Image Evaluation and Exposure Dose with the Application of Tube Voltage and Adaptive Statistical Iterative Reconstruction of Low Dose Computed Tomography (저 선량 전산화단층촬영의 관전압과 적응식 통계적 반복 재구성법 적용에 따른 영상평가 및 피폭선량)

  • The study has attempted to evaluate and compare the image evaluation and exposure dose by respectively applying filter back projection (FBP), the existing test method, and adaptive statistical iterative reconstruction (ASIR) with different values of tube voltage during the low dose computed tomography (LDCT). With the image reconstruction method as basis, chest phantom was utilized with the FBP and ASIR set at 10%, 20% respectively, and the change of tube voltage (100 kVp, 120 kVp). For image evaluation, back ground noise, signal-noise ratio (SNR) and contrast-noise ratio (CNR) were measured, and, for dose assessment, CTDIvol and DLP were measured respectively. In terms of image evaluation, there was significant difference in ascending aorta (AA) SNR and inpraspinatus muscle (IM) SNR with the different amount of tube voltage (p < 0.05). In terms of CTDIvol, the measured values with the same tube voltage of 120 kVp were 2.6 mGy with no-ASIR and 2.17 mGy with 20%-ASIR respectively, decreased by 0.43 mGy, and the values with 100 kVp were 1.61 mGy with no-ASIR and 1.34 mGy with 20%-ASIR, decreased by 0.27 mGy. In terms of DLP, the measured values with 120 kVp were $103.21mGy{\cdot}cm$ with no-ASIR and $85.94mGy{\cdot}cm$ with 20%-ASIR, decreased by $17.27mGy{\cdot}cm$ (about 16.7%), and the values with 100 kVp were $63.84mGy{\cdot}cm$ with no-ASIR and $53.25mGy{\cdot}cm$ with 20%-ASIR, a decrease by $10.62mGy{\cdot}cm$ (about 16.7%). At lower tube voltage, the rate of dose significantly decreased, but the negative effects on image evaluation was shown due to the increase of noise.

Evaluation on Usefulness of BMI Application to Urological CT Examination (비뇨기계 CT 검사 시 체질량 지수 적용의 유용성 평가)

  • The purpose of this study was to evaluate the usefulness of BMI application to Urography CT by applying different tube voltages in accordance with body mass index. Group A (n = 38) with body mass index of lower than 25 was examined with tube voltage of 100 kVp while Group B (n = 45) with a BMI of 25 and higher was examined with tube voltage of 120 kVp. C group (n = 37) with body mass index (BMI) of lower than 25 was examined with tube voltage of 120kVp. Although the difference in average dose between group A (100 kVp) and group C (120 kVp) with low body mass index (BMI) of lower than 25 was $214.8mGy{\cdot}cm$, there was no significant difference in qualitative evaluation and, compared with patient group with body mass index of 25 and higher, results obtained were rather good. Therefore, this study verified that the tube voltage of lower than 100 kVp does not have adverse effect on the quality of image for patients with body mass index (BMI) of lower than 25.

cDNA Cloning and Expression of Human Rotavirus Outer Capsid Protein VP7 in Insect Cells

  • Rotavirus is a major cause of severe gastroenteritis in young children and animals throughout the world. The VP7 of rotavirus is thought to induce the synthesis of neutralizing antibodies and to be responsible for determining viral serotypes. The cDNA coding for the VP7 capsid protein of human rotavirus, obtained from Korean patients (HRV-Y14), was cloned and its nucleotide sequence was determined. Comparative analysis of the nucleotide sequences between VP7 of Y14 and that of other foreign isolates showed $92.7~95.2\%$ homology to G1 serotypes (RV-4, KU, K8, WA), $74.2\%$ homolgy to G2 serotype HU-5, $76.4\%$ homology to G3 serotype SA-11, and $77.6\%$ homology to G4 serotype A01321. These data suggest that HRV-Y14 can be classified as a G1 serotype. cDNA coding for VP7 of HRV-YI4 was subcloned into the baculovirus vector and the VP7 glycoprotein was expressed in insect cells. The expressed proteins in Sf9 cell extract and tissue culture fluid were separated on SDS-PAGE, and Western blot analysis with monoclonal antibody raised against the synthetic peptide containing 21 amino acids within the VP7 conserved region was performed. The molecular weight of recombinant VP7 was estimated to be 36 kDa which is about the same size as the native VP7. Addition of tunicamycin in the culture media caused a reduction of the molecular weight of the recombinant VP7 indicating that the expressed protein was glycosylated.

