Mortality of the Horned Turban Shell, Batillus cornutus Caused by Vibrio spp (소라(Batillus cornutus)의 비브리오균 감염에 의한 폐사)

  • Mass mortality of the horned turban shell, Batillus cornutus was investigated using histological and bacteriological methods. Some pathogenic bacteria were isolated from mortal or inactive individuals. The pathogenic agents causing mortality of the horned turban shells were as Vibrio alginolyticus and V. anguillarum. Laboratory experiment indicated that optimal growth temperature of two bacteria was 25 to 30$^{\circ}C$ and 3% of NaCl. Histological examination of the horned turban shells showed that gill necrosis is one of the major symptom of infected individuals. It was believed that sudden increase of those two bacterial agents due to environmental change cause mortality of horned turban shells.

Detection and Identification of Vibrio Species Using Whole-Cell Protein Pattern Analysis

  • Outbreaks of foodborne diseases associated with Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae frequently occur in countries having a dietary habit of raw seafood consumption. For rapid identification of different Vibrio species involved in foodborne diseases, whole-cell protein pattern analysis for 13 type strains of 12 Vibrio species was performed using SDS-PAGE analysis. Pathogenic Vibrio species such as V. parahaemolyticus, V. vulnificus, V. cholerae, V. alginolyticus, V. fluvialis, and V. mimicus were included in the 12 Vibrio species used in this study. Each of the 12 Vibrio species showed clearly specific band patterns of its own. Two different strains of V. parahaemolyticus showed two different SDS-PAGE whole-cell protein patterns, giving the possibility of categorizing isolated strains in the same V. parahaemolyticus species into two subgroups. The 36 Vibrio isolates collected from sushi restaurants in Busan were all identified as V. parahaemolyticus by comparing their protein patterns with those of Vibrio type strains. The identified isolates were categorized into two different subgroups of V. parahaemolyticus. The whole-cell protein pattern analysis by SDS-PAGE can be used as a specific, rapid, and simple identification method for Vibrio spp. involved in foodborne diseases at the subspecies level.

  • 어패류를 날 것으로 즐겨먹는 우리나라와 일본에서는 매년 여름철이면 병원성 비브리오균에 의한 식중독 사고가 많이 발생한다. 현재까지 알려져 있는 40 여종의 비브리오균 중 12종이 사람에게 질병을 일으키는 것으로 알려져 있으며, 우리나라에서 주로 문제를 일으키는 것은 Vibrio parahaemolytius, V. vulnificus 등 이다. 그러나 최근에는 V. cholerae non-Ol의 병원성에 관한 연구도 많으며, 특히 2001년에는 우리나라에서 발생율이 희박하던 V. cholerae Ol에 의한 환자가 140명을 상회하는 대형사고가 발생하기도 하였다.   • Diarrheal diseases are a major cause of both illness and death in developing countries and are caused by rotavirus, Shigella spp., Salmonella spp., enterotoxigenic Escherichia coli (ETEC), and Vibrio spp. In this study, for the development of vaccine against diarrheal diseases caused by Shigella sonei, Salmonella typhimurium, E. coli O157, and Vibrio cholerae, cloning and nucleotide sequence analysis of genes and characteristics of their gene products in E. coli were performed. For construction of attenuated strain of S. sonnei KNIH104 and Salmonella typhimurium KNIH100, the aroA genes were cloned, respectively. The recombinant plasmid $_pJP{\Delta}A45$ containing aroA deleted region and suicide vector $(_pJP5603)$ was constructed. The aroA gene deleted mutants were constructed using this recombinant plasmid. For cloning gene encoding antigenic region of E. coli O157 KNIH317, the O-antigen synthesis gene cluster and sit gene was cloned. The E. coli XL1-Blue cells harboring this recombinant plasmid showed cytotoxicity in Vero cells. The ctx gene was cloned for tile purpose of antigenic region against V. cholerae KNIH002. Sequence analysis confirmed that the virulence gene cassette was consisted of ace, zot, ctxA and ctxB genes.

Isolation and Degradation Activity of a TBTCl (Tributyltin Chloride) Resistant Bacteriain Gwangyang Bay (광양만에서 TBTCl (Tributyltin Chloride) 내성세균의 분리 및 분해활성)

  • BACKGROUND: Tributyltin chloride is among the most toxic compounds known for aquatic ecosystems. Microorganisms are responsible for removal of TBTCl. Nevertheless, only a limited number of marine bacteria were investigated for biodegradation of TBTCl in Korea. METHODS AND RESULTS: The number of TBTCl resistant bacteria ranged from $2.5{\times}10^3$ to $3.8{\times}10^3$ cfu/mL in the seawater, and ranged from $3.2{\times}10^5$ to $9.1{\times}10^5$ cfu/g in the surface sediment, respectively. The morphological, physiological, and biochemical characteristics of TBTCl resistant bacteria were investigated by API 20NE and other tests. The most abundant species of TBTCl resistant bacteria were Vibrio spp. (19.2%), Bacillus spp. (16.2%), Aeromonas spp. (15.2%), and Pseudomonas spp. (13.1%), etc. Eleven TBTCl resistant isolates also had a resistance to heavy metals (Cd, Cu, Hg, and Zn). Among them, isolate T7 showing the strong TBTCl-resistance was selected. This isolate was identified as the genus Pantoea by 16S rRNA gene sequencing and designated as Pantoea sp. T7. In addition, this bacterium was cultivated up to the growth of 50.7% after 60 hrs at TBTCl concentration of $500{\mu}M$. TBTCl-degrading activity of Pantoea sp. T7 was measured by GC-FPD analysis. As a result of biological TBTCl-degradation at TBTCl concentration of $100{\mu}M$, TBTCl-removal efficiency of Pantoeasp. T7 was 62.7% after 40 hrs. CONCLUSION(S): These results suggest that Pantoea sp. T7 is potentially useful for the bioremediation of TBT contamination.

