무창 분만 ${\cdot}$ 자돈사 환기 형태가 돈사내 환경에 미치는 영향 (Effects of Ventilation Types on Interior Environment of the Enclosed Farrowing-Nursery Pig House)

  • This study was conducted to collect basic data about the effects of ventilation types on the interior environment of the enclosed farrowing-nursery pig house in Anseong, Icheon and Jeungpyong. Surveyed ventilation types in the enclosed farrowing-nursery pig house are classified in to 4 types. In V1 type, air enters through a planar slot inlet placed on the juncture of the entering wall and exit through the chimney fan outlet; in V2 type, air enters through a perforated ceiling inlet and exits chimney fan outlet(V2); in V3 type, air enters through a circular duct inlet and exit chimney fan outlet(V3); in V4 type, enters through a circular duct inlet and exits side wall exhaust fan outlet(V4). Temperature, relative humidity, air velocity and ammonia concentration($NH_3$) were measured in the interior of swine building in the summer. Interior temperature was not remarkably different in all ventilation types in this study. However, temperature of the V4 was somewhat lower than that of the other types. Air velocity of the V4 was higher and $NH_3$ concentration of the V4 was lower than those of other ventilation types. It is suggested that the V4 ventilation type be applicable in the enclosed farrowing-nursery pig house in Korea.

  • 감귤 과수원의 식물기생선충을 조사하던 중, 한국미기록 선충인 Paratrichodorus minor가 처음으로 제주도 감귤과수원 토양에서 발견되었다. Paratrichodorus minor는 크기가 $428.34{\sim}506.22{\mu}m$이고, a = $12.08{\sim}22.79$, b = $4.02{\sim}5.02$, V(%) = $38.46{\sim}60.68$이었으며, 배설공(excretory pore)이 후부식도부에 위치하는 특징을 가지고 있다. 또한, 1976년에 한국에서 보고된 P. porosus의 형태적 특징에 관하여 재기술 하였다.

Vibrio fluvialis 유래의 hemolysin 정제와 생화학적 특성 (Purification and characterization of biochemical properties of hemolysin from Vibrio fluvialis)

  • 병원성 미생물인 Vibrio fluvialis로부터 hemolysin을 분리정제 하였다. 즉, hemolysin의 정제는 균 배양액을 황산암모늄, ion exchange chromatography를 행하였으며 SDS-PAGE를 통해 그 단백질의 크기가 약 79kDa임을 알게되었다. 정제된 hemolysin (VFH)는 35$^{\circ}C$에서 최적활성을 나타내었고, 4$0^{\circ}C$에서는 그 활성이 감소되었다. RTG-2 어류유래 세포주에 hemolysin (VFH)의 처리로 세포독성을 측정한 결과 50$\mu\textrm{g}$의 VFH가 약 80%의 cell line을 사멸시켰다. 또한 현미경을 통한 관찰에서도 세포의 형태변화를 관찰할 수 있었다.

체외수정 유래 소 핵이식란의 배양에 관한 연구 (Studies on the culture of bovine nuclear transplant embryos derived in vitro fertilization)

  • The fusion rates of nuclear transplant embryos with various DC voltage were 55.6-79.2%. The significantly higher fusion rates of nuclear transplant embryos were achieved at the electric field strenght of DC 1.0-2.0kV/cm(72.0-79.2%) than DC 0.75kV/cm(55.6%, P<0.05). No significant differences in the percentage of embryos that cleaved(48.1, 55.4 and 42.6% respectively) and the percentage of cleaved embryos that developed to morulae/blastocyst(1.9, 5.3 and 1.9% respectively) could be found among the three types of in vitro culture system (Granulosa cell, BOEC co-culture and SOF, P>0.01). The age of the recipient cytoplast(30 vs 40hr) had no effect on the fusion rates and the rates of cleavage development(36.9 vs 44.1%, P>0.01).

