Quality of Stored Grape (Vitis labruscana) Treated with Ethylene-Absorbent and Activated Charcoal (에틸렌 흡착제와 활성탄 처리에 의한 저장 포도의 품질 특성)

  • In order to ensure a stable supply of grapes for consumers and to extend grape storage, we investigated changes in the quality of grapes (Vitis labruscana) stored after treatment with ethylene absorbent (EA) or activated charcoal (AC). We treated harvested 'Sheridan' and 'Muscat Bailey A' grapes with EA and with 0.1, 0.5, 1.0, and 2.0% (w/w) AC, and stored them at $0{\pm}1^{\circ}C$, $90{\pm}2%$ humidity for 60 days. After the EA or AC treatment, the storability of the 'Sheridan' and 'Muscat Bailey A' grapes was increased, as compared with non treatment, and 'Sheridan' kept better than 'Muscat Bailey A'. The quality of the stored grapes maintained with EA or the amount of AC, which decreased the rate of weight loss, abnormal fruit development, berry abscission, saccharinity, and growth of fungi. The free sugar and anthocyanin contents in the pericarp increased gradually in both 'Sheridan' and 'Muscat Bailey A', and increased more in 'Sheridan'. Conversely, the total acidity of 'Sheridan' and 'Muscat Bailey A' decreased with storage, and decreased less in 'Sheridan'. There were no differences in the free sugar content, anthocyanin content, and total acidity among treatments. The changes in grape quality with storage were delayed in the order of EA, 2% AC, 1% AC, 0.5% AC, and 0.1% AC. Ultimately, we postulate that EA and 2% AC best maintain the qualities of grape, and extend the grape storage period.

Biological Activities of Ethanol Extracts and Fractions of Black Olympia Grape(Vitis Labruscana L.) (거봉 포도종의 에탄올 추출물 및 분획물에 대한 생리활성 효능)

  • This study was conducted to determine biological activities, such as lipid peroxidation inhibition and cytotoxic effect of ethanol extracts of Black Olympia grape seeds and skins, and of organic solvent fractionated ethanol extracts obtained from grape seeds and skins at different temperatures. Among different extraction temperatures, the ethanol extract of grape seed obtained at 30$\^{C}$ had the strongest lipid oxidation inhibition of 60.1%, while the strongest lipid oxidation inhibitory effect of 71.2% was observed in the presence of 20 $\mu\textrm{g}$/㎖ ethylacetate fraction obtained from ethanol extract of grape seeds at 30$\^{C}$. The ethanol extract of grape seeds showed more strong lipid oxidation inhibition than that of skin extracts. Similar results were observed in cytotoxic effects. The ethanol extract of grape seeds at 30$\^{C}$ exhibited more strong cytotoxicity than that of skin extracts on MCF-7, Hep3B, and A549 cell lines. Among organic solvent fractions extracted from the ethanol extracts of gape seeds and skins, the hexane fraction showed the strongest cytotoxic inhibition of 75.15% and 62.50% on MCF-7 and Hep3B cell in the presence of 1.0 $\mu\textrm{g}$/㎖ respectively. On the other hand, the water fraction showed the strongest cytotoxic inhibition of 65.41% on A549 cell in the presence of 1.0 $\mu\textrm{g}$/㎖. Overall, the ethanol extracts and their fractions of Black Olympia grape seeds showed strong lipid oxidation inhibition and cytotoxicity than those of grape skins.