Microbiological Pollution Investigation of Lowland Spring Water at the Jeju City Waterfront, Jeju Island (제주도 내 제주시 해안가 저지대용천수의 미생물학적 오염도 조사)

  • Bacteriological examination of spring water in Jeju City was conducted. A total of 21 spring water samples were collected from January to April, 2010. During the study period, the range of temperature was 0.6 to $15.4^{\circ}C$, and the results of the analyses showed that hydrogen ion concentrations (pH) for spring water were 0.43 to 7.9. Salinity levels for the samples averaged from 3.0 to 1.64%, and levels of water-dissolved oxygen were 1.85 to 6.06 mg/l. The range of total coliforms in spring water samples at 21 stations located in the designated spring water were <1.8->1,600 MPN/100 ml. Furthermore, the range of geometric means of total coliforms was 9.9-151.6 MPN/100 ml, while the range of fecal coliforms in spring water samples at 21 stations located in the designated spring water area was <1.8->1,600 MPN/100 ml. Finally, the range of geometric mean of fecal coliforms was 3.1-151.6 MPN/100 ml. The level of microbial contamination was examined in 21 samples for indications of bacterial contamination such as heterotrophic bacteria, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, and Shigella spp. were frequently detected in the spring water. Salmonella spp., Shigella spp., Vibrio parahaemolyticus, and S. aureus were detected in the range of $0-0.5{\times}10^1$, $0-0.1{\times}10^1$, $0-0.1{\times}10^1$, and $0-0.3{\times}10^1$ CFU/ml, respectively, while E. coli O157:H7 was not detected in the examined spring water samples.

Effects of Pre-treatment Method on the Surface Microbes of Radish (Raphanus sativus L.) leaves (전처리 방법이 무청의 표면 미생물 변화에 미치는 영향)

  • It was investigated the effects of pre-treatment method on the microbes on the surface of radish (Raphanus sativus L.) leaves. Independent variables put in water washing ($X_1$), microwave treatment ($X_2$) and steam treatment ($X_3$) using central composite design and response surface analysis. It was not detected in the pathogenic microbes, Samonella spp., Camphylobacter spp., Vibrio spp., Shigella spp., Staphyloccocus spp., on the surface of collected radish leaves without pre-treatment. But general microbes showed $3.90{\times}10^5{\sim}1.20{\times}10^7CFU/g$ of total microbes, $1.10{\times}10^2{\sim}2.00{\times}10^5CFU/g$ of E. coli, $2.40{\times}10^3{\sim}3.55{\times}10^6CFU/g$ of yeast/mold on the surface of various radish leaves and lactic acid bacteria was detected or not according to collected samples. The best method of pre-treatment was steam treatment on the microbe reduction effect of samples surface. Also, the multiplex regression coefficients analysis was calculated three independent variables ($X_1,\;X_2,\;X_3$) and dependent variables (total microbes, lactic acid bacteria and yeast/molds). It showed high correlation $R^2$, 0.89, 0.87, 0.85, respectively. For effective reduction of surface microbes, the best method was water washing with microwave or steam treatment at the same time.

Comparative study of Photobacterium damselae subsp. damselae and Vibrios on pathogenicity in vitro (In vitro에서 Photobacterium damselae subsp. damselae와 Vibrio 속 세균의 병원성 비교)

  • Photobacterium damselae subsp. damselae and 4 Vibrio spp.(V. anguillarum, V. splendidus, V. harveyi and V. ordalii) were isolated from the diseased olive flounders, Paralichthys olivaceus. The isolates were tested on the pathogenicity in vitro. The properties of extracellular products(ECPs) were investigated with enzymatic activities, hemolytic activities toward the sheep and olive flounder erythrocytes, and cytotoxicity activities on the cell-line. And potential signal transduction pathways of the bacterial internalization were detected by using signal transduction inhibitors. P. damselae was high in phospholipase activity, hemolytic activity to olive flounder erythrocytes and cytotoxicity activity. And P. damselae had diversified internalizing pathways as compared to isolated vibrios. Therefore, these activities may be related with pathogenicity of P. damselae.

  • Monthly distribution of marine bacteria and phytoplankton in Suyeong Bay were investigated with laboratory experiment of dominant algal species and bacterial flora. During the periods of study, the highest density of phytoplankton and bacteria occurred in May with the number of $3.3{\times}10^6cells/l$ and $1.93{\times}10^8cells/ml$, respectively. 10 genera of bacteria and 22 genera of phytoplankton were isolated and identified. In May when phytoplankton bloom occurred, dominant species of bacteria were Acinetobacter calcoaceticus($29.1\%$) and Bacillus subtilis($22.9\%$), and dominant species of phytoplankton were Chaetoceros spp.($62.8\%$) and Skeletonema spp. ($19.4\%$). Pseudomonas spp., which was the most abundant bacterial species during the study periods, were rapidly decreased in May. In laboratory studies of culturing bacteria and phytoplankton isloated in May, the growth of Pseudomonas vesicularis seems to be influenced by the concentrations of excretion matter of Chaetoceros spp. To examine the result colsely, the problem of pure isolation for phytoplankton must be solved and more experimental process have to be conducted.