Vaccination of Shrimp (Penaeus chinensis) against White Spot Syndrome Virus (WSSV)

  • Two structural protein genes, VP19 and VP466, of white spot syndrome virus (WSSV) were cloned and expressed in Sf21 insect cells using a baculovirus expression system for the development of injection and oral feeding vaccines against WSSV for shrimps. The cumulative mortalities of the shrimps vaccinated by the injection of rVP19 and rVP466 at 15 days after the challenge with WSSV were 50.2% and 51.8%, respectively. For the vaccination by oral feeding of rVP19 and rVP466, the cumulative mortalities were 49.2% and 89.2%, respectively. These results show that protection against WSSV can be generated in the shrimp, using the viral structural protein as a protein vaccine.

  • We prepared polystyrene-b-poly(2-vinyl pyridine) (PSb-P2VP) lamellar films which is hydrophobic block-hydrophilic polyelectrolyte block polymer have 57 kg/mol-b-57 kg/mol. The result of UV-visible absorption spectra supported that effect of poly(ethylene oxide) on the band gap tuning of PS-P2VP photonic gel like salt effect.

Molecular diversity of the VP2 of Carnivore protoparvovirus 1 (CPV-2) of fecal samples from Bogotá

  • Background: Carnivore protoparvovirus 1, also known as canine parvovirus type 2 (CPV-2), is the main pathogen in hemorrhagic gastroenteritis in dogs, with a high mortality rate. Three subtypes (a, b, c) have been described based on VP2 residue 426, where 2a, 2b, and 2c have asparagine, aspartic acid, and glutamic acid, respectively. Objectives: This study examined the presence of CPV-2 variants in the fecal samples of dogs diagnosed with canine parvovirus in Bogotá. Methods: Fecal samples were collected from 54 puppies and young dogs (< 1 year) that tested positive for the CPV through rapid antigen test detection between 2014-2018. Molecular screening was developed for VP1 because primers 555 for VP2 do not amplify, it was necessary to design a primer set for VP2 amplification of 982 nt. All samples that were amplified were sequenced by Sanger. Phylogenetics and structural analysis was carried out, focusing on residue 426. Results: As a result 47 out of 54 samples tested positive for VP1 screening, and 34/47 samples tested positive for VP2 980 primers as subtype 2a (n = 30) or 2b (n = 4); subtype 2c was not detected. All VP2 sequences had the amino acid, T, at 440, and most Colombian sequences showed an S514A substitution, which in the structural modeling is located in an antigenic region, together with the 426 residue. Conclusions: The 2c variant was not detected, and these findings suggest that Colombian strains of CPV-2 might be under an antigenic drift.

Effects of Radiation Dose and Image Quality at the Coronary Angiography (관상동맥검사에서 선량과 화질에 관한 연구)

  • The aim of this study was to assess the effect of exposure factors such as kVp and mA applied by BMI on the image quality and patients absorbed dose of Coronary angiography in CT. Each data sets were into 4groups with different exposure values : Group A at 100kVp, 240mAs, Group B at 120kVp, 240mAs, Group C at 100kVp, 270mAs and Group D at 120kVp, 270mAs, and the mean of the scores of 4 groups was calculated for image quality as 4grades that is, 1(poor), 2(fair), 3(good) and 4(very good). Patient absorbed dose was calculated as DLP on the monitor. In case of absorbed dose, deviation in 2groups at 100kVp was 5.6 $mGy{\cdot}cm$, 11 $mGy{\cdot}cm$, was at 120kVp(DLP) with p<0.05. There was rather difference between groups with 100kVp or 120kVp respectively but the gaps were very little. No significant correlation was found between exposure factors and image quality in any images assessed(p>0.05), and the image quality was sufficient for diagnosis. As we applying coronary angiography, the selection of adequate exposure factors considering BMI identified might be effective for reduction of patient absorbed dose, improvement of image quality and diagnostic accuracy.

