Response of Monodehydroascorbate Reductase (MDHAR) in Lettuce (Lactuca sativa L.) Leaves Subjected to Water Deficit Stress (수분 부족 스트레스 처리시 Monodehydroascorbate Reductase (MDHAR)의 반응)

  • The relationship between water deficit stress and monodehydroascorbate reductase (MDHAR) activity was determined in lettuce (Lactuca sativa L.) leaves under water stress condition imposed by with-holding water for 72 hrs. Relative water content determined in water deficit stressed lettuce leaves gradually reduced from 91.29% to 74.58%, and water content of medium drastically decreased 4.73% after quitting of irrigation. Hydrogen peroxide content in leaves subjected to water deficit stress rapidly increased, but soluble protein content rapidly decreased when those were compared to control plant. The relationship between relative water content and hydrogen peroxide content in stressed leaves positively correlated with $R^2$=0.8851, but soluble protein content reversely correlated with $R^2$=0.9826. Total chlorophyll content in stressed plant leaves was higher than that of control plant, and increased rapidly in early stage of treatment of both stressed and control plants. Carotenoid content was higher than that of control plant, and the ratio of carotenoid to total chlorophyll in stressed plant was higher as compared to control plant. As water deficit stress continued progressively, total ascorbate content in stressed plant leaves was a little higher than that of control plant. But dehydroascorbate (DHA) content within 6 hr of water deficit stress was higher than that of control plant, and then, content of control plant in 12 hr of stress treatment higher than that of stressed leaves. The activity of monodehydroascorbate reductase of cytosolic and chloroplastic tractions increased dramatically, and mRNA of MDHAR was highly detected by probing $^{32}P$-labeled single stranded MDHAR RNA of lettuce plant leaves subjected to water deficit stress. Relationship between MDHAR activity and relative water content and hydrogen peroxide highly correlated with $R^2$=0.9937 and 0.8645, respectively.

Variability of Soil Water Content, Temperature, and Electrical Conductivity in Strawberry and Tomato Greenhouses in Winter

  • Purpose: Monitoring and control of environmental condition is highly important for optimum control of the conditions, especially in greenhouses and plant factories, and the condition is not uniform within the facility. Objectives of the study were to investigate variability in soil water content and to provide information useful for better irrigation control. Methods: Experiments were conducted in a strawberry-growing greenhouse (greenhouse 1) and a cherry tomato-growing greenhouse (greenhouse 2) in winter. Soil water content, electrical conductivity (EC), and temperature were measured over the entire area, at different distances from an irrigation pump, and on ridge and furrow areas. Results: When measured over the entire greenhouse area, soil water content decreased and temperature and electrical conductivity increased over time from morning to afternoon after irrigation. Water content decreased by distance from the irrigation pump up to 70 m and increased after that, and temperature showed an inverse pattern. Soil water contents on the ridge were lower than those on the furrow, and the differences were 10.2~18.4%, indicating considerable variability. The lowest EC were observed on the furrow and highest values were observed on the ridge. Soil water contents were less and temperature levels were greater at the window side than in the center locations. Conclusions: Selection of number and location of soil water content sensor would be the first step for better water content monitoring and irrigation control. Results of the study would provide basic data useful for optimum sensor location and control for underground greenhouse environment.

Development of an Automatic Water Control System for Greenhouse Soil Water Content Management (시설재배 토양의 수분 조절을 위한 자동 수분제어시스템 개발)

  • This study was conducted to develop an automatic soil water content control system for greenhouse, which consisted of drip irrigation nozzles, soil water content sensors, an on/off valve, a servo-motor assembly and a control program. The control logic adopted in the system was Ziegler-Nichols algorithm and rising time, time constant and over/undershoot ratio as control variables in the system was selected and determined by various control experiments to maintain small delay time and low overshoot. Based on the experimental results, it was concluded that the control system developed in the study could replace the unreliable conventional greenhouse soil water management.

  • In this study, air meter method and capacitance measurement method to apply economically at quality control of ready-mixed concrete among various unit water content measurement technique was selected. Then, it was evaluated estimating performance of unit water content according to the change of water-binder ratio and unit water content. Also, it was examined influence about error occurrence of unit water content by change of properties of used materials. Finally, based on this study, it was proposed fundamental data to utilize measurement technique of unit water content to quality control. of ready-mixed concrete in construction field.

Study on the Growth and the Drought Resistance of Amorpha fructicosa under the Control of Water Supply (수분공급조절에 의한 족제비싸리의 생장과 내건성에 관한 연구)

  • The growth and the resistance of Amorpha fructicosa L. under water control was experimented in frames out of doors. The plant grew in a wagner's pot under water control. The soil moisture content was controlled at 5%, 10%, 15% and 20%. The growth of leaf, stem and root in the groups of 5% and 10% soil moisture content were different from values in the groups of 15% and 20%. The T/R ratio in the groups of 5% and 10% soil moisture content were different from the ratio in the groups of 15% and 20% soil moisture content. The T/R ratio of former was lower than the latter, but the C/F ratio of the former was higher than the latter. RGR and NAR of Amorpha fructicosa decreased in 5% and 10% soil moisture content but increased in 15% and 20% soil moisture content during growing period. The maximum values of RGR and NAR were respectively 0.089 and 0.080 at 20% soil moisture content. The highest value of LAR was 1.560 at 5% soil moisture content. RGR and NAR were comparatively affected by soil moisture content.