상수리(상실(橡實))의 각종 용매 추출물이 linoleic acid의 항산화력에 미치는 영향 (Antioxidative Activity of Various Solvent Extracts of Quercisemen to Linoleic Acid)

  • The extracts of Quercisemen(Quereus acutissima Carruthers seed) powder was obtained by the extraction with distilled water, water : acetonet(1:1, v/v), ethanol and ethyl ether, respectively. As a result this experiment, the antioxidative activity of each solvent extracts on linoleic acid were examined as follows: 1. Each fraction extracted by the acetone : water(1:1, v/v), water and ethanol respectively showed high antioxidative activity. 2. A fraction of the tannin extracted from the solvent, acetone, water(1:1, v/v) showed even more antioxidative activity than that of the ${\alpha}$-tocopherol or BHT. 3. Acceleration of peroxide reaction by $Fe^{++}$ and $Cu^{++}$ on the linoleic acid was strongly inhibited by adding the tannin, 4. Organic acid, such as malic acid, citiric acid and tartaric acid with the tannin were showed the synergistic effect fo the antioxidation reaction.

시판 어패류에서 분리한 장염 비브리오균의 항생제 감수성 특성 (Antibiotic Susceptibility of Vibrio parahaemolyticus Isolated from Commercial Marine Products)

  • 2006년부터 2008년까지 3년간 시판 수산물 2,239건에서 분리한 V. parahaemolyticus에 대한 항생제 감수성 결과는 다음과 같았다. 어류 789건을 검사하여 42주(5.3%)의 V. parahaemolyticus가 분리 동정되었으며, 패류 735건 중 27건(3.7%), 연체류 387건 중 12건(3.1%), 기타 수산물 328건 중 12건(3.7%), 총 93건(4.2%)에서 V. parahaemolyticus가 분리 동정되었다. 내성률이 높은 항생제는 ampicillin 93.5%, cephalothin 90.3%, streptomycin 87.1%, ticarcillin 55.9%, 그리고 amikacin 40.9%이었으며, 감수성률이 높은 항생제는 nalidixic acid, chloramphenicol 및 ampicillin/sulbactam trimethoprim/sulfamethoxazole 98.9%, gentamicin 및 tetracycline 82.8%, ceftriaxone 63.4%이었다. 중등도 내성률은 kanamycin 79.6%, ciprofloxacin 64.5%, amikacin 47.3%, cefoxitin 43.0%순이었다. 항생제 내성양상은 3제 내성이 37.6%, 4제 내성 24.7%, 5제 내성 17.2%, 6제 내성 11.8%, 2제 내성6.5%, 7제 내성 2.2% 순이었으며, 가장 많은 내성 양상은 AM-CF-S의 3제 내성(22.6%), AM-CF-S-TIC 4제 내성(18.3%), AM-AN-CF-S-TIC 5제 내성(9.7%) 순이었다.

유색미 도입 유전자원의 생육 및 품질특성 변이 다변량 분석 (Multivariate Analysis of Variation of Growth and Quality Characteristics in Colored Rice Germplasm)

  • 우리나라에 도입된 유색미 유전자원 178점의 생육 및 품질특성 변이를 평가하고 주성분 분석을 통해 유전자원을 분류하여 벼 육종의 기초자료로 활용하고자 연구를 수행하였다. 1. 도입 유색미 유전자원 178점의 변이계수는 이삭길이, 단백질 함량이 가장 높았고, 장폭비, 천립중, 간장, 아밀로스 함량 순으로 높았고, 수량구성요소인 주당수수가 가장 낮았다. 2. 유색미 유전자원의 생육 및 품질특성에 대한 주성분 분석 결과, 각 주성분의 고유값과 기여율은 제1주성분 2.06개, 29.49%, 제2주성분 1.31개, 18.75%, 제3주성분 1.21개, 17.36% 및 제4주성분 1.01개, 14.38%로 4개의 주성분 고유값이 1 이상이며, 누적기여율이 79.98%로 도입 유색미 유전자원 평가가 가능하였다. 3. 군집분석 결과, I군집은 79자원이 포함되었으며, II, III, IV, V, VI, VII군집은 각각 46자원, 19자원, 13자원, 4자원, 8자원 및 9자원이 분포하였다. I군집, II군집에는 짙은갈색의 자원이 많이 분포되었고, V군집, VI군집, VII군집에는 자색 계열의 종피색을 가진 자원이 많았다. 특히, V군집은 암자색과 자색만으로 자색계열의 종피색을 가진 자원만이 포함되어 있다. VII군집에서 간장, 이삭길이, 장폭비가 다른 군집에 비해 평균값이 적었으며, V군집에서는 천립중 평균이 다른 군집에 비해 가장 낮았다. IV군집에서는 평균 주당수수 가장 낮았으나, 천립중 평균은 가장 높게 나타났다.

