Porphyromonas gingivalis, one of the major periodontal pathogens, is implicated in the initiation and progression of periodontal disease. The initial stages of periodontal inflammation are accompanied by vascular hyperpermeability. In our present study, we report that the P. gingivalis lipopolysaccharide (LPS) increases the mRNA expression of interleukin-8 (IL-8), a major inducer of vascular permeability, in vascular endothelial cells. P. gingivalis LPS also stimulated the induction of IL-8 secretion in endothelial cells. The P. gingivalis LPS-induced expression of IL-8 was primarily modulated by nuclear factor-${\kappa}$B(NF-${\kappa}$B). P. gingivalis LPS significantly enhanced the vascular permeability both in vitro and in vivo, and a blockade of the IL-8 receptor decreased the P. gingivalis LPS-induced vascular permeability. Taken together, these results suggest that P. gingivalis LPS increases vascular permeability through the NF-${\kappa}$B-dependent production of IL-8 in vascular endothelial cells.
The antihistamine effects of the triprolidine were studied in rats to determine the feasibility of their enhanced transdermal delivery from the poly (4-methyl-1-pentene) (TPX) matrix system containing penetration enhancer and plasticizer. The antihistamine effects were determined by the Evans blue dye procedure by comparing the changes in vascular permeability increase following the transdermal administration. The vascular permeability increase was significantly reduced by transdermal administration of the triprolidine-TPX system containing triethyl citrate (TEC) and polyoxyethylene-2-oleyl ether (POE). Both the plasticizer and penetration enhancer played an important role in the skin permeation of triprolidine and increased the antihistamine effects. These results showed that the triprolidine-TPX matrix system containing plasticizer and penetration enhancer could be a transdermal delivery system providing the increased antihistamine effects.
These studies were conducted to investigate the antiinflammatory effects and the mechanism of action of natural products. We used the methods of "carrageenan induced foot edema" , "PAF (platelet activating factor) induced foot edema" , "inhibition test of vascular permeability" , "inhibition test of white blood cell migration" , "formation of granuloma" and "adjuvant induced arthritis" to examine the antiinflammatory erects of Angelica gigas, Ledebouriella seseloides, Ginkgo biloba and Bamboo salt (Jukyom). The oral administration of the water extract of Angelica gigantis radix, the methanolic extract of Ginkgo folium and the aqueous solution of Bamboo salt showed antiinflammatory effect on carrageenan and PAF induced foot edema in SD rat at a dose of 1 g/kg. The same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in mice. The aqueous solution of Bamboo salt inhibited the formation of ganuloma in SD rats at a oral dose of 1 g/kg. Angelica gigantis radix seems to have antiinflammatory effect by inhibition of PAF.
Mungbean trypsin inhibitor (MBTI) was isolated and purified from Mung bean which has been used as a galenic and traditional food. MBTI and poly(ethylene glycol) were conjugated by using water soluble carbodiimide. We evaluated the therapeutic value of the MBTI and MBTI-PEG conjugate using animal models, sublethal septic shock model in guinea pig caused by pseudomonal elastase, shock model in rat caused by lipopolysaccharide, and the vascular permeability test by using pseudomonal elastase. In two shock model in guinea p Is and in rat, hypotesion shock was inhibited by pretreatment of MBTI. And also the vascular permeability caused by pseudomonal elastase reduced by pretreatment of MBTI. Also, therapeutic value of the MBTI-PEG conjugate was evaluated by using the sublethal septic shock model caused by pseudomonal elastase. The MBTI-PEG conjugate was more effective than native MBTI against pseudomonal elastase induced septic shock in guinea pig model.