  • Three soybean cultivars, Hwangkeum, Tanyeob and Enrei were planted in the same pot under glasshouse conditions to investigate the influence of the different soil water content such as pF 1.4(wet), 2.1(control) and 3.6(dry) on the transpiration rate, dry matter production and water requirement. The transpiration rate remained the high constant rates under the wet soil condition and the control than the dry condition, and showed a linear correlation between transpiration rate and solar radiation under the all condition of soil water. The transpiration rate highly increased in the morning, but dramatically decreased in the other time in a day. The dry matter production was higher under the conditions of wet soil and the control than that under the dry condition. Also, the dry matter production Tanyeob was higher than other cultivars under all soil water content. The water requirement was higher for Enrei and lower Tanyeob than the control.

Water Absorption Characteristics of Substrate with Physical Properties of wick in Subirrigation System Using wick (심지형 저면관수시스템의 심지의 물리적 성질에 따른 수분흡수 특성)

  • The objectives of this study were to investigate the effect of the physical properties of wick on the water absorption of substrate. Physical properties of wick in this study were cotton composition, width and length. The water Infiltration rate through the wick was 0.24 ㎝/s at 90 -95% cotton content, which was faster than at 80-85% (0.13 cm/s) and 70-75% (0.08 cm/s). As the cotton content increased, the water absorption of substrate became greater : the amount of absorbed water was about 5-7g higher at 90-95% than at 80-85% and 70-75% at a wick width of 1 ㎝, the velocity of water absorption through the wick was fastest with 0.25 ㎝ㆍs/sup -1/. The amount of absorbed water was higher at 3 ㎝ than at 1 and 2 ㎝. However, the water absorption rate through the cross - sectional area of wick (g H₂O /㎠/hr) was higher at a wick width of 2 ㎝ than at those of 1 and 3 ㎝. The amount of absorbed water in the substrate was higher at 2 : 1 than at 1 : 1 (length in substrate : length out of substrate). Absorbed water amount was larger at 30-40% initial moisture content than any other treatment.

  • In this study, it was selected that in order to make an investigation into the status of the unit water content control of the ready-mixed concrete plants in the country, Capacitance Measurement Method out of various methods which are able to gauge the amount of unit water content in fresh concrete. Then, it were estimated that the quantity of unit water in fresh concrete and the technical standard of every mixture design of the six ready-mixed concrete plants chosen at random in the country. Finally, based on this study, it was proposed as fundamental data to utilize measurement techniques of the quantity of unit water for the quality control of the ready-mixed concrete in construction field.

Embedded System-Based Fast Fourier Transform Method for Measuring Water Content in Crude Oil

  • The moisture content of crude oil notably affects various aspects of oil production, storage, transportation, and exploration. However, accurately measuring this moisture content is challenging because of numerous influencing factors, leading to a lack of precision in existing detection methods. This inadequacy hinders the progress of China's petroleum industry. To overcome these challenges, this paper proposes a conductivity-based method for measuring crude oil moisture content. By employing an embedded system, we designed a sensor comprising five electrodes. Additionally, we developed signal excitation and signal processing circuits. Moreover, a software program was designed to analyze and compute the output signal using fast Fourier transform operations. This facilitated the identification of flow patterns, computation of relevant flow rates, and establishment of correlation rates based on frequency spectral characteristics. Based on experimental results, we established a functional relationship between measurement parameters and crude oil moisture content. This study enhanced the precision of moisture content measurement, thereby addressing existing limitations and fostering the advancement of China's petroleum industry.

A Methodology for Compaction Control of Crushed-Rock-Soil-Fills (암버럭-토사 성토 노반의 다짐 관리 방안)

  • More strict construction control of railway roadbeds is demanded in high speed railway system because of heavier repeated dynamic loading than conventional railways. The aim of this study is to propose a compaction control methodology of crushed-rock-soil-fills including as large particles as $200\sim300mm$ in diameter, which are easily encountered in high speed railway roadbed. Field tensity evaluation and in turn compaction control of such crushed-rock-soil-fills are almost impossible by conventional methods such as in-situ density measurements or plate loading tests. The proposed method consists of shear wave measurements of compaction specimens in laboratory and in-situ measurements of fills. In other words, compaction control can be carried out by comparing laboratory and field shear wave velocities using as a compaction control parameter. The proposed method was implemented at a soil site in the beginning and will be expanded to crushed-rock-soil-fills in future. One interesting result is that similar relationship of shear wave velocity and water content was obtained as that of density and water content with the maximum value at the optimum moisture content.
