Flaviviruses Induce Pro-inflammatory and Anti-inflammatory Cytokines from Murine Dendritic Cells through MyD88-dependent Pathway

  • Background: The genus Flavivirus consists of many emerging arboviruses, including Dengue virus (DV), Japanese encephalitis virus (JEV) and West Nile virus (WNV). Effective preventive vaccines remain elusive for these diseases. Mice are being increasingly used as the animal model for vaccine studies. However, the pathogenic mechanisms of these viruses are not clearly understood. Here, we investigated the interaction of DV and JEV with murine bone marrow-derived dendritic cells (bmDC). Methods: ELISA and FACS analysis were employed to investigate cytokine production and phenotypic changes of DCs obtained from bone marrow following flavivirus infection. Results: We observed that these viruses altered the cytokine profile and phenotypic markers. Although both viruses belong to the same family, JEV-infected bmDC produced anti-inflammatory cytokine (IL-10) along with pro-inflammatory cytokines, whereas DV infection induced production of large amounts of pro-inflammatory cytokines (IL-6 and TNF-${\alpha}$) and no IL-10 from murine bmDCs. Both flaviviruses also up-regulated the expression of co-stimulatory molecules such as CD40, CD80 and CD86. JEV infection led to down-regulation of MHC II expression on infected bmDCs. We also found that cytokine production induced by JEV and DV is MyD88-dependent. This dependence was complete for DV, as cytokine production was completely abolished in the absence of MyD88. With regard to JEV, the absence of MyD88 led to a partial reduction in cytokine levels. Conclusion: Here, we demonstrate that MyD88 plays an important role in the pathogenesis of flaviviruses. Our study provides insight into the pathogenesis of JEV and DV in the murine model.

Immune Response of the Japanese Eel(Anguilla japonica) to Vibrio anguillarum (Vibrio균에 대한 뱀장어 (Anguilla japonica)의 면역반응)

  • Some eels Anguilla japonica, ranging from 16 to 23 g in their weight(average: 20 g), were sampled at the private eel farming company equipped with water recycling system, located at Kimhae city, Kyungnam Province, Korea. Three kinds of vaccine were prepared with Vibrio anguillarum (EPM-8406) isolated at National Fisheries University in Korea for the immune response experiment against eels; those vaccines were made by inactivating the strain with $0.3\%$ formalin for 24 hrs at $25^{\circ}C$, heating for 3 mins or for 15 mins at $121^{\circ}C$, respectively. The various optimal vaccination conditions for the control of vibriosis in the fish were investigated based on the cultivation temperature, vaccination concentration and booster effect. The maximum titer rapidly increased with higher temperature up to $23^{\circ}C$, but there were little differences between $23^{\circ}C\;and\;28^{\circ}C$. The formalin-killed vaccine showed good efficacy at the injection concentration of above $10^8$ cells per fish and little effect at the below $10^7$ cells. The booster effect on the vaccination showed good efficacy above twice-injections with little difference between the numbers of injection.

Stability Evaluation of National Reference Standards for Blood Products in Korea

  • National reference standards (NRSs) for biologics are established through potency estimation by a multi-center joint study of standard materials used in the approval process for national lot release and quality control of vaccines, blood products, and other biologics. In this study, a stability evaluation was conducted to determine whether the potency of NRSs for six blood products was being maintained at a consistent level in Korea. The present study conducted real-time stability tests via in-vivo/in-vitro bioassay on NRSs for blood coagulation factor VIII concentrate (2nd standard), antithrombin concentrate, prekallikrein activator, anti-hepatitis B immunoglobulin, blood coagulation factor IX concentrate, and anti-tetanus human immunoglobulin, as well as a trend analysis using cumulative annual results. The real-time stability test results showed that the mean potency of six NRSs was all within the control limit. In the trend analysis, the potency of NRS for blood coagulation factor VIII concentrate (2nd standard) showed a decreasing trend, while the potency of all other products had been stably maintained. The present study confirmed that the mean potency of NRSs for six blood products had been stably maintained in Korea. The findings of the present study establish a foundation that can ensure the quality of NRSs for biologics in Korea, and it is expected to make a major contribution to the supply of high-quality biologics.

Gene Cloning, Expression and Immunogenicity of the Protective Antigen Subolesin in Dermacentor silvarum

