Prediction of Rice Embryo Proteins using EST-Databases

  • An attempt was made to link rice embryo proteins to DNA sequences and to understand their functions. One hundred of the 700 spots detected on the embryo 2-DE gels were microsequenced. Of these, 28% of the embryo proteins were matched to DNA sequences with known functions, but 72% of the proteins were unknown in functions as previously reported (Woo et al. 2002). In addition, twenty-four protein spots with 100% of homology and nine with over 80% were matched to ESTs (expressed sequence tags) after expanding the amino acid sequences of the protein spots by Database searches using the available rice EST databases at the NCBI (http://www/ncbi.nlm.nih.gov/) and DDBJ (http://www.ddbj.nig.ac.jp/). The chromosomal location of some proteins were also obtained from the rice genetic map provided by Japanese Rice Genome Research Program (http://rgp.dna.affrc.go.jp). The DNA sequence databases including EST have been reported for rice (Oryza sativa L.) now provides whole or partial gene sequence, and recent advances in protein characterization allow the linking proteins to DNA sequences in the functional analysis. This work shows that proteome analysis could be a useful tool strategy to link sequence information and to functional genomics.

Prediction of Slope Failure Using Control Chart Method (통계관리도 기법을 적용한 사면붕괴 예측)

  • In this study, a field model experiment was performed to analyze the bahavior of slope during failure. It was analyzed through x-MR control chart method with inverse displacement and K-value. As a result, the portent was confirmed at 4 minutes before slope failure in Case 1. The change of the control limit line according to moving range was analyzed and it was effective to apply K = 3. Use of the inverse displacement and x-MR control chart method will be useful for the prediction of abnormal behavior through quick and objective judgment. Prediction of slope failure using control chart method can be used as basic data of slope measurement management standard, and it can contribute in reduction of life and property damage caused by slope disaster.

  • In this study, a prognostic technique for cavitation erosion that is applicable to centrifugal pumps is devised. To estimate the erosion states of pumps, damage rates are calculated based on cavitation noise measurements. The accumulated damage is predicted by using Miner's rule and the estimated damage undergone when coping with particular operating conditions. The remaining useful life (RUL) of the pump impellers is estimated according to the accumulated damage prediction and based on the assumption of future operating conditions. A Monte Carlo simulation is applied to obtain a prognostic uncertainty. The comparison of the prediction and the test results demonstrates that the developed method can be applied to predict cavitation erosion states and RUL estimates.

Durability Prediction for Concrete Structures Exposed to Carbonation Using a Bayesian Approach (베이지안 기법을 이용한 중성화에 노출된 콘크리트 구조물의 내구성 예측)

  • This paper provides a new approach for predicting the corrosion resistivity of reinforced concrete structures exposed to carbonation. In this method, the prediction can be updated successively by a Bayesian theory when additional data are available. The stochastic properties of model parameters are explicitly taken into account into the model. To simplify the procedure of the model, the probability of the durability limit is determined from the samples obtained from the Latin hypercube sampling technique. The new method may be very useful in designing important concrete structures and help to predict the remaining service life of existing concrete structures which have been monitored.

  • In order to efficeintly manage a battery, it is important to accurately estimate and manage the SOH(State of Health) and RUL(Remaining Useful Life) of the batteries. Even if the batteries are of the same type, the characteristics such as facility capacity and voltage are different, and when the battery for the training model and the battery for prediction through the model are different, there is a limit to measuring the accuracy. In this paper, We proposed the entropy index using voltage distribution and discharge time is generalized, and four batteries are defined as a training set and a test set alternately one by one to predict the health status of batteries through linear regression analysis of machine learning. The proposed method showed a high accuracy of more than 95% using the MAPE(Mean Absolute Percentage Error).

  • Dementia is a major burden for many countries including South Korea, where life expectancy is continuously growing and the proportion of aged people is rapidly growing. Neurodegenerative disorders, such as, Alzheimer disease, dementia with Lewy bodies, frontotemporal dementia, Parkinson disease, progressive supranuclear palsy, corticobasal degeneration, Huntington disease, can cause dementia, and cerebrovascular disease also can cause dementia. Depression or hypothyroidism also can cause cognitive deficits, but they are reversible by management of underlying cause unlike the forementioned dementias. Therefore these are called pseudodementia. We are entering an era of dementia care that will be based upon the identification of potentially modifiable risk factors and early disease markers, and the application of new drugs postpone progression of dementias or target specific proteins that cause dementia. Efficient pharmacologic treatment of dementia needs not only to distinguish underlying causes of dementia but also to be installed as soon as possible. Therefore, differential diagnosis and early diagnosis of dementia are utmost importance. F-18 FDG PET is useful for clarifying dementing diseases and is also useful for early detection of the diseases. Purpose of this article is to review the current value of FDG PET for dementing diseases including differential diagnosis of dementia and prediction of evolving dementia.

