This study was carried out to analyze the major chemical component of the leaf and stem of dried Petasites japonicus S. et Z. Max.. Comparing proximate composition of leaf and stem of Petasites japonicus as dry matter basis, stem contained higher moisture, crude fat, crude ash and carbohydrate, with less crude protein. The main component of free sugar and disaccharide in both leaf and stem were fructose and sucrose, respectively. Total amino acids of leaf and stem were 6,853.32 mg% and 2,350.61 mg% respectively. Although the amino acid composition of leaf and stem were different glutamic acid and aspartic acid were the major amino acids in samples. The major fatty acids of total lipids were linolenic acid in leaf and linoleic acid in stem. The ratios of unsaturated fatty acids to saturated fatty acid were 3.93 in leaf and 3.44 in stem. The unsaturated fatty acid content of samples were 3 times higher than those of saturated fatty acid contents. Oxalic acid was the major organic acids in leaf and stem. The contents of vitamin A, C and E were higher in leaf than in stem. The mined compositions of both leaf and stem were composed in order of K, Mg, Ca, Fe, Na, and Zn.
While the contents of protein, fat, fiber, sodium, vitamin A, and vitamin D in dry dog food were 22.0%, 7.8%. 5.4%, 0.36%, 2035 IU/100g and 201 IU/100g, respectevely, those in dry cat food were 29.8%, 6.5%, 4.5%, 0.38%, 1543 IU/100g and 163 IU/100g, respectively. Concerning the lipid contents of pet food, the concentration of unsturated fatty acid was slightly higher of dry dog food(59.2%) than that of dry cat food(55.9%), but the differences was statistically not significant. The lipid peroxide concentration in dry dog food after storage in a refrigerator at $4^{\circ}C$ or dark place of room temperture at $20{\pm}2^{\circ}C$ for 30 days(8.0 nmol/g and 7.8 nmol/g) was not different from the value at the beginning of storage. However, upon storage in direct sunlight at $20{\pm}2^{\circ}C$ of room temperture, the concentration increased 4.9-fold(35.5 nmol/g) after 5 days and 10.2-fold(73.6 nmol/g) after 30 days. These results suggest that pet food may be refrigerated or stored in a dark place after unsealing.
The purpose of this experiment was performed for ascorbic acid contents of market milk in Seoul city area during the period of two seasons from spring (April, 13 to 30) to summer (July, 5 to 16) 1976. The samples were collected into five groups (A,B,C.D and E) according to factory products of market milks. The results obtained were as follows : 1. On an average, the value in spring was significantly lower than in summer, that is $0.228{\pm}0.222\;and\;0.374{\pm}0.028\;mg/100ml,$ respectively. 2. The differences among the groups of each factory product showed highest in C,A group and lower in E, B and D group. Thus, there were statistically significant (p<0.01) among-them.

A kojic acid derivative, kojic acid 7-O-$\beta$-D-tetraacetylglucopyranoside(KTG) was synthesized. Regio-and stereo-selective glycosylation at 7-postion in kojic acid with $\beta$-D-pentaacetylglucose was achieved with high yield(80%) by the use of Lewis acid and organic base in nonpolar solvent. KTG was hydrolyzed in methanol by the aid of sodium methoxide to give kojic acid 7-O-$\beta$-D-glucopyranoside(KGP). KGP is freely soluble in water and soluble in methanol and ethanol. Its structure was comfirmed by $^1$H-NMR and $^{13}$ C-NMR. Tyrosinase activity inhibition of KGP was measured with mushroom tyrosinase compared with ascorbic acid, kojic acid and arbutin. KGP showed higher tyrosinase inhibition activity($IC_{50}$/=33.3 $\mu\textrm{g}$/ml) than ascorbic acid(63.2 $\mu\textrm{g}$/ml) and arbutin(91.8 $\mu\textrm{g}$/ml) but lower inhibition activity than kojic acid(8.3 $\mu\textrm{g}$/ml). To test free-radical scavenging activity, we used 1, 1-diphenyl-2-picrylhydrazyl(DPPH) as a free-radical source. Free-radical scavenging activity of KGP was very low($SC_{50}$/>1000 $\mu\textrm{g}$/ml) compared with ascorbic acid($SC_{50}$/=2.68 $\mu\textrm{g}$/ml) and arbutin($SC_{50}$/=180$\mu\textrm{g}$/ml). Melanin formation inhibition of KGP was measured in B16 melanoma, compared with kojic acid, arbutin and Vitamin C. Inhibition activity of KGP for melanin formation was not found within test concentrations.

