율무 추출물의 마우스 비장세포와 대식세포 활성 효과 (Effects of Job's Tears(Yul-Moo) Extracts on Mouse Splenocyte and Macrophage Cell Activation)

  • Job's Tears(Yul-Moo) is a grass crop long-used as a traditional medicine; it is also a nourishing food. There are reports of its anti-inflammatory, stomachic, antiallergic activity, and antispastic effects and Job's Tears has been used in China to treat rheumatism, and neuralgia although its warts, rheumanism remains unclear. Thus, the present study was performed to investigate the in vitro effect of Job's Tears extracts on immune function. Here mouse splenocyte proliferation and cytokine production$(IL-1{\beta},\;IL-6,\;TNF-{\alpha})$ by peritoneal macrophages cultured with ethanol and water extracts of Job's Tears were examined. splenocytes proliferation increased with Job's Tears water extracts supplement at concentrations investigated The cytokine production$(IL-1{\beta},\;IL-6,\;TNF-{\alpha})$ by ELISA using a cytokine kit And $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production increased water extracts supplementation. This in vitro study suggests that supplementation with Job's Tears water extracts may enhance immune function by regulating the splenocyte proliferation and enhancing cytokine production of activated macrophages.

한국산 버섯추출물의 항진균 및 항세균활성 검색(II) (The Screening of Antifungal and Antibacterial Activities of Extracts from Mushrooms in Korea (II))

  • 한국산 버섯들의 항균활성을 검색하기 위하여 흰주름버섯외 35종의 버섯자실체 분말을 석유에테르, 80% 에탄올 및 멸균증류수로 각각 추출한 108종의 각 추출물들에 대하여 항균활성을 검색하였다. 이로서 진갈색주름버섯의 에탄올 추출물 등 5종의 추출물들이 효모에 대하여, 독우산광대버섯의 물 추출물 등 7종의 추출물들이 진균에 대하여, 마귀광대버섯의 물 추출물 등 9종의 추출물들이 그람음성 세균에 대하여 그리고 말불버섯의 물 추출물등 25종의 추출물들이 그람양성세균에 대하여 각각 항균활성을 보였다.

새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

대추 메탄올 추출물의 페놀성 화합물 함량과 항산화 효능 (Antioxidative Capacity and Total Phenolic Compounds of Methanol Extract from Zizyphus jujuba)

  • 대추의 페놀화합물 함량과 항산화 효능을 알아보기 위하여 특초(大棗: 대조), 상초(中棗: 중조), 약초(小棗: 소조) 세 종류의 대추를 메탄올에 추출한 용액과 물에 침 지한 후 메탄올로 추출한 용액을 동결 건조하여 총 페놀화합물 함량, 전자공여능, 산패유도기간을 측정하였다. 대추의 총 페놀화합 물 함량은 메탄올에 추출한 약초, 상초, 특초가 각각 $326.46\;mg\%,\;223.13\;mg\%,\;158.06\;mg\%$이었고 물에 침 지한 후 메탄올에 추출한 약초,상초,특초의 함량은 각각 $113.23\;mg\%,\;81.46\;mg\%,\;72.78\;mg\%$이었다. 전자공여능은 BHT $0.02\%$ 첨가구보다 더 우수한 것은 물에 침 지한 후 메탄올로 추출한 특초의 1.5, 2.0, 2.5, 3.0$\%$ 첨가구와 상초와 약초의 각 1.0, 1.5, 2.0, 2.5, $3.0\%$ 첨가구 등 총 22개의 시료이 었다. 그리고 $\delta-tocopherol\;0.02\%$ 첨가구와 같거나 더 우수한 시료는 물에 침 지한 후 메탄을 추출한 상초 1.5, $2.0\%$와 약초 1.0, $1.5\%$등 총 8개의 시료이었다 산패유도기간은 대추 시료의 첨가 농도가 증가함에 따라 산패유도기간도 증가하였으며 BHT $0.02\%$ 첨가구보다 산패유도기간이 더 우수하거나 비슷한 시료는 물에 침지한 후 메탄올에 추출한 상초 1.5, 2.0$\%$와 약초 1.0, 1.5, $2.0\%$를 첨가한 것과 메탄을 추출물인 특초 $2.0\%$와 약초를 $2.0\%$첨가한 것이었다 대추의 항산화 효능은 물에 침 지한 후 메탄올 추출한 약초가 가장 우수하였으며 약초추출물의 동결건조 시료는 식품첨가물로 사용할 수 있는 가능성이 있다고 생각된다.