  • Subolesin (4D8), the ortholog of insect akirins, is a highly conserved protective antigen and thus has the potential for development of a broad-spectrum vaccine against ticks and mosquitoes. To date, no protective antigens have been characterized nor tested as candidate vaccines against Dermacentor silvarum bites and transmission of associated pathogens. In this study, we cloned the open reading frame (ORF) of D. silvarum 4D8 cDNA (Ds4D8), which consisted of 498 bp encoding 165 amino acid residues. The results of sequence alignments and phylogenetic analysis demonstrated that D. silvarum 4D8 (Ds4D8) is highly conserved showing more than 81% identity of amino acid sequences with those of other hard ticks. Additionally, Ds4D8 containing restriction sites was ligated into the pET-32(a+) expression vector and the recombinant plasmid was transformed into Escherichia coli rosetta. The recombinant Ds4D8 (rDs4D8) was induced by isopropyl ${\beta}$-D-thiogalactopyranoside (IPTG) and purified using Ni affinity chromatography. The SDS-PAGE results showed that the molecular weight of rDs4D8 was 40 kDa, which was consistent with the expected molecular mass considering 22 kDa histidine-tagged thioredoxin (TRX) protein from the expression vector. Western blot results showed that rabbit anti-D. silvarum serum recognized the expressed rDs4D8, suggesting an immune response against rDs4D8. These results provided the basis for developing a candidate vaccine against D. silvarum ticks and transmission of associated pathogens.

  • Purpose: To compare differences and changes in the parents' awareness regarding influenza and influenza vaccination for their children according to educational intervention. Methods: This study involved 1,034 parents with children aged 6-59 months, who lived in five areas in northern Seoul and Gyeonggi Province in 2015; the study was conducted using the questionnaire survey method. Survey items included awareness regarding influenza and influenza vaccination for children and future intentions of influenza vaccination and the underlying reasons. The participants were divided into 3 groups and differences according to education (group A: no education, group B: provided education) and the changes before and after education (group C) were analyzed. Results: Differences according to education between groups A and B were seen in the recognition of high-risk of influenza in their children, effectiveness of influenza vaccination, and cost of the influenza vaccine. Changes before and after education in group C were seen in the recognition of high-risk of influenza in their children, safety of influenza vaccination, cost of the influenza vaccine, and negativity toward the influenza vaccine. Conclusions: Educated parents were more aware regarding influenza and influenza vaccination for their children aged 6-59 months.

A Case of Miliary Tuberculosis Associated with Multiple Intracranial Tuberculoma (다발성 뇌결핵종을 동반한 속립결핵 1례)

  • The incidence of tuberculosis has been decreased, and especially the incidence of severe complicated tuberculosis has been markedly decreased as the result of widely used BCG vaccines. But tuberculosis is still an important community accquired infectiouse disease in the world despite continued worldwide efforts to control the disease. Miliary tuberculosis, the most serious complicated tuberculosis, can be occurred by lymphohematogenous dissemination of tuberculosis, and intracranial tuberculoma with or without tuberculosis meningitis can be developed in case of miliary tuberculosis. In general, serious tuberculosis infections such as miliary tuberculosis and CNS tuberculosis are developed especially in young infants and children in cases of delayed diagnosis and treatment despite receiving BCG vaccination, and usually those patients have contact sources. Intrcranial tuberculoma in children are usually found near infratentorial site at the base of cerebellum, and clinically symptoms and signs of increased intracranial pressure developed before treatment. Serial brain CT or MRI is a good non-invasive diagnostic modality of intracranial tuberculoma. Although surgical intervention was initially advocated as the mainstay of intracranial tuberculoma therapy, but many recent clinical studies indicate that intracranial tuberculoma can be cured with medical treatment alone. We experienced a case of 3 months old male patient, who was diagnosed as having miliary tuberculosis associated with multiple intracranial tuberculoma. He received BCG vaccination at 4 weeks after birth, and his father was confirmed as active pulmonary tuberculosis patient after this patient's admission. We report this case with a review of related literatures.

Factors Influencing Influenza Vaccination Coverage in Korean Adolescents: Analysis of the Korea National Health and Nutrition Examination Survey IV to VI (국내 청소년의 인플루엔자 예방접종률에 영향을미치는 요인: 국민건강영양실태조사 4-6기 자료 분석)

  • Purpose: Influenza vaccination coverage in adolescents is an important goal of informed vaccination policies and programs. This study aimed to estimate the influenza vaccination coverage rate and investigate the factors influencing influenza vaccination coverage in Korean adolescents. Methods: The study population consisted of 5,213 adolescents (aged 12 to 18 years) who participated in the Korea National Health and Nutrition Examination Survey from 2007 to 2014 (except for 2013). We analyzed influenza vaccination coverage in relation to the demographics, lifestyle, and medical characteristics of the participants. Results: The influenza vaccination coverage rate, during the study period, was 23.2% (range, 21.1% to 24.7%). Logistic regression analysis revealed that factors influencing influenza vaccination were elementary school age (odds ratio [OR], 1.706; 95% confidence interval [CI], 1.526 to 1.906), good self-rated health status (OR, 1.192; 95% CI, 1.057 to 1.344), a drinking status of non-drinker (OR, 1.769; 95% CI, 1.474 to 2.122), a smoking status of non-smoker (OR, 1.459; 95% CI, 1.144 to 1.860), and a past diagnosis of pneumonia (OR, 1.469; 95% CI, 1.076 to 2.006). Conclusions: Influenza vaccination coverage in Korean adolescents is relatively low. Special efforts are needed to increase vaccination coverage for adolescent groups with low vaccination rates including adolescent smokers and drinkers, middle and high school age adolescents, and adolescents with a poor self-rated health status.