Lipid nanoparticles (LNPs) are a stable and an effective system that protects cell-impermeable biologically active compounds such as nucleic acids, proteins, and peptides against degradation caused by subtle environmental changes. This study focuses on developing LNPs encapsulating gallic acid (GA), an antioxidant, to effectively prolong the half-life of tetrahexyldecyl ascorbate (THDC), a oil-soluble vitamin C derivative. These LNPs were synthesized in small, uniform sizes at room temperature and pressure conditions using a microfluidics chip. Compared to liposomes manufactured under high pressure and high temperature conditions through conventional microfluidizers, LNPs manufactured through microfluidics chips had excellent dispersion and temperature stability, and improved skin absorption as well as improved oxidative stability of fat-soluble vitamin C derivatives. Future studies will focus on ex vivo and in vivo evaluations to study skin improvement to further validate these results.
A $2{\times}4$ factorial study was carried out to investigate the interaction of aflatoxin and vitamin $D_3$ in broiler chicks. The day-old 336 chicks were allocated to triplicate 8 treatments. The 0 or 0.5 ppm of aflatoxin $B_1$ (AFB) and 0, 500, 1,000 or 1,500 IU/Kg of vitamin $D_3$ (VD) were supplemented to the basal diet. There were no significant differences among treatments in respect to the body weight gain, feed intake, feed conversion, shank color, mortality and incidence of weak legs. The utilization efficiencies of dry matter, crude protein, ether extract, N-free extract and crude ash showed also no significant differences among treatments, respectively. The mean utilization efficiency of crude fiber in AFB group was lower than that in normal groups (P<.01). However, no significant difference was found among groups fed different levels of VD, and no interaction between AFB and VD was found. The utilization efficiency of Ca in AFB group was somewhat higher than that in normal group without statistical significance, and the similar values were found among groups fed different VD. The utilization efficiencies of P and Na were not significantly different among treatments, respectively. The tibia ash appeared to be similar among treatments fed different levels of AFB and VD. However, the Ca content in tibia of birds fed 0.5 ppm of AFB was higher than that of normal chicks (P<.05). The slightly increasing trend was shown in Ca contents when fed increasing revel of VD, and the interaction between AFB and VD was recognized(P<.01). The P content of tibia was increased by feeding AFB(P<.05). However, there was no significant difference among groups fed different level of VD and no interaction between AFB and VD in respect to the P content of tibia. Feeding AFB did not affect the Na content in tibia. However, there was a highly significant difference among groups fed different levels of VD(P<.01), the highest values were at 1,000 IU/Kg group, and the interaction between AFB and VD was not significant. The Ca content in serum of birds fed AFB was higher than in control group (P<.01). The Ca of serum increased when fed more VD, although no significance was found among groups, and there was an interaction between AFB and VD(P<.05). The P content of serum showed no significant difference among treatments. The alkaline phosphatase activity in serum of chicks fed AFB was higher than that of control group (P<.01). The enzyme activity increased slightly with increasing level of VD, however, there was no interaction between AFB and VD.
Bone marrow culture systems are widely used to differentiate osteoclast-like cells in vitro using several osteotropic hormones. In this study, we isolated and cultured the mouse bone marrow cells with or without some osteotropic hormones such as parathyroid hormone(PTH), prostaglandin $E_2(PGE_2)$ and $l,25(OH)_2-vitamin$$D_3$(Vit. $D_3$). We confirmed the formation of osteoclast-like cells morphologically and functionally by the expression of tartrate-resistant acid phosphatase(TRAP) and by their capability to resorb dentin slices. We also studied the effects of transforming growth $factor-{\beta}(TGF-{\beta})$ and epidermal growth factor(EGF) on the Vit. $D_3-induced$ osteoclast-like cell formation. In control, a few multinucleated cells were formed whereas PTH and $PGE_2$ increased the number of multinucleated cells. PTH, $PGE_2$ and Vit. $D_3$ induced the formation of TRAP-positive multinucleated cells. After culture of mouse bone marrow cells on the dentin slices with or without osteotropic hormones, giant cells with diverse morphology were found on the dentin slices under the scanning electronmicroscopy. After removing the attached cells, resorption pits were identified on the dentin slices, and the shape of resorption pits was variable. EGF increased the osteoclast-like cell formation induced by Vit. $D_3$, however, $TGF-{\beta}$ showed biphasic effect, which at low concentration, increased and at high concentration, decreased the osteoclast-like cell formation induced by Vit. $D_3$.