Seungmagalgeuntang-gamibang has long been known to have anti-allergic effect. However, the mechanism of action of Seungmagalgeuntang-gamibang is not well investigated. The author analysed the effects of Seungmagalgeuntang-gamibang on the vascular permeability, delayed-type and contact hypersensitivities, and phagocytic function, the results obtained are as follows: 1. Administration of Seungmagalgeungtang-gamibang decreased the vascular permeability induced by serotonin in the mouse. 2. Administration of Seungmagalgeuntang-gamibang decreased the vascular permeability induced by histamine without statistical significant. 3. Administration of Seungmagalgeuntang-gamibang decreased the delayed-type hypersensitivity induced by sheep red blood cells. 4. Administration of Seungmagalgeungtang-gamibang decreased the contact hypersensitivity induced by dinitrochlorobenzene. 5. Seungmagalgeungtang-gamibang increased the phagocytic-activities of macrophages in vitro and in vivo. 6. Seungmagalgeungtang-gamibang enhanced the formation of reactive oxygen intermediates in vitro and in vivo. The above results demonstrate that Seungmagalgeuntang-gamibang suppresses the hypersensitivity reactions with increasing the phagocytic functions and formations of reactive oxygen intermediates from macrophages.

Vascular endothelial growth factor (VEGF) plays a pivotal role in pathologic ocular neovascularization and vascular leakage via activation of VEGF receptor 2 (VEGFR2). This study was undertaken to evaluate the therapeutic mechanisms and effects of the tetrapeptide Arg-Leu-Tyr-Glu (RLYE), a VEGFR2 inhibitor, in the development of vascular permeability and choroidal neovascularization (CNV). In cultured human retinal microvascular endothelial cells (HRMECs), treatment with RLYE blocked VEGF-A-induced phosphorylation of VEGFR2, Akt, ERK, and endothelial nitric oxide synthase (eNOS), leading to suppression of VEGF-A-mediated hyper-production of NO. Treatment with RLYE also inhibited VEGF-A-stimulated angiogenic processes (migration, proliferation, and tube formation) and the hyperpermeability of HRMECs, in addition to attenuating VEGF-A-induced angiogenesis and vascular permeability in mice. The anti-vascular permeability activity of RLYE was correlated with enhanced stability and positioning of the junction proteins VE-cadherin, ${\beta}$-catenin, claudin-5, and ZO-1, critical components of the cortical actin ring structure and retinal endothelial barrier, at the boundary between HRMECs stimulated with VEGF-A. Furthermore, intravitreally injected RLYE bound to retinal microvascular endothelium and inhibited laser-induced CNV in mice. These findings suggest that RLYE has potential as a therapeutic drug for the treatment of CNV by preventing VEGFR2-mediated vascular leakage and angiogenesis.