청고추, 쇠비름, 표고버섯의 물 추출물 및 당침액의 항산화 활성 및 ${\alpha}$-Glucosidase Inhibition 활성 비교 (Comparison of Antioxidant and ${\alpha}$-Glucosidase Inhibition Activities among Water Extracts and Sugar Immersion Extracts of Green Pepper, Purslane and Shiitake)

  • 본 연구에서는 최근 건강 관리를 위해 다양하게 상용되고 있는 당침액의 오 남용을 막기 위해 과학적으로 물 추출물과 당침액의 특성을 규명하고자 하였다. 그 결과, 일반 물 추출물과 비교하였을 때 당침액의 총 폴리페놀 및 플라보노이드 함량이 크게 감소하는 것을 확인하였으며, 그에 따라 항산화 활성 또한 감소한 것을 확인할 수 있었다. 이는 물 추출물과 비교하여 당침액의 건조 분말에는 설탕 또는 그 분해물들이 많은 부분을 차지하기 때문이라 판단된다. 그러나 혈당강하 효능의 target 효소로 연구되고 있는 ${\alpha}$-amylase 및 ${\alpha}$-glucosidase의 활성을 측정한 결과, 청고추, 쇠비름, 표고버섯의 물 추출물과 당침액은 ${\alpha}$-amylase 억제 활성을 나타내지 않았지만, ${\alpha}$-glucosidase 억제 활성은 의약품인 acarbose에는 미치지 못하였으나, 물 추출물에 비해 당침액이 농도 의존적으로 높은 억제 활성을 나타내었다. 또한 세포 독성을 HepG2 cell에서 확인한 결과, 물 추출물과 비교하여 당침액의 독성이 약 1/10 감소한 것을 확인할 수 있었다. 이와 같은 결과는 민간에서의 믿음처럼 당침액이 물 추출물과 비교하였을 때 모든 부분에서 우수한 기능을 가진다고 할 수 없으며, 활용할 용도에 따라 소재를 물 추출하거나 당침액을 제조하는 등의 가공 과정을 거치는 것이 목표한 기능을 효과적으로 확보하는 방법임을 의미한다. 따라서 민간에서 맹목적으로 믿는 당침액의 효능에 대한 과학적인 연구 접근법은 당침액의 적절한 사용 목적과 방법을 설정하는 기초 자료로 활용이 가능할 것이다.

교감단(交感丹)의 투여가 STRESS에 의한 면역반응의 억제에 미치는 영향 (Effect of Gyogamdan Administration on the Stress-Induced Immunosuppression in the Mouse)

  • This study was done to know the effects of the water extracts of Gyogamdan(GGD) on the function of macrophages, the most important cells of the innate immune system, and the rosette forming ability of splenocytes in the mouse under stress. The effects of GGD on the immunosuppression induced by noise stress are as follows. 1. Administration of GGD water extracts normalized the bo요 weight which might be decreased by noise stress. 2. Administration of GGD water extracts increased the production of the such reactive oxygen intermediates as superoxide and hydrogen peroxide from macrophsges in vivo & in vitro which were decreased by noise stress. 3. Administration of GGS water extracts did not affect the production of reactive nitrogen intermediates. 4. Administration of GGD water extracts increased the rosette forming ability of splenocytes which was decreased by noise stress. The above effects of GGD might be useful for the treatment of stress-induced infections diseases which could be caused by the suppression of immune responeses which are initiated by the functions of macrophages of the innate immune system.

