레티놀 결합 단백질(retinol-binding protein, RBP)은 고등 척추동물에서 혈류를 통해 특이적으로 레티놀을 표적세포에 운반해 주는 중요한 역할을 한다. 우리나라의 연안에 서식하고 있으며 산업적으로 중요한 조피볼락(Sebastes schlegeli)을 대상으로 4-nonylphenol(NP)가 RBP mRNA 발현에 미치는 영향을 구명하기 위해 간으로부터 cDNA library를 제작하고 RBP 단편의 염기서열을 분석하였다. 분석된 RBP mRNA 서열로부터 아미노산 서열을 추정하여 다른 종의 RBP 아미노산 서열과 비교한 결과 Sparus aurata와는 80%, 무지개 송어(Oncorhynchus mykiss)와는 72%, 유럽 뱀장어(Anguilla anguilla)와는 78%의 높은 상동성을 보였다. 조피볼락에서 4-NP에 대한 RBP와 VTG mRNAs 발현에 미치는 영향을 northern blot 분석 방법에 의해 조사하였다. 4-NP를 10 mg/kg BW로 주사한 실험구에서는 암수 모두 VTG mRNA 발현에 영향을 주지 않았으나, RBP mRNA 발현은 수컷에서 48시간 후에 감소하였다. 4-NP를 25 mg/kg BW으로 주사한 실험구에서는 24시간 후에 암수 모두 VTG mRNA 발현이 증가하였으며, RBP mRNA 발현은 48시간 후에 암수 모두 감소하였다. 이들 결과로부터 조피볼락에서는 4-NP 등의 에스트로겐 유사물질이 RBP와 VTG 유전자 발현에 상반되는 효과를 유도함을 알 수 있었다.
비스페놀A(BPA)는 대표적인 내분비계 교란물질로 광범위한 사용으로 인해 환경 내에서 지속적으로 검출됨에 따라 인간을 비롯한 다양한 생물에서 성장, 발생, 생식 등에 유해한 영향을 미치는 것으로 알려져 있다. 따라서 BPA를 대체하기 위한 구조 유사체들이 개발되어 널리 사용되고 있으나 이러한 대체제들이 내분비계 교란 작용을 갖는지에 대한 연구가 필요하다. 본 연구에서는 BPA와 그 구조 유사체인 BPS와 BPF에 노출시킨 기수산 물벼룩 Diaphanosoma celebensis에서 탈피과정에 관여하는 ecdysteroid 합성(nvd, cyp314a1), receptors (EcRA, EcRB, USP, ERR), 그리고 하위 경로에 있는 유전자(HR3, E75, Vtg, VtgR)의 시간 별 발현 변화를 조사하였다. nvd와 cyp314a1 유전자의 발현은 BPA 보다 BPF에서 6시간 일찍 발현이 증가하는 양상을 보인 반면, BPS의 경우에는 이들 유전자의 발현이 24시간 내내 감소하는 양상을 보였다. BPA와 BPF 노출 시 EcR 유전자들의 발현 양상도 이와 유사한 경향을 보였다. ERR 유전자의 발현은 BPF와 BPS에서 BPA 보다 6시간 일찍 발현이 증가하는 양상을 보였고, HR3, E75, VtgR의 유전자 발현도 노출군에서 시간 차이는 있지만 유의하게 증가하는 양상을 보였다. 반면 Vtg는 24시간 이내에서는 크게 증가하지는 않았다. 이러한 결과는 BPA 뿐 아니라 BPF와 BPS도 탈피에 관여하는 호르몬의 합성 및 조절 경로의 유전자의 발현을 조절할 수 있으며, 서로 다른 기전으로 기수산 물벼룩의 내분비계를 교란시킬 수 있는 능력을 갖는다고 볼 수 있다. 본 연구는 비스페놀 구조 유사체가 기수산 물벼룩의 탈피과정에 관여하는 분자 경로 어떻게 영향을 미치는지를 이해하는데 도움이 될 것이다.
We tested biomarker systems [enzyme-linked immunosorbent assay (ELISA) and immunochromatography assay (ICG) kits] for the screening of endocrine-disrupting chemicals in contaminated environments using antibodies resulting from $17{\beta}$-Estradiol-induced vitellogenin (Vtg) in the wild scorpion fish Sebastiscus marmoratus. Monoclonal antibodies of two clones (S28 and S15) were used as capture and tracer antibodies for ELISA and ICG assays. ELISA detected Vtg at levels greater than $0.1{\mu}g/mL$, while ICG detected Vtg at levels greater than $1{\mu}g/mL$. However, the ICG system was able to detect antibodies from $17{\beta}$-Estradiol-induced Vtg serum that had been diluted 1,000 times. Our results suggest that previously developed biomarker assays can be used as detection systems to detect known endocrine-disrupting chemicals in contaminated environments, and to measure their activity.

