Evaluation of Organic Matter and Trace Metal Contaminations of Intertidal Sediments from Coastal Islands in the Southern Region of Jeollanam Province (전남 남부 도서갯벌 퇴적물의 유기물 및 미량금속 오염 평가)

  • We measured the grain size, ignition loss (IL), chemical oxygen demand (COD), acid volatile sulfide (AVS) and trace metals (Al, Fe, Cu, Pb, Zn, Cd, Cr, Mn, Hg, and As) of intertidal surface sediment collected from 11 islands (62 stations) in the southern region of Jeollanam Province. The objective of this research was to evaluate the organic matter and trace metals contaminations of sediments from coastal island tidal flats. Surface sediment texture was characterized as follows: mud, sandy silt, muddy sand, and slightly gravelly sand facies. The finer sediments are mainly dominated in the northern part of each island. The concentrations of IL, COD, AVS and some trace metals (Al, Fe, Zn, Cr, Cu, and Hg) were higher in the northwestern part of Wan Island and the area between Gogeum and Sinji Islands, and were associated with relatively finer sediment, as compared to other locations. The concentrations of Mn, Pb, Cd, and As were higher in the northwestern and southeastern parts of Geoguem and Pyungil Islands, but were not correlated with mean grain size. Based on sediment quality guidelines (SQGs), the concentrations of trace metals were lower than the values of effect range low (ERL), used in United States, and threshold effects level (TEL), used in Korea, with exception of As. Similarly, the intertidal sediments were moderately contaminated with As, based on the the enrichment factor (EF) and the geoaccumulation index ($I_{geo}$). The high concentration of As in intertidal sediments from this study region may be due to the input of naturally or artificially contaminated submarine groundwater, contaminated waste from seaweed aquaculture operations and/or land-based seaweed processing facilities. Further studies are needed to identify the sources of As in this study region, and to determine the effects of As contamination on coastal ecosystem.

Duckweed as a Protein Source for Fine-Wool Merino Sheep: Its Edibility and Effects on Wool Yield and Characteristics

  • Two experiments were carried out to investigate whether duckweed is useful as a dietary protein source for fine-wool Merino sheep and to evaluate its effects on wool yield and characteristics. In Experiment 1, the sheep were given one of three maintenance diets consisting of oaten chaff (520-700 g/d) supplemented with 16-32 g crude protein/d in the form of fresh (1 kg/d) or sun-dried (50-100 g/d) duckweed. Each ration was estimated to provide 5.4 MJ (1.3 Mcal)/d of metabolisable energy (ME). The sheep readily ingested the fresh or dried duckweed. None of the wool measures (yield, rate of fibre elongation, fibre diameter) differed (p>0.05) between dietary treatments. In Experiment 2, oaten-chaff-based diets (800 g/d) supplying 6.5-7.2 MJ (1.6-1.7 Mcal)/d of ME were supplemented with iso-nitrogenous amounts (4-5 g N) either of urea (8 g), cottonseed meal (60 g) or dried duckweed (100 g). In this experiment, the rate of wool fibre elongation, thought to be related to intestinal amino acid absorption, was lower (p<0.05) for sheep given the oaten chaff/urea diet than for those given either oaten chaff/cottonseed meal or oaten chaff/duckweed for which the rates did not differ (p>0.05). Fibre diameter, which ranged from 16.0-16.7 mm, did not differ (p>0.05) between diets, but tended to be lower on the oaten chaff/urea diet so that volume of wool produced was also significantly lower (p<0.05) on this diet than on the diets containing duckweed or cottonseed meal. Rumen ammonia concentrations at 4.5 and 7.5 h after feeding were higher (p<0.05) for sheep given the oaten chaff/urea diet than for those given the other two diets. A comparison of the rumen ammonia concentrations, wool growth rate and predicted flows of amino acids from the rumen of sheep supplemented with duckweed rather than cottonseed meal suggested that duckweed is a valuable source of 'escape protein' for ruminants.

