Correlations Between Climate Change-Related Infectious Diseases and Meteorological Factors in Korea (국내 기후변화 관련 감염병과 기상요인간의 상관성)

  • Objectives: Infectious diseases are known to be affected by climate change. We investigated if the infectious diseases were related to meteorological factors in Korea. Methods: Scrub typhus, hemorrhagic fever with renal syndrome (HFRS), leptospirosis, malaria and Vibrio vulnificus sepsis among the National Notifiable Infectious Diseases were selected as the climate change-related infectious diseases. Temperature, relative humidity and precipitation were used as meteorological factors. The study period was from 2001 through 2008. We examined the seasonality of the diseases and those correlations with meteorological factors. We also analyzed the correlations between the incidences of the diseases during the outbreak periods and monthly meteorological factors in the hyper-endemic regions. Results: All of the investigated diseases showed strong seasonality; malaria and V. vulnificus sepsis were prevalent in summer and scrub typhus, HFRS and leptospirosis were prevalent in the autumn. There were significant correlations between the monthly numbers of cases and all the meteorological factors for malaria and V. vulnificus sepsis, but there were no correlation for the other diseases. However, the incidence of scrub typhus in hyper-endemic region during the outbreak period was positively correlated with temperature and humidity during the summer. The incidences of HFRS and leptospirosis had positive correlations with precipitation in November and temperature and humidity in February, respectively. V. vulnificus sepsis showed positive correlations with precipitation in April/May/July. Conclusions: In Korea, the incidences of the infectious diseases were correlated with meteorological factors, and this implies that the incidences could be influenced by climate change.

Identification of the Vibrios Isolated from a Shellfish, Sunset Shell, (Soletellina olivacea) (빛조개(Soletellina olivacea)로부터 분리된 비브리오의 생화학적 성상)

  • This study was conducted to investigate the vibrio flora in an edible shellfish. sunset shelfish. Soletelliim olivacen. which were collected in the estuarine area. Dadaepo near Nakdong River in Korea lkoin January 1997 to November 1997. Including five pathogemc vibrios (Vibrio alginolyticus, Vibrio pamhaemol~~licz~s, Vibrio cholerae non-01. Vibrio vulnificus, and Vihrio jl~~vinlis), a lotal of eight species of vlbr~os (Vi61-io splendidrrs biovar I, Vibrio splendidus biovar 11, Vibrio snlrnonicida and Vibrio tr,~biasllii) were identified from the sunset shellfish by heir biochemical characters. The isolation of Vihrio pamhaemolyricns, which is known not to grow below $15^{\circ}C$, in winter season indicates that the sunset shelllish is one oT the natural owl.- wintering hosts for Vibrio parahuemolyticus.

Antibacterial Effects of Natural Essential Oils from Ginger and Mustard against Vibrio Species Inoculated on Sliced Raw Flatfish

  • In order to extend the shelf life of sliced raw flatfish, the antimicrobial effects of natural essential oil from mustard and a mixture of ginger and mustard essential oils were tested at various temperatures. In addition, volatile components of the mixed essential oils were analyzed using gas chromatography and gas chromatography mass spectrometry. The viable cell counts of Vibrio parahaemolyticus treated with mixed essential oils from ginger and mustard was 0.7-1.3 log CFU/g lower than those of other treatments during storage at $5^{\circ}C$. During storage at $20^{\circ}C$, the viable cell counts of V. parahaemolyticus, V. vulnificus 01, and V. vulnificus 02 treated with the essential oils increased slightly from 6.53-6.64 log CFU/g at initial stages to 6.77-7.72 log CFU/g after 24-hr of storage, however they were 1.38-1.97 log CFU/g lower than those of the control group (8.74-9.10 log CFU/g). These results show that the growth of V. parahaemolyticus and V. vulnificus inoculated on sliced raw flatfish could be inhibited by treatment with natural essential oils from ginger and mustard at $5^{\circ}C$ of storage. However, the antibacterial effects of the essential oils on Vibrio species observed in this study were not sufficient to merit their use in sliced raw flatfish at temperatures exceeding $20^{\circ}C$.

Studies on the hemolysin produced by Vibrio Vulnificus ys-1 (Vibrio vulnificus ys-1이 생산하는 hemolysin에 관한 연구)

  • We isolated 100 Vobrio sp. from marine products and sea from July to September, 1997. We attemped on purification of hemolysin produced by Vibrio sp. The growth, hemolysin production patterns by the 100 strains of Vibrio sp. showed identical, in general. V. unlnificus ys-1 produced hemolysis as the higtest titer. The optimal culture conditions for the hemolysin production by the V. vunificus ys-1 are followings; 1. Hemolysin production was optimal dering the late exponetial phage. 2. Maximal growth, hemolysin production were in heart infusion broth. 3. Maximal yields of hemolysin was obtained when the heart infusion broth had an intial pH of 8.0, 3$0^{\circ}C$, 3% NaCL. Hemolysin was purified from culture filtrate of the strain by ammonium sulfate recipitation, ion exchange and hydrophobic interaction chromatography. The results were as follows; 1. Hemogeneity of the purified hemolysin was demonstrated by revealing single band on SDS-PAGE. The molecular weight of purified hemolysin was 45KDa. 2. The absorbance rattern in ultraviolet wsa typical of those seen with most proteinb with 280nm. 3. Purified hemolysin was atable at 5$0^{\circ}C$ but 7$0^{\circ}C$ of the acivity was lost by heating for 30 min at 6$0^{\circ}C$/ Optimal temperature of purified hemolysin was 35$^{\circ}C$. 4. Purified hemolysin was stable at the pH range of 6~9, but in the less the pH5.0. above the pH 9.0, the hemolysin activity was lost completely.

