Morphological and phylogeny of Plenodomus sinensis and P. collinsoniae, two unreported species isolated from soil in Korea

  • Two unreported fungal isolates, KNU-GW1901 and KNU-AP100C, were collected from soil sample in Gyeongsangbuk-do, Korea. Their cultural and morphological characteristics were examined after 4 weeks of incubation at 25℃ on potato dextrose agar (PDA), malt extract agar (MEA), and oatmeal agar (OA). The conidial shape of KNU-GW1901 was aseptate, hyaline, globose to ellipsoidal, oblong, and reniform to pyriform and 2.61-4.97×1.93-3.61 ㎛ in size, whereas no conidial structures were observed in KNU-AP100C. The internal transcribed spacer (ITS) regions, large subunit (LSU), and small subunit (SSU) sequences were used to determine the taxonomic positions of the strains using the maximum likelihood phylogenetic tree. The isolate KNU-GW1901 was closely clustered with Plenodomus sinensis MFLUCC 17-0767, and KNU-AP100C was closely matched with P. collinsoniae CBS 120227. Based on the findings of morphological, cultural, and phylogenetic analysis, the isolates KNU-GW1901 and KNU-AP100C were identical to the previously described P. sinensis and P. collinsoniae isolates, respectively, which are first reported in Korea.

청미래덩굴의 근권에서 분리된 2종의 Glomeromycota 미기록종 (Two Unreported Glomeromycota Fungi Isolated from Rhizospheres of Smilax china)

  • 청미래덩굴의 근권 토양을 온실에서 배양하여 Glomeromycota에 속하는 균류의 포자를 분리하였다. 분리된 포자는 18S partial rDNA 영역의 DNA 염기서열 분석과, 더 정확한 동정을 위해 추가적으로 ITS 영역과 28S rDNA 영역까지 포함하는 $Kr{\ddot{u}}ger$ fragment 지역의 염기서열을 분석하여 동정하였다. 동정 과정에서 2종의 국내 미기록종 Glomeromycota 균류의 포자를 확인하였으며, 확인된 종은 Diversispora eburnea, Paraglomus laccatum이다. 확인된 미기록종 포자의 형태적 특성 및 계통적 분석의 결과에 대해 기술하였다.

우포 습지에 분포하는 수지상균근균 포자의 다양성 (Spore Diversity of Arbuscular Mycorrhizal Fungi in Upo Wetland)

  • 우포 습지에 서식하는 식물의 근권 토양에서 수지상균근균(AMF) 포자를 추출하였다. 추출한 포자는 형태적 특성과 18S rDNA 염기서열의 분자적 분석을 이용해 동정하였다. 그 결과 6속 9종의 AMF 포자가 분리된 것을 확인하였으며, 종 다양성 지수를 계산하여 분석한 결과 습지에서 AMF 포자의 종 다양성은 식물에 의해 영향을 받는 것을 확인하였다. 연구 과정에서 국내 미기록종 1종을 확인하였으며, 확인된 종은 Diversispora epigaea이다. 미기록종 AMF 포자의 형태적 특성 및 분자적 분석의 결과에 대해 서술하였다.

Unreported Post-harvest Disease of Apples Caused by Plenodomus collinsoniae in Korea

  • This study was conducted to isolate and identify the fungal pathogen caused unreported post-harvest disease on apples (cv. Fuji) fruit in Korea. The disease symptoms on apples appeared as irregular, light to dark brown, slightly sunken spots. The three fungal strains were isolated from infected tissues of apple fruits and their cultural and morphological characteristics were completely consistent with those of Plenodomus collinsoniae. The phylogenetic analysis using the internal transcribed spacer (ITS) regions, beta-tubulin (TUB), and the second largest subunit of RNA polymerase II (RPB2) sequences revealed the closest relationship of the isolates with Plenodomus collinsoniae at the species level. The pathogenicity test showed the same dark brown spots on Fuji apple cultivar. Therefore, P. collinsoniae is a newly reported fungal agent causing post-harvest disease on apples in Korea.