An application of the electrostatic spray technology to increase scrubbing efficiency of SO$_{2}$ emitted from thermal systems (열시스템에서 생성된 SO$_{2}$ 가스의 배출저감을 위한 정전기 분무 원리의 응용)

  • Emission control of acid exhaust gases from coal-fired power plants and waste incinerators has become an increasing concern of both industries and regulators. Among those gaseous emissions, SO$_{2}$ has been eliminated by a Spray Drying Absorber (SDA) system, where the exhaust gas is mixed with atomized limestone-water slurry droplets and then the chemical reaction of SO$_{2}$ with alkaline components of the liquid feed forms sulfates. Liquid atomization is necessary because it maximizes the reaction efficiency by increasing the total surface area of the alkaline components. An experimental study was performed with a laboratory scale SDA to investigate whether the scrubbing efficiency for SO$_{2}$ reduction increased or not with the application of a DC electric field to the limestone-water slurry. For a selected experimental condition SO$_{2}$ concentrations exited from the reactor were measured with various applied voltages and liquid flow rates. The applied voltage varied from -10 to 10 kV by 1 kV, and the volume flow rate of slurry was set to 15, 25, 35 ml/min which were within the range of emission mode. Consequently, the SO$_{2}$ scrubbing efficiency increased with increasing the applied voltage but was independent of the polarity of the applied voltage. For the electrical and flow conditions considered a theoretical study of estimating average size and charge of the atomized droplets was carried out based on the measured current-voltage characteristics. The droplet charge to mass ratio increased and the droplet diameter decreased as the strength of the applied voltage increased.

Sphingopyxis granuli sp. nov., a $\beta$-Glucosidase-Producing Bacterium in the Family Sphingomonadaceae in $\alpha$-4 Subclass of the Proteobacteria

  • Strain Kw07$^T$, a Gram-negative, non-spore-forming, rod-shaped bacterium, was isolated from granules in an Up-flow Anaerobic Sludge Blanket (UASB) bioreactor used in the treatment of brewery waste­water. 16S rRNA gene sequence analysis revealed that strain Kw07T belongs to the a-4 subclass of the Proteobacteria, and the highest degree of sequence similarity was determined to be to Sphingopyxis macrogoltabida IFO 15033T (97.8%). Chemotaxonomic data revealed that strain Kw07T possesses a quinone system with the predominant compound Q-I0, the predominant fatty acid C,s:, OJ7c, and sphingolipids, aU of which corroborated our assignment ofthe strain to the Sphingopyxis genus. The results of DNA-DNA hybridization and physiological and biochemical tests clearly demonstrated that strain Kw07T represents a distinct species. Based on these data, Kw07T (= KCTC 12209T = NBRC 100800T) should be classified as the type strain for a novel Sphingopyxis species, for which the name Sphingopyxis granuli sp. novo has been proposed.

A Biological Study on the Methanol-Utilizing Bacteria (Methanol 자화세균에 관한 생물학적 연구)

  • By the successive enrichment culture, more than 250 methanol-utilizing bacteria were isolated from various samples such as soil, waste water and sewage. Two strains of which were selected and tentatively identified as Acinetobacter sp. and Pseudomonas sp. experiments were carried out to determine the growth conditions for the higher biomass yield and to demonstrate the difference to protein composition dependent upon carbon sources of these two species. the results were as follows ; 1. the optimum pH was determined as 8 in the both species. The optimum temperature in Acinetobacter sp. was $25^{\circ}C{\sim}30^{\circ}C$ and pseudomonas sp. was $30^{\circ}C-35^{\circ}C$. The optimum initial concentration of mthanol was determined as 1-2% in Acinetobacter sp. and 2-3% in pseudomonas sp. 2. The optimum concnetrations of nitrogen source, micro-elements, and vitamins such as biotin and thiamine-HCl in Acnetobactar sp. were 1g $(NH_4)_3SO4,\;1{\sim}3mg\;Mn^{++},\;4mg\;Fe^{++},\;10{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine-HCl per liter medium. In the Pseudomonas sp., 2g $(NH_4)_3SO4,\;1mg\;Mn^{++},\;trace\;amounts\;of\;Fe^{++},\;5{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine HCl per liter were effective. Maximum biomass yield was 2.5g/l in Acinetobacter sp. and 4.8g/l in Pseudomonas sp. 3. Protein composition of the two strains exhibited that alkai-labile protein was higher than alkali-stable protein. In Pseudomonas sp., the contents of acid soluble fraction and alkali-stable protein of the cells grown in the methanol medium were higher than in sucrose medium. On the other hand, in Acinetobacter sp., alkalilabile protein of the cells grown in sucrose medium was higher than in methanol medium.

