Viruliferous Rate of the Small Brown Planthopper in the Maize Field Infected with Rice black-streaked dwarf virus (옥수수 재배지의 벼검은줄오갈병 매개충인 애멸구의 보독충율)

  • The Rice black-streaked dwarf virus (RBSDV) infected 99-100% of a $1000\;m^2$-maize field in Mungyeong City in 2007. Adjacent to the area is a Persimmon orchard where barley crops were grown under the trees as green manure crops and for soil amendments. The barley acted as winter host to the small brown plant hoppers (SBPH) enabling the insects to survive and pass the winter season. The existence of RBSDV was detected and confirmed by RT-PCR using S9 specific primer. Samples of the insect vector SBPH were collected in the area on May 3, June 7 and, August 4 and the results of the RT-PCR analysis revealed viruliferous insect rates of 2.9, 4.8, and 4.4%, respectively. These observed viruliferous insect rates were similar with those detected in RBSDV infected rice fields.

Production of Transgenic Birdsfoot trefoil Plants by Introduction of 'SWPA2 Promoter + AtNDPK2 Gene' ('SWPA2프로모터+AtNDPK2유전자' 도입에 의한 버즈풋 트레포일 형질전환체 생산)

  • To develop transgenic birdsfoot trefoil (Lotus corniculatus L.) plants tolerant to environmental stress, Arabidopsis NDPK gene (AtNDPK) was introduced into birdsfoot trefoil plants using Agrobacterium-mediated transformation and expressed powerfully under the control of the SWPA2 promoter. The expression vector, pCAMBIA2300 was used for introduction of AtNDPK gene into birdsfoot trefoil plants. The transformed calli were selected on kanamycin containing medium and then regenerated. The transformed birdsfoot trefoil plants were cultivated fur 4 months on BOi2Y medium. Genomic DNA PCR and Southern blot analysis confirmed the incorporation of AtNDPK into the birdsfoot trefoil genome.

An Analysis on the Influence of the Financial Market Fluctuations on the Housing Market before and after the Global Financial Crisis (글로벌 금융위기 전후 금융시장 변동이 주택시장에 미치는 영향 분석)

  • As the subprime mortgage crisis spread globally, it depressed not only the financial market, but also the construction business in Korea. In fact, according to CERIK, the BSI of the construction businesses plunged from 80 points in December 2006 to 14.6 points in November 2008, and the extent of the depression in the housing sector was particularly serious. In this respect, this paper analyzes the influence of the financial market fluctuation on the housing market before and after the Global Financial Crisis using VECM. The periods from January 2000 to December 2007 and January 2008 to October 2015, before and after the financial crisis, were set as Models 1 and 2, respectively. The results are as follows. First, when the economy is good, the Gangnam housing market is an attractive one for investment. However, when it is depressed, the Gangnam housing market changes in response to the macroeconomic fluctuations. Second, the Gangbuk and Gangnam housing markets showed different responses to fluctuations in the financial market. Third, when the economy is bad, the effect of low interest rates is limited, due to the housing market risk.

Acquisition of Thermotolerance in the Transgenic Plants with BcHSP17.6 cDNA (BcHSP17.6 cDNA의 도입에 의한 형질전환된 식물의 내열성 획득)

  • Recombinant plasmid, pBKH4, containing NPT II and P35S-BcHSP17.6 was constructed by ligation of Bum H I -digested pBKSl-l and BcHSP 17.6 (thermotolerance gene) 6om pBLH4. The tobacco leaf disc was cocultivated with transformed Agmbacterium tumefaciens bearing pBKH4 for 24 hours and transformed shoots were selected on MS-n/B medium containing $100\;{\mu\textrm{g}}/ml$ of kanamycin. Heat-killing temperature of Nicotima tabacum was $50^{\circ}$ for >15min, and transformed tobacco plants with BcHSP17.6 cDNA exhibited thermotolerance at the heat-killing temperature. The transgenic plants were analyzed by Southern blot hybridization with the probe of ${\alpha}^{_32}P$ labelled BcHSP17.6 cDNA. Transcription and expression level of BcHSP17.6 cDNA were also continued by Northern blot analysis and Ouchterlony double immunodiffusion assay. In this study, we suggest that the BcHSP17.6 cDNA introduced to tobacco plant is related to thenuoto-lerance and 17.6-kD LMW HSP acts as a protector from heat damage in plants.

