Characteristics of Watermelon Mosaic Virus Transmission Occurring in Korean Ginseng (인삼에서 발생하는 수박모자이크바이러스의 감염 특성)

  • Korean ginseng (Panax ginseng) is the most popular herb for medical purpose in Korea. Recently, viral diseases from Korean ginseng showing various degrees of severe mottling, variegation and mosaic symptoms have caused quantity losses of Korean ginseng in a large number of farms. Watermelon mosaic virus (named WMV-gin) was identified as a causal agent for the disease of Korean ginseng. Interestingly, WMV-gin failed to infect both Korean ginseng plant and susceptible host species including cucurbitaceous plants by mechanical inoculation. However, WMV-gin could successfully infect Korean ginseng by transmission of two aphid species (Myzus persicae and Aphis gossypii). It is likely that transmission of WMV-gin was done by both the aphid species during feeding behavior of the two aphid species on Korean ginseng, though the aphids dislike feeding in Korea ginseng. Similarly, a strain of WMV (WMV-wm) isolated from watermelon was transmitted successfully to Korean ginseng plant by the two aphid species, but not by mechanical inoculations. Transmission assays using M. persicae and A. gossypii clearly showed both WMV-gin and WMV-wm were not transmitted from infected Korean ginseng plant to cucurbit species that are good host species for WMV. These results suggest WMV disease occurring in Korean ginseng plant can be controlled by ecological approaches.

  • This study was conducted to investigate the occurrence of Soil borne wheat mosaic virus(SbWMV) in barley fields in Korea and to examine the host pathogenicity of SbWMV. By using the ELISA test, SbWMV was detected in the six regions : Suwon, Milyang, Jinju, Youngkwang, Iksan, and Chonju. SbWMV was isolated from the two strains, Albori strain from Jinju and Eunpamil strain from Milyang. SbWMV was collected from leaves showing mosaic, yellowing and necrosis stripes. SbWMV was inoculated mechanically on 1∼1.5 leaf stages with leaf-rubbing to identify the host pathogenicity of 36 Korean barley cultivars, a wheat cultivar, two rye cultivars, three Japanese barley cultivars and Chenopodium amaranticola. Viral sympoms of inoculated leaves appeared on moulted loaves about 4 to 6 weeks of inoculation. Baegdong and Tapgolbori, infected from Albori strain and Eunpamil strain infected from Samdobori showed much higher susceptibility than C. amaranticola and C. quinoa which showed ring spots and chlorotic spots respectively. Virus particles were observed by the electron microscope. They were rod-shapes, which are bipartite, of 142 nm or 281 nm in length with 20 nm diameter on infected leaves. Specific detection and identification of SbWMV was set up using the RT-PCR. PCR fragments of SbWMV(0.5kb) were obtained by using the designed primers for SbWMV RNA 2.

  • WMV(Winoow Media Video)는 ASF(Advanced Streaming Format)파일 포맷을 기반으로 하는 마이크로 소프트사의 대표적인 스트리밍 파일 포맷이다. WMV 파일 포맷은 MPEG-4를 기반으로 내용 기반 부호화를 통해 영상을 객체 단위로 나누어 처리하므로 사용자의 의도에 따라 다양한 조작과 디스플레이가 가능하다. 또한 영상 데이터를 패킷에 붙여 네트워크를 통해 스트리밍 서비스도 가능하다. 따라서 본 논문에서는 WMV파일 포맷 분석을 통해 영상의 가장 중요한 Hey-Frame을 추출과 데이터 헤더 부분을 제외한 실제 영상 스트림의 시작 위치를 찾는 알고리즘을 설계 및 구현하였다. 이로써 실제 스트리ald 서비스외에도 다양한 멀티디미어 서비스에 적용 가능한 방법을 마련하였다.

Ultrastructural Aspects of the Mixed Infections of Watermelon Mosaic Potyvirus and Cucumber Green Mottle Mosaic Tobamovirus Isolated from Watermelon

  • Symptoms on 4 varieties of watermelons inoculated with watermelon mosaic potyvirus II isolated from watermelon (WMV-W) were severe mosaic and leaf malformation while those inoculated with cucumber green mottle mosaic tobamovirus from watermelon (CGMMV-W) were mild mosaic and chlorotic spots. Inoculation of the mixture of WMV-W and CGMMV-W produced extremely severe mosaic along with necrotic spots and general necrosis. Doubly infected plants were also stunted. Cells infected with WMV-W or CGMMV-W alone exhibited the intrinsically ultra-structural properties of each virus infection. WMV-W induced potyvirus-characteristic cylindrical inclusions in the cytosol. Virus particles were orderly aligned along the tonoplasts. CGMMV-W induced tobamovirus-characteristic stacked crystalline arrays of virus particles in the cytosol. Cells infected doubly with WMV-W and CGMMV-W contained striking cytopathic effects that were not present in single infection of each virus. The unique ring structure, nonagon, was that a single potyvirus particle was surrounded by 9 CGMMV-W tobamovirus particles.