향나무 심재 추출물로 염색된 직물의 방미성과 집먼지 진드기 기피효과 (Antifungal activity and house dust mite repellent effect of fabric dyed with Juniperus chinensis heartwood extracts)

  • This study was carried out to investigate the antifungal activity and house dust mite repellent effect of cotton and silk fabrics dyed with Juniperus chinensis heartwood extracts by hot-water and methanol. The chemical composition of Juniperus chinensis heartwood extracts by hot-water and methanol was analyzed by GC-MS. In the case of antifungal activity, dyed fabrics with hot water extract had no effect against chaetomium globosum fungus. But showing no growth in fabrics dyed with methanol extract, dyed fabrics with methanol extract showed very excellent antifungal activity. Silk fabric dyed with methanol extract showed excellent house dust mite repellent effect of 94.3%~96.0% against dermatophagodes farinae. The composition of Juniperus chinensis heartwood extracts from hot-water and methanol was different. There were no terpene and its analogue peaks in hot-water extract. However there were terpene and its analogue peaks in methanol extract. From these GC-MS results, terpene was identified in methanol extract of Juniperus chinensis heartwood. Efficient ingredient of antifungal activity and house dust mite repellent effect was assumed terpene.

민들레 뿌리 물 추출물의 마우스 선천 및 획득 면역계에 미치는 효과 (Effect of Water Extracts from Root of Taraxacum officinale on Innate and Adaptive Immune Responses in Mice)

  • Hot-water($100^{\circ}C$) and cold-water($4^{\circ}C$) extracts of Taraxacum officinale root were assessed for the effects of innate and adaptive immune responses in mice. Hot water extracts(TO-100) and cold water extracts(TO-4) did not affect the viability of macrophages at concentrations below to 18 mg/ml and 8 mg/ml, respectively. The thioglycollate-induced macrophages cultured with TO-100 and TO-4 produced a significantly higher quantity of various cytokines, such as IL-6 and IL-12, than those treated with medium. This shows that the extracts potently stimulated the innate immune response. When mice were subcutaneously immunized(sc) with OVA+FIA(Freund's incomplete adjuvant)-emulsified TO-100, TO-100 did not affect the production of IgE, but enhanced the production of IgG1, IgG2a and IgG2b. The culture supernatant obtained from the splenocytes of mice treated with OVA+FIA-emulsified TO-100 also evidenced elevated levels of both OVA-specific Th1-type(IFN-$\gamma$) and Th2-type cytokines(IL-4, IL-6 and IL-10). These results suggested that TO-100 can modulate the immune responses to allergens in mice.

  • Rehmannia Radix is a thick tuberous root about 3-6 cm diameter. The tuberous root is repared for medicine. The material in the fresh state is shengdihuang (生地黃). The material in the dried state is gandihuang (乾地黃). Shudihuang (熱地黃) is made by taking juicy roots, washing them in millet wine, steaming on a willow frame in a percelain vessel, drying, and resteaming and redrying nine times. The aim of this experiments was to investigate the effect of Rehmannia Radix Water extracts on the plasma renin activity and plasma levels of aldosterone and atrial natriuretic peptide (ANP) in rats. The results of study were as follows: Plasma renin activity was not changed after administration of Rehmannia Radix water extracts. Plasma levels of aldosterone was decreased significantly after administration of Shudihuang (熱地黃) water extracts. Plasma levels of atrial natriuretic peptide was increased significantly after administration of Shudihuang (熱地黃) water extracts.

순무의 이화학적 및 기능적 특성 (Physicochemical and Functional Properties of Turnip)

  • This study was conducted to investigate physicochemical and functional properties of turnip. The concentrations of antocyanin in the rind of root was 3.24mg%, which was about 3.5 times higher than those in the flesh. Among free amino acids in each part of turnip, glutamic acid showed the highest concentrations(132.5mg%) in the root followed by alanine(25.0mg%), valine (23.3mg%), and serine (20.7mg%). Contents of minerals in the root of turnip were 395.7mg% for potassium, 187mg% for calcium, 53.6mg% for phosphate, and 40.7mg% for magnesium. According to dynamic heasdspace analysis, dimethyl disulfide was the most abundant flavor components from the juice of fresh turnip and other volatile components such as dimethyl trisulfide, 1 hexanal, 2 methyl 1 butamine, and 1 penten 3 ol were also identified. From the blanched turnip, however, 3 isothiocyanato 1 propene and 4 isothiocyanato 1 butene were mainly detected. Fresh and blanched turnips were extracted with water or 70% acetone to investigate percent yield. The concentrations of total polyphenols in the extracts of fresh turnip were higher than those of blanched turnip. The nitrite scavenging effects of water and 70% acetone extracts from fresh or blanched turnip were decreased as pH was increased. Between two water extracts, the 30oC water extracts showed the higher values of electron donating ability than 95$^{\circ}C$ water extracts.