Characterization of Lipases from Staphylococcus aureus and Staphylococcus epidermidis Isolated from Human Facial Sebaceous Skin

  • Two staphylococcal lipases were obtained from Staphylococcus epidermidis S2 and Staphylococcus aureus S11 isolated from sebaceous areas on the skin of the human face. The molecular mass of both enzymes was estimated to be 45 kDa by SDS-PAGE. S2 lipase displayed its highest activity in the hydrolysis of olive oil at $32^{\circ}C$ and pH 8, whereas S11 lipase showed optimal activity at $31^{\circ}C$ and pH 8.5. The S2 lipase showed the property of cold-adaptation, with activation energy of 6.52 kcal/mol. In contrast, S11 lipase's activation energy, at 21 kcal/mol, was more characteristic of mesophilic lipases. S2 lipase was stable up to $45^{\circ}C$ and within the pH range from 5 to 9, whereas S11 lipase was stable up to $50^{\circ}C$ and from pH 6 to 10. Both enzymes had high activity against tributyrin, waste soybean oil, and fish oil. Sequence analysis of the S2 lipase gene showed an open reading frame of 2,067 bp encoding a signal peptide (35 aa), a pro-peptide (267 aa), and a mature enzyme (386 aa); the S11 lipase gene, at 2,076 bp, also encoded a signal peptide (37 aa), pro-peptide (255 aa), and mature enzyme (399 aa). The two enzymes maintained amino acid sequence identity of 98-99% with other similar staphylococcal lipases. Their microbial origins and biochemical properties may make these staphylococcal lipases isolated from facial sebaceous skin suitable for use as catalysts in the cosmetic, medicinal, food, or detergent industries.

A Study on the Tetrafluoroborate Decomposition Reaction and Removal of Fluoride Using Aluminum (알루미늄을 이용한 불화붕산염의 분해 반응 및 불소 처리에 관한 연구)

  • The fluorine-containing waste water tends to show a higher removal efficiency through the coagulative precipitation process with calcium. However the tetrafluoroborate produced from the etching process is difficult to remaval due to it's low reactivity with calcium. The objective of this study is improving the efficiency of fluoride ion removal in tetrafluoroborate through decomposing. Research on tetrafluoroborate decomposition depending on reaction pH, temperature, and aluminum dosage were conducted, using a laboratory-scale reactor. The result shows that the reaction of tetrafluoroborate with aluminum is faster with lower pH, higher water temperature, and higher Al/T-F (Aluminum/Total Fluoride) mole ratio. It is found that there was no big change in concentration after over 120 minutes of reaction. This study is to be able to improve the efficiency of tetrafluoroborate and fluoride wastewater treatment by using aluminum.