The follow up study after massive outbreak of Akabane and bovine ephemeral fever viruses in Korea

  • Since a large number of Akabane and bovine ephemeral fever (BEF) infection occurred in the southern part of Korea in 2010, recent information about seroprevalence of Akabane virus (AKAV) and bovine ephemeral fever virus (BEFV) has been required for preventing both diseases. In this study, serological assay against AKAV and BEFV using virus neutralization assay was conducted using 1,743 bovine sera collected from Namwon, Miryang, Yeongju and Uljin which located in Southern part of Korea from March to May in 2012. The overall seropositive rates for AKAV and BEFV were found to be 49.8% and 1.2%, respectively. The regional distribution of seroprevalence for AKAV ranged from 18.1% to 63.7%. Seroprevalences of AKAV were 63.7% in Miryang, 62.3% in Uljin, 50.7% in Namwon, and 18.1% in Yeongju. The seropositive rates for AKAV in southern part of Korea were higher than the annual average at the national level. On the other hand, seropositive rates of BEFV in four regions were from 0.3 to 3.1%. In detail, regional seroprevalences were 3.1% in Miryang, 2.0% in Uljin, and 1.7% in Yeongju, and 0.3% in Namwon. Even only one year after massive outbreaks, overall seropositive rates were very low, similar to the annual average at the nation level. This result indicates that many number of cattle infected with BEFV may be replaced by new born calf or cattle in farm may not be immunized with vaccines. To prevent another epidemic, a national wide warning should be issued and more aggressive control measure must be implied. Recent global warming phenomenon could lead to more vigorous activity of haematophagous vectors and it is possible that arboviral diseases such as AKAV and BEFV are increased. Therefore, continuous sero-monitoring and extensive vaccination combined with control of haematophagous vectors are important to effectively prevent and control diseases caused by AKAV and BEFV.

Analysis of foot-and-mouth disease virus structural protein antibody positive rates according to ages in cattle and pigs (소와 돼지의 연령별 구제역 백신 항체 양성률 분석)

  • The best way to prevent foot-and-mouth disease (FMD) constantly occurring in Korea will be vaccination. In this study, FMD vaccines were given to Korean native cattle (Hanwoo), dairy cattle, and pigs to investigate the antibody positive rate of FMD vaccine by age in year and month. Hanwoo, dairy cattle, and pigs showed antibody positive rates of 99.5%, 97.7%, and 95.9%, respectively. High antibody positive rates more than 95% were found in Hanwoo and dairy cattle. In particular, high antibody positive rates were found in Hanwoo and dairy cattle regardless of age. Pigs showed a relatively low antibody positive rate of 57.6% at 3 months of age and then constantly maintained a high antibody positive rate of above 95.0% after 4 months of age. As a result of this study, high antibody positive rates were found when regular FMD vaccination was given to newborn calves and piglets after FMD vaccination twice to them. Therefore, it is considered the most important to receive vaccination thoroughly according to vaccination plan in order to prevent FMD.

Pathogenicity of Salmonella gallinarum isolated from chickens in Korea (국내 분리주 Salmonella gallinarum의 닭에 대한 병원성)

  • Fowl typhoid(FT) caused by Salmonella gallinarum is an infectious, egg-transmitted disease and characterized by swollen bronze liver, greenish-yellow diarrhea and high mortality in growing and adult chickens. Since 1992 the outbreak of FT has been increased. Several problems have been occurred such as absence of appropriate vaccines and lack of useful therapeutic methods. In these studies we investigated the pathogenicity of S gallinarum isolated in chickens. To compare the pathogenicity among the species of chickens, all chickens were challenged intramuscularly or orally with $1{\times}10^7$ CFU of S gallinarum. The brown-colored layers were more susceptible and white leghone chickens were more resistant than other species. In the brown layer chickens orally challenged, lethal doses ($LD_{50}$) of the isolates were inoculated at 1 day, 2 weeks, 4 weeks and 8 weeks old chickens with amount of $10^{4.2}$, $10^{4.7}$, $10^{7.0}$ and $10^{7.6}$ CFU, respectively. The chickens which were intramuscularly challenged with the less amount than $10^2$ CFU showed higher mortality than that of the chickens orally inoculated with same dose. Also, we investigated the recovery rates of bacteria from various organs of survival chickens which were challenged orally with $5{\times}l0^7$ CFU of S gallinarum. The bacteria was more frequently and isolated earlier from the liver and spleen than from any other ogans. In the pathogenicity test, the white-leghorn chickens which were known as resistant-strain against Salmonella were artificially immunosuppressed using bursectomy and/or dexamethasone treatment. Mortality of chickens with both bursectomized and treated with dexamethasone was higher(90%) than that of the control group(10%), the bursectomized chickens(10%) and the dexamethasone only treated group(20%). It was suggested that the protective mechanism in chickens against S gallinarum may be required both the functions of B-cells and T-cells.
