Pharmacognostical Studies on the Folk 'Medicine MeoRuIp' (민간약 머루잎의 생약학적 연구)

  • Korean folk medicine 'MeoRuIp' has been used to cure cough, rheumatism and abdominal pain after child birth. There has been no pharmacognostical confirmation on the botanical origin of the crude drug. To clarify the botanical origin of 'MeoRuIp', the morphological and anatomical characteristics of the leaves of Vitis and Ampelopsis species growing in Korea, i.e. Vitis amurensis, V. amurensis forma. glabrescens, V. flexuosa, V. thunbergii var. sinuata and Ampelopsis brevipedunculata var. heterophylla were studied. As a result, it was clarified that 'MeoRuIp' was the leaf of Vitis amurensis and Vitis flexuosa.

DNA Topoisomerase I Inhibitory Activity of Stilbenes and Oligostilbenes from Leaf and Stem of Vitis amurensis

  • The DNA Topoisomerase I (DNA Topo I) inhibitory effect of ten isolated compounds (1.10) from the leaf and stem of Vitis amurensis were examined. Among them, amurensin G (5) and r-2-viniferin (7) showed high potent inhibitory activity against DNA Topo I. DNA Topo I, an important target for anticancer drugs, can cause DNA breaks and play a key role during cell proliferation, transcription and repair. Thus, the results suggest that the selected compounds (5 and 7) from Vitis amurensis have a possibility as DNA Topo I-targeting anticancer agents.

Pollen Characteristics of Flower Type and Cross Compatibility with Table Grape in Vitis amurensis Rupr. (왕머루의 화형에 따른 화분특성 및 포도의 교배화합성)

  • This study was conducted to identify cross compatibility with table grape and pollen characteristics by flower type in Vitis amurensis Rupr. (GW-22, GW-45, GW-56, GW-200, GW-202 and GW-300) collected in Gangwon area. Flower structure, germinability and morphology of pollen, and cross compatibility with table grape were examined. Flowers of GW-22 and GW-56 had an aborted gynoecium and five erect fdaments, and all of them was male. The percentage of pollen germination of GW-22 and GW-56 on the agar medium was 27.6-29.8%. Flowers of GW-45, GW-200, GW-202 and GW-300 had well-developed pistils and five stamens with curled filaments. The percentage of pollen germination of female plant on the agar medium was 0%. Pollen grail)s from GW-22 and GW-56 (male flower) have symmetrically distributed three furrows on their surface and each furrow contains a germ pore at the equatorial level (Tricolporated). GW-45, GW-200, GW202, and GW-300 (Female flower) produced pollen grains with no germinative structures (acolporated). In self-pollina-tion of female flowers, berries of clusters were no berry and no seed. Therefore, flower structure in V. amurensis proved to be that of other dioecious Vitis species. In cross combination of female and male flower of V. amurensis made normal seeds. In cross combination of table grape and V. amurensis of male flower made normal seeds. But, in cross combination of female flower of V. amurensis did not make seeds. Therefore, V. amurensis was cross compatible with vitis spp, and V. amurensis varieties may contribute to breeding studies of Vitis spp.

Growth and Fruit Characteristics of Vitis amurensis Rupr. Collected in Gangwondo (강원도에서 수집된 왕머루의 생장과 과실특성)

  • This study was carried out to evaluate the fruit and growth characteristics of selecting Vitis amurensis through functional material analysis and sensory evaluation in V. amurensis collected in Gangwondo. For evaluation of growth characteristics in V. amurensis, experiments were carried out by compared with the two grape cultivars 'Campbell Early' and 'Kyoho'. The full bloom and verasion time in V. amurensis was investigated faster than those of examined cultivar grapes, while harvest time was investigated latter than those of examined cultivar grapes, but agronomic characteristics was not thought significantly difference between cultivar grapes and V. amurensis. For evaluation of shoot growth phase, the growth curve was very similar to cultivar grapes. The berry size in V. amurensis showed that increases rapidly between 3 and 4 days after full bloom time, and approximately doubles between the second growth period and harvest time, and the berry development phase investigated that consist of two sigmoid growth periods separated by a lag phase. The berry weight and soluble solids in V. amurensis increased with the tree age, but acidity and total sugar contents decreased, and showed a special quality and stable growth according to vine age. To investigation of functional materials, the anthocyanin content in V. amurensis ranged from 16.6 to 50.2 mg/100 g, and the resveratrol content ranged from 0.143 to $0.236{\mu}g/100\;g$ which was higher than those of cultivar grapes. These result indicated that V. amurensis tended to have the useful material larger than cultivar grapes. Therefore, other edibility factors of V. amurensis collected in Gangwondo may contribute to breeding studies in Vitis. spp.

Protective Effect of Vitis amurensis Stems and Leaves Extract on Hydrogen Peroxide-induced Oxidative Neuronal Cell Damage in Cultured Neurons (과산화수소수로 유도된 배양 뇌신경세포손상에 대한 왕머루 잎과 줄기 추출물의 보호효과)

  • Vitis amurensis (VA; Vitaceae) has long been used in oriental herbal medicine. It has been reported that roots and seeds of VA have anti-inflammatory and antioxidant effects. In the present study, the protective effect of ethanol extract from stems and leaves of VA on hydrogen peroxide (${H_2}{O_2}$) (100 ${\mu}M$)-induced neuronal cell damage was examined in primary cultured rat cortical neurons. VA (10-100 ${\mu}g$/ml) concentration-dependently inhibited ${H_2}{O_2}$-induced apoptotic neuronal cell death measured by 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. VA inhibited ${H_2}{O_2}$-induced elevation of intracellular $Ca^{2+}$ concentration (${[Ca^{2+}]}_i$) and generation of reactive oxygen species (ROS), which were measured by fluorescent dyes. Pretreatment of VA also prevented glutamate release into medium induced by 100 ${\mu}M$ ${H_2}{O_2}$, which was measured by HPLC. These results suggest that VA showed a neuroprotective effect on ${H_2}{O_2}$-induced neuronal cell death by interfering with ${H_2}{O_2}$-induced elevation of ${[Ca^{2+}]}_i$, glutamate release, and ROS generation. This has a significant meaning of finding a new pharmacological activity of stems and leaves of VA in the CNS.