Effect of extracellular calcium in vitellogenin (VTG) production in response to estradiol-17 $\beta$$(E_2,\;2\times10^{-6}M)$ was examined in primary hepatocyte culture of rainbow trout, Onchorhynchus mykiss. Total calcium in estrogenized sera significantly increased, compared with the control, while diffusible calcium was insignificant. However, diffusible calcium in the incubation medium with $E_2$ was significantly reduced, compared with the control. The uptake of extracellular calcium by cultured hepatocytes signifIcantly increased 90 min after $E_2$ addition. Moreover, the accumulation of intracellular calcium increased in the cultures with $E_2$, regardless of the calcium concentrations in the incubation media. In addition, $E_2-primed $ VTG production was significantly decreased by withdrawal of E_2$ from the incubation medium. Moreover, VTG production by $E_2-primed$ hepatocytes was reduced by removing calcium from the incubation medium with or without $E_2$. These results suggest that the entry of extracellular calcium into the cytoplasm is an important step for VTG production in primary hepatocyte cultures in rainbow trout.
Effects of aluminum (Al) on plasma vitellogenin (VTG), alkaline-labile phosphorus (ALPP), calcium (Ca), glutamate pyruvate transaminase (GPT), the hepatosomatic index (HSI), and hepatic Al concentration were examined in estradiol-$17{\beta}$ ($E_2$)-administered immature rockfish Sebastes schlegeli. Fish were injected intraperitoneally with $E_2$ (5 mg/kg body weight [BW]) and/or Al (0, 0.1, 1, 5, and 10 mg/kg BW) and plasma and liver samples were extracted 7 days later. After sodium dodecyl sulfate polyacrylamide gel electrophoresis, the relative amount of VTG was determined by integrated optical density. VTG accounted for 23.6% of the total proteins in the control group, but this value decreased with increasing Al administration. Al reduced the concentrations of ALPP and Ca in a concentration-dependent manner and significant reduction occurred at Al concentrations greater than 5 mg/kg. The concentration of GPT increased in a concentration-dependent manner in all Al-administered rockfish. The concentrations of Al in the liver also increased, and HSI was decreased, in a concentration-dependent manner. These results suggest that Al inhibits $E_2$-induced VTG production by being toxic to hepatocytes in marine fish.
무지개송어의 초대배양 간세포에 있어서 Vitellogenin (VTG)의 합성 유도는 전기영동적 수법에 의해 행하였다. 간세포는 7일 동안 phositively charged dish를 이용한 배양으로 단층 확산되었다. 배양 7일의 간세포 생존율은 $E_2$의 유무에 따라 각각 20.7% 및 23.6%가 감소하였으며, DNA함유량도 각각 13.7% 및 14.0%가 감소하였다. $E_2$ 첨가군과 대조군간에는 생존율과 DNA 함유량에 있어서는 유의한 차이가 나타나지 않았다. 그리고, 총 단백질에 대한 VTG의 비율은 $E_2$$10^{-6}$ M의 첨가시에 최대치를 나타내었다. 그러나, 고농도($10^{-5}$M)에서는 오히려 감소하는 경향을 나타내었다.
Effects of cyclic (c) AMP and G-protein related reagents (3-isobutyl-l-methyxanthine (IBMX), Forskolin (FSK), cholera toxin (CTX), and pertussis toxin (PTX≫ on estradiol-17$\beta$ induced vitellogenin (VTG) induction were examined in primary hepatocyte cultures in rainbow trout Oncorhynchus mykiss. The addition of IBMX, FSK, or CTX to the incubation medium markedly increased VTG production, while PTX was not effective in stimulating the production. It is well known that cAMP regulates phosphorylation and dephosphorylation through mediation of protein kinase A. These results suggest that VTG production is highly dependent on cAMP state in hepatocytes because of its highly phosphorylated nature.

This study was conducted to determine the endorcrine disruption effects of the several major pharmaceutical residues in water using adult Japanese medaka (Oryzias latipes). Four frequently used pharmaceuticals including caffeine, ketoconazole, acetaminophen, and diltiazem were investigated for the vitellogenin(Vtg) induction in the medaka using Western blotting and ELISA. $17\beta$,-estradiol was used as a positive control. Vtg was qualified and quantified through Western blot and ELISA. Following SDS gel electrophoresis, the dominant protein band was identified to molecular weight approximately 205 kDa in whole body samples of vitellogenic female. With female medaka exposed to $17\beta,-estradiol$, no significant difference in total protein induction was noted. In contrast, three to five day exposure of male fish to $17\beta,-estradiol$ resulted in $63.07\%o$, increase of total protein comparing to that of control males (p<0.01). Vtg induction in male fish was observed with all the test pharmaceuticals: At concentrations greater than 1ppm of diltiazem, 2 ppm of caffeine, 4 ppm of acetaminophen, and 10 ppm of ketoconazole, Vtg induction was monotonously increased in a dose dependent manner. This study is one of the first reports suggesting potential endocrine disruption mechanism of common human pharmaceutical products in aquatic ecosystem. Although the effect concentrations obtained from this investigation are environmentally unrealistically high, endocrine disruption should be considered as one of the important consequences of pharmaceutical pollution in aquatic environment, and warrants due attention in future researches.
