Establishment of rapid discrimination system of leguminous plants at metabolic level using FT-IR spectroscopy with multivariate analysis (FT-IR 스펙트럼 기반 다변량통계분석기법에 의한 두과작물의 대사체 수준 식별체계 확립)

  • To determine whether FT-IR spectroscopy combined with multivariate analysis for whole cell extracts can be used to discriminate major leguminous plant at metabolic level, seed extracts of six leguminous plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from seed extracts were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). The PCA could not fully discriminate six leguminous plants, however PLS-DA could successfully discriminate six leguminous plants. The hierarchical dendrogram based on PLS-DA separated the six leguminous plants into four branches. The first branch was consisted of all three Vigna species including Vigna radiata var. radiate, Vigna angularis var. angularis and Vigna unguiculata subsp. Unguiculata. Whereas Pisum sativum var. sativum, Glycine max L and Phaseolus vulgaris var. vulgaris were clustered into a separate branch respectively. The overall results showed that metabolic discrimination system were in accordance with known phylogenic taxonomy. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from seed extracts represented the most probable chemotaxonomical relationship between six leguminous plants.

Comparison of Chemical Constituents in Mung bean (Vigna radiata L.) Flour between Cultivation Regions and Seeding Dates

  • Legumes are one of the largest families of crop plants and are widely consumed and produced for their nutritional and commercial benefits. Mung bean (Vigna radiata L.) is a legume crop that contains various functional compounds ; moreover, it has strong antioxidant properties and is becoming an increasingly important food crop. However, most previous studies on mung beans have focused on their primary metabolites. In this study, we investigated the composition and contents of phenolic compounds, fatty acids, soyasapogenol and tocopherol in mung beans cultivated in different regions and cultivated at different seeding dates. Material analysis was conducted using the following methods: LC-MS/MS, GC-FID and HPLC-ELSD. In total, 57 different samples were analyzed. Thirteen phenolic compounds were detected in mung beans. Of these, vitexin and isovitexin were the most abundant compounds, accounting for approximately 99% of phenolic compounds. The difference in phenol compounds according to the seeding dates of mung bean was not statistically significant. The total fatty acid content in beans was the highest in Pyeongchang. Significant differences in total fatty acid content were found according to the cultivation regions. Crops grown in Sohyeon and Dahyeon showed the highest soyasapogenol B content in the Suwon region, and these were the lowest in Jeonju. The total tocopherol content of beans cultivated in Dahyeon and Sohyeon was the lowest and highest in Pyeongchang. Soyasapogenol B and total tocopherol content were not significantly different according to seeding dates. This study was conducted to obtain basic data for the cultivation of mung beans with a high content of various functional materials in terms of regional specialization and optimal seeding time.

Variation of Vitexin and Isovitexin Contents in Mungbean (Vigna radiata (L.) Wilczek) Germplasms (녹두 유전자원의 Vitexin 및 Isovitexin 함량 변이)

  • In this study, the selected 789 lines having agronomic values out of over 2,500 mungbean (Vigna radiata (L.) Wilczek) germplasms, examined the 27 characters including hypocotyl color and leaf size of 401 lines in 2005 and of 388 lines in 2006, and analyzed the contents of vitexin and isovitexin in these mungbean germplasms. The average contents of vitexin and isovitexin in mungbean of the 401 lines used in 2005 were 8.71($1.1{\sim}13.4$) and 9.54($0.9{\sim}15.9$) mg/g, respectively, and the correlation coefficiency ($R^2$) of the contents of vitexin and isovitexin was 0.958. Nine lines including VC3890B were selected to be the ones containing high vitexin and isovitexin. The average contents of vitexin and isovitexin in mungbean of the 388 lines used in 2006 were 10.17($2.0{\sim}15.9$) and 10.64($0.2{\sim}17.6$) mg/g, respectively, and the correlation coefficiency ($R^2$) of the contents of vitexin and isovitexin was 0.958. Six lines including VC4096-2B-4-B-2-B were selected to be the ones containing high vitexin and isovitexin. The contents of vitexin and isovitexin were higher in the mungbean with larger leaves and longer ripening period out of the main characters.

