Effects of Combined Treatment of Gamma Irradiation and Addition of Fucoidan/laminarin on Ready-to-eat Pork Patty (감마선 조사와 푸코이단/라미나린 병용처리가 돈육 패티의 품질에 미치는 영향)

  • The objective of this study was investigated to apply the fucoidan and laminarin extract from low-grade Undaria pinnatifida to real food product, a pork patty. Pork patties added with fucoidan and laminarin showed lower lipid oxidation and the inhibition against lipid oxidation was shown to be dependent on the irradiation doses. The Hunter color values of pork patty added with fucoidan increased significantly with an increase of irradiation dose. The hardness profiles of the patties with fucoidan and laminarin was decreased, but the amount of water in the patties was increased. Also, the combination of gamma irradiation and addition of fucoidan and laminarin was shown to be effective for the microbiological control. These results suggested that gamma irradiation and fucoidan and laminarin treatment showed the positive effect on microbial stability and quality of the pork patty.

  • The chemical composition of Undaria pinnatifida Laver (U.P. Laver) were determind. 1. Proximate compositions of U.P. Laver were 22.1% crude protein, 1.3% crude fat, 10.2% crude ash, 66.4% N.F.E, 3 0% crude fiber and 22.8% alginic acid. 2. U.P. Laver protein was chiefly composed of glutamic acid (3.24%) and aspartic acid (2.22%) and considerable quantities of leucine (1.98%), alanine (1.56%), valine (1.32%), lysine (1.22%) and phenylalanine (1.15%). 3. The major fatty acid contents of the lipid extracted from U.P. Laver were linolenic acid (45.2%), palmitic acid (26.14%) and linoleic acid (11.27%) and the minor fatty acid were myristic acid (8.41%), oleic acid (8.11%), and stearic acid (0.87%), respectively. The saturated and unsaturated fatty acid ratio of the oil extracted from U.P. Laver was 35.42/64.58 4. The vitamin contents of U. P. Laver were $28.1{\mu}g/g$ niacin, $25.0{\mu}g/g$ vit. E, $18.8{\mu}g/g$ vit. C. The minerals were composed 1.52% calcium, 0.32% phosphorus, 0.88% magnesium, 0.49% potassium and the others were small quantities.

  • The purpose of this study is screening of microfloras involved in hydrolysis of sea tangle (Laminaria japonica) and sea mustard ( Undaria pinnatifida), This is a part of studies on the hydrolysis of seaweed using microorganisms. General process is difficult to extract of the useful constituent parts as intercellular mucilage, storage polysaccharide and mineral from seafood. It was screening to thirty-three microflora samples as destructed of tissue in sea tangle and sea mustard to about two hundred microflora samples from mountain, rice field, dry field, sea, seaside and fish market etc. in the neighborhood of Yeosu. Sufficient results of the naked eye observation were obtained at eight microflora samples as a feces of bull, a decayed pine tree, a soil of dry field, the mud of the banks in a rice Held, the water of a ditch in a rice Held, the weed of the banks in a rice field, the water in a rice field and leaved in the air, And the value of chemical analysis of the sample is much better in comparison with control. Accordingly, the hydrolysis of seaweed using microorganisms in the inside of these microflora samples can possibility.

Isolation and Identification of an Antibacterial Substance from Sea Mustard, Undaria pinnatifida, for Streptococcus mutans (미역 추출물로부터 충치 원인균, Streptococcus mutans에 대한 항균물질의 분리 및 동정)

  • An antibacterial substance to the Streptococcus mutans, a causative bacterium for decayed teeth, was isolated from the dried sea mustard, Undaria pinnatifida, and identified by GC and GC/MS. Acetone extract from the sea mustard (10.4 kg), was evaporated and partitioned to 4 fractions such as hexane, chloroform, butanol and water. The most active chloroform fraction were further purified through basic alumina, silicic acid and ODS column, successively, and finally, 3 antibacterial substances were isolated on the HPLC attached ODS column by using 95% MeOH and guided with UV detector (254 nm). Antibacterial substances (total 160mg, yield $1.5\times10^{-3}$%) had the same Rf value (0.42) on the TLC developed hexane diethyl ether acetic acid (80:30:1) and those methyl esters moved to 0.95. They were identified as the same unsaturated fatty acid, $C_{18:4,\;n-3}$ (3,6,9,12-octadecatetraenoic acid, stearidonic acid) compared relative retention times (15.5 min) with authentic fatty acid on the GC chromatogram. It was further confirmed unambiguously on the GC/MS giving molecular ion peak at m/z 290 which coincided with its methyl ester.

