동재하시험을 이용한 모래지반의 말뚝지지력 산정 (Evaluation of Bearing Capacity of Piles in Sand Using Pile Driving Analyzer)

  • 말뚝재하시험방법 중 정적재하시험은 시험에 소요되는 시간과 비용측면에서 불리하여 이를 개선하기 위한 여러 가지의 시험방법이 개발되어 소개되고 있다. 국내에 소개된 시험방법으로는 동재하시험으로 불리는 항타분석기 (PDA : Pile Driving Analyzer)를 이용한 방법을 비롯하여 STATNAMIC. Osterberg cell등이 있다. 이들 중 동재하시험의 사용은 상당히 활성화되어 있는 실정이나 지반조건과 말뚝시공방법에 따라서는 지지력산정시 추가적인 해석모델을 사용하여야 보다 정확한 지지력을 평가 할 수 있는 것으로 알려져 있다. 본 논문에서는 현장에서 수행한 정적 및 동적 재하시험결과의 비교1분석으로 부터 주로 SIP말뚝에 대한 동재하시험시 발생할수 있는 기하감쇠(geometrical damping)의 영향을 고찰하였다. 해석결과로 부터 기하감쇠가 발생한 지반의 경우 CAPWAP에 의해 산정된 지지력이 정재하시험에 의한 지지력에 비해 30~60% 정도 과소평가 되었으며 이때 말뚝주면에 대한 Smith의 감쇠계수 (SSkn)가 1.0 sec/m를 초과 하였다. CAPWAP해석시 기하감쇠를 고려한 해석모델을 사용함으로써 정재하 시험결과와 근사한 지지력을 얻을 수 있었으며 SSkn값도 0.7sec/m이하의 일반적인 범위로 해석되었다.

1999년 봉화군 일개 중.고등학교에서 발생한 세균성이질에 관한 역학조사 (Epidemiologic Investigation of an Outbreak of Shigella sonnei among Students in Bonghwa, 1999)

  • Objectives : This study was carried out to investigate the sources of infection and modes of transmission of an outbreak of shigellosis that occurred among students of B middle and high school in Bonghwa, Korea from May 1 to 21, 1999. Methods : We conducted questionnaires to 468 students, 38 stalls and 9 food handlers twice times (May 6, May 21) for follow up and secondary attack rate. Personal details and history of illness and exposure to particular foods were sought. And we conducted rectal swab for culture to 243 students, 33 staffs and 9 food handlers. Bacteriological examinations of water in the school were done. Cases were identified as subjects who had diarrhea (two or more loose stools in a 24-hour periods) on or after May 1. Results : A total of 307 cases (attack rate: 59.6%) of 515 subject were identified, including 50 confirmed (46 students and 4 staffs) by S. sonnei. All 9 food handlers denied illness and were had rectal swab for culture at May 6 that were negative for S. sonnei. 146 of 307 reported fever, 156 had tenesmus, 44 reported vomiting, and only 5 of 307 reported blood in the stool. The median duration of diarrhea was 4 days (range: 1-18 days). The mean incubation period until onset of diarrhea was 63 hours (range: 46-144 hours) and the secondary attack rate was 2.8% (43 cases of 1,561 family members). Risk for illness was higher among students who had eaten watered kimchi at March 30 than among those who did not [301(72.7%) of 417 versus 5(9.6%) of 52; RR=7.51;95% CI=3.26-17.31]. Conclusion : The source of infection was estimated to be contaminated watered kimchi by ore or two food hardier who is presumed to be carrier.

Prevalence and Microbial Flora of Chicken Slaughtering and Processing Procedure

  • This study has been performed to measure the prevalence and microbial flora on chicken slaughtering as well as the processing process from the months of October to November. Whole-chicken rinsing technique was used in order to analyze the incidence of microorganisms on chicken carcass at the stage before chilling (after evisceration), after chilling and after cutting. The swab technique was used on processing the processed samples, such as working plates and cutting knives. Brine and cooling water from four cooling tubs were taken from each processing processes and were used as samples. Furthermore, the matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) for whole cell fingerprinting in combination with a dedicated bioinformatic software tool was used to identify the isolated microorganisms. Of the tested samples and processes, brine ($4.50{\pm}0.64$ Log CFU/mL) and chicken carcass before chilling ($4.15{\pm}0.46$ Log CFU/mL) showed the highest population of microorganisms; the predominant microbial flora of them were Moellerella wisconsensis (54.84%), a member of the Enterobacteriaceae family, and Escherichia coli (60.36%), respectively. However, the predominant microbial flora of cut carcass was changed to Staphylococcus aureus (27.32%), which is a kind of pathogenic microorganism that can cause a food-borne illness. Therefore, the slaughtering and processing procedure of chicken are needed to be controlled more hygienically.