Cloning and Sequence Analysis of Glyceraldehyde-3-Phosphate Dehydrogenase Gene in Yak

  • In order to study the biological function of gapdh gene in yak, and prove whether the gapdh gene was a useful intra-reference gene that can be given an important role in molecular biology research of yak, the cDNA sequence encoding glyceraldehyde-3-phosphate dehydrogenase from yak was cloned by the RT-PCR method using gene specific PCR primers. The sequence results indicated that the cloned cDNA fragment (1,008 bp) contained a 1,002 bp open reading frame, encoding 333 amino acids (AAs) with a molecular mass of 35.753 kDa. The deduced amino acids sequence showed a high level of sequence identity to Bos Taurus (99.70%), Xenopus laevis (94.29%), Homo sapiens (97.01%), Mus musculus (97.90%) and Sus scrofa (98.20%). The expression of yak's gapdh gene in heart, spleen, kidney and brain tissues was also detected; the results showed that the gapdh gene was expressed in all these tissues. Further analysis of yak GAPDH amino acid sequence implied that it contained a complete glyceraldehyde-3-phosphate dehydrogenase active site (ASCTTNCL) which ranged from 148 to 155 amino acid residues. It also contained two conserved domains, a NAD binding domain in its N-terminal and a complete catalytic domain of sugar transport in its C-terminal. The phylogenetic analysis showed that yak and Bos taurus were the closest species. The prediction of secondary structures indicated that GAPDH of yak had a similar secondary structure to other isolated GAPDH. The results of this study suggested that the gapdh gene of yak was similar to other species and could be used as the intra-reference to analyze the expression of other genes in yak.

Analysis of Hypertension Risk Factors by Life Cycle Based on Machine Learning (머신러닝 기반 생애주기별 고혈압 위험 요인 분석)

  • Chronic diseases such as hypertension require a differentiated approach according to age and life cycle. Chronic diseases such as hypertension require differentiated management according to the life cycle. It is also known that the cause of hypertension is a combination of various factors. This study uses machine learning prediction techniques to analyze various factors affecting hypertension by life cycle. To this end, a total of 35 variables were used through preprocessing and variable selection processes for the National Health and Nutrition Survey data of the Korea Centers for Disease Control and Prevention. As a result of the study, among the tree-based machine learning models, XGBoost was found to have high predictive performance in both middle and old age. Looking at the risk factors for hypertension by life cycle, individual characteristic factors, genetic factors, and nutritional intake factors were found to be risk factors for hypertension in the middle age, and nutritional intake factors, dietary factors, and lifestyle factors were derived as risk factors for hypertension. The results of this study are expected to be used as basic data useful for hypertension management by life cycle.

Computational Prediction of Solvation Free Energies of Amino Acids with Genetic Algorithm

  • We propose an improved solvent contact model to estimate the solvation free energies of amino acids from individual atomic contributions. The modification of the solvation model involves the optimization of three kinds of parameters in the solvation free energy function: atomic fragmental volume, maximum atomic occupancy, and atomic solvation parameters. All of these atomic parameters for 17 atom types are developed by the operation of a standard genetic algorithm in such a way to minimize the difference between experimental and calculated solvation free energies. The present solvation model is able to predict the experimental solvation free energies of amino acids with the squared correlation coefficients of 0.94 and 0.93 for the parameterization with Gaussian and screened Coulomb potential as the envelope functions, respectively. This result indicates that the improved solvent contact model with the newly developed atomic parameters would be a useful tool for the estimation of the molecular solvation free energy of a protein in aqueous solution.

  • A technique on the detection of tool wear based on the ultrasonic pulse-echo method in turning process is presented. The change in amount of the reflected energy from nose and flank of the tool can be related to the level of tool wear and mechanical integrity of the tool, that is, there exists an excellent correlation between the ultrasonic measurement and tool wear. As a results, the method is very useful for the prediction of cutting tool life and the determination of tool exchange period.