  • 미역의 뿌리부분인 미역귀에는 알긴산과 후코이단 성분이 풍부하고 고혈압과 당뇨등과 같은 성인병을 예방하는데 도움이 된다. 후코이단은 미역과 다시마 같은 갈조류에서 추출된 천연물질로 특히 면역력을 활성화하고 항암작용으로 잘 알려져 있다. 알긴산 등 유용성분이 함유된 것으로 알려진 미역귀의 유용성분을 분석하여, 향후 미역귀를 활용한 제품개발의 기초자료를 제공하고자 본 연구를 수행하였다. 본 연구에 사용된 미역귀는 2015년 12월 완도 금일수산에서 제공받아 연구재료로 활용하였다. AOAC방법에 따라 분석한 미역귀의 일반성분은 수분이 대부분을 차지하였으며, 조섬유와 가용성 무질소물 함량이 높게 나타났다. 이는 시료로 사용한 미역귀가 수확 후 바로 냉동보관 되어서 수분함량이 높은 것으로 보여진다. 유리당은 미역귀 1g에 증류수를 가하여 $60^{\circ}C$에서 4시간 추출 한 후, 원심분리(3,000 rpm, 30 min)하여 $0.45{\mu}m$ membrane filter (Millipore Co., USA)로 여과한 여액을 HPLC (High Performance Liquid Chromatography)를 이용하여 분석한 결과, 미역귀의 유리당은 Galactose 1종만이 검출되었다. 미역귀의 구성아미노산과 유리아미노산은 $0.45{\mu}m$ membrane filter로 여과하여 얻은 여액을 일정량 취하여 AccQ-Tag 시약을 사용하여 유도체화 시킨 후 HPLC로 분석하였다. 미역귀의 구성아미노산은 glutamic acid, aspartic acid, alanine, lysine, threonine, valine, arginine 순으로 높은 함량을 나타내었으며, 유리아미노산은 glutamic acid, alanine, threonine, arginine, lysine 순으로 높은 함량을 나타내었다. 미역귀의 유리아미노산함량은 표고에 비하여 현저히 낮은 함량이 나타났으며, 주요아미노산은 해조류의 풍미를 대표하는 아미노산인 glutamic acid와 alanine으로 나타났다.

Effects of diet with brown algae on the quality characteristics of eggs (갈조류 첨가 사료로 식이한 산란계 계란의 품질 특성)

  • This study was performed to determine the effect of brown algae on the egg quality of laying hens fed a non-supplemented diet (Control) and a diet supplemented with 10% Undaria pinnatifida (A), Laminaria religiosa (B), and Hijikia fusiforme (C), respectively. Isa Brown (28 weeks old) were raised for 8 weeks. The darkness, redness, and blueness of the egg yolks increased with the addition of brown algae, expecially Undaria pinnatifida (A). The fucoxanthin contents were observed to be only 21.3-34.8 mg% level in the brown algae but they were nearly traced in the egg yolks. Among the analyzed minerals, the Ca, Fe, K contents increased to 391.2, 6.3, and 450.5 mg%, respectively, but the amount of P was reduced to 110.5 mg%. The total mineral contents were higher in the egg yolk with Hijikia fusiforme (C) (1,155.1 mg%) than with any other supplement. The brown algae diet reduced the saturated fatty acid level from 38.3 to 32.1% but increased the unsaturated fatty acid level from 61.5 to 69.6% owing to the rise of the level of polyunsaturated fatty acids.