Characteristics of Ice Wine Fermentation of Freeze-Concentrated Campbell Early Grape Juice by S. cerevisiae S13 and D8 Isolated from Korean Grapes (포도로부터 분리한 S. cerevisiae S13 및 D8에 의한 캠벨 얼리 동결농축 과즙의 아이스와인 발효 특성)

  • Cryoextraction (a freeze concentration using an instrument) can increase the sugar concentration in grape juice by reducing its water content, similar to the natural freezing of grapes for natural ice wine. In this study, fermentation of freeze-concentrated Campbell Early grape (Vitis labruscana) juice to 36 $^{\circ}Bx$ was carried out using Saccharomyces cerevisiae strains D8 and S13 isolated from Korean grapes. During the fermentation, strains S13 and D8 showed rapid sugar reduction and alcohol production compared with S. cerevisiae Fermivin$^{(R)}$ used as a control. After nine-day fermentation, the residual sugar contents were lower in W13(9.77%) and D8 wine(9.07) than that in Fermivin$^{(R)}$ wine(14.0%). Total acid content was high in the D8>S13>Fermivin$^{(R)}$ wine, in that order. The acetaldehyde content was highest in the D8 wine and lowest in the Fermivin$^{(R)}$ wine, among the three. The methanol content was slightly higher in the S13 and D8 wines than in the Fermivin$^{(R)}$ wine. In the sensory evaluation, the S13 wine exhibited the highest score in flavor and taste among the three wines. Both the two S13 and D8 wines exhibited higher scores than Fermivin$^{(R)}$ wine in overall preference.

Effects of Streptomycin Sulfate on Parthenocarpy Induction in Grapes (Streptomycin을 이용(利用)한 포도(葡萄)의 단위결과(單爲結果) 유기(誘起)에 관(關)한 연구(硏究))

  • Several seeded cultivars of Vitis labruscana showing various responses to GA-induced seedlessness were tested to study the effects of streptomycin sulfate on induction of parthenocarpy. The results were summarized as follows. 1. Prebloom dipping of streptomycin sulfate at 200 ppm induced 90~100 % parthenocarpy in 'Schuyler', 'Delaware', 'Ohira-Dela.' and 'Tanored' cultivars and also stimulated maturity but did not show any toxic effects. 2. In most cultivars, the addition of 25 ppm GA to streptomycin tended to increase berry setting and induction of parthenocarpy. 3. Dipping of strepto mycin alone in 'Campbell Early' severely induced berry abscission but the addition of calcium acetate or BA to streptomycin slightly increased setting of berries. 4. In 'Kyoho' and 'Pione' cultivars, streptomycin successfully induced parthenocarpy. Berry setting by streptomycin treatment was variable according to environmental conditions. 5. In 'Campbell Early' and 'Tanored' cultivars, streptomycin severely reduced viability of pollen.

Free Radical Scavenging Effect of Seed and Skin Extracts from Campbell Early Grape (Vitis labruscana B.) (국내산 포도 캠벨종의 종자 및 과피 추출물의 Free Radical 소거능 탐색)

  • This study was conducted to determine optimal extraction condition and free radical scavenging effect of ethanol extracts of Campell Early grape seeds and skins at different temperatures, and of organic solvent fractionated from ethanol extracts of grape seeds and skins. The free radical scavenging effect was tested using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and expressed as dose required for a 50% reduction of DPPH radical (RC$_{50}$). The extraction yields were increased with increasing the extraction temperature but it was not related with ethanol concentrations. The ethanol extract of grape seeds obtained at 5$0^{\circ}C$ showed the more stronger RC$_{50}$ (16.8 $\mu\textrm{g}$/mL) than those of both 3$0^{\circ}C$ and 78$^{\circ}C$. The ethylacetate fraction obtained from ethanol extract of grape seed showed the strongest RC$_{50}$ (15.4 $\mu\textrm{g}$/mL). Overall, both ethanol extracts and their fractions of grape seeds showed significantly stronger free radical scavenging effect than those of skin extracts.racts.

