Effect of Cadmium Dose Injection on Peroxidative Damage in Rat Liver (카드뮴 투여가 흰쥐 간조직의 과산화적 손상에 미치는 영향)

  • In order to investigate the liver damage and hepatic protective systems in cadmium(Dd) administered rats, five different levels of Cd were injected intraperitoneally to male rats of sprague-Dawley strains weighing $250{\pm}15g.$ Levels of daily Cd administration were 0(control), 0.625(A), 1.25(B), 2.5(C) and 5mg(D)/kg of body weight and single inhection per day was done for consecutive two days. With increasing Cd dosed, serum glutamic oxaloacetic transaminase, glutamic pyruvic transaminase and alkaline phosphatase activities were increased. And at the same time, hepatic reduced glutathione contents were decreased, whereas the levels of oxidized form were increased. Liver lipid peroxide levels of A, B, C and D groups were 1.1, 1.5, 1.8 activities and vitamin E contents were progressively reduced in accordance with the increase in Cd dose. However, liver superoxide dismutase activities were not different between control and A group although it was higher in B and lower in C and D groups compared with control.

Effects of Vitamin E Supplementation on Renal Glycosylation Products in Diabetic KK Mice (당뇨 KK마우스에서 비타민 E 보강식이가 신장 당화단백질 생성에 미치는 영향)

  • We investigated the effects of vitamin E supplementation on protein glycosylation in early and end stage product, and light microscopic studies were done on the renal glomeruli of KK-mice of various ages and various duration of diabetes. Weaned KK-mice were fed high fat diets containing 20% corn oil(wt/wt), and sacrificed at 4,6, and 9 months of age. The high vitamin E diet was a high fit diet supplemented with an excess amount of d1-$\alpha$-tocopheryl acetate (2080IU/kg diet). We measured Hemoglobin $A_{IC}$ (Hb $A_{IC}$) as a glycosylation early product, and renal collagen-linked fluorescence as a glycosylation end product. In the diabetic group, levels of Hb $A_{IC}$ were increased within 2 months after onset of diabetes and remained at a constant level for the duration of experiment. 5 months after onset of diabetes, renal collagen linked fluorescence(CLF) was markedly increased. A quantative, morphologically demonstratable, progressive thickening of the basement membrane and calcification occured in the diabetic KK-mice. There is a statiscally positive correlation between CLF and histologic grade of diabetic nephropathy. Hepatic vitamin E levels correlated with those of Hb $A_{IC}$, renal CLF, and renal calcification. Treatment with vitamin I did not modify the level of blood glucose. However, we observered a significant lowering of CLF and Hb $A_{IC}$ in diabetic mice. Supplementation of vitamin E was found to delay the progression of diabetic nephropathy. Immune Response and Plasma Alpha Tocopherol and Selenium Status of Male Buffalo (Bubalus bubalis) Calves Supplemented with Vitamin E and Selenium

  • An experiment was conducted using 20 male buffalo calves to study the effect of vitamin E and selenium supplementation on their immune response and plasma ${\alpha}$-tocopherol and selenium status. These buffalo calves (10-12 months old, average body weight $75.30{\pm}2.20 $ kg) were randomly allotted to four treatments on the basis of their body weights and were fed on wheat straw and concentrate mixture to meet their nutrient requirements of 500 g/d body weight gain. The buffalo calves were fed either a control diet (neither supplemented with Se nor VE) or diets supplemented with Se at 0.3 ppm (+Se), DL-alpha tocopheryl acetate at 300 IU (+VE), and both DL-alpha tocopheryl acetate at 300 IU and Se at 0.3 ppm (+Se+VE). These experimental diets were fed for 180 days. Blood samples were collected at day 0 and subsequently at 45 day intervals up to 180 days of experimental feeding to monitor plasma ${\alpha}$-tocopherol and Se concentrations. To assess humoral immune response, all calves were sensitized with formalin inactivated Pasteurella multocida antigen at 135 days of experimental feeding and blood was collected on 0, 7, 14, 21 and 28 days post vaccination (DPV) to measure antibody production using indirect ELISA. Cell mediated immune response of calves was assessed after 180 days of experimental feeding by in vivo delayed type hypersensitivity (DTH) reaction using phytohaemaglutinin-P (PHA-P) as a mitogen. Results revealed that feeding of VE and Se improved the plasma levels of these nutrients. Plasma levels of Se were affected by supplementation of both VE (p<0.001) and Se (p<0.001); however, no interaction ($Se{\times}VE$) was observed. Supplementation of Se improved the humoral immune response (p<0.008), whereas, VE showed a tendency towards improvement in cell mediated immune response (p<0.064). It was concluded that vitamin E and Se supplementation improved the status of these micronutrients and humoral immune response in buffalo calves.

