인천연안 Vibrio parahaemolyticus의 분포 및 유산과 과산화수소 처리에 의한 항균효과 (Antimicrobial Effect of Lactic acid and Hydrogen Peroxide and Distribution of Vibrio parahaemolyticus from the Incheon Adjacent Sea)

  • This study was carried out to investigate the distribution of Vibrio parahaemolyticus in the Incheon adjacent sea, and antimicrobial effect on growth of Vibrio parahaemolyticus in lactic acid and hydrogen peroxide and combination of lactic acid and hydrogen peroxide. The detected strains were compared geographical, months and sample types. The distribution of Vibrio parahaemolyticus was high at Ganghwa county with 66.1%(336 samples), on 7-9 months with 72.4%(386 samples) and from tireland with 75.0%(90 samples), respectively. The minimun inhibitory concentration (MIC) of lactic acid in Vibrio parahaemolyticus were 1250 ppm at pH 6.5 and 7.0, 625 ppm at pH 6.0. respectively. The minimun inhibitory concentration (MIC) of hydrogen peroxide in Vibrio parahaemolyticus were 25 ppm at pH 6.5 and 7.0, 12.5 ppm at pH 6.0, respectively. MICs of combined treatment of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were 625 ppm of lactic acid with 12.5 ppm of hydrogen peroxide. The correlations between MICs of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were obtained through the coefficient of determination($R^2$). $R^2$ value were 1.0000. The antimicrobial effect of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus could be confirmed from the result of this experiment.

  • 세균 16S rRNA 유전자 염기서열은 분자계통분류, 진화역사 규명, 미생물 검출 등 다양한 목적으로 이용되어 왔다. 세균 제놈(genome)은 multiple rRNA 오페론을 갖고 있으며, 이들 유전자 염기서열은 일부 변이가 있는 것으로 알려져 있다. 본 연구에서는 Vibrio 속의 16S rRNA 유전자 염기서열을 이용하여 세포 내 16S rRNA의 이질성을 규명하였다. 분석은 GenBank 자료 중에서 제놈 염기서열 annotation이 완료된 V. cholerae, V. harveyi, V. parahaemolyticus, V. splendidus, V. vulnificus를 이용하여 실시하였다. Vibrio 속은 1번 염색체에 7~10개의 16S rRNA 유전자 copy를 갖고 있으며, 이들의 세포 내 유전자 변이는 0.9% 이하 상이성(99.1%이상 DNA 상동성)을 보였다. 2번 염색체에서는 16S rRNA 유전자가 1개 이하로 존재하였다. 유전체내 16S rRNA 유전형은 최소 5개(V. vulnificus #CMCP6)에서 최대 8개(V. parahaemolyticus #RIMD 2210633, V. harveyi #ATCC BAA-1116)로 조사되었다. 본 결과는 Vibrio 속의 16S rRNA 유전자 염기서열이 높은 이질성을 갖는 것을 제시해 준다.

Risk assessment of vibriosis by Vibrio cholerae and Vibrio vulnificus in whip-arm octopus consumption in South Korea

  • This study evaluated the risk of foodborne illness from highly pathogenic Vibrio spp. (Vibrio vulnificus and V. cholerae) by raw whip-arm octopus (Octopus minor) consumption. In total 180 samples of raw whip-arm octopus were collected from markets and examined for the prevalence of V. vulnificus and V. cholerae. Predictive models describing the kinetic behavior of Vibrio spp. in raw whip-arm octopus were developed, and the data on amounts and frequency of raw whip-arm octopus consumption were collected. Using the collected data, a risk assessment simulation was conducted to estimate the probability of foodborne illness raw whip-arm octopus consumption using @RISK. Initial contamination levels of Vibrio spp. in raw whip-arm octopus were -3.9 Log colony-forming unit/g, as estimated by beta distribution fitting. The developed predictive models were appropriate to describe Vibrio spp. in raw whip-arm octopus during distribution and storage with R2 values of 0.946-0.964. The consumption frequency and daily consumption amounts of raw whip-arm octopus per person were 0.47% and 57.65 g, respectively. The probability of foodborne illness from raw whip-arm octopus consumption was estimated to be 8.71 × 10-15 for V. vulnificus and 7.08 × 10-13 for V. cholerae. These results suggest that the risk of Vibrio spp. infection from raw whip-arm octopus consumption is low in South Korea.