Effect of GA3 and Thidiazuron on Seedlessness and Fruit Quality of 'Kyoho' Grapes (GA3와 Thidiazuron 처리가 '거봉' 포도의 무핵화와 과실품질에 미치는 영향)

  • This experiment was conducted to find the effects of a $GA_3$ and thidiazuron (TDZ) on seedless rate, harvest time, fruit cracking and fruit quality in 'Kyoho' grapes over two years from 2008 to 2009. In 2008, fruit clusters were dip treated with $GA_3$ $25.0mg{\cdot}L^{-1}$ twice at full bloom (FB) and 14 days after full bloom (DAFB) in a combination with TDZ 0 or $2.5mg{\cdot}L^{-1}$. Berry seedless rate and berry enlargement were slightly improved only when TDZ was added to the second $GA_3$ treatment at 14 DAFB, compared to $GA_3$ + TDZ treatments at both FB and 14 DAFB. However, berry cracking rate was significantly increased by any plant growth regulator (PGR) treatments compared to non treatment. In 2009, $GA_3$ at $12.5mg{\cdot}L^{-1}$ and $25.0mg{\cdot}L^{-1}$ was dip treated twice at FB and 14 DAFB while TDZ $2.5mg{\cdot}L^{-1}$ was treated only at 14 DAFB. Berry cracking rate was depended on the concentration of $GA_3$ applied. The higher concentration at $25.0mg{\cdot}L^{-1}$ significantly increased berry cracking rate while the lower concentration at $12.5mg{\cdot}L^{-1}$ had no effect. Also, the addition of TDZ to $GA_3$ $25.0mg{\cdot}L^{-1}$ at 14 DAFB, substantially decreased the cracking rate to the level of untreated control. Although all PGR treatments advanced fruit maturity, the most significant advance occurred when TDZ was added to $GA_3$ $12.5mg{\cdot}L^{-1}$ only at the second dip. Considering the overall aspects related to fruit maturity and quality, we concluded that the double applications of $12.5mg{\cdot}L^{-1}$ $GA_3$ at FB and 14 DAFB with addition of $2.5mg{\cdot}L^{-1}$ TDZ only at 14 DAFB was appropriate to produce about 400-500 g size of seedless 'Kyoho' grape cluster having 35-40 berries.

Characterization of the Effects of Different Wavelengths of Night-break Lighting on the Fruit Quality and Yield of 'Kyoho' Grapes (파장별 야간 조사에 따른 '거봉' 포도의 품질 및 생산량)

  • This study was conducted to estimate the light pollution damage caused by night-break lighting in 'Kyoho' grapes. Night irradiation was performed every night for four hours (10 p.m. to 2 a.m.) from the full bloom to the harvest stage using red, blue, and white lamps as artificial light. Fruit yield, soluble solid content, anthocyanin content, and chlorophyll a content were affected by night irradiation. The soluble solid content of the control was $17.5^{\circ}Brix$, approximately $1^{\circ}Brix$ higher than the red ($16.4^{\circ}Brix$), blue ($16.2^{\circ}Brix$), and white light treated grapes ($16.3^{\circ}Brix$). The anthocyanin content of the skin was also higher in the control at $4.08{\mu}g{\cdot}cm^{-2}$ compared to the red ($3.14{\mu}g{\cdot}cm^{-2}$), blue ($2.47{\mu}g{\cdot}cm^{-2}$), and white ($2.82{\mu}g{\cdot}cm^{-2}$) light treated samples. On the other hand, the chlorophyll a content of the control was the lowest at $0.268{\mu}g{\cdot}cm^{-2}$ as compared with the red ($0.339{\mu}g{\cdot}cm^{-2}$), blue ($0.345{\mu}g{\cdot}cm^{-2}$), and white ($0.372{\mu}g{\cdot}cm^{-2}$) light treated samples. Considering that higher soluble solid contents, higher accumulation of anthocyanin, and lower chlorophyll a contents are factors involved in fruit maturation, night irradiation may delay fruit maturation and red light treatment may result in decreased yield. Our results confirmed that night-break lighting regardless of the wavelength provoked light pollution in 'Kyoho' grapes. Therefore, fruit maturation may be poor in the presence of artificial light, including streetlamps, in neighboring vineyards.

