Reduction of Ambiguity in Phosphorylation-site Localization in Large-scale Phosphopeptide Profiling by Data Filter using Unique Mass Class Information

  • The rapid development of shotgun proteomics is paving the way for extensive proteome profiling, while providing extensive information on various post translational modifications (PTMs) that occur to a proteome of interest. For example, the current phosphoproteomic methods can yield more than 10,000 phosphopeptides identified from a proteome sample. Despite these developments, it remains a challenging issue to pinpoint the true phosphorylation sites, especially when multiple sites are possible for phosphorylation in the peptides. We developed the Phospho-UMC filter, which is a simple method of localizing the site of phosphorylation using unique mass classes (UMCs) information to differentiate phosphopeptides with different phosphorylation sites and increase the confidence in phosphorylation site localization. The method was applied to large scale phosphopeptide profiling data and was demonstrated to be effective in the reducing ambiguity associated with the tandem mass spectrometric data analysis of phosphopeptides.

Complex organic molecules detected in twelve high mass star forming regions with ALMA

  • One of the key questions on star formation is how the organic molecules are synthesized and delivered to the planets and comets since they are the building blocks of prebiotic molecules such as amino acid, which is thought to contribute to bringing life on Earth. Recent astrochemical models and experiments have explained that complex organic molecules (COMs; molecules composed of six or more atoms) are produced on the dust grain mantles in cold and dense gas in prestellar cores. However, the chemical networks and the roles of physical conditions on chemistry are not still understood well. To address this question, hot (> 100 K) cores in high mass young stellar objects (M > 8 Msun) are great laboratories due to their strong emissions and larger samples than those of low-mass counterparts. In addition, CH3OH masers, which have been mostly found in high mass star forming regions, can provide constraints due to their very unique emerging mechanisms. We investigate twelve high mass star forming regions in ALMA band 6 observation. They are associated with 44/95 GHz Class I and 6.7 GHz Class II CH3OH masers, implying that the active accretion processes are ongoing. For these previously unresolved regions, 66 continuum peaks are detected. Among them, we found 28 cores emitting COMs and specified 10 cores associated with 6.7 GHz Class II CH3OH masers. The chemical diversity of COMs is found in cores in terms of richness and complexity; we identified up to 19 COMs including oxygen- and nitrogen-bearing molecules and their isotopologues in a core. Oxygen-bearing molecules appear to be abundant and more complex than nitrogen-bearing species. On the other hand, the COMs detection rate steeply grows with the gas column density, which can be attributed to the effective COMs formation in dense cores.

  • The primary purpose of this study is to examine consumers'probing actions to see what information sources consumers search for medical information when there are diverse medical service information channels, and classify consumers by information source. Its secondary purpose is to understand trust of information and attitude toward information by consumer type, value of medical service, satisfaction with medical service, and word-of-mouth intention. This study will concretely identify information utilization patterns of medical consumers, and explain the unique characteristics and behavior of segmented types of medical consumers. The significance of this study lies in the search for ways to establish information channels trusted by consumers for building an efficient medical service market in the future. The results of this study show that consumers were classified by the latent class analysis(LCA) into 5 types: low-level information seekers, word-of-mouth information seekers, mass media information seekers, digital information seekers and diverse information seekers. The reliability of information sources by type of medical consumer was statistically significant, and in the analysis of differences in consumer attitude, there was a statistically significant difference in cognitive responses. The value of medical service was statistically significant in health recovery and medical service word-of-mouth intention.

Sesquiterpene-Neolignans from the Stem Bark of Magnolia obovata and Their Cytotoxic Activity

  • Three sesquiterpene-lignans, eudeshonokiol B (1), eudesobovatol B (2), and clovanemagnolol (3), were isolated from the stem bark of Magnolia obovata, together with magnolol (4), honokiol (5), and obovatol (6) on the basis of spectroscopic and physicochemical analyses including 2D NMR and Mass. Compounds 1 - 3 were belongs to a unique class of natural products made up of a sesquiterpene and biphenyl-type neolignan via an ether bond. All the isolated compounds were tested in vitro for their cytotoxic activity against the HeLa, A549, and HCTll6 cancer cell lines. Compounds 1 - 6 showed the cytotoxic activity against tested cancer cell lines, with $IC_{50}$ values ranging from 7.1 to 14.4 ${\mu}g/mL$.

