Extensive Rib Osteosarcoma with Intraperitoneal Invasion in a Small Breed Dog: Clinical and Histopathologic Findings

  • An 11-year-old mixed breed male dog was presented with a thoracoabdominal wall mass of 3-year duration. Initial tentative diagnosis of osteosarcoma was made. Despite chemotherapy treatment, 72 days following the date of presentation, the dog was euthanized. Based upon necropsy and histopathologic findings, the tumor was definitely diagnosed as a combined type osteosarcoma of the rib. At necropsy examination, the tumor extended the left kidney and diaphragm, but distant metastasis was not found. The tumor's weight was 2.3kg and that was 38.3% of the dog's weight. This case report describes the clinicopathological, computed tomographic, histopathologic and immunohistochemical findings of extensive rib osteosarcoma in a small breed dog.

In Vitro Screening for Compounds Derived from Traditional Chinese Medicines with Antiviral Activities Against Porcine Reproductive and Respiratory Syndrome Virus

  • Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol-2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration ($CC_{50}$) and 50% effective concentration ($EC_{50}$) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The $EC_{50}$ values were $270.8{\pm}14.6{\mu}g/ml$ and $28.21{\pm}26.0{\mu}g/ml$ and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

Peripheral Vestibular Syndrome in a Cat with Foreign Body Otitis Media/Interna

  • A 15-month-old, intact male, domestic short hair cat was presented with ataxia, protrusion of the right third eyelid, and anorexia. Clinical signs were firstly noted after ear cleaning at home. Symptoms of the Homer's syndrome were evident. However, postural and proprioceptive reaction deficits were not detected on neurologic examination. Otoscopic examination revealed foreign body in right ear canal. A diagnosis of peripheral vestibular syndrome was made based on results of physical examination including neurologic and otic examination, blood work, and magnetic resonance imaging (MRI). Based on these examinations, the present patient was definitely diagnosed as otitis medial interna induced by foreign body. The vestibular signs were resolved 10 days after removal of cotton tips in right ear canal. This case report indicates that home-care cotton swab can iatrogenically induce otitis media/interna in cats.

  • An analytical method for the simultaneous determination of veterinary medicines (amprolium and decoquinate) in cattle and chicken's muscle by HPLC/UV-vis was established. Samples were extracted by a HLB (Hydrophilic-Liphophilic Balance) cartridge with acetonitrile and methanol. Prior to HPLC injection, a mixture solvent (Water:MeOH, 1:1) was utilized as a reconstitution solvent. Chromatographic separation was achieved with a C18 column ($250{\times}4.6mm$, $5{\mu}m$) using gradient elution with 20 mM HFBA and MeOH:ACN (1:1.8). The calibration curves from the spiked blank matrix showed good linearity (above $r^2$=0.997) in the concentration range of $0.13-12.0mg\;kg^{-1}$. The relative recovery (accuracy) and limit of quantitation (LOQ) were in the range of 78.5-107.1% and $0.13-0.42mg\;kg^{-1}$, respectively. The developed method can be used to determine under the MRL (Maximum Residue Limits) levels of veterinary medicines in animal tissues.

Topical Application of 0.1% Tacrolimus Ointment for Treatment of Sinus Refractory Pododermatitis Secondary to Atopy in a Dog

  • An 8-year-old, castrated male Shih-tzu was referred due to relapsing pododermatitis and generalized pruritus. On physical examination, right forepaw showed swelling and serosanguinous exudates from sinus tract on dorsal paw. There were no remarkable findings on complete blood count (CBC), serum chemistry, and radiologic examination. On cytological examination of exudates from sinus tract, phagocyted bacteria and numerous degenerative neutrophils were noted. Results of deep skin scraping and plucking hair examination were unremarkable. Skin biopsy was performed and ruled out other skin diseases. Histopathology showed epidermal hyperplasia and diffuse mononuclear cell inflammation in dermal layer. Moreover, pyogranulomatous inflammation was demonstrated in subcutaneous layer. This case was clinically diagnosed as pododermatitis secondary to atopic dermatitis (AD). Clinical signs of pododermatitis were gradually improved following topical application of tacrolimus 0.1% ointment. This case report describes that tacrolimus ointment can be applicable for the treatment of refractory pododermatitis.

Cerebrospinal fluid analysis in 13 clinically healthy Beagle dogs; hematological, biochemical and electrophoretic findings

  • The purpose of this study is to define the normal findings of cerebrospinal fluid (CSF) of the clinically healthy Beagle dogs and to provide basic information in diagnosis of neurologic disorders. CSF obtained from 13 clinically healthy dogs was examined for total and differential cell counts, total protein concentration, glucose and lactate dehydrogenase (LDH) concentration, specific gravity, turbidity, and protein electrophoresis. On gross examination, CSF samples evaluated were clear and colorless. Few red blood cells and nucleated cells were present. The mean concentration of glucose and LDH examined were 65.8 mg/dl and 2.7 mg/dl, respectively. The cellular components of CSF samples based on differential counts were monocytes (41.9%), activated macrophages (35.8%), lymphocytes (20.0%), neutrophils (1.6%), and eosinophils (0.7%). The fractions of electrophoretic protein in CSF were albumin (52.7%), alpha-globulin (16.5%), beta-globulin (24.8%), and gamma-globulin (3.0%). Results of albumin quota were ranged from 0.15 to 0.38. In conclusion, this study provided normal composition of CSF in Beagle dogs.