  • This study was conducted to investigate the content of antioxidant and GST activity of the oxyfluorfen-tolerant and -susceptible rice cultivars with barnyardgrass, a typical susceptible weed in accordance by oxyfluorfen treatment. The content of vitamin C was higher in the tolerant rice cultivar than in the susceptible rice cultivar and barnyardgrass. The contents of vitamin E, carotenoid, glutathione(GSH, GSSG, total glutathione) were not different between the tolerant and susceptible plants. In the case of the content of vitamin C due to the treatment of oxyfluorfen, the tolerant rice cultivars, Hawon and Baru decreased less than the susceptible rice cultivars and barnyardgrass. After the treatment of oxyfluorfen the contents of vitamin E and GSH in the tolerant rice cultivars were higher than in the susceptible rice cultivars or barnyardgrass. But in the content of carotenoid was greater in the tolerant rice cultivars than in the susceptible rice cultivars but they didn`t have any difference in comparison with the susceptible barnyardgrass. And there was no difference in the content of GSSG between the tolerant and susceptible plants. When CDNB or oxyfluorfen were used as substrate, the GST activity, was higher in the tolerant rice cultivars than in the susceptible rice cultivars or batnyardgrass. After the treatment of oxyfluorfen the GST activity was not induced in the rice, but was induced in the barnyardgrass. Even after the treatment of acifluorfen, bifenox and oxadiazon the GST activity was not induced in the rice cultivars.

Influence of UV Irradiation Procedures on the Concentration of Vitamin $D_{3}$ and 25-Hydroxyvitamin $D_{3}$ in the Liver and Skeleton of Broiler Chicks (자외선 조사방법이 육계 병아리의 간장과 골격중 Vitamin $D_{3}$ 및 25-Hydroxyvitamin $D_{3}$ 농도에 미치는 영향)

  • This study was carried out to elucidate the time course variation of vitamin $D_{3}$ (V$D_{3}$) and 25-hydroxyvitamin $D_{3}$ [25(OH)$D_{3}$] contents in the liver and skeleton of 3-wk old broiler chicks when treated with different UV irradiation procedure. Day-old Hubbard chicks were fed vitamin D deficient diet for 3 wk and exposed to medium wave ultraviolet(UVB) light with different irradiation procedures. Procedure I was 30 min continuous irradiation(O.204 mJ /$cm^{2}$) and Procedure II was three seperate 10 min irradiation with 12 h intervals, and Procedure III was three seperate 10 min irradiation with 24 h intervals. The liver and skeleton samples were collected at 10 different times between 0000~2400 h after the last irradiation. The V$D_{3}$ and 25(OH)$D_{3}$ fractions wereseparated by Sep-Pak silica cartridge and the concentrations were determined by normal phase HPLC. The mean content of V$D_{3}$ in the liver of the birds treated by Procedure II was 6.68 ng /g, which was higher than 5.60 and 5.30 ng /g from Procedure I and Ill, respectively(P<.O5). With regard to the effect of elapsed time after UVB irradiation on the V$D_{3}$ concentration of the liver, 96 h treatment showed the highest value(13.08 ng/g)(P<.05). There was a significant(P$D_{3}$ were significantly(P$D_{3}$ in the skeleton of tibia and femur, there were no significant differences among Procedure I, II and III, but significant differences were found among those from various elapsed time after irradiation, The highest value was shown at 96 h(O.99 ng /g) treatment, and interaction between irradiation procedure and elapsed time was not significant. With regard to the mean content of 25(OH)$D_{3}$ in bone, the Procedure II(18.79 ng /g) and III(17.73 ng /g) showed higher values than Procedure I did (P<.05), and the 12 h elapsed time showed the highest value(31.17 ng /g) among 10 treatments (P<.05), however, there was no significant interaction between exposing procedure and elapsed time. In conclusion, the Procedure II would he more desirable than Procedure I or III to produce more V$D_{3}$ and 25(OH)$D_{3}$ in the liver and skeleton of birds by exposing to the UVB light. Also, it was verified that 25(OH)$D_{3}$ increases more quickly than V$D_{3}$ in both tissues tested and is utilized more quickly to recover from the rickets of chicks.