Objective: This study aimed to determine whether there are regional differences in the blood-brain barrier (BBB) permeability of cognitively normal elderly participants and to identify factors influencing BBB permeability with a clinically feasible, 10-minute dynamic contrast-enhanced (DCE) MRI protocol. Materials and Methods: This IRB-approved prospective study recruited 35 cognitively normal adults (26 women; mean age, 64.5 ± 5.6 years) who underwent DCE T1-weighted imaging. Permeability maps (Ktrans) were coregistered with masks to calculate the mean regional values. The paired t test and Friedman test were used to compare Ktrans between different regions. The relationships between Ktrans and the factors of age, sex, education, cognition score, vascular risk burden, vascular factors on imaging, and medial temporal lobar atrophy were assessed using Pearson correlation and the Spearman rank test. Results: The mean permeability rates of the right and left hippocampi, as assessed with automatic segmentation, were 0.529 ± 0.472 and 0.585 ± 0.515 (Ktrans, x 10-3 min-1), respectively. Concerning the deep gray matter, the Ktrans of the thalamus was significantly greater than those of the putamen and hippocampus (p = 0.007, p = 0.041). Regarding the white matter, the Ktrans value of the occipital white matter was significantly greater than those of the frontal, cingulate, and temporal white matter (p < 0.0001, p = 0.0007, p = 0.0002). The variations in Ktrans across brain regions were not related to age, cognitive score, vascular risk burden, vascular risk factors on imaging, or medial temporal lobar atrophy in the study group. Conclusion: Our study demonstrated regional differences in BBB permeability (Ktrans) in cognitively normal elderly adults using a clinically acceptable 10-minutes DCE imaging protocol. The regional differences suggest that the integrity of the BBB varies across the brains of cognitively normal elderly adults. We recommend considering regional differences in Ktrans values when evaluating BBB permeability in patients with neurodegenerative diseases.
Actions for 48-hour homolgous passive cutaneous anaphylaxis (48-hr PCA) and chemical mediators were investigated in mice and rats. The hyaluronidase activity, which was used in the in vitro screening test of the antiallergic action, was significantly inhibited by Magnoiliae Flos, Achyranthis Radix, Forsythiae Fructus, Alpiniae Fructus, Anemarrhenae Rhizoma and Ponciri Fructus among twelve medicinal plants and tranilast as a comparative drug of the antiallergic action. In the mouse ear, 48-hr PCA was significantly inhibited by intraperitoneal (i.p.) pretreatment with Magnoliae Flos, Achyranthis Radix, Alpiniae Fructus, Anemarrhenae Rhizoma, Ponciri Fructus, Ledebouriellae Radix and tranilast. And also, the increment of vascular permeability induced by histamine or serotoin was inhibited significantly by i.p. pretreatment with Magnoliae Flos, Achyranthis Radix, Alpiniae Fructus, Anemarrheuae rhizoma, Zizyphi Fructus and tranilast. In the rat dorsal skin, the increment of vascular permeability induced by histamine or serotonin was significantly inhibited by i.p. pretreatment with Magnoliae Flos, Acyranthis Radix, Alpiniae Fructus, Anemarrhenae Rhizoma and tranilast. And also, the increment of vascular permeability induced by compound 48/80 or calcium ionophore A 23187 was significantly inhibited by i.p. pretreatment with Magnoliae Flos, Achyranthis Radix, Alpiniae Fructus, Amemarrhenae Rhizoma, Zizyphi Fructus, Ledebouriellae Radix, Lithospermi Radix and tranilast. These results suggest that each water extracts of Magnoliae Flos, Achyranthis Radix, Alpiniae Fructus and Anemarrhenae Rhizoma have especially antiallergic activities.

Pulmonary edema is a major cause of mortality due to acute lung injury (ALI). The involvement of protein kinase C-${\delta}$ (PKC-${\delta}$) in ALI has been a controversial topic. Here we investigated PKC-${\delta}$ function in ALI using PKC-${\delta}$ knockout (KO) mice and PKC inhibitors. Our results indicated that although the ability to produce proinflammatory mediators in response to LPS injury in PKC-${\delta}$ KO mice was similar to that of control mice, they showed enhanced recruitment of neutrophils to the lung and more severe pulmonary edema. PKC-${\delta}$ inhibition promoted barrier dysfunction in an endothelial cell layer in vitro, and administration of a PKC-${\delta}$-specific inhibitor significantly increased steady state vascular permeability. A neutrophil transmigration assay indicated that the PKC-${\delta}$ inhibition increased neutrophil transmigration through an endothelial monolayer. This suggests that PKC-${\delta}$ inhibition induces structural changes in endothelial cells, allowing extravasation of proteins and neutrophils.

Allergic conjunctivitis is the most common allergic disease. These diseases are severe & frequent which requires search of new treatments. The aim of the study was to investigate the effects of Tinospora cordifolia (TC), Rubia cordifolia Linn. (RC) on experimentally induced allergic conjunctivitis in rats. In this study, dried water soluble extracts of TC and RC. (250 and 500 mg/kg, p.o. for 7 days) were evaluated for their antiallergic activity in Wistar rats. They were tested for inhibition of egg albumin-induced vascular permeability, inhibition of histamine release from the rat conjunctiva as well as in histamine content in tears. TC and RC showed significant (P < 0.05) inhibition in vascular permeability, inhibition in histamine release from the rat conjunctiva which is reflected by reduced level of histamine content in tears. The activities were found to be comparable to azelastine hydrochloride. These results suggest that the inhibitory effect on egg albumin-induced experimental allergic conjunctivitis in rat may be due to the antihistaminic activity of TC and RC. Our studies provide evidence that TC and RC may be beneficial in the treatment of allergic conjunctivitis.