NAG Vibrio의 검사에 관한 소고 (Study on the Identification Methods of the Non-agglutinating Vibrio)

  • The genus Vibrio contains some of the most important intestinal pathogens of humans, including Vibrio cholerae, the cause of epidemic Asiatic cholera. A group of organisms which have been reffered to as the non-agglutinating vibrio (NAG) do not agglutinate in the Vibrio cholerae 0 group 1 antisera, but are indistinguishable from the 0-1 group both chemically and genetically. Non-O-l Vibrio cholerae can cause isolated as well as focal outbreaks of diarrhea, but the volume of fluid loss does not approach that of classic cholera, and the disease is usually self-limiting. These free-living organisms are found world-widely distributed in the environment including sewage, contaminated water, estuaries, seafood and animals. These strains involved in several cases were isolated from the environment and some patients of diarrhea, and a few epidemiologic reports indicated the wide distribution of the strains throughout the country, giving an attention to the role the organisms may play in an outbreak of diarrhea in Korea. More research on the epidemiology, serologic typing and virulence of the group of organisms, should be, therefore, done to obtain a complete understanding of their role in human disease.

장염 Vibrio 식중독의 세균학적 연구 (Studies on Vibrio Parahaemolyticus Food Poisoning)

  • The author was carried out bacteriological identification, and in order to evaluate the sensitivity of the different chemotherapeutic agents including chloramphenicol to Vibrio parahaemolyticus isolated from the stool of the patient's diarrhea. The results obtained were as follows: 1) Biochemical properties of Vibrio parahaemolyticus strains isolated from patients with diarrheal food poisoning was showed Table 1. 2) The sensitivity pattern of the isolated strains of Vibrio parahaemolyticus were sensitive to chloramphenicol, sulfonamide, kanamycin and colistin. But tetracycline, penicillin and leucomycin were resistant.

비브리오속 균주들에서외 세포의 효소의 분포 (Distribution of Extracellular Proteases from Various Vibrio Species)

  • The members of the genus Vibrio include harmless aquatic strain as well as strains capable of causing infections in human and fish. Pathogenic mechanisms are only understood for Vibrio cholerae O1 and O139 and not for the majority of Vibrio species. Twelve clinical and nonclinical strains were examined by in vitro and in vivo experiments for the importance of extracellular enzymes as a virulence determinant of Vibrio species. In vivo cytotoxicity assay was performed by injecting approximately $10^{8}$ cells/mL into mice (BALB/c). V. harvyi and V. vulnificus showed 100% lethality within 3hr after bacterial injection. V. fluvialis and four strains of V. parahaemolyticus showed 50% lethality within 4hr. V. mimicus, V. alginolyticus and V. furnissii revealed 30% lethality within 9hr. Nonclinical strains, V. campbellii and V. ordalii, did not show any lethality. In vitro protease and hemolytic activities were also good indicators for clinical and nonclinical strains of Vibrio species. The clinical strains showed much higher activities than nonclinical strains. The activity of some clinical strains of re-isolates was evidently increased. Most clinical strains had $\beta$ hemolytic activity. The results demonstrate that the prevalent distribution of extracellular proteases in pathogenic Vibrio sp. implies their importance as a virulence determinant.