Isolation of ten unrecorded yeasts from soil in Korea

  • In 2020, as a subset study to discover indigenous yeast species in Korea, a total of 22 yeast species were isolated from soil samples collected in Gwangju-si, Gyeonggi-do. Among them, 10 strains were unreported species. From the high 26S rRNA gene sequence similarity and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged independent and predefined yeast species. The 22 strains were assigned to the genera Dothiora (1 strain), Sarocladium (1 strain), Tetrapisispora (1 strain) and Torulaspora (2 strains) of the phylum Ascomycota; the genera Erythrobasidium (1 strain), Leucosporidium (4 strains), Ustilentyloma (2 strains), Fellozyma (1 strain), Sampaiozyma (2 strains), Filobasidium (1 strain), Solicoccozyma (2 strains) and Vishniacozyma (4 strains) of the phylum Basidiomycota. This is the first official report of the following species in Korea: Dothiora cannabinae (1 strain), Sarocladium strictum (1 strain), Fellozyma inositophila (1 strain), Filobasidium magnum (1 strain), Solicoccozyma phenolicus (1 strain), Solicoccozyma terreus(1 strain), Vishniacozyma tephrensis(1 strain) and Vishniacozyma victoriae (3 strains). Twelve unrecorded UV-resistant bacterial species isolated in 2020

  • In 2020, a total of 12 bacterial strains were isolated from soil after a comprehensive investigation of indigenous prokaryotic species in Korea. It was determined that each strain belonged to independent and predefined bacterial species, with high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species. This study identified four families in the phylum Actinobacteria, two families in the phylum Proteobacteria, one family in the phylum Bacteroidetes one family in the phylum Firmicutes; and four species in the family Nocardiaceae, two species in the family Nocardioidaceae, one species in the family Cellulomonadaceae, one species in the family Hymenobacter, one species in the family Methylobacteriaceae, one species in the family Microbacteriaceae, one species in the family Bacillaceae and one species in the family Sphingomonadaceae. There is no official report of these 12 species in Korea, so they are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are included in the species description section.

  • An investigation of the freshwater algal flora of Nakdong river estuary, Jinyang Lake and Danjang stream was carried out from May to November in 1993. This paper deals with a total 16 taxa belonging to 5 genera which are recorded for the first time in Korean freshwater algal flora. Among them, the 5 genera Coronastrum Thompson, Echinospaeridium Lemmermann, Dichtomococcus Korsikov, Dicloster Jao et al. and Siderocystopsis Swale are newly added to the freshwater algal flora of Korea.

  • 1972年 5, 6月 全北 內藏山一帶이 파리類 調査를 行한 바, 다음과 같은 우리나라 未記錄種이 發見되었다. 1. Parasarcophaga unguitigris Rohd. 1983 2. Protocalliphora azurea (Fallen, 1816) 3. Dichaetomyia japonica Hori et Kurahashi, 1967 4. Hydrotaea occulta (Meigen, 1826) 5. Phaonia crassipalpis Shinonage et Kano, 1971 6. Megophyro multisetosa Shinonaga, 1970 Dichaetomyia, Megophyra, Protocalliphora의 3屬은 우리나라에서 처음으로 記錄된다. 위 種들이 追加되어 우리나라 집파리科는 23種, 검정파리科는 14種, 쉬파리科는 36種이 된다.

Two Species of Endophytic Cladosporium in Pine Trees in Korea

  • During our studies on the diverse endophytic fungi resident on conifer needles, many species of Cladosporium previously unreported in Korea were encountered. In this paper, we report on two species of Cladosporium from the needles of pine trees (Pinus spp.). Based on analyses of internal transcribed spacer gene sequence, and cultural and micromorphological characteristics, they were identified as C. oxysporum and C. sphaerospermum. Both species have not been hitherto reported in Korea.
