Endocrine Disruption Induced by Some Sulfa Drugs and Tetracyclines on Oryzias latipes

  • There has been increasing concern in recent years about the occurrence, fate and toxicity of pharmaceutical products in aquatic environment. Although these compounds have been detected in a wide variety of environmental samples including sewage effluent, surface waters, ground water and drinking water, their concentrations generally range from the low ppt to ppb levels. It is therefore often thought to be unlikely that pharmaceuticals will have a detrimental effect on the environment. This study was conducted to determine the endocrine disruption effects of the several pharmaceutical residues in water using adult Japanese medaka (Oryzias latipes). The common antibiotics were used sulfa durgs (sulfamethoxazole and sulfamethazine) and tetracycline drugs (oxytetracycline and tetracycline). Positive control that was induced Vtg (vitellogenin) in male fish was used $17\beta$-estradiol. Vtg was qualified and quantified through Western blotting and ELISA. After SDS gel electrophoresis, the dominant protein band was identified to molecular weight approximately 205 kDa in whole body samples of vitellogenic female. In female medaka exposed to $17\beta$estradiol, there was no significant difference in total protein induction. In contrast, three to five day exposure of male fish to $17\beta$-estradiol resulted in more than 60.0% increase of total protein compared to that of control males (p<0.01). In case of antibiotics, female fish didn't show significant difference, but male fish was showed significant difference. In addition, Vtg induction in male fish was observed with all the test chemicals. On concentrations greater than 0.1 ppm of sulfamethoxazole, 1 ppm of sulfamethazine, 1 ppm of oxytetracycline and 20 ppm of tetracycline, Vtg induction was increased in a dose response manner. This study is one of the early reports suggesting potential endocrine disruption mechanism of antibiotic pharmaceutical products in aquatic ecosystem. Although the effect concentrations obtained from this study were high as unrealistically as in environments, it is endocrine disruption that we should be considered as one of the important consequences of pharmaceutical contamination at water environment, and warrants due attention in future researches.

Aluminium and Cadmium Interfere with the Estrogen Receptor Level in the Primary Culture of Hepatocytes in the Rainbow Trout Oncorhynchus mykiss

  • Al and Cd-induced inhibition of vitellogenin (VTG) production was examined at the estrogen receptor (ER) level in rainbow trout Oncorhynchus mykiss hepatocytes. The binding of $[^3H]$ $estradiol-17\beta\;(E_2)$ to hepatocytes reached a plateau 3 days after addition of $E_2\;(2\times\;10^{-6} M)$to the medium. The binding activity was linearly reduced with the increased concentrations $(-10^{-5}\;M)$ of 4-hydroxy-tamoxifen (4-OHT) and specific binding linearly increased with the increased doses of $[^3H]\;E_2$, indicating that the radioligand bound to ER. Al $(-10^{-4}\;M)$and Cd $(10^{-6}\;M)$ as well as 4-OHT $(10^{-6}\;M)$ significantly reduced the $[^3H]\;E_2$-binding activity by $30­40\%$, while they completely inhibited VTG production. Al and Cd had no effect on $E_2-human$ $ER\alpha$ binding activity at any concentrations used $(-10^5\;nM\;each)$. These results suggested that Al and Cd inhibited VTG production in part by interfereing with the ER level. Inhibitory effects of these metals on the $E_z-dependent$ upregulation of ER activity are also discussed.

Profiles of Plasma Sex Steroid Hormone and Vitellogenin According to Ovarian Development of the Oblong Rockfish Sebastes oblongus (황점볼락 난소 발달에 따른 혈중 성호르몬과 난황단백전구체의 변동)

  • To understand the steroidogenic activities and plasma vitellogenin (VTG) profiles according to the reproductive phases in the oblong rockfish Sebastes oblongus, we examined changes in sex steroid hormones and plasma vitellogenin. Plasma levels of testosterone (T) was significantly higher value in only ovulation stage (P<0.05). In vitellogenesis, plasma estradiol-$17{\beta}$ ($E_2$) had a high level in August which was a similar higher level until ovulation than other ovarian development stages (P<0.05). However, $E_2$ was significantly decreased after embryo stage (P<0.05). This indicates that variability in $E_2$ at different stage is associated with the development of the oocytes. Plasma levels of $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) were significantly high at the stages of vitellogenesis and ovulation (P<0.001). It is assumed that DHP plays an important role in vitellogenesis. Also, We determined the plasma levels of vitellogenin (VTG) divided the development stage into four steps: immaturation, vitellogenesis, and ovulation and parturition. A significant lower levels of VTG were shown in immaturation and parturition (P<0.05), which did not discriminate between them. However, in vitellogenesis and ovulation were shown in a remarkable higher levels of VTG(P<0.05), but not significantly different between them. Consequently, plasma VTG levels were considerably increased after October and maintained a higher concentration until ovulation, but significantly decreased after ovulation. It is suggested that VTG plays also an important role in the development of vitellogenesis and oogenesis.