Serological Detection of Antibodies against Anaplasma spp. in Cattle Reared in the Gyeongsangbuk-do, Korea

  • Anaplasmosis is a tick-borne, non-contagious, zoonotic disease caused by Anaplasma spp., which include Anaplasma marginale, A. centrale, A. phagocytophilum, A. platys, A. ovis, and A. bovis. Recently, in Korea, the prevalence of Anaplasma spp. has been investigated in some animals, such as dogs, horses, goats, cats, and Korean water deer. In cattle, A. marginale is the most virulent species and regarded as the typical type of species. However, data on the seroprevalence of Anaplasma spp. in cattle in Korea during the last decade is limited. This study was designed to investigate the seroprevalence of bovine anaplasmosis in Korea. From 2010 to 2013, blood samples were collected from 568 cattle. Forty animals (7.0%) tested seropositive for Anaplasma spp. by cELISA. Despite that current bovine anaplasmosis seropositivity rate in the Gyeongsangbuk-do is lower than those in tropical countries, anaplasmosis needs to be regarded as a concerning disease. The identification of the specific Anaplasma species infecting cattle in this province requires additional molecular studies. Moreover, further monitoring and control programs for bovine anaplasmosis is required, and the information from this study will be beneficial to develop these programs.

서울지역 학교급식 위생관리 수행수준 평가 (Assessment of Food Safety Management Performance for School Food Service in the Seoul Area)

  • Effective and systematic sanitation management programs are necessary to prevent foodborne disease outbreaks in school foodservice operations. The purpose of this study was to identify the elements to improve in order to ensure the safety of school food service by evaluating sanitation management practices implemented under HACCP-based programs. The survey was designed to assess the level of hygiene practices of school food service by using an inspection checklist of food hygiene and safety. Fifty-four school foodservice establishments considered as poor sanitation practice groups from two year inspections by Seoul Metropolitan Office of Education were surveyed from September to December in 2005. Inspection checklists consisted of seven categories with 50 checkpoints; facilities and equipment management, personal hygiene, ingredient control, process control, environmental sanitation management, HACCP system and safety management. Surveyed schools scored $68.0{\pm}12.42$ points out of 100 on average. The average score (% of compliance) of each field was 10.7/20 (53.3%) for facilities and equipment management, 7.4/11 (67.2%) for personal hygiene, 7.4/11 (74.1%) for ingredient control, 22.4/32 (69.8%) for process control, 8.9/12 (73.8%) for environmental sanitation management, 4.2/7 (59.7%) for HACCP systems management, and 7.2/8 (89.7%) for safety management, respectively. The field to be improved first was the sanitation control of facilities and equipment. The elements to improve this category were unprofessional consultation for kitchen layout, improper compartment of the kitchen area, lacks of pest control, inadequate water supply, poor ventilation system, and insufficient hand-washing facilities. To elevate the overall performance level of sanitation management, prerequisite programs prior to HACCP plan implementation should be stressed on the school officials, specifically principals, for the integration of the system.

Identification and spectral analysis of the CIBER/LRS detected stars

  • CIBER (Cosmic Infrared Background ExpeRiment) is a sounding-rocket borne experiment which is designed to find the evidence of the First stars (Pop.III stars) in the universe. They are expected to be formed between the recombination era at z ~ 1100 and the most distant quasar (z ~ 8). They have never been directly detected due to its faintness so far, but can be observed as a background radiation at around $1{\mu}m$ which is called the Cosmic Near-Infrared Background (CNB). The CIBER is successfully launched on July 10, 2010 at White Sands Missile Range, New Mexico, USA. It consists of three kinds of instruments. One of them is a LRS (Low Resolution Spectrometer) which is a refractive telescope of 5.5 cm aperture with spectral resolution of 20 ~ 30 and wavelength coverage of 0.7 to $2.0{\mu}m$ to measure the spectrum of the CNB. Since LRS detects not only CNB but also stellar components, we can study their spectral features with the broad band advantage especially at around $1{\mu}m$ which is difficult at ground observations because of the atmospheric absorption by water vapor. I identified around 300 stars from observed six fields. If we can classify their spectral types with SED fitting, we can study their physical conditions of the stellar atmosphere as well as making a stellar catalogue of continuous stellar spectrum.

