne/cm. The properties of biosurfactants that were purified from Pseudomonas sp. SW1 were investigated. The purification procedure included acid precipitation from culture supernatant, silica gel G60 column chromatography, and Sephadex G-150 gel filtration. The biosurfactants were separated into two different types, viz., types I and II. Biosurfactant type Isignificantly reduced the surface tension of water from 72 to 27 dyne/cm at concentration levels above 30 mg/l. The surface tension of water was reduced to a minimum of approximately 30 dyne/cm by biosurfactant type II at concentration levels over 80 mg/l. The biosurfactants were effective in a wide range of pHs, at NaCl concentrations of up to 4%, at $CaCl_2$ concentration up to 100 mM, and at temperatures up to $200^{\circ}C$ for 8 h.

The Effect of Vacuum Pressure in Membrane Filtration Systems for the Efficient Detection of Bacteria from Natural Mineral Water

  • The procedures currently used for determining microbiological quality of natural mineral water recommend filtration through membrane filters. In this study, we evaluated the effect of vacuum pressure for the accurate detection of bacteria from water samples seeded with Escherichia coli. We observed that the number of E. coli detected increased with increasing vacuum pressure. In order to examine the retention rate of bacteria in the holes of the membranes under the different pressures, the membrane filters were removed after filtration, washed with sterile water by vortexing, and placed on m-Endo agar plates. With all the filters tested, the number of E. coli retained within the filters at negative 600 mmHg was approximately 10 to $20\%$ higher than that obtained with 100 mmHg. These results demonstrate that the vacuum pressure exerted during the filtration procedure may affect the fixation of bacteria into some portions of openings in the membrane filter.

Isolation and Characterization of Biofouling Bacteria in Ultra-high Purity Water Used in the Semiconductor Manufacturing Process

  • Bacteria were isolated and identified from an advanced high-purity water system that supplies ultra-high purity water (UHPW) for 16-megabyte DRAM semiconductor manufacturing. Scanning electron microscopic and microbiological observations revealed that the primary source of the bacteria isolated from the UHPW was detached cells from biofilms developed on the pipe wall through which the UHPW, a man-made and extremely nutrient poor environment, was passing. About 63-65% of the bacteria isolated from the UHPW and the pipe wall were Gram-positive, whereas only 10% of the bacteria isolated from the feed water were Gram-positive. The of Gram-positive bacteria and seven genera of Gram-negative bacteria. Strains of the UHPW bacteria effectively adhered to and formed a biofilm on the surface of polyvinyl chloride (PVC) pipe.