Objectives : Deer antler Water Extract(DAE), prepared from the pilose antler of Cervus korean TEMMINCK var. mantchuricus Swinhoe (Nokyong), a traditional immuno-suppressive and immuno-activating Korean herbal-acupuncture, is thought to play an important role in human bone remodeling. Methods : To determine whether DAE can induce the differentiation of resting zone chondrocytes(RC) or not, confluent cell cultures were pretreated for 24, 36, 48, 72, and 120hrs with DAE. At the end of pretreatment, the media were replaced with new media containing $10^{-10}{\sim}10^{-8}M\;1,25-(OH)_2D_3$ and the cells incubated for an additional 24hrs. Results : This second treatment was chosen because prior studies had shown that only the more mature growth zone chondrocytes(GC) respond to this vitamin $D_3$ metabolite. The effect of DAE pretreatment on cell maturation was confirmed by measuring alkaline phosphatase (ALPase)-specific activity. Changes in matrix protein synthesis were examined by measuring collagen synthesis, as well as $^{35}SO_4$ incorporation into proteoglycans. When RC cells were pretreated for 120h with DAE, treatment with $1,25-(OH)_2D_3$ caused a dose-dependent increase in ALPase-specific activity and collagen synthesis, however, the proteoglycan production was not affected. RC cells pretreated with $1,25-(OH)_2D_3$ responded like RC cells that had not received any pretreatment. Conclusion : These results indicate that DAE directly regulates the maturation of RC chondrocytes into GC chondrocytes. Therefore it was indicated that DAE may play a significant role in regulating chondrocyte maturation during endochondral ossification.
Diabetes mellitus is a multifactorial disease. Particularly, diabetic nephropathy is a serious complication for diabetic patients, yet the precise mechanisms that underline the initial stage of diabetic renal inflammation remain unknown. However, oxidative stress induced by hyperglycemia in diabetes is implicated in diabetic renal disease. We hypothesized that dietary supplementation of antioxidants either VCE (0.5% VC + 0.5% VE) or Comb (0.5% VC + 0.5% VE + 2.5% N-acetylcysteine) improves acute diabetic renal inflammation through modulation of blood glucose levels and antioxidant and anti-inflammatory responses. Experimental animals (5.5 weeks old female ICR) used were treated with alloxan (180 mg/kg) once. When fasting blood glucose levels were higher than 250 mg/dL, mice were divided into 3 groups fed different levels of antioxidant supplementation, DM (diabetic mice fed AIN 93G purified rodent diet); VCE (diabetic mice fed 0.5% vitamin C and 0.5% vitamin E supplemented diet); Comb (diabetic mice fed 0.5% vitamin C, 0.5% vitamin E and 2.5% N-acetylcysteine supplemented diet), for 10 days and then sacrificed. Body weights were measured once a week and blood glucose levels were monitored twice a week. Lipid peroxidation products, thiobarbituric acid reacting substances were measured in kidney. NF-${\kappa}B$ activation was indirectly demonstrated by pI${\kappa}B$-${\alpna}$ and expressions of selective inflammatory and oxidative stress markers including antioxidant enzymes were also determined. Dietary antioxidant supplementation improved levels of blood glucose as well as kidney lipid peroxi-dation. Dietary antioxidant supplementation improved NF-${\kappa}B$ activation and protein expression of HO-1, but not mRNA expression levels in diabetic mice fed Comb diet. In contrast, the mRNA and protein expression of CuZnSOD was decreased in diabetic mice fed Comb diet. However, antioxidant supplementation did not improve mRNA and protein expressions of IL-$1{\beta}$ and MnSOD in diabetic mice. These findings demonstrate that acute diabetic renal inflammation was associated with altered inflammatory and antioxidant responses and suggest that antioxidant cocktail supplementation may have beneficial effects on early stage of diabetic nephropathy through modulation of blood glucose levels and antioxidant enzyme expressions.

Bone cells produce multiple growth factors and cytokines that have effects on bone metabolism and can be incorporated into the bone matrix. The present study was designed to extend these observations by examining the interactions between transforming growth factor-$\beta$(TGF-$\beta$) or interleukin-$1\beta$(rhIL-$1\beta$) and bone cells in a rat long bone culture model. IL-$1\beta$ regulates several activities of the osteoblast cells derived from rat long bone explants in vitro. IL-$1\beta$ stimulated cellular proliferation as well as the synthesis of prostaglandin $E_2$ and Plasminogen activator activity in the cultured cells in a dose-dependent manner. TGF-$\beta$ is present in the bone matrix and potentially released during bone resorption. TGF-$\beta$ reduced basal bone resorption and inhibited vitamin $D_3[1,25(OH)_2D_3]$-induced bone resorption in rat long bone cells. These results support the role of IL-$1\beta$ in the pathological modulation of bone cell metabolism, with regard to implication in the Pathogenesis of osteoporosis by IL-$1\beta$, and that TGF-$\beta$ positively inhibits the bone resorption.