Mungbean Cultivar, 'Munpyeong', with a Short Stem and High Yield (단경 다수성 녹두 품종 '문평')

  • A mungbean cultivar "Munpyeong" (Vigna radiata (L.) Wilczek) was derived as a cross between "Keumseong" and "IT163157" at the Jeollanamdo Agricultural Research and Extension Services (JARES) in 2013. "Munpyeong" has an erect growth habit, heart leaflet, green hypocotyls, black and straight pods when matured, and a dull green seed surface. The stem length of "Munpyeong" was 60 cm, which was 6 cm shorter than that of the control cultivar "Owool." The pod number per plant was 25, which was 3 more than that of the control cultivar. The 1000 seed weight of "Munpyeong" was 45 g, which was 4 g lighter than that of the control cultivar "Owool." Its field resistance to mungbean mottle virus, cercospora leaf spot, and powdery mildew are similar to the control cultivar, whereas its field resistance to lodging was slightly stronger. The hard seed rate of "Munpyeong" was 3.2%, which was similar to that of the control cultivar. The sprout yield ratio was 7.54 times, which was 9% more than that of the control cultivar. The average yield of "Munpyeong" was 1.93 ton/㏊, which was 21% more than that of the control cultivar "Owool" (Cultivar registration number: 5878).

Changes of Germination Rate of Pulses Seed Germplasm after Long-term Conservation

  • The seeds of soybean (Glycine max), adzuki bean (Vigna angularis), mung bean (Vigna radiata), and kidney bean (Phaseolus vulgaris L.) were examined the germination rate after 10 years of long-term storage ($-18^{\circ}C$) conservation. For soybean seeds, 2,313 accessions were examined and germination rate of 1,082 accessions was decreased with below 15% of initial germination rate. For 227 accessions of soybean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 589 accessions was increased and showed no change for 415 accessions after 10 years of long-term storage. For adzuki bean seeds, 2,058 accessions were examined and germination rate of 739 accessions was decreased with below 15% of initial germination rate. For 63 accessions of adzuki bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 535 accessions was increased and showed no change for 721 accessions after 10 years of long-term storage. For mung bean seeds, 438 accessions were examined and germination rate of 139 accessions was decreased with below 15% of initial germination rate. For 5 accessions of mung bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 155 accessions was increased and showed no change for 139 accessions after 10 years of long-term storage. For kdney bean seeds, 366 accessions were examined and germination rate of 7 accessions was decreased with below 15% of initial germination rate. For 65 accessions of kidney bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 201 accessions was increased and showed no change for 93 accessions after 10 years of long-term storage.

Biochemical Characterization of 1-Aminocyclopropane-1-Carboxylate Oxidase in Mung Bean Hypocotyls

  • The final step in ethylene biosynthesis is catalyzed by the enzyme 1-aminocyclopropane-1-carboxylate (ACC) oxidase. ACC oxidase was extracted from mung bean hypocotyls and its biochemical characteristics were determined. In vitro ACC oxidase activity required ascorbate and $Fe^{2+}$, and was enhanced by sodium bicarbonate. Maximum specific activity (approximately 20 nl ethylene $h^{-1}$ mg $protein^{-1}$) was obtained in an assay medium containing 100 mM MOPS (pH 7.5), $25\;{\mu}M$ $FeSO_4$, 6 mM sodium ascorbate, 1 mM ACC, 5 mM sodium bicarbonate and 10% glycerol. The apparent $K_m$ for ACC was $80{\pm}3\;{\mu}M$. Pretreating mung bean hypocotyls with ethylene increased in vitro ACC oxidase activity twofold. ACC oxidase activity was strongly inhibited by metal ions such as $Co^{2+}$, $Cu^{2+}$, $Zn^{2+}$, and $Mn^{2+}$, and by salicylic acid. Inactivation of ACC oxidase by salicylic acid could be overcome by increasing the $Fe^{2+}$ concentration of the assay medium. The possible mode of inhibition of ACC oxidase activity by salicylic acid is discussed.

Responses of Mungbean Varieties to Rhizobium Inoculation in respect of Nodulation, Nitrogenase Activity, Dry Matter Yield, and Nitrogen Uptake

  • The responses of six mungbean [Vigna radiata (L.) Wilczek] varieties to Rhizobium inoculation on number and dry weight of nodules, nitrogenase activity of root nodule bacteria, dry weight of shoot and root, nitrogen content, and uptake by shoot were investigated. The mungbean varieties were BARI Mung-2, BARI Mung-3, BARI Mung-4, BARI Mung-5, BINA Moog-2, and BU Mung-1. Two-third seeds of each variety were inoculated with Rhizobium inoculant and the remaining one-third seeds were kept uninoculated. Rhizobium strains TAL 169 and TAL 441 were used for inoculation of seeds. Inoculation of seeds with Rhizobium strains significantly increased nodulation, nitrogenases activity, dry matter production, nitrogen content, and uptake by shoot of the crop compared to uninoculated control. There was positive correlation among the number and dry weight of nodules, nitrogenase activity, dry weight of shoot and root, nitrogen content, and uptake by shoot of the crop. It was concluded that BARI Mung-4 in association with Rhizobium strain TAL 169 performed best in recording nodulation, nitrogenase activity, dry matter production, and nitrogen uptake by shoot of mungbean.