Comparison of Health Status, Dietary Behavior and Nutrient Intakes According to Family Types of the Elderly in Rural Areas (농촌지역 노인에서 동거유형에 따른 건강상태, 식행동 및 영양소 섭취 수준의 변화 양상 비교)

  • This study was conducted to compare of health status, dietary behaviors and nutrient intakes according to family types of the elderly in rural areas. Family types were divided into three types-elderly living alone, elderly living with spouse, and elderly living with spouse and children. Subjects were 119 persons aged over 65 years (34 male, 85 females) living in rural areas and period of survey was from 29 January 2007 to 2 February. General environmental factors, health status, dietary behaviors and nutrient intakes were compared according to family types, the elderly living along showed a significantly lower in monthly income(p<0.01), self perceived health status(p<0.001), Instrumental Activity of Daily Living(IADL) score, and General Self Efficacy Scale(GSES) score compared to the elderly living with spouse and those living with spouse and children. However, Center for Epidemiologic Studies Depression Scale(CES-D) was higher than those of the elderly living family members (p<0.05). In addition, sum of dietary behavior score was the lowest in the elderly living alone (22.3 in elderly living alone, 24.1 in elderly living with spouse, 23.4 in elderly living with spouse and children, p<0.001) and nutrient intakes of potassium, zinc, vitamin C(p<0.01 respectively), calcium, phosphorus, iron, vitamin A, vitamin E, vitamin B6 and folic acid(p<0.05 respectively) also reported to be significantly lower in the elderly living alone than in others of two types. From these results, health status, dietary behavior pattern and nutrient intakes of the elderly living alone were found to be inadequate overall, so measures to deal with these health and nutritional status were needed.

Microencapsulated Iron for Drink Yogurt Fortification

  • This study was designed to examine the effect of icroencapsulated iron fortified drink yogurt and vitamin C as a bioavailable helper of iron on chemical and sensory aspects during 20 d storage. Coating material was polyglycerol monostearate (PGMS), and ferric ammonium sulfate and vit C were selected as core materials. The highest efficiency of microencapsulation of iron and vit C were 73% and 95%, respectively, with 5:1:50 ratio (w/w/v) as coating to core material to distilled water. Iron fortification did not affect the fermentation time required for the drink yogurt to reach pH 4.2. The addition of uncapsulated iron decreased the pH during storage. TBA absorbance was significantly lower in capsulated treatments than in uncapsulated treatments during storage. In sensory aspect, the yogurt sample added with uncapsulated iron and vit C, regardless of capsulation, showed a significantly high score of astringency, compared with those of control and other groups. A significantly strong sourness was observed in treatment containing capsulated iron and uncapsulated vitamin C at every time interval. The present study provides evidence that microencapsulation of iron with PGMS is effective for iron fortification in drink yogurt.