Effects of 4-Nonlyphenol Exposure on $P.$ $olivaceus$ and $S.$ $schlegeli$ Vitellogenesis

  • The effects of the estrogenic compound 4-nonylphenol (4-NP) on vitellogenesis in juvenile olive flounder ($Paralichthys$ $olivaceus$) and rockfish ($Sebastes$ $schlegeli$) exposed continuously at 10, 50 and $100{\mu}g{\ell}^{-1}$ levels for 7 days were compared. The expression of VTG mRNA level and protein using specific probes were examined. The levels of plasma estradiol-17 ${\beta}(E_2)$ and testosterone (T) were assessed by radioimmunoassay (RIA). Plasma $E_2$ concentrations increased significantly in two female fish species exposed to $100{\mu}g{\ell}^{-1}$ of 4-NP over concentrations in control fish. Plasma T concentrations increased in $P.$ $olivaceus$. Four days after exposure, the level of VTG mRNA expression increased in $P.$ $olivaceus$ and $S.$ $schlegeli$ exposed to $20{\mu}g{\ell}^{-1}$ of 4-NP. In addition, plasma VTG protein expression was seen in $P.$ $olivaceus$ and $S.$ $schlegeli$. In $S.$ $schlegeli$ and $P.$ $olivaceus$ exposed to 4-NP, the changes were noticed mainly in hepatocytic vaculation after 7 days of exposure. Thus, 4-NP may disrupt vitellogenesis in immature fish both directly and indirectly via disrupted steroidogenesis and liver pathology. Immature $S.$ $schlegeli$ were the most sensitive to 4-NP exposure in vitellogenesis.

Plasma Sex Steroid Hormone and Vitellogenin Profiles during Ovarian Development of the Wild Marbled Sole (Limanda yokohamae) (자연산 문치가자미 (Limanda yokohamae)의 난소 발달에 따른 혈중 성호르몬과 난황단백전구체 농도 변동)

  • This study correlated changes in estradiol-l7$\beta$ ($E_2$), testosterone (T), 17$\alpha$,20-dihydroxy-4-pregnen-3-one (DHP), and vitellogenin (VTG) levels with changes in the gonadosomatic index (GSI) and ovarian histology during the annual reproductive cycle of the wild marbled sole, Limanda yokohamae. Synchronous oocyte development occurs in this fish. Ovary maturity was classified into four periods, based on histological observations: the spawning (December to February), post-spawning (February to April), recovery (May to August), and vitellogenic (September to November) periods. Seasonal changes in the GSI were inversely correlated with water temperatures and reflected the degree of ovarian maturity. Plasma VTG levels were correlated with changes in the GSI, which increased from September to a peak in January, and levels remained comparatively high until February. Estradiol-17$\beta$ was at baseline levels (<0.11 ng/mL) during the spring and summer, and peaked rapidly (1.55$\pm$0.445 ng/mL) from October to January. Plasma T and DHP levels had a similar profile; they rose markedly during the spawning period and remained low (or were not detectable) from spring through autumn. These data indicate that changes in plsama steroid hormones and VTG levels are correlated with the annual ovarian activity of the marbled sole. Based on these results and published reports, it appears that in this species DHP is the most important maturation-inducing steroid and that T is also related to final maturation.

Effect of 17β-estradiol on Ecdysteroid Pathway Related Genes in the Brackish Water Flea Diaphanosoma celebensis (17β-estradiol이 기수산 물벼룩의 Ecdysteroid 경로에 미치는 영향)

  • 17β-estradiol (E2) is a natural hormone secreted by ovary, and continuously discharged from household and livestock wastewater into aquatic environment. Due to its strong estrogenic activity, it has adverse effects on development and reproduction in crustacean as an endocrine disrupting chemical. Although ecdysteroid signaling pathway play a key role in development in crustacean, little information on transcriptional modulation of ecdysteroid-related genes in response to E2 is available in small crustacean. Here, we investigated the acute toxicity of E2 to obtain 24-h LCx values in the brackish water flea Diaphanosoma celebensis. Time-dependent expression patterns of seven ecdysteroid pathway - related genes (CYP314a1, EcRA, EcRB, USP, ERR, Vtg, VtgR) were further examined using quantitative real time reverse transcriptase polymerase chain reaction (qRT-PCR). As results, 24-h LC50 and LC10 values were 9.581 mg/l and 4.842 mg/l, respectively. The mRNA expression of CYP314a1, EcRA, USP, VtgR was significantly up-regulated at 12 or 24 h after exposure to E2. These findings indicate that E2 can affect their molting and reproduction by modulating the expression of ecdysteroid pathway - related in D. celebensis. This study will be useful for better understanding of molecular mode of action of endocrine disrupting chemicals on molting process in small crustacean.