Characteristics of Vibrio vulnificus Isolated in Incheon (인천지역에서 분리된 비브리오 패혈증균의 특성)

  • We performed the biochemical characteristics, molecular epidemiologocal analysis, and drug susceptibility test on V. vulnificus isolated from environmental sources in Incheon. For this study, 233 strains were isolated from seawater, sediment, shellfish. V. vulnificus isolates were divided into 15 biochemical groups, which were positive for ONPG and Amygdalin test. Among the 209 strains, 206 (98.6%) strains and 110 (52.6%) strains revealed positive for vvhA and viuB gene, and the viuB gene detection rates of V. vulnificus from seawater, shellfish and sediment were 48%, 48.5% and 61.6%, respectively. From disc diffusion test on 175 isolates, most of strains were sensitive to Imipenem (100.0%), Sulfamethoxazole/trimethoprim (98.9%), Tetracycline, Ciprofloxacin (98.3%), Ampicillin/sulbactam (97.1%), Ohloramphenicol (96.6%), Cefepime (94.9%) and Ceftriaxone (94.8%), multi-drug resistance rates was 31.5% of seawater, 34.4% of sediment and 29.2% of shellfish. PFGE was performed on 233 V. vulnificus isolates with the objective of investigating the extent of genetic diversity of these isolates in our region. We could find that at least 126 different PFGE patterns were generated according by 90% of similarity and 13 clusters by 58% of similarity. The major cluster was type I (44.6%) during the most of the year, and type J was frequent pattern in June and October. There were 9 distinct PFGE types in July, 8 types in August, 7 types in June, 6 types in September, 5 types in October 3 types in May and 1 type in March.

The Virulence of Vibrio vulnificus is Affected by the Cellular Level of Superoxide Dismutase Activity

  • The virulence of superoxide dismutase (SOD) mutants of Vibrio vulnificus, as tested by intraperitoneal injection into mice, decreases in the order of sodC mutant, sodA mutant, and sodB mutant lacking CuZnSOD, MnSOD, and FeSOD, respectively. The survival of SOD mutants under superoxide stress also decreases in the same order. The virulence of soxR mutant, which is unable to induce MnSOD in response to superoxide, is similar to that of the sodA mutant, as the survival of the soxR mutant under superoxide stress is similar to that of the sodA mutant. Consistently, the lowered survival of the soxR mutant is complemented not only with soxR but also with sodA. Thus, the virulence of V. vulnificus is significantly affected by the cellular level of SOD activity, and an increase in SOD level through MnSOD induction by SoxR under superoxide stress is essential for virulence.

Distribution and Antibiotic Resistance of Vibrio vulnificus isolated in Incheon Coastal Area (인천지역 해양환경에서 분리된 비브리오패혈증군의 분포 및 항생제 내성 양성)

  • We investigated the distribution and drug susceptibility test of V. vulnificus isolated from environmental sources in Incheon. In this survey, total 4,302 samples were obtained from different sites of the Incheon coastal area during the periods from march 2004 to November 2006. Among the 4,302 samples, 310 strains were isolated. The isolation rates of Vibrio vulnificus from fish, shellfish, estuarine water and sediment were 6.7%, 4.7%, 12.4% and 23.2%, respectively. The highest isolation rate was 14.6% in September and in spite of low temperature the isolation rate was 5.1% in November. The highest rate of antibiotic resistance was observed against streptomycin(15.3%), cefazolin(8.5%), cephalothin(8.3%), amikacin(8.3%), cefoxitin(6.7%) and nalidixic acid(6.7%). Seventeen percent of isolates were observed to be resistant to two or more of the antibiotics tested.

Regulatory Characteristics of the Vibrio vulnificus putAP Operon Encoding Proline Dehydrogenase and Proline Permease

  • The proline utilization (put) operon of Vibrio vulnificus consists of the putAP genes encoding a proline dehydrogenase and proline permease. The result of put-lux transcriptional fusion analysis suggests that the V vulnificus putAP operon is not autoregulated by the PutA protein. A putR null mutation decreased proline dehydrogenase activity and the level of the put transcripts, indicating that transcription of putAP is under the positive control of PutR. The deduced amino acid sequence of the putR was similar to those reported from other bacteria with high levels of identity. Chromatin IP and GST pull-down assays revealed that PutR specifically binds to the putAP promoter region in vivo, and interacts with CRP in vitro. Taken together, the results suggested that PutR exerts its effect on putAP expression by directly interacting with CRP bound to the upstream region of P$_{put}$.

Development of piezoelectric immunosensor for the rapid detection of marine derived pathogenic bacteria, Vibrio vulnificus

  • Biosensors consist of biochemical recognition agents like antibodies immobilized on the surfaces of transducers that change the recognition into a measurable electronic signal. Here we report a piezoelectric immunosensor made to detect Vibrio vulnificus. A 9MHz AT-cut piezoelectric wafer attached with two gold electrodes of 5mm diameter was used as the transducer of the QCM biosensor with a reproducibility of ${\pm}0.1Hz$ in frequency response. We have tried different approaches to immobilize antibody on the sensor chip. Concerning the orientation of antibody for the best antigen binding capacity, the antibody was immobilized by specific binding to protein G or by cross-linking through hydrazine. In addition, protein G was cross-linked on glutaraldehyde activated immine layer (PEI) or EDC/NHS activated sulfide monolayer (MPA). PEI was found to be more effective to immobilize protein G following glutaraldehyde activation than MPA. However, hydrazine chip showed a better capability to immobilize more IgG than protein G chip and a higher sensitivity. The sensor system was able to detect V. vulnificus in dose dependent manner and was able to detect bacterial cells within 5 minutes by monitoring frequency shifts in real time. The detection limit can be improved by preincubation to enrich the bacterial cell number.