곰소만 및 변산 해역 해수에 병원성 비브리오균(Vibrios spp.)의 분포 (Distribution of Pathogenic Vibrio Species in Seawater in Gomso Bay and Byeonsan, West Coast of Korea)

  • The pathogenic Vibrio genus contains halophilic bacteria that are distributed in marine and freshwater environments. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are potent human pathogens and leading causes of septicemia, wound infection, and seafood-borne gastroenteritis. The aim of this study was to investigate the presence of pathogenic Vibrio species in seawater off the west coast of Korea. Sixty-four seawater samples were obtained from different sites in Gomso Bay and Byeonsan from April 2018 to November 2018. Pathogenic Vibrio species were detected using a combination of most probable number (MPN)-polymerase chain reaction methods. V. cholerae, V. vulnificus, and V. parahaemolyticus were found in 0.0%, 20.3%, and 65.6% of seawater samples, respectively. Quantitative results revealed 3.6-23 MPN/100 mL of V. vulnificus, and 3.6-930 MPN/100 mL of V. parahaemolyticus in the samples. Overall, these results provide new insight into the necessity for seawater sanitation in Gomso Bay and Byunsan; they also provide evidence that will help reduce outbreaks of seafood-borne illness caused by pathogenic Vibrio species.

16S-23S rRNA Intergenic Spacer Region을 이용한 Vibrio ichthyoenteri Species-specific Primer 개발 (Use of 16S-23S rRNA Intergenic Spacer Region for Species-specific Primer Developed of Vibrio Ichthyoenteri)

  • Rotifer와 병든 넙치 자어로부터 분리된 2개의 균주는 표현형적인 특성 확인 결과 Vibrio ichthyoenteri로 확인이 되었다. V. ichthyoenteri를 검출하기 위래 고감도 PCR 방법 개발을 하기 위래 V. ichthyoenteri 16S-23S rRNA intergenic spacer region(ISR)을 분석하였고, V. ichthyoenteri중 특이적 primer를 개발하였다. V. ichthyoenteri 의 ISR를 분석한 결과 1개의 다형성 ISR type서열을 포함하고 있었다. ISR서열은 길이는 348bp이며 tRNA gene을 가지고 있지 않았다. 이 서열을 가지고 이미 알려진 다른 Vibrio 종의 ISR 서열과 mutiple alignment를 수행한 결과 여러 영역에서 높은 가변성을 나타내어 가변 부위를 표적으로 하여 V. ichthyoenteri를 검출하기 위한 종 특이적 primer를 제작하였다. 제작된 primer의 특이성을 확인하기 위해 Vibrio 표준균주 19종의 genomic DNA와 분리균주 18 group에 genomic DNA 그리고 V. ichthyoenteri와 가장 유사한 서열을 가지고 있다고 알려진 Vibrio 종의 genomic DNA를 가지고 시험하였다. 그 결과 본 연구에서 제작된 종 특이적 primer를가지고 PCR 반응을 하면 V. ichthyoenteri를 검출 할 수가 있다.

솔잎 추출물과 고추냉이의 Vibrio에 대한 항균활성 (Antimicrobial Activity of Pine Needle Extract and Horseradish on the Growth of Vibrio)

  • 시판 꽃게와 가자미에서 Vibrio를 분리하여 Api 20E kit로 동정한 결과 V. parahaemolyticus HY I과 V. vulnificus FST I으로 동정되었다. 분리 Vibrio와 V.parahaemolyticus ATCC 17802에 대한 솔잎 추출물의 항균활성 검색을 실시한 결과 솔잎 추출물 1%를 첨가하였을 경우 뚜렷한 성장억제현상을 나타내었다. 시판 고추냉이와 솔잎 ethanol 추출물을 혼합하여 각 Vibrio에 대한 성장억제효과를 검토한 결과 l%고추냉이 단독 첨가구보다 1% 솔잎 추출물과 혼합치리구가 뚜렷한 성장억제효과를 나타내었다. 시판 고추냉이를 간장에 첨가하여 고추냉이 간장을 제조한 다음 Vibrio균의 생존을 실험한 결과 솔잎 추출물 1%를 첨가한 고추냉이 간장에서 Vibrio 생균수의 감소는 뚜렷하였다 고추냉이 간장을 제조 한 다음 관능검사를 실 시 한 결과 5% 수준에서 대조구와 솔잎 추출물 첨가구 간의 유의적인 차이는 나타나지 않았다.