Effects of the Elevated Temperature and Carbon Dioxide on Vine Growth and Fruit Quality of 'Campbell Early' Grapevines (Vitis labruscana) (온도와 이산화탄소의 상승처리가 포도 '캠벨얼리'의 수체생육과 과실품질에 미치는 영향)

  • The effects of elevated temperature and $CO_2$ concentration on vine growth and characteristics of fruits of three-year-old 'Campbell Early' grapevine were investigated. The treatment groups consisted of a control group (ambient temperature and $390{\mu}L{\cdot}L^{-1}\;CO_2$), an elevated temperature group (ambient temperature + $4.0^{\circ}C$ and $390{\mu}L{\cdot}L^{-1}\;CO_2$), an elevated $CO_2$ group (ambient temperature and $700{\mu}L{\cdot}L^{-1}\;CO_2$), and an elevated $CO_2$/temperature group (ambient temperature + $4.0^{\circ}C$ and $700{\mu}L{\cdot}L^{-1}\;CO_2$). The average shoot length was 312.6 cm in the elevated $CO_2$/temperature group, which was higher than the other groups; with 206.2 cm in the control group and 255.6 cm and 224.8 cm in the elevated temperature group and elevated $CO_2$ group respectively. However, the shoot diameter showed a tendency of decreasing in the elevated temperature and elevated $CO_2$/temperature groups. The equatorial diameter of berries was increased in the higher carbon dioxide concentration, and the soluble solid content was the highest in the elevated $CO_2$ group, with $14.6^{\circ}Brix$ among all treatment groups and the lowest in the elevated temperature group ($13.9^{\circ}Brix$). The harvest date was approximately 11 d earlier in the elevated $CO_2$/temperature group and 4 to 2 days earlier in the elevated $CO_2$ group and elevated temperature group, respectively. Regarding the rate of photosynthesis and transpiration during the growth period, higher photosynthetic rates were observed in the elevated $CO_2$ group and the elevated $CO_2$/temperature group during the early stage of growth; however the photosynthetic rate was reduced dramatically in summer, which was contrary to transpiration.

Effects of Night Temperature at Veraison on Berry Skin Coloration of 'Kyoho' Grapevines (포도 '거봉' 품종의 변색기 야간 온도 처리가 과피 착색에 미치는 영향)

  • We analyzed berry skin coloration, anthocyanin accumulation, and plant hormone contents in berry skins to determine the effect of night temperature at veraison on berry skin coloration in 'Kyoho' grapevines (Vitis labruscana L.). Vines were grown under 21, 24, and 27℃ at night for 20 days at veraison, from 40 to 60 days after full bloom (DAFB). Berry skin coloration of 'Kyoho' grapes was more suppressed in 27℃ treated vines, followed by that in 24℃ treated vines, than that in 21℃ treated vines. Cluster and berry weight and soluble solids content was lower in 24 and 27℃ treated vines than in 21℃ treated vines. Anthocyanin started to accumulate from 60 DAFB in berry skin of 21℃ treated vines, and malvidin and total anthocyanin content increased until 100 DAFB. The total and most of the individual anthocyanins decreased in 24 and 27℃ treated vines; however, peonidin did not decrease in 24℃ treated vines compared to that in 21℃ treated vines. Abscisic acid (ABA) peaked at veraison in berry skins of 21℃ treated vines and decreased thereafter until 100 DAFB. The increase in ABA content was inhibited in berry skins of 24 and 27℃ treated vines. Gibberellin (GA) content in berry skins decreased rapidly at veraison, with the decrease being slower under 27℃ than under 21℃. ABA/GA in berry skins of 21℃ treated vines peaked at 60 DAFB and decreased thereafter until 100 DAFB. However, ABA/GA decreased in berry skins of 24 and 27℃ treated vines, with reduced anthocyanin accumulation. Therefore, high night temperature (above 24℃) at veraison suppressed the berry skin coloration of 'Kyoho' grapes with changes in anthocyanin contents and composition due to the decrease in ABA/GA ratio and fruit soluble solids contents.