  • Polymers are the major constructive material of pharmaceutical formulations that play a prime role in designing effective drug-delivery systems and releasing drugs at their sites of application. Polymers are composed of multiple repeating units of high molecular mass components with attendant properties. Most synthetic polymers are non-biocompatible, expensive, and extremely inclined to deliver adverse impacts. Meanwhile, edible polymers obtained from natural sources have gained remarkable recognition for their promising use in modern medicine. Moreover, polymers derived from natural sources are generally preferred due to certain of their unique features such as abundant availability, biocompatibility, nontoxicity, economical, safe, and effective functions that fit the purpose. Polysaccharides including starch, cellulose, hemicellulose, pectin, and mucilage are identified as a major class of naturally obtained molecules that have a substantial role as functional polymers. This review summarizes the potential role of polysaccharides derived from vegetable sources such as adhesives, anticaking agents, binders, disintegrants, emulsifiers, film-framing agents, and thickeners. This is simply an opportunity to abandon synthetic excipients that hurt our bodies and think back to nature from where we originate.

Candidates for the young stellar outflows: Water and Methanol masers from young stellar objects

  • We conducted simultaneous 22GHz water maser and 44GHz class I methanol maser surveys of newly-identified 282 H2 emission features from the $2.122{\mu}m$ H2 narrowband image survey in the Galactic plane (UWISH2 project) using Korea VLBI Network (KVN) 21-m radio telescopes. We detected 16 and 13 new water and methanol maser sources, respectively. This result indicates that at least ~5% of the H2 emission features originate from young stellar objects (YSOs) that are in the right physical condition to produce the water and methanol masers. The masers are closely related to the current outflow activities in the Galactic plane. The power sources of these 23 diffused/collimated H2 emission features (six sources are detected for both masers) are likely to be intermediate-to high-mass YSOs, based on a comparison with the maser luminosities of other well-studied YSOs. Both maser velocities are mostly close to their own systemic velocities within 5 km/s, even though water masers generally show larger variabilities in the line intensities, velocities, and shapes than methanol masers. We also discovered three new water maser sources with high-velocity components: ~25 km/s red-shifted CMHO019, ~50 km/s blue-shifted CMHO132, and ~120 km/s blue-shifted CMHO182. In particular, we propose that the dominant blue-shifted water maser of CHMO182 could become a unique laboratory for the study of high-mass stellar jet and their accelerations.

Function of the Tethered rec-eCG in Rat and Equine Receptors

  • The glycoprotein hormone family represents a class of heterodimers, that includes the placental hormone equine chorionic gonadotropin (eCG) and the anterior pituitary hormones- follitropin (FSH), lutropin (LH), and thyrotropin (TSH). The 4 hormones are heterodimers, with a common $\alpha$-subunit and unique $\beta$-subunits. eCG is the most heavily glycosylated of the known pituitary and placental glycoprotein hormones. Recent observations using single chain glycoprotein hormone analogs in which, the $\beta$-and $\alpha$-subunits are linked, implied that heterodimeric-like quaternary configuration is not a prerequisite for receptor/signal transduction. To study the function and signal transduction of tethered rec-eCG, a single chain eCG molecule was constructed and rec-eCG protein was produced. Molecular mass of the single chain is about 45 kDa. All mice were ovulated by tethered rec-eCG treatment. The dual activity of tethered rec-eCG was determined in receptor cell lines of nonequid species; in fact, this dual activity was proven in species other than horse. Tethered rec-eCG in equids does not bind to FSH receptors, suggesting that eCG is primarily an LH-like hormone in the horse. Taken together, these data suggest that tethered rec-eCG has dual activity in nonequid species in vitro. However, it has only LH-like activity in equid species in vitro.

Socialist Pop After Cultural Revolution (문화혁명기 이후의 중국의 사회주의 팝아트)