Agronomical traits of Korean Mungbean Landrace

  • Mungbean(Vigna radiata (L.) R. Wilczek) is one of legume cultivated in south and east Asia. It well cultivated in high temperature and drought condition. In order to establish pure line for genomic studies, the agronomical characteristics of Korean mungbean landraces were evaluated. A total of 324 acessions derived from National Agrobiodiversity Center were selfed two times in 2021 nersery. Tree times of selfed from 324 accessions were characterized. The color of seed coat was divided into three groups: green, yellow and brown. Among them green color was the most common with 91.7%. Flower colors were divided into four groups: yellow, purplish yellow, greenish yellow and purplish green. Among them purplish yellow color had the highest with 77.8%. Out of 324 resources, only 4 showed compound leaves, and all resources had a heart-shaped leaf. The days to first flowering was 31 to 78 days. About 63% of the accessions bloomed 35 to 50 days after sowing. 324 Korean landraces are expected to be used as a population of the Korean mungbean core collection and serve as a basis for genomic breeding materials for mungbean.

Renal and Salivary Excretions of Plasma Purine Derivatives in Swamp Buffaloes and Zebu Cattle

  • This study compared the recovery rate of intrajugular-administered allantoin in the urine and saliva between swamp buffaloes and zebu cattle to examine whether it could explain the lower excretion rate of urinary purine derivatives (PD) in the buffaloes. Three male swamp buffalo yearlings, with an average body weight of $349{\pm}40.35$ kg, and three Thai native cattle ($154{\pm}3.26$ kg) of similar age and sex were used in the study. Animals were kept in individual pens and fed at a maintenance energy level with a diet containing 65% monk bean husk (Vigna radiata) as roughage and 35% concentrates. Allantoin solution was infused into the jugular vein in four incremental rates equivalent to 0, 5, 10 and 15 mmol/d and urine was collected daily in acidified form. Daily PD excretion was linearly correlated with intrajugular allantoin infusion in both species. The relationship between daily urinary PD excretion (Y, mmol/d) and intrajugular allantoin infused (X, mmol/d) was $Y=0.75{\pm}0.318X+22.45{\pm}2.98$ ($r^2$ = 0.36, n = 12, MSE = 38.02, CV = 21.9, p<0.01) for swamp buffaloes and $Y=0.96{\pm}0.10X+15.93{\pm}0.92$ ($r^2$ = 0.91, n = 12, MSE = 3.60, CV = 8.27, p<0.01) for zebu cattle. The salivary PD concentration was not correlated with intrajugular allantoin infusion in both species, with values for buffaloes numerically lower than those for cattle. The present study reconfirmed previous studies that buffaloes have a lower plasma PD excretion rate via the renal route and a significant proportion (22%) of the plasma PD loss is via the saliva. However, results of our present and previous studies suggest that differences in purine base (PB) metabolism between buffaloes and zebu cattle occur before the purine compounds reach the plasma pool.

Oviposition Preference of The Bean Bug, Riptortus clavatus (Thunberg) (Hemiptera: Alydidae), on Soybean and Mungbean Plants (콩 및 녹두에서 톱다리개미허리노린재[Riptortus clavatus (Thunberg)](노린재목: 호리허리노린재과)의 산란선호성)

  • Oviposition preference of the bean bug, Riptortus clavatus to sites on a plant and within a field, to plants at different developmental stages, and to different leguminous germplasms was observed. The insect layed its eggs mainly on the back surface of leaf in the upper half of plant in both observation from oviposition cage and soybean field. The egg number in fields were observed at a statistically-same rate in marginal and inside area, and at a median value of one egg per leaf in oviposited leaves. Full seed stage of soybean hardly affected oviposition preference of the insect. A statistically-significant difference in oviposition to different leguminous germplasms was observed.