Construction of a Full-length cDNA Library from Korean Stewartia (Stewartia koreana Nakai) and Characterization of EST Dataset (노각나무(Stewartia koreana Nakai)의 cDNA library 제작 및 EST 분석)

  • In this study, we report the generation and analysis of 1,392 expressed sequence tags (ESTs) from Korean Stewartia (Stewartia koreana Nakai). A cDNA library was generated from the young leaf tissue and a total of 1,392 cDNA were partially sequenced. EST and unigene sequence quality were determined by computational filtering, manual review, and BLAST analyses. Finally, 1,301 ESTs were acquired after the removal of the vector sequence and filtering over a minimum length 100 nucleotides. A total of 893 unigene, consisting of 150 contigs and 743 singletons, was identified after assembling. Also, we identified 95 new microsatellite-containing sequences from the unigenes and classified the structure according to their repeat unit. According to homology search with BLASTX against the NCBI database, 65% of ESTs were homologous with known function and 11.6% of ESTs were matched with putative or unknown function. The remaining 23.2% of ESTs showed no significant similarity to any protein sequences found in the public database. Annotation based searches against multiple databases including wine grape and populus sequences helped to identify putative functions of ESTs and unigenes. Gene ontology (GO) classification showed that the most abundant GO terms were transport, nucleotide binding, plastid, in terms biological process, molecular function and cellular component, respectively. The sequence data will be used to characterize potential roles of new genes in Stewartia and provided for the useful tools as a genetic resource.

  • This paper proposes algorithm to reduce the computing time in a neural network that reduces transmission of data for tracking mobile objects in surveillance networks in terms of detection and communication load. Object Detection can be defined as follows : Given image sequence, which can forom a digitalized image, the goal of object detection is to determine whether or not there is any object in the image, and if present, returns its location, direction, size, and so on. But object in an given image is considerably difficult because location, size, light conditions, obstacle and so on change the overall appearance of objects, thereby making it difficult to detect them rapidly and exactly. Therefore, this paper proposes fast and exact object detection which overcomes some restrictions by using neural network. Proposed system can be object detection irrelevant to obstacle, background and pose rapidly. And neural network calculation time is decreased by reducing input vector size of neural network. Principle Component Analysis can reduce the dimension of data. In the video input in real time from a CCTV was experimented and in case of color segment, the result shows different success rate depending on camera settings. Experimental results show proposed method attains 30% higher recognition performance than the conventional method.

Promoter Cloning of Human SETDB1 Gene Utilizing Bioinformatic Programs (생물정보 프로그램을 활용한 SETDB1 유전자 프로모터 클로닝)

  • Eukaryotic gene expression is an important process, which is initiated by several transcription factors and RNA polymerases that occupy the promoter region of genomic DNA. Although there are many experiments to identify the promoter region in a gene, it is time and labor consuming to finalize it. In this study, we utilized bioinformatic programs, including Ensembl, NCBI, and CpG plots, to identify the cloning promoter region in SETDB1 genomic DNA. We performed PCR amplification to obtain the SETDB1 promoter on an approximately 2 kb region upstream from the TSS named SETDB1-P1. The PCR product was ligated with TA cloning vectors, and we confirmed the insert size using restriction enzyme digestion. Sequentially, the insert was subcloned into a pGL3-luc vector to produce pGL3-SETDB1- P1-luc and then confirmed by DNA sequencing. We also obtained a fragmented PCR product called P2 and P3 and performed a luciferase assay using pGL3-SETDB1-P1-luc transfection. We found that several anticancer drugs, including taxol, 4-FU, and doxorubicin, decreased the promoter activity of SETDB1. We obtained consistent data on the regulation of SETDB1 gene expression after anticancer drug treatment using Western blot analysis and RT-PCR. Our results suggest that promoter cloning of the human SETDB1 gene utilizing bioinformatics is a very useful and timesaving approach to study gene expression.