  • This thesis examines contemporary Chinese painting after the Cultural Revolution(1966~76), focusing upon so-called "Chinese Pop art", which I termed as "Socialist Pop art". I considered the art of this period within the broader context of social changes especially after the Tienanmen incident of 1989. After the Cultural Revolution during which idolization of Chairman Mao was at its peak, one of the major changes in communist China was that an anti-Mao wave was generated in almost every social class. For example, novels that revealed the hardships during the Cultural Revolution were published. Posters that openly criticized the Maoism were also produced and displayed on the walls, and demand for democracy spurred widespread activist movements among young generations. These broad social changes were also reflected in art. A variety of art movements were introduced from the West to China, and after a period of experimentation with the new imported styles, artists began to apply the new artistic idiom to their works in order to visualize their own social and political realities they lived in. It was a shift from earlier Socialist Realism to a new expression either directly or indirectly, "Socialist Pop", an amalgam of Socialist Realism and Pop art tradition. After the 1989 crackdown of Tienanmen Square protest, when communist government quelled with brutal measures the students, workers, and ordinary people who rose for democracy, greater urge to protest the Deng Xiaoping regime emerged. This time coincided with the gradual emergence of art using Pop art vocabulary to satirize the social reality, the Socialist Pop art, along with many other art forms all with avant-garde spirit. One of the most frequent subjects of Chinese Pop art was visual images of Chairman Mao and his Cultural Revolution, and new China that was saturated with capitalism, which tainted the Chinese way of life with a Western way of consumerism and commercialism. The reason for the popularity of Mao's image was spurred by the "Mao Craze" in the early 1990's. People suddenly began to fall in a kind of nostalgia for the past, and once again, Mao Zedong was idolized as an entity who can heal the problems of modern China who had been marching towards their ultimate destination, the economic development. But this time Chairman Mao was no more an idol but just a popular, commercial product. He is no more an object of worship of almost religious nature but he has become an iconography symbolizing the complex nature of present Chinese society. During this process of depicting the social reality, Chinese artists are making the authority and sanctity of Maoism ineffective. Dealing with this new trend of contemporary Chinese art in view of "Socialist Pop art" two manners of re-creating Pop art can be illustrated: one that incorporates the propaganda posters of the Cultural Revolution; the other borrows from Chinese traditional popular imagery or mass media, such as photos taken during Mao era. What is worth mentioning is that these posters and photos of the Cultural Revolution can be identified as 'popular' media, as they were directed to educate the popular mass, thus combination of this ingenuous pop media with Western Pop art can be fully justified as a genre unique to China. Through this genre, we can discover a new chapter of the Chinese contemporary painting and its society, as their Pop art can be considered as self-portraits true to their present appearances.

GIS Based Real-Time Transit Information Integration and Its Transit Planning Implications

  • Over the years, Advanced Public Transportation System (APTS) has been implemented to manage and operate public transportation system. With the expanding mass spatio-temporal data such as comprehensive spatial information of each individual passenger and public transportation vehicle, it has been required to consolidate and analyze these multiple data sets from various sources. This paper demonstrates how GIS is utilized for the consolidation of massive transit related spatio-temporal information. And it presents effective applications to improve transit planning process and support transit related decision-making activities. GIS based system is used to combine multi-agents' data in the areas of transit operation and individual transit ride and transfer management. Due to the unique comprehensiveness and the level of detail of the data provide by the Seoul Transit system, this GIS based information consolidation is the first in its class. Based on the integrated database, this paper describes the effective and efficient GIS based analysis deployed in a transportation system planning process. The data integration systems and analytic models developed in this paper can be transferred and applied by any municipal governments provided that the appropriate data is available.

Extragalactic Sciences from SPICA/FPC-S

  • The SPICA (SPace Infrared Telescope for Cosmology & Astrophysics) project is a next-generation infrared space telescope optimized for mid- and far-infrared observation with a cryogenically cooled 3m-class telescope. The focal plane instruments onboard SPICA will enable us to resolve many astronomical key issues from the formation and evolution of galaxies to the planetary formation. The FPC-S (Focal Plane Camera - Sciecne) is a near-infrared instrument proposed by Korea as an international collaboration. Owing to the capability of both low-resolution imaging spectroscopy and wide-band imaging with a field of view of $5^{\prime}{\times}5^{\prime}$, it has large throughput as well as high sensitivity for diffuse light compared with JWST. In order to strengthen advantages of the FPC-S, we propose the studies of probing population III stars by the measurement of cosmic near-infrared background radiation and the star formation history at high redshift by the discoveries of active star-forming galaxies. In addition to the major scientific targets, to survey large area opens a new parameter space to investigate the deep Universe. The good survey capability in the parallel imaging mode allows us to study the rare, bright objects such as quasars, bright star-forming galaxies in the early Universe as a way to understand the formation of the first objects in the Universe, and ultra-cool brown dwarfs. Observations in the warm mission will give us a unique chance to detect high-z supernovae, ices in young stellar objects (YSOs) even with low mass, the $3.3{\mu}$ feature of shocked circumstance in supernova remnants. Here, we report the current status of SPICA/FPC project and its extragalactic sciences.