Mass Outbreak of Hypervitaminosis A in Foxes after Prolonged Feeding of Pig Liver (여우에 있어서 돼지 간 급여에 의한 비타민 A 과잉증의 집단발생)

  • Hypervitaminosis A occurred in about 30% of foxes of five farms in Choongcheongbukdo-province from October 1989 to February 1990. Clinical signs were loss of appetite, retarded growth, lameness. Radiological changes were decreased density of the long bones, especially fibula, and osteophytes developed either on the lateral condyle of humerus or ulnar just below the semilunar notch. Pathological change was the development of exotosis as is evidenced by radiological examination. Pig liver and lung consisted 30∼55% of feeds from March 1989 to February 1990. The serum concentrations of vitamin A in 2 affected foxes were 45,900 and 47,300 IU/d$\ell$ and the concentrations of vitamin A in the liver were 8,760 and 10,842 IU/gm respectively. It is concluded that the high level of vitamin A in the pig liver and the large amount of pig liver in the feed composition are the etiology of the disease.

Effects of Different Oil Sources and Vitamin E in Breeder Diet on Egg Quality, Hatchability and Development of the Neonatal Offspring

  • Effects of polyunsaturated fatty acid (PUFA) and vitamin E (VE) supplementation in the diet of breeder hens on the egg quality and hatchability, lipid peroxides of the egg yolk, and development of the newly-hatched offspring chicks were investigated. A total of 800 Avian 48, 28 wk-old broiler breeders were assigned randomly to 4 groups with 4 replicates of 45 females and 5 males. Each group was fed one of the following four diets with different oil sources and levels of VE: corn oil (CO), fish oil (FO), CO+VE and FO+VE. The results showed that: i) Addition of FO in the breeder diet reduced the whole egg weight, yolk weight, albumen weight, yolk color score and neonatal offspring chick body weight without affecting the hatchability as compared to the CO treatment. ii) Addition of VE efficiently reduced the lipid peroxides of egg yolk from hens fed diets containing FO. iii) VE in the breeder diet significantly promoted the development of liver and heart of the chick offspring.

Effect of Feeding Aflatoxin $B_1$ and Vitamin $D_3$ on Metatarsus Bone Minerals of Broiler Chicks (Aflatoxin $B_1$과 Vitamin $D_3$ 결여가 Broiler병아리의 중족골 무기물함량에 미치는 영향)

  • This study was conducted to investigate the interaction of aflatoxin B$_1$($AFB_1$) and vitamin D$_3$($VD_3$) in broiler chicks. The 336 broiler chicks(Hubbard line) of equally mixed sex were allocated to triplicate 8(2$\times$4 factorial) treatment groups. The 0 or 1ppm of AFB$_1$and 0, 500, 1,000 or 1,500IU/kg of VD$_3$ were supplemented to the basal diet Fourteen broilers of equally mixed sex were allocated to each replica and 24 groups were arranged in a randomized block design After 3 weeks of feeding the metatarsus were collected from the right and left legs of 4 chicks (2 for each sex) per group. The bone ash and minerals were measured. 1. In respect to the fresh weight of metatarsus bone no significant difference was found between 0 and 1ppm $AFB_1$ treatments, however, decreasing trend was recognized when fed increasing level of $VD_3$(P<.01). 2. The ash content in non-fat dry metatarsus bone decreased when fed 1ppm $AFB_1$(P<.01). However, that increased according to the increasing amount of $VD_3$(P<.01). Although there was no interaction between $AFB_1$ and $VD_3$ it was shown that the 1500IU/kg of $VD_3$ was neccessary to cover the decrease in ash content of metatarsus. when fed 1ppm of $AFB_1$. 3. The Ca contents in metatarsus were not influenced by feeding $AFB_1$ but an increasing trend was verified by feeding increasing levels of $VD_3$(P<.05). 4. The P content decreased as $AFB_1$ was fed(P<.01), while no response was found when fed'different levels of $VD_3$ 5. The Cu content decreased when fed $AFB_1$(P<.05). 6. The Na, Mg, K, Zn, Fe and Mn contents were not affected by feeding $AFB_1$ and /or $VD_3$.