Betula Platyphylla var. Japonica Extract Prevent Ultraviolet C Light-induced Cell Damage in Chinese Hamster Fibroblast (V79-4) Cells

  • The present study reports the protective properties of a total methanol extract of B. platyphylla var. japonica against ultraviolet (UV)-C irradiation. Pretreatment of Chinese hamster fibroblast (V79-4) cells with a total methanol extract significantly increased cell survival following $300\;J/m^2$ of UV-C irradiation. The total methanol extract was further fractionated into 5 fractions: n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these fractions, B. platyphylla var. japonica ethylacetate, butanol and water fractions showed significant protective effects against the cellular damage induced by UV-C irradiation. In order to elucidate the mechanism underlying this protective effect, DPPH (Editor note: abbreviations should be spelled out at first use.) radical scavenging and lipid peroxidation inhibitory activity were measured. Significant radical scavenging and lipid peroxidation inhibitory activities were observed for the ethylacetate fraction. In summary, the present data demonstrate that an extract of B. platyphylla var. japonica has a significant protective effect against UV-C irradiation. The underlying mechanism of this protective effect may involve radical scavenging and inhibition of lipid peroxidation by the B. platyphylla var. japonica extract.

An Electrochemical Approach for Fabricating Organic Thin Film Photoelectrodes Consisting of Gold Nanoparticles and Polythiophene

  • A novel method of fabricating polythiophene-gold nanoparticle composite film electrodes for photoelectric conversion is demonstrated. The method includes electrodeposition of gold and electropolymerization of 2,2'-bithiophene onto an indium-tin-oxide (ITO) electrode. First, electrodeposition of gold onto the ITO electrode was carried out with various repetition times of pulsed applied potential (0.25 s at -2.0 V vs. Ag/AgCl) in an aqueous solution of $HAuCl_4$. Significant progress of the number density of deposited gold nanoparticles was confirmed from scanning electron micrographs, from 4 (1 time) to 25% (15 times). Next, electropolymerization of 2,2'-bithiophene onto the above ITO electrode was performed under controlled charge condition (+1.4 V vs. Ag wire, 15 $mC/cm^2$). Structural characterization of as-fabricated films were carried out by spectroscopic and electron micrographic methods. Photocurrent responses from the sample film electrodes were investigated in the presence of electron acceptors (methyl viologen and oxygen). Photocurrent intensities increased with increasing the density of deposited gold nanoparticles up to ~10%, and tended to decrease above it. It suggests that the surplus gold nanoparticles exhibit quenching effects rather than enhancement effects based on localized electric fields induced by surface plasmon resonance of the deposited gold nanoparticles.

Induction of Antioxidant Enzymes in Phloroglucinol Treated Cells

  • We investigated the cytoprotective effect of phloroglucinol, which was isolated from Ecklonia cava (brown seaweed), against oxidative stress induced cell damage in Chinese hamster lung fibroblast(V79-4) cells. Phloroglucinol was found to scavenge intracellular reactive oxygen species (ROS) generated by $\gamma-ray$ radiation. In addition, Phloroglucinol inhibited cell damage induced by radiation through scavenging ROS. Phloroglucinol increased the superoxide dismutase and glutathione peroxidase activity, Taken together, the results suggest that phloroglucinol protectes V79-4 cells against oxidative damage by enhancing the cellular antioxidant enzymes activity.