The Expression of Leptin, Estrogen Receptors, and Vitellogenin mRNAs in Migrating Female Chum Salmon, Oncorhynchus keta: The Effects of Hypo-osmotic Environmental Changes

  • Leptin plays an important role in energy homeostasis and reproductive function in fish, especially in reproduction. Migrating fish, such as salmonoids, are affected by external environmental factors, and salinity changes are a particularly important influence on spawning migrations. The aim of this study was to test whether changes in salinity affect the expression of leptin, estrogen receptors (ERs), and vitellogenin (VTG) in chum salmon (Oncorhynchus keta). The expression and activity of leptin, the expression of ERs and VTG, and the levels of estradiol-$17{\beta}$ and cortisol increased after the fish were transferred to FW, demonstrating that changes in salinity stimulate the HPG axis in migrating female chum salmon. These findings reveal details about the role of elevated leptin levels and sex steroid hormones in stimulating sexual maturation and reproduction in response to salinity changes in chum salmon.

Real time-PCR을 이용한 내분비계장애 생체지표의 발현감수성 비교

  • 에스트로젠을 처리한 송사리의 간으로부터 choriogenin vitellogenin estrogen receptor의 발현량을 전사수준에서 Real-time을 사용하여 정량.비교하였다. 시험어종으로는 부화 후 5개월 이상된 성숙한 수컷 송사리(Oryzias latipes)를(체중 약 250mg/마리)를 사용하여 17$\beta$-estradiol(25ppt, 50ppt, 100ppt)에 24시간 노출시켰다. Fluorescence dye는 choriogenin vitellogenin estrogen receptor의 경우 FAM (6-carboxyfluorescein)을 사용하였으며, $\beta$-actin의 경우는 VIC를 사용하였다. 프로브에 사용하는 quencher dye는 TAMRA(6-carboxy-N',N',N',N'-tetramethyl rhodamine)을 사용하였다. Internal control로 사용된 $\beta$-actin은 17$\beta$-estradiol의 농도에 상관 없이 0~10pM 범위에서 일정하게 발현됨을 보여주었다. vitellogenin choriogenin L 및 choriogenin H는 17$\beta$-estradiol의 농도에 의존하여 발현이 증가되는 용량-반응양상(Dose-dependent)을 나타내었다. 반면, estrogen receptor는 모든 처리군에서 $10^{-2}$pM 정도로 발혐됨에 따라 본 시험농도의 17$\beta$-estradiol에 의해서는 거의 유도발현이 되지 않음을 보여주었다. choriogenin L, choriogenin H, vitellogenin I 및 estrogen receptor 발현감수성을 비교한 결과, 25ppt 및 50ppt의 17$\beta$-esoadiol 농도에서는 ChgL > ChgH > VTG I >ER의 순으로 감수성이 높았으며, 100ppt 노출에서는 ChgL > VTG I > Chg H > ER의 순으로 감수성이 높게 나타났다. 결론적으로 choriogenin이 에스트로젠물질에 의한 가장 민감한 생체지표임을 알 수 있었다.

The Endocrine Disruption Induced by Ampicillin and Amoxicillin in Japanese Medaka (Oryzias latipes)

  • The study was designed to determine the estrogenic effect of some penicillins on endocrine function in adult Japanese medaka (Oryzias latipes). Vitellogenin (Vtg) produced in male fish has been used for a biomarker to study endocrine disrupters. $17\beta-estradiol\;(E_2)$ was used a positive control that was induced Vtg in male fish. Result of total protein qantification and ELISA for female and male fish were exposed to $17\beta-estradiol$ 10ng/ml for $3\sim5$ days. As a result, male fish exposed to amoxicillin respectively appeared 0.75, 0.23, 8.21 and $9.36\%_{\circ}$ of 1, 10, 100 and 1000 ppm respectively, that value was elevated compared with control male fish. Male fish exposed to ampicillin respectively appeared 1.85, 4.68, 0.85 and $39.59\%_{\circ}$ of 1, 10, 100 and 1000 ppm respectively, that value was elevated compared with control male fish. This study is one of the first reports suggesting potential endocrine disruption of some penicillins in aquatic ecosystem. These results suggest that vitellogenin and estrogen receptor induction patterns alter in male medaka treated with selected estrogenic compounds, and that these results may be useful molecular biomarkers for screening estrogenic EDCs (endocrine-disrupting chemicals) in the shortest possible time.