Challenges of stem cell therapies in companion animal practice

  • Regenerative medicine using stem cells from various sources are emerging treatment modality in several refractory diseases in veterinary medicine. It is well-known that stem cells can differentiate into specific cell types, self-renew, and regenerate. In addition, the unique immunomodulatory effects of stem cells have made stem cell transplantation a promising option for treating a wide range of disease and injuries. Recently, the medical demands for companion animals have been rapidly increasing, and certain disease conditions require alternative treatment options. In this review, we focused on stem cell application research in companion animals including experimental models, case reports and clinical trials in dogs and cats. The clinical studies and therapeutic protocols were categorized, evaluated and summarized according to the organ systems involved. The results indicate that evidence for the effectiveness of cell-based treatment in specific diseases or organ systems is not yet conclusive. Nonetheless, stem cell therapy may be a realistic treatment option in the near future, therefore, considerable efforts are needed to find optimized cell sources, cell numbers and delivery methods in order to standardize treatment methods and evaluation processes.

Study on Prevention of Adhesion Formation by Use of Sodium Carboxymethylcellulose and Dextran 70 II. Effect of Administration of Sodium Carboxymethylcellulose and Dextran 70 Solutions on Peripheral Blood Pictures in Adhesion-induced Rabbits (Sodium Carboxymethylcellulose 및 Dextran 70을 이용한 유착형성 방지에 관한 연구 II. 유착자극후 Sodium Carboxymethylcellulose 및 Dextran 70의 투여가 토끼의 혈액상에 미치는 영향)

  • The effectiveness of sodium carboxymethylcellulose(SCMC) and dextran 70 in the prevention of adhesion formation in abdominal cavity of rabbits following artificial injuries was elucidated and the effects -of these medicines on blood pictures were also examined. After abrasion treatment on jejunum in gonadotropins primed rabbits, 1, 2 and 3% of SCMC solutions, 6 and 10% dextran 70 solutions and a synthetic solution of 1% SCMC and loft dextran 70 in 0.9% saline solution were infused into the abdominal cavity. Four weeks later the abdominal cavities were reopened under general anesthesia. The synthetic solution showed the highest adhesion reduction rate(100%), while 1% SCMC, 6 and loft dextran solutions showed no significantly evident effect of adhesion prevention. The SCMC solutions showed better adhesion reduction effect than dextran 70 solutions. After infusion of these adhesion preventive medicines, the changes of total leucocytes, erythrocytes, lymphocytes, PCV, plasma fibrinogen and protein contents were examined. No remarkable difference in blood pictures was shown between the synthetic solution and the other medicines. Therefore, it can be suggested that the synthetic solution of 1% SCMC and 10% dextran 70 in 0.9% saline solution at dose of 5$m\ell$/kg of body weight is prominently effective in the prevention of postoperative adhesion formation and wolf be safe in animals and human.

Effects of Oldenlandidae Herba as an Alternative Medicine on the Swine Productivity (대체의학으로서 백화사설초 급여가 돼지 생산성에 미치는 효과)

  • As control of swine consumptive disease is the key of success in pig farming business, the effect of Oldenlandidae Herba on prevention of swine diseases as an alternative medicine was investigated in-vivo. As it contains substances such as ${\beta}$-sitisterol, ursolic acid and 3-O-${\beta}$-D-giucoside, it is effective on suppression of colonic tumor in rats and also, is widely used as an preventative medicine of cancer and inflammation in Chinese medicine. In this experiment, sows and piglets on two different farms were treated with Oldenlandidae Herba, and they recoded weaning rate greater than 80%, which were higher than national average 70% (PSY 68%). Meanwhile, neither organ accumulation nor toxicity has been detected on the basis of hematology and serum chemistry. In conclusion, the extracts of Oldenlandidae Herba is expected to be an good alternative medicines for swine productivity.

Granulosa Cell Tumor of Ovary in a Yorkshire terrier Dog (요크셔테리어에서 발생한 난소의 Granulosa cell tumor)

  • A 12-year-old, intact female Yorkshire terrier was presented with progressive abdominal distention. On radiographic and ultrasonographic evaluation, a large mass was detected in right upper abdomen and it had several discrete anechoic follicles and large cysts. The mass was removed by complete ovariohysterectomy and the resected specimen was histopathologically examined. Based on the gross and histopathologic findings, the mass was definitely diagnosed as malignant granulosa cell tumor (GCT). Until now, the dog has been successfully managed more than a year without complications.