Characterization of Carotenoid Biosynthetic Pathway Using Viviparous Mutant Embryos in Maize ( Zea mays L. )

  • Carotenoid compounds in embryos of wild-type(WT) and viviparous mutants of maize(Zea mays L.) were analyzed using high performance liquid ehromatography (HPLC) with a photodiode array detector. Zeaxanthin accumulates in WT embryos as the major carotenoid. Phytoene accumulates in vp2 and vp5. Phytofluene in w3 and ${\xi}$-carotene in the vp9 mutant embryos. This indicates that the vp2 and vp5 mutants impair phytoene desaturase from 15-cis-phytoene to 15-cis-phytofluene. The w3 mutant has neither an isomerase from 15-cis-phytofluene to all-trans-phytofuene nor phytofluene desaturase from phytofluene to ${\xi}$-carotene. The vp9 mutant does not have the ${\xi}$-carotene desaturase from ${\xi}$-carotene to lycopene. Our analysis shows that the terminal carotenoid. ${\gamma}$-carotene(${\beta},{\Psi}$-carotene), accumulates in the vp7 mutant embryos. The ${\varepsilon}$-carotene(${\varepsilon},{\varepsilon}$-carotene), a product of ${\delta}$-carotene(${\varepsilon},{\Psi}$-carotene) in some plants, however, has not been found in maize embryos. The vp7 mutant impairs a cyclization step from ${\gamma}$-carotene to both ${\beta}$-carotene and ${\alpha}$-carotene. We suggest that monocyclic ${\gamma}$-carotene is the sole precursor of both bicyclic ${\beta}$-carotene(${\beta},{\beta}$-carotene) and ${\alpha}$-carotene(${\beta},{\varepsilon}$-carotene) in maize.

Dose and Image Evaluation according to Changes in Tube Voltage during Chest X-ray Examination according to Automatic Exposure Control (자동노출제어장치 유·무에 따른 흉부 후·전방향 검사 시 관전압 변화에 따른 선량 및 영상평가)

  • This study was conducted to improve the problems of exposure dose and image reading applied to patients due to the incorrect use of AEC during chest radiography. Images were acquired by dividing the case where AEC was used as the test condition and the case where AEC was not used. As a result of the study, the dose was reduced by 1.17% in 110 kVp without AEC than with AEC, 17.2% decrease at 100 kVp, 30.19% decrease at 90 kVp, and 46.45% decrease at 80 kVp. There was a significant difference in the statistical values according to the tube voltage change in the lung, trachea, and heart SNR average values with AEC and without AEC 110 kVp, but the difference in image quality was insignificant in actual images. When AEC was not applied at the same tube voltage, the dose could be reduced by 17.2% while maintaining the image quality similar to that of with AEC at 100 kVp without AEC. Therefore, rather than relying on AE conditions during chest radiographic examination, it is considered that the conditions should be considered for the examination while lowering the dose by selecting an appropriate tube voltage.

Selective Permeate Transport Characteristics of Iodine ion at Cell Membrane Model of Thyroid which Irradiated by High Energy X-Ray (고에너지 엑스선을 조사한 갑상선의 세포막모델에서 요오드이온의 선택적 투과성 전달 특성)

  • The selective permeate transport characteristics of iodine ion at follicle cell membrane model in thyroid which irradiated by high energy x-ray(linac 6 MV) was investigated. The follicle cell membrane model used in this experiment was a polysulfonated copolymerized membrane of poly(4-vinylpyridine-co-acrylonitrile:VP-AN). The difference of membrane thickness [2 mole AN%(w/w)], fixed carrier concentration[VP-AN%(w/w)], OH- concentration were occurred at difference of I- concentration and quantity of thyroid hormone, respectively. The tensile strength in fixed carrier concentration[VP-AN% (w/w): 0-62 %] of irradiated membrane was found to be decreased about 1.2-1.8 times than non-irradiated membrane. The I- selective permeate initial flux with increase of membrane thickness [2mole AN%(w/w)], fixed carrier concentration[VP-AN%(w/ w)], OH- concentration in irradiated membrane were found to be decreased about 2.1-4.5 times, about 2.2-2.5 times, about 2.1-2.67 times than non-irradiated membrane, respectively. As a result, the quantity of thyroid hormone was decreased at irradiated membrane than non-irradiated membrane. The decrease of thyroid hormone was occurred at hypothyroidism and hyperthyroidism, thyroid cancer, and so on. As the thyroid hormone in cell membrane model were abnormal, cell damages were appeared at cell.