Inhibitory Effect of an Ethanol Extract Mixture of Vitis amurensis, Aralia cordata, and Glycyrrhizae radix on Amyloid β Protein (25-35)-Induced Neurotoxicity (머루전초, 독활전초, 감초 혼합추출물의 Amyloid β Protein (25-35) 유발 신경 독성에 대한 억제효과)

  • The present study investigated an ethanol extract (SSB) of a mixture of three medicinal plants of Vitis amurensis, Aralia cordata, and Glycyrrhizae radix for possible neuroprotective effects on neurotoxicity induced by Amyloid ${\beta}$ protein ($A{\beta}$) (25-35) in cultured rat cortical neurons and antidementia activity in mice. Exposure of cultured cortical neurons to $15{\mu}M$ $A{\beta}$ (25-35) for 36 h induced neuronal apoptotic death. At $1-30{\mu}g/m{\ell}$, SSB inhibited neuronal death, elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) induced by $A{\beta}$ (25-35) in cultured cortical neurons. Memory impairment and increase of acetylcholinesterase activity induced by intracerebroventricular injection of mice with 16 nmol $A{\beta}$ (25-35) was inhibited by chronic treatment with SSB (25, 50 and 100 mg/kg, p.o., for 8 days). From these results, it is suggested that antidementia effect of SSB is due to its neuroprotective effect against $A{\beta}$ (25-35)-induced neurotoxicity and that SSB may have a therapeutic role in preventing the progression of Alzheimer's disease.

Antiadipogenic Effect of Vitis amurensis Root Methanol Extract and Its Solvent Fractions in 3T3-L1 Preadipocytes (머루근 추출물 및 분획물의 항비만 활성)

  • Vitis amurensis Rupreche, a sort of grape, grows naturally in Asian countries. It is known for important biological effects such as antioxidation, anti-inflammation, neuroprotection, and angiogenesis inhibition. Although its root is used as a traditional folk medicine in Korea, the root's biological activities are poorly studied. In the present study, the effects of V. amurensis root methanol extract (VARM) and its solvent fractions on adipocyte differentiation and adipogenesis in 3T3-L1 preadipocytes were investigated. The VARM significantly suppressed adipocyte differentiation, lipid accumulation, and the triglyceride (TG) content of 3T3-L1 preadipocytes in a dose-dependent manner, without cytotoxicity. To identify active molecules, the VARM was fractionated with a series of organic solvents including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH). All the fractions also showed inhibition of lipid accumulation in the 3T3-L1 preadipocytes. The $CH_2Cl_2$ fraction showed the most powerful anti-obesity effect through the modulation of cytidine-cytidine-adenosine-adenosinethymidine (CCAAT)/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$, peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) gene and protein expression. Oleanolic acid was one of the main active compounds involved in the anti-obesity activity of the V. amurensis root. These results provide important new insight into the potential potent anti-adipogenic effect of the V. amurensis root and illustrate that one of the main compounds involved in this effect is oleanolic acid.

Vegetation and Soil Properties of Scrophularia takesimensis Population in Ulleung Island (울릉도 섬현삼(Scrophularia takesimensis Nakai) 개체군의 식생과 토양특성)

  • This study was carried out to analyze vegetation and soil characteristic, and ordination of Scrophularia takesimensis population, Ulleung Island. The Scrophularia takesimensis population was classified into Aster spathulifolius for. oharai dominant population, Vitis amurensis dominant population and Scrophularia takesimensis typical population. The Scrophularia takesimensis population was located in elevation of 1m to 6m, in Ulleung Island. In the study sites, soil organic matter, nitrogen, available phosphorous, exchangeable potassium, exchangeable calcium, exchangeable magnesium and exchangeable sodium concentration, and cation exchange capacity, and soil pH were 9.1~19.1%, 0.19~0.52%, 87.1~196.7mg/kg, 2.0~2.8cmol+/kg, 5.4~5.9cmol+/kg, 5.9~8.8cmol+/kg, 4.4~4.8cmol+/kg, 20.3~26.7cmol+/kg, and 6.8~8.0, respectively. The Vitis amurensis dominant population was mainly found in the high percentage of total nitrogen, organic matter, CEC in comparison with Aster spathulifolius for. oharai dominant population.

Fermentation Characteristics of Wild Grape (Vitis amurensis) Wine Prepared with Reed (Phragmites communis) Root (갈대뿌리를 첨가하여 제조한 산머루주의 발효 특성)

  • This study investigated the fermentation characteristics of wild grape(Vitis amurensis) wine prepared with reed(Phragmites communis) root. The reed root was added to the fermentation of wild grape wine in the quantities of 0.5%, 1.0% and 2.0%. An addition of reed root during yeast fermentation of wild grape wine decreased the acidity and increased pH of fermentation culture, thus enhancing the consumption of sugar and production of ethanol. Especially, the addition of 2% reed root led to production of about 13% ethanol in 4 day of fermentation time. The sensory evaluation showed that the wild grape wine fermented with 2.0% reed root was most acceptable.