The In Vitro and In Vivo Efficacy of Hen IgY Against Vibrio parahaemolyticus and Vibrio vulnificus

  • The inhibitory effect of IgY against Vibrio parahaemolyticus and Vibrio vulnificus responsible for seafood-borne diseases was investigated in this study. Water-soluble fractions (WSF) of protein containing IgYs were isolated from the egg yolk of hens initially immunized with formalin-inactivated V. parahaemolyticus or V. vulnificus. Protein, total and specific IgY contents of the WSF were determined. The inhibitory and protective effects of IgYs on the growth of V. parahaemolyticus and V. vulnificus were assayed in liquid medium and in mice. IgYs showed high affinity to their corresponding antigens with high titer from day 28 onwards. Protein contents and total IgY concentrations remained stable throughout the immunization period, whereas specific IgY concentrations increased steadily and reached a plateau at day 49. Specific IgY powder (150 mg/ml) significantly inhibited further multiplication of both V. parahaemolyticus and V. vulnificus in liquid medium as compared with the control IgY. The bacteria count in mice feces was lower in mice pretreated with specific IgYs than in those pretreated with PBS or control IgY. Higher survival of mice was observed in the experimental groups pretreated with either anti-V. parahaemolyticus (75% survival) or anti-V. vulnificus (87% survival) IgYs, compared with those in the control groups pretreated with PBS or nonspecific IgY. All mice in the control groups died within three days after bacteria inoculation; hence, the protective effect of specific IgYs against infection caused by V. parahaemolyticus and V. vulnificus was demonstrated.

2013-2016년 경남 연안 해수 및 패류에서 병원성 비브리오균의 계절적 및 지역적 변동 (Seasonal and Spatial Variation of Pathogenic Vibrio Species Isolated from Seawater and Shellfish off the Gyeongnam Coast of Korea in 2013-2016)

  • The seasonal and spatial variation of pathogenic Vibrio species, such as V. parahaemolyticus, V. vulnificus, V. alginolyticus, and V. cholerae were investigated in seawater and in bivalves off the Gyeongnam coast of Korea, which is an important area for shellfish production, during the period 2013-2016. V. parahaemolyticus, V. vulnificus, V. alginolyticus, and V. cholerae were detected in 12.1%, 5.2%, 15.4%, and 0.9% of seawater samples, respectively. V. parahaemolyticus, V. vulnificus, V. alginolyticus, and V. cholera were detected in 21.9%, 7.1%, 12.2%, and 0.0% of shellfish samples, respectively. The Vibrio spp. in seawater and bivalve samples were detected at high levels during the summer to early autumn; however, the levels were low during the winter. Therefore, their occurrence was seasonally dependent and correlated with high water temperature, which is also the biggest factor contributing to foodborne outbreaks associated with Vibrio. Relatively high detection rates of the strains were also found in the sea area that was continually exposed to inland wastewater. Our findings show that continuous monitoring is needed to reveal the patterns of occurrence of these pathogens from marine samples collected off the Korean coast, to reduce seafood-borne outbreaks caused by Vibrio.

  • 수계 압력감응점착제(PSA)는 유해성 휘발성유기용매 사용을 최소화할 수 있는 친환경 기술로써 학계와 산업계에서 큰 관심을 받아왔다. 수계 PSA의 합성은 유화중합을 이용하는데 이 때 이용되는 페놀계 계면활성제는 환경 및 인체유해성이 높아 이를 대체할 수 있는 신규 계면활성제 개발이 산업적으로 요구된다. 본 논문에서는 환경유해성 페놀계 CO-436 계면활성제를 대체할 수 있는 스티렌페놀계 계면활성제를 이용하여 수계 PSA를 합성하고 스티렌페놀계 계면활성제의 종류 및 농도에 따른 수계 PSA의 물성과 산업계 응용 가능성에 대해 알아보고자 한다.

Bactericidal Efficacy of Oxidized Silver against Biofilms Formed by Curtobacterium flaccumfaciens pv. flaccumfaciens

  • Bacterial wilt is a re-emerging disease on dry bean and can affect many other crop species within the Fabaceae. The causal agent, Curtobacterium flaccumfaciens pv. flaccumfaciens (CFF), is a small, Gram-positive, rod-shaped bacterium that is seed-transmitted. Infections in the host become systemic, leading to wilting and economic loss. Clean seed programs and bactericidal seed treatments are two critical management tools. This study characterizes the efficacies of five bactericidal chemicals against CFF. It was hypothesized that this bacterium was capable of forming biofilms, and that the cells within biofilms would be more tolerant to bactericidal treatments. The minimum biocide eradication concentration assay protocol was used to grow CFF biofilms, expose the biofilms to bactericides, and enumerate survivors compared to a non-treated control (water). Streptomycin and oxysilver bisulfate had EC95 values at the lowest concentrations and are likely the best candidates for seed treatment products for controlling seed-borne bacterial wilt of bean. The results showed that CFF formed biofilms during at least two phases of the bacterial wilt disease cycle, and the biofilms were much more difficult to eradicate than their planktonic counterparts. Overall, biofilm formation by CFF is an important part of the bacterial wilt disease cycle in dry edible bean and antibiofilm bactericides such as streptomycin and oxysilver bisulfate may be best suited for use in disease management.