Expression Analysis of the Ligand to Ly-6E.1 Mouse Hematopoietic Stem Cell Antigen

  • Ly-6E.1 antigen was proposed as a regulatory molecule of T lymphocyte activation, a hematopoietic stem cell marker, a memory cell marker, and an adhesion molecule. Though there were several reports suggesting the presence of Ly-6 ligand, the characterization of the ligand was not yet performed, As an attempt to screen the expression of Ly-6E.1 ligand, we prepared a probe for detecting Ly-6E.1 ligand by producing a fusion protein between Ly-6E.1 and $hlgC_{r1}$, A mammalian cell expression vector with Ly-6E.$1/hlgC_{r1}$ chimeric cDNA was transfected in SP2/0-Ag14 myeloma cells, and stable transfectants were selected. The fusion protein was produced as a dimer and maintained the epitopes for monoclonal antibodies specific for Ly-6E.1 and for anti-human lgG antibody. The purified fusion protein through Gammabind G column was used for FACS analyses for the expression of Ly-6E.1 ligand. The fusion protein interacted with several cell lines originating from B cells, T cells, or monocytes. The fusion Protein also strongly stained bone marrow, lymph node, and spleen cells, but thymic cells weakly, if any. The staining was more obvious in C57BL/6 $(Ly-6^b)$ than Balb/c $(Ly-6^a)$ mice. These results suggest that the interaction of Ly-6E.1 with Ly-6E.1 ligand may function both in the stem cell environment and in the activation of mature lymphocytes. The fusion protein may be a valuable tool in characterization of biochemical properties of the Ly-6E.1 ligand and, further, in isolating its cDNA.

Report on Forests Damaged Due to Cutting Trees Infected by Pine Wilt Disease (소나무재선충 고사목 제거에 따른 산림훼손 보고)

  • Despite various control methods for pine wilt disease, the disease has been increasing at an alarming rate every year in Korea. Still there is a lack of research on the problems of the current methods used to curb the disease. One main method to control pine wilt disease is fumigation for eliminating the larva of the pine sawyer beetle (Monochamus alternatus) which is known as the vector insect for pine wilt disease. We surveyed and analyzed the amount of destroyed uninfected trees in the fumigated area by studying 32 survey plots ($100m^2$) in Miryang city. The destroyed trees' crown area was estimated by induced regression between root collar area and crown area of the remaining trees (coniferous tree: $R^2=57.4$, deciduous broad-leaved tree: $R^2=63.8$). According to the analysis results, the infected trees (cut trees) were 18.7% and cut trees which were not infected were 35.5% of the total tree population of the study area on the basis of total crown area. In case of pine trees, 8.4% of uninfected trees were cut off but 62% of uninfected deciduous broad-leaved trees including Quercus spp. were affected and damaged during the fumigation process. Since these broad-leaved trees dominate the sub-canopy layer of the forest, this indiscriminate interruption of broad-leaved trees has the adverse effect of depriving the public benefits such as shadow, a cooler weather, controlling humidity, etc.

Isogeometric Shape Design Optimization of Power Flow Problems at High Frequencies (고주파수 파워흐름 문제의 아이소-지오메트릭 형상 최적설계)

  • Using an isogeometric approach, a continuum-based shape design optimization method is developed for steady state power flow problems at high frequencies. In case the isogeometric method is employed to the shape design optimization, the NURBS basis functions used in CAD geometric modeling are directly utilized to embed the exact geometry into the computational framework so that the design parameterization for shape optimization is much easier than that in the finite element method and consequently provides the enhanced smoothness of design perturbations. Thus, exact geometric models can be used in both the response and the shape sensitivity analyses, where normal vector and curvature are continuous over the whole design space so that enhanced shape sensitivity can be expected. Through numerical examples, the developed isogeometric sensitivity is compared with finite difference one to provide excellent agreement. Also, it turns out that the proposed method works very well in the shape optimization problems.
