  • The purpose of this study is to investigate the effect of water extract from Artemisiae Argi Folium Fermented with Lactobacillus pentosus (AFL) on viability of human hepatocyte HepG2 cells treated with hepatotoxicants such as EtOH, gallic acid, nicotine, acetaminophen, acetaldehyde, and lipopolysaccharide. AFL (0~400 ug/mL) was treated with EtOH, gallic acid, nicotine, acetaminophen, acetaldehyde, and lipopolysaccharide. And the viability of HepG2 cells was measured by MTT assay. AFL at the high concentration such as 400 ug/mL showed to increase significantly viabilities of HepG2 cells compared with hepatotoxicants (EtOH, gallic acid, nicotine, acetaminophen, and lipopolysaccharide) only (p<0.05). AFL could be supposed to have the hepatoprotective effect against hepatotoxicants such as gallic acid, EtOH, nicotine, acetaminophen, and lipopolysaccharide at the high concentration.

  • The effects of the members of the same nuclear receptor superfamily (all-trans retinoic acid (RA), thyroid hormone(T3) or hydrocortisone) on proliferation and differentiation in the SK-N-SH neuroblastoma (NB) cell lines were studied. NB cells were treated with RA, T3, or hydrocortisone at concentration of 10$^{-6}$ M or 10$^{-8}$ M for 3 days or 7 days. RA induced concentration- and time-dependent morphologic differentiation(neurite outgrowth and microtubule-associated protein expression) and growth inhibition in NB cells. Treatment of 10$^{-7}$ M T3 for 7 days increased viability and differentiation of NB cells. Treatment of 10$^{-6}$ M hydrocortisone for 7 days increased viability of NB cells. Although these three effectors are members of the same receptor superfamily, the regulation of brain development may be carried out in a different manner.

  • Microorganisms and their by-products are considered to be the major causes of pulpal and periapical pathosis. The role of microorganisms in endodotic infection has been studied regarding the prevalence of particular organisms found in root canal and periapical lesions. The aim of this study was to investigate the effect of culture supernatants of several oral microorganims isolated from infected root canals on the viability of cultured cell lines using colorimetirc assay. S. simulans, S. sciuri, E. faecium, S. intermedius, S. mitis, S. sanguis and S. uberis were incubated in Todd-Hewitt broth for 16 hours. 20 and 100ul of filtered bacterial cell culture supernatants were added to MK and Hep-2 cells. Cell viability was measured using MIT colorimetric assay. 20ul and 100u1 of S. sanguis supernatant showed significant cytotoxicity compared to control on MK cells. 100ul of S. sanguis supernatant significantly depressed viability of HEp-2 cells. E. faecium and S. intermedius did not affect the viability of MK and HEp-2 cells.

  • In order to clarify effects of phytohormones on the viability and the cell wall regeneration of protoplasts isolated from Nicotiana tobacum L. var. BY4, protoplasts isolated from mesophyll tissue were cultured on the Murashige-Skoog liquid media supplemented with auxin(2, 4-D, NAA, IAA) and/or cytokinin (kinetin, BAP, 2ip). Viability of protopplasts was higher in the culture medium containing auxin and cytokinin, especially in the combination of 2, 4-D and BAP. The effectual cell wall regeneration of protolasts was observed when theprotoplasts were cultrued on the medium supplemented with auxin alone, especially with IAA. Cell wall regernation started from 2-3 days after culture and was not detected at budding regions. When the protoplasts were cultured on the phytohormone-free medium, the viability of protoplasts dramatically decreased 4 days after culture.

  • The aim of the present studies was to analyze the physiological background of ethanol inhibition in Zumomonas mobilis. The experiments were carried out with a number of continuous culture to give steady state concentration of ethanol. The domposition of fatty acids in the cells obtained from various conditions was analyzed and cell viability was also estimated. As results, it was found that vaccenic acid was the mafor fatty acid in the cell of Z. mobilis and the concentration was changed apparently to increase as increasing the concentration of ethanol produced from substrate utilization. Finally it was observed also that cell viability was decreased remarkably at the elevated ethanol concentration. Those changes might play important roles in the ethanol fermentation to give more complex phenomena observed at high concentration of ethanol.

  • After the short-term dynamics due to the major disturbance are over, the power system may lead to viability crisis state wherein there is possibility of cascading damage. This paper presents an emergency control algorithm to alleviate the obstacles of system frequency or bus voltage during the viability crisis state. The algorithm considers the effects of controlling reactive power sources for load shedding and generation reallocation in order to alleviate the obstacles. The problem is decomposed into a subproblem I and a subproblem II. The former minimizes system frequency deviation from nominal value and the latter voltage violation of load buses. The optimization problem is solved by a reduced gradient technique which can handle a great number of inequality constraints more easily. It has been verified that the use of the proposed algorithm for IEEE 14 bus system alleviates the obstacles efficiently during the viability crisis.

  • Kudoa septempunctata have been reported as a causative agent for acute transient gastrointestinal troubles after eating raw olive flounder (Paralichthys olivaceus). It raised public health concerns and quarantine control in several countries. Quantitative evaluation on viability of K. septempunctata is crucial to develop effective chemotherapeutics against it. A cytometry using fluorescent stains was employed to assess effect of three compounds on viability of K. septempunctata. Epigallocatechin gallate reduced markedly viability of K. septempunctata at 0.5 mM or more, and damaged K. septempunctata spores by producing cracks.

  • Seed viability testing provides valuable information for assessing seed lot germinability. However, most testing methods require destruction of seed prior to test. Because the dissected seeds for viability test cannot be used further evaluation, the nondestructively X-ray photography technique that can be applied for the evaluation of seed quality has been developed. In order to know the validity and accuracy of X-ray photography technique in seed evaluation test that conducted to remove the abnormal seed from a seed lot, we have compared the results from tetrazolium viability test, germination test and X-ray contrast method in cotton. Metallic salts treatment increased the efficiency of X-ray photographic method by enhancing the penetration of X-ray in abnormal or damaged seeds rather than normal seeds that have strong and well-organized tissues in seed. Cotton seeds presoaked for 16 hr in distilled water followed by soaking into metallic salt solution (5% NaI in water) for 60 min were easily classified seeds into dead seed and viable seed based on the radiography images obtained by X-ray radiation. We concluded that soft X-ray photography was reliable to find out the various defective characters due to heat and mechanical damage of seeds.
