Assessment of Factors Influencing Agrobacterium Mediated Transformation in Cucumber (Cucumis sativus L)

  • Five day old cotyledon explants of Cucumber (Cucumis sativus L) cv Poinsett 76 were cocultivated with two Agrobacterium strains (EHA105 and LBA 4404) each carrying GUS as the reporter gene and npt-II as the selection marker gene in the T-DNA region of the vector. Transformed shoots were selected at 150 mg/L kanamycin. A two day cocultivation coupled with $20\;{\mu}M$ acetosyringone increased the frequency (8.2 and 15.4 shoots) of GUS expression in the shoots of transformed plant. Among the two Agrobacterium strains, EHA 105 performed better than LBA 4404 in bringing two-fold increase in transformation efficiency (14%) than LBA 4404 (7.4%). PCR analysis was done to confirm the integration of T-DNA into cucumber genome.

  • The RGB Bayer filter system consists of mosaic R, G, and B filters on the grid of photo sensors with which typical commercial DSLR (Digital Single Lens Reflex) cameras and CCD/CMOS cameras are taken. Many unique astronomical data taken with RGB Bayer filter systems are available, including transient objects, e.g., supernovae, variable stars, and solar system bodies. The utilization of such data in scientific research strongly requires reliable photometry transformation methods. In this work, we develop a series of formulae to derive magnitudes in the Johnson-Cousins BVR filter system from those in the RGB Bayer filter system.

  • The ability to transform the chloroplast of Cyanidioschyzon merolae was limited by lack of confirmed and reliable promoter sequences (among other reasons), capable of delivering stable or modulated DNA transcription followed by protein synthesis. Our research has confirmed the applicability of three selected chloroplast promoters in C. merolae chloroplast overexpression of the exogenous protein (i.e., chloramphenicol acetyltransferase) and genetic transformation. These results might facilitate further research on genetically modified strains of C. merolae to envisage yet unknown aspect of cellular and plastic physiology as well as C. merolae potential applications as bio-factories or sources of useful chemicals.

  • The aim of this article is to derive twenty five transformation formulas in the form of a single result for the triple hypergeometric series $X_8$ introduced earlier by Exton. The results are derived with the help of generalized Watson#s theorem obtained earlier by Lavoie et al. An interesting special cases are also pointed out.

  • We construct a stationary, properly ordered Bratteli diagram B=(V,E,$\geq$) so that its Vershik map is isomorphic to Chacon's homeomorphism. It is the simplest stationary, properly ordered Bratteli diagram isomorphic to (Xc, Tc). We also find a primitive and proper substitution of Chacon's transformation.

Factors Determining Intention to Continue Using E-HRM

  • The development of information technology has promoted organizational transformation through the utilization of an electronic information system. This research aimed to identify factors that influence continuous intention to use E-HRM. This empirical research applies the Technology Acceptance Model and Cognitive Model for identifying significantly impacted areas of continuous intention to use E-HRM in a highly dynamic environment. The data were collected using questionnaires delivered directly to respondents. The sample was 100 employees of ESQ Group selected through random sampling. The variables used were subjective norms (X1), perceived behavioral control (X2), perceived innovativeness (Y1), cognitive absorption (Y2), satisfaction (Y3), and continuous intention to use E-HRM (Y4). Statistical analysis using Structural Equation Modelling (SEM) with Smart PLS was applied. The results revealed that behavioral control (X2) did not influence the continuous intention to use E-HRM (Y4) and that cognitive absorption (Y2) and satisfaction (Y3) did not significantly influence continuous intention to use E-HRM (Y4). Subjective norms (X1) significantly influenced both perceived innovativeness (Y1) and continuous intention to use E-HRM (Y4), perceived behavioral control (X2) significantly influenced both perceived innovativeness (Y1) and cognitive absorption (Y2), and perceived innovativeness (Y1) significantly influenced both satisfaction (Y3) and continuous intention to use E-HRM (Y4).

Transformation of Fuji Apple Plant Harboring the Coat Protein Gene of Cucumber mosaic virus

  • Transformation of Fuji apple (Malus domestica 'Fuji') was performed using Agrobacterium tumefaciens harboring a coat protein (CP) gene of Cucumber mosaic virus (CMV). A plasmid DNA containing the virus CP and NPT II genes was introduced into the loaves of apple by th e Agrobacterium - mediated transformation procedure. Regenerated transformants of the apple were obtained by kanamycin resistance conferred by the introduced NPT II gene. PCR analysis showed that 3 out of 20 putatively selected R0 plant lines contain the CMV-CP gene. Nine putative transgenic lines out of 20 lines were investigated with the PCR analysis; 5 regenerants produced a 450 bp DNA band and 3 regenerants showed a 671 bp DNA band for the NPT II and CMV-CP genes, respectively. Southern hybyidization results demonstrate the successful integration of the CMV-CP gene into the genome of the apple. This is the first report on the generation of useful vius resistance source of transgenic apple for molecular breeding program.

  • When the half infinite crack in the orthotropic material strip with a large anisotropic ratio(E11>>E22) propagates with constant velocity, dynamic stress component $\sigma$y occurre d along the $\chi$ axis is derived by using the Fourier transformation and Wiener-Hopf technique, and the dynamic stress intensity factor is derived. The dynamic stress intensity factor depends on a crack velocity, mechanical properties and specimen hight. The normalized dynamic stress intensity factors approach the maximum values when normalized time(=Cs/a) is about 2. They have the constant values when the normalized time is greater than or equal to about 2, and decrease with increasing a/h(h: specimen hight, a: crack length) and the normalized crack propagation velocity( = c/Cs, Cs: shear wave velocity, c: crack propagation velocity).

Expression of Tunicamycin Resistance in Bacillus subtilsls by Several Transfroming Plasmids

  • pSp-Si (1.6kbp) was originally found in pediococcus halophilus to be a cryptic multicopy-plasmid. Hoping that the plasmid can also replicate in Bacillus subtilis, protoplast transformation of strain 207-25 (recE) was performed using pSP-Sl onto which was added the marker of tmrB8 (on 4.9 kbp EcoRI fragment ) or tmrB+ (on 0.9 kbp xbaI fragment) gene. Though the tmrB8 gene can expres tunicamycin-resistance at the single copy state, and the tmrB+ gene exerts the resistance only at the multicopy state, we could not confirm the replication of pSP-Sl (tmrB8) or pSP-Sl(tmrB+) in B. subtilis. During the experiment, however, we unexpectedly found that the circularized 0.9 kbp xgaI fragment (tmrB+) itself, which had no replication origin, could transform strain 207-25 to tunicamycin-resistant by protoplast transformation. Southern hybridization analyses with tmrB+ and other probes revealed the integration of the fragment at a single copy state into a position other than the homologous tmrB gene. This recE independent integration of another tmrB+ gene into the chromosome may contribute to the tunicamycinresistance in the transformants.