  • In order to utilize effectively sea mustard(Undaria pinnatifida) which is excellent in nutrition and medical action, powdered sea mustard juice was prepared and then its chemical composition and the stability of pigments were examined. Powdered sea mustard was made by washing fresh sea mustard with tap water to remove clay and sand, blanching at $85^{\circ}C$ for 20 sec in mixing solution(3% salt+1% $MgCO_3$), hot air drying($50-55^{\circ}C$, 3 hrs) after draining and pulverizing dried sea mustard to 200mesh. And then powdered sea mustard mixtures for juice type beverage was made by adding 0.75% of salt, 1.25% of sugar, 0.2% of ascorbic acid, 0.25% of flour of roasted barley and 0.8% of pulverized sea mustard to 100ml of water. Chemical composition of product was not scarcely changed during processing while amino-nitrogen content was increased and alginic acid and ash contents were decreased. The retention of chlorophyll and total carotenoid pigments of product against fresh sea mustard were 91.6% and 89.5% respectively. Judging from sensory evaluation, color, flavor, taste and dispersibility of powdered sea mustard juice were excellent and undesirable flavor of product was masked by addition of flour of roasted barley.

  • The effect of three different diets (formulated diet, FD; Undaria pinnatifida, UP; Laminaria japonica, LJ) on growth in the Haliotis discus hannai for 90 days was investigated. The shell length of UP ($80.62{\pm}1.92$ mm) and LJ ($81.14{\pm}1.16$ mm) were significantly faster than those of FD ($79.38{\pm}1.69$ mm) (P < 0.05). However, shell breadth and total weight were no significant difference among three diets. On the other hand, the weight gain of FD (16.65% for 0-45day and 25.71% for 45-90day) tend to have higher than those of UP (14.57% and 23.30%) and LJ (12.65% and 24.51%). This results shows that seaweed diets (UP and LJ) help the shell growth of abalone and formulated diet (FD) help the weight gain of abalone. Therefore, the growth of shell and muscle will depend on different diets.

Inhibition of Xanthine Oxidase by Seaweed Extracts (해조류 추출물의 Xanthine Oxidase 저해작용)

  • Inhibition of xanthine oxidase by seaweed extracts obtained from Undaria pinnatifida, Ecklonia stolonifera, Ecklonia cava, Laminaria japonica, Sargassum, Codiumfragile, Enteromorpha compressa and Porphyra tenera were investigated. Extracts of E. stolonifera and E. mua remarkably inhibited xanthine oxidase activity compared to those of other seaweed. The xanthine oxidase inhibitory activity of E. cava was higher than that of E. stolonifera. Diethyl ether extract from E. cava was more effective in the inhibition of xanthine oxidase than other solvent extracts. Two xanthine oxidase inhibitors(A-1 and A-2) from diethyl ether extract were isolated and purified by silica gel column chromatography, thin layer chromatography and high performance liquid chromatography. Xanthine oxidase inhibitory activities of these compounds were 27.8 and 48.1% per 0.4mg, respectively. The active compound A-2 had absorption peak at 420nm, 456nm and 467nm, which can be considered as siphonaxanthine.

Algal Consumption and Preference of Sea Urchins, Strongylocentrotus nudus, S. intermedius and Abalone, Haliotis discus hannai (둥근성게, 북쪽말똥성게 및 참전복의 일일 먹이소비량 및 해조류 먹이선택성)

  • In 2006, we studied herbivore consumption rate and preference for algal species using laboratory experiments. In the no-choice feeding experiment, three herbivores (two sea urchins, Stronglyocentrotus nudus and Stronglyocentrotus intermedius and an abalone, Haliotis discus hannai) showed significantly (P<0.05) distinct preferences for four algae (Laminaria japonica, Undaria pinnatifida, Costaria costata and Ecklonia stolonifera). The alga preferred by S. nudus was L. japonica followed by U. pinnatifida, C. costata and E. stolonifera with respective daily feeding rates of $5.7{\pm}:0.85\;g,\;4.4{\pm}0.89\;g,\;3.1{\pm}0.39\;g\;and\;2.1{\pm}0.32\;g(mean{\pm}SE)$. A similar trend was found for S. intermedius. Interestingly, the herbivore consumption rates for the algae differed with or without competition. The competition stimulated the feeding activity for S. nudus, but lowered the activity for S. intermedius. The multi-choice feeding experiment confirmed that L. japonica was significantly preferred by two herbivores: S. nudus and H. discus hannai (Duncan's multiple range test, ANOVA, p<0.05).