  • Mixed infection of watermelon mosaic potyvirus II isolated from pumpkin (WMV-P) and cucumber green mottle mosaic tobamovirus from watermelon (CGMMV-W) caused extremely severe symptoms such as progressive silting and death of watermelon plants. Single infections of either WMV-P or CGMMV-W on the same hosts produced only vein clearing and/or mosaic on the upper leaves. In cells infected with WMV-P, potyvirus-characteristic inclusions of pinwheels, scrolls and cylindrical inclusions were present in the cytosol. Parallel arrays of virus particles in the tonoplast were also common. In cells infected with CGMMV-W, virus particles occurred as stacked-bands of scattered randomly in the cytosol and vacuoles in all type cells. Many cells also contained vesiculated mitochondria with fibril-containing vesicles. Cells infected mixedly with WMV-P and CGMMV-W contained structural features that were not observed in cells infected singly with the two viruses. A particle of WMV-P potyvirus was surrounded by evenly spaced nine particles of CGMMV-W tobamovirus, which made a unique nonagon ring. The angled layers having $60^{\circ}$-$135^{\circ}$ were alternating layer, herringbone, crosshatching and ladder figures.

  • As content serviced for IP set-top boxes is streamed over IP network, the existing hacking tools for IP network can be used to capture the streamed content. Until recently, most of the content serviced on IP set-top boxes has been MPEG-2 TS. However, this content will be gradually moved to WMV, MPEG-4 or H.264 because of the relatively low compression efficiency and overhead of the TS packet. In this paper, we propose a DRM scheme other than WMRM for streamed WMV content. Our approach is to design a DRM scheme independent to the existing WMV streaming system. We also design this scheme in order to provide the feature for controlling the DRM processing time considering device performance. We verified it through the experiment.

  • Due to the development of information telecommunication technology, e-learning industry is rapidly expanding its scope along with its production technology. The recent trend of e-learning program is likely converted from Wmv(Window Media Video) of Microsoft to Flv(Flash video), which has less capacity but better quality than other image file. It has successfully drawn the users attention since Flv can operate at most OS environments and browsers let alone with window and Lenux without extra players and codec setup. However, there is no accurate data on comparative analysis between Wmv and Flv regarding capacity, quality and production time. Therefore, the study shows the comparative data analysis on Wmv and Flv so as to set out production platform up to its idiosyncrasy.

Incidence of Watermelon Mosaic Virus in Cucurbits (박과 작물에 발생하는 Watermelon Mosaic Virus에 관한 연구)

  • Cucurbits including pumpkin (Cucurbita pepo), gourd (Lagenariaa siceraria), cucumber (Cucumis sativus), melon(Cucumis melo) and watermelon(Cucurbita anguria) were diseased with mosaic symptoms. The causal virus was identified as watermelon mosaic virus(WMV). The WMV was transmitted by Myzus persicae Sulzer, and no seed borne virus was found. The virus caused large local lesions on the inoculated leaves of the Chenopodium amaranticolor and mosaic symptom on the upper leaves of Cucumis melo, Cucumis sativus, Lagenaria siceraria, Cucurbita anguria and Cucurbita pepo. There were no symptoms on the inoculated leaves of the Nicotiana tabacum var. Bright yellow, Nicotiana glutinosa, Vigna unguiculata. Petunia hybrida and Datura stramonium. Thermal inactivation point was $55\~65^{\circ}C$, dilution end point was $10^{-4}\;10^{-5}$ and longevity in vitro of the virus was $7\~8$ days. The virus showed positive reaction against watermelon mosaic virus antiserum in microprecipitin tests. The virus particles were flexuous rods in size of 750 nm.

  • A disease, supposedly caused by a virus, was observed from Insam (Panax ginseng) fields of Punggi in year 2006. It has long believed to be a physiological disorder. However, the incidence of the disease has increased every year. When several samples were observed under electron microscope, filamentous virus-like particles were observed. The nucleotide sequences of the virus were analyzed by RT-PCR with specific primer sets derived from the results of DNA chip. The results indicated that the disease was caused by Watermelon mosaic virus (WMV). It revealed that the result of the biological assay by the virus was different from that of WMV previously found in other crops. Therefore, this is the first report that WMV causes the disease in P. ginseng and the virus is named to be WMV-Insam.

  • 오늘날 우리 생활에 영상이나 이미지는 우리 실생활에 아주 밀접하게 연관되어 있다. 카메라, 휴대폰, TV, 영상 및 이미지 관련 기기들이 증가하고 이로 인해 영상이나 이미지 관련 서비스의 기술적인 요소들이 중요시되고 있다. 이러한 영상에서 기본적으로 사용하는 압축방식인 DCT는 직교 변환 방식의 국제 표준으로써, 정지 이미지나 동영상의 압축 파일등에서 사용된다. DCT(Discrete Cosine Transform) 알고리즘은 음성 및 영상 압축 등 많은 디지털 신호처리 분야에서 사용되고 있다. 본 논문에서는 WMV의 $4{\times}4$, $4{\times}8$,$8{\times}4$, $8{\times}8$ 4가지 모드에 대해 DCT를 지원할 수 있도록 C언어를 통해 상위 수준의 검증을 수행하고, 이를 HDL을 사용하여 코딩하고, Modelsim SE6.1을 사용해 회로 검증하였다.