Rcovery of Nitric acid, Copper and Nickel from Plating Waste of Automobile (자동차 휠 도금박리 폐액으로부터 용매추출법을 이용한 질산과 구리추출 및 니켈회수)

  • 자동차용 고광택 크롬 도금 박리액에는 질산과 유가금속인 구리 및 니켈이 다량 함유되어 있는 것으로 알려져 있다. 질산($HNO_3$) 및 유가금속은 고가이며 유독하므로 경제적 및 친환경적으로 반드시 회수하여 재활용하여야 한다. 본 연구에서는 도금박리액으로부터 질산과 구리, 니켈을 용매 추출법을 이용하여 분리하였다. 수상에 존재하는 질산의 농도는 0.01 ~ 1N NaOH를 이용하여 적정하여 분석하고, 금속의 농도는 ICP-MS 및 ICP-AES 등을 이용하여 분석하였다. 도금 박리액을 분석한 결과 Cu(76850mg/L), Ni(51990 mg/L)이 함유되어 있음을 알 수 있었다. 용액 내 질산의 양을 NaOH 용액을 이용하여 적정법으로 측정하였을 때, 질산의 양은 대략 1.02 M 임을 알 수 있었다. 50 % Tributylphosphate (TBP)를 이용하여 3단 추출한 유기층의 용액을 증류수를 이용하여 3회의 역추출을 하였을 때, 원액으로부터 48.1 %의 질산을 회수할 수 있음을 알 수 있었으며, 순도는 99.5% 이상이었다. 질산 회수 후 용액 내에 남은 구리와 니켈은 ISE-106로 구리를 추출하여 니켈을 분리한 후 황산을 이용해 역추출 하였다. 회수된 구리는 NaOH를 이용하여 pH를 조절하고 수산화구리 형태로 침전시킨 후 $N_2H_4$를 이용하여 환원시켰고, 온도와 pH 및 환원제를 이용하여 다양한 조건 하에 구리 분말을 제조하였다. 구리를 추출하여 라피네이트 용액으로 분리된 니켈은 $NaBH_4$를 이용하여 환원시켰고, 다양한 조건 하에서 니켈 분말을 제조하였다. 환원 된 분말은 분석결과 99%의 순수한 분말임을 알 수 있었다.

  • Biodiesel is synthesized by transesterification of vegetable or animal oils with alcohols. Since it has similar characteristic with diesel fuel, it can be used as a fuel by mixing with diesel fuel. Moreover, it is advantageous that biodiesel can reduce air pollution emitted from fuel combustion and is produced from sustainable energy, biomass. Recently, many researchers have investigated biodiesel synthesis using supercritical methanol since it is economical due to shorter reaction time and simple separation/purification process, compared with conventional alkali- or acid-catalyzed process. By the development of biodiesel production process from waste edible oil using supercritical methanol, it can be expected to utilize potential energy resources, reduce carbon dioxide emission, and improve environmental conditions.

Parametric Optimization of Feruloyl Esterase Production from Aspergillus terreus Strain GA2 Isolated from Tropical Agro-Ecosystems Cultivating Sweet Sorghum

  • A fungal strain, Aspergillus terreus strain GA2, isolated from an agricultural field cultivating sweet sorghum, produced feruloyl esterase using maize bran. In order to obtain maximum yields of feruloyl esterase, the solid state fermentation (SSF) conditions for enzyme production were standardized. Effective feruloyl esterase production was observed with maize bran as substrate followed by wheat bran, coconut husk, and rice husk among the tested agro-waste crop residues. Optimum particle size of 0.71-0.3 mm and moisture content of 80% favored enzyme production. Moreover, optimum feruloyl esterase production was observed at pH 6.0 and a temperature of $30^{\circ}C$. Supplementation of potato starch (0.6%) as the carbon source and casein (1%) as the nitrogen source favored enzyme production. Furthermore, the culture produced the enzyme after 7 days of incubation when the C:N ratio was 5. Optimization of the SSF conditions revealed that maximum enzyme activity (1,162 U/gds) was observed after 7 days in a production medium of 80% moisture content and pH 6.0 containing 16 g maize bran [25% (w/v)] of particle size of 0.71-0.3 mm, 0.6% potato starch, 3.0% casein, and 64 ml of formulated basal salt solution. Overall, the enzyme production was enhanced by 3.2-fold as compared with un-optimized conditions.
