Effects of Serial Passage on the Characteristics and Chondrogenic Differentiation of Canine Umbilical Cord Matrix Derived Mesenchymal Stem Cells

  • Mesenchymal stem cells (MSCs) are often known to have a therapeutic potential in the cell-mediated repair for fatal or incurable diseases. In this study, canine umbilical cord MSCs (cUC-MSCs) were isolated from umbilical cord matrix (n = 3) and subjected to proliferative culture for 5 consecutive passages. The cells at each passage were characterized for multipotent MSC properties such as proliferation kinetics, expression patterns of MSC surface markers and self-renewal associated markers, and chondrogenic differentiation. In results, the proliferation of the cells as determined by the cumulative population doubling level was observed at its peak on passage 3 and stopped after passage 5, whereas cell doubling time dramatically increased after passage 4. Expression of MSC surface markers (CD44, CD54, CD61, CD80, CD90 and Flk-1), molecule (HMGA2) and pluripotent markers (sox2, nanog) associated with self-renewal was negatively correlated with the number of passages. However, MSC surface marker (CD105) and pluripotent marker (Oct3/4) decreased with increasing the number of subpassage. cUC-MSCs at passage 1 to 5 underwent chondrogenesis under specific culture conditions, but percentage of chondrogenic differentiation decreased with increasing the number of subpassage. Collectively, the present study suggested that sequential subpassage could affect multipotent properties of cUC-MSCs and needs to be addressed before clinical applications.

시험관내 배양된 제대혈 모세포에서의 신경항원 발현 (Neural Antigen Expressions in Cultured Human Umbilical Cord Blood Stem Cells in vitro)

  • Objectives : Cord blood stem cells have been widely used as donor cells for bone marrow transplantation recently. These cells can give rise to a variety of hematopoietic lineages to repopulate the blood. Recent observations reveal that some bone marrow cells and bone marrow stromal cells(MSCs) can grow to become either neurons or glial cells. It is, however, unclear whether or not there exists stems cells which can differentiate into neurons in the blood during the early stages of postnatal life. Methods : Human cord blood stem cells were prepared from human placenta after full term delivery. To induce neuronal differentiation of stem cells, ${\beta}$-mercaptoethanol was treated. To confirm the neuro-glial characteristics of differentiated stem cells, immunocytochemical stain for NeuN, neurofilament, glial fibrillary acidic protein(GFAP), microtubule associated protein2(MAP2) was performed. RT-PCR was performed for detecting nestin mRNA and MAP2 mRNA. Results : We showed in this experiment that neuro-glial markers(NeuN, neurofilament, MAP2, GFAP) were expressed and axon-like cytoplasmic processes are elaborated in the cultured human cord blood stem cells prepared from new born placenta after full term delivery. Nestin mRNA was also detected in fresh cord blood monocytes. Conclusions : These results suggest that human cord blood derived stem cells may be potential sources of neurons in early postnatal life.

Identification of MFGE8 in mesenchymal stem cell secretome as an anti-fibrotic factor in liver fibrosis

  • The beneficial paracrine roles of mesenchymal stem cells (MSCs) in tissue repair have potential in therapeutic strategies against various diseases. However, the key therapeutic factors secreted from MSCs and their exact molecular mechanisms of action remain unclear. In this study, the cell-free secretome of umbilical cord-derived MSCs showed significant anti-fibrotic activity in the mouse models of liver fibrosis. The involved action mechanism was the regulation of hepatic stellate cell activation by direct inhibition of the $TGF{\beta}$/Smad-signaling. Antagonizing the milk fat globule-EGF factor 8 (MFGE8) activity blocked the anti-fibrotic effects of the MSC secretome in vitro and in vivo. Moreover, MFGE8 was secreted by MSCs from the umbilical cord as well as other tissues, including teeth and bone marrow. Administration of recombinant MFGE8 protein alone had a significant anti-fibrotic effect in two different models of liver fibrosis. Additionally, MFGE8 downregulated $TGF{\beta}$ type I receptor expression by binding to ${\alpha}v{\beta}3$ integrin on HSCs. These findings revealed the potential role of MFGE8 in modulating $TGF{\beta}$-signaling. Thus, MFGE8 could serve as a novel therapeutic agent for liver fibrosis.

Mesenchymal stem cells for restoration of ovarian function

  • With the progress of regenerative medicine, mesenchymal stem cells (MSCs) have received attention as a way to restore ovarian function. It has been reported that MSCs derived from bone marrow, adipose, umbilical cord blood, menstrual blood, and amniotic fluid improved ovarian function. In light of previous studies and advances in this field, there are increased expectations regarding the utilization of MSCs to restore ovarian function. This review summarizes recent research into potential applications of MSCs in women with infertility or primary ovarian insufficiency, including cases where these conditions are induced by anticancer therapy.

탯줄유래 줄기세포의 계대배양에 따른 특성 변화의 분석 (Characterization of Umbilical Cord-derived Stem Cells during Expansion in Vitro)

  • 목 적: 중간엽 줄기세포를 임상에 적용하기 위해서는 체외 배양을 통한 세포증식 과정이 필요하나, 오랜 기간 동안 체외 배양을 하게 되면 노화되어 특성이 변하고 분화 능력 또한 감소하게 된다. 따라서 현재까지는 초기 계대배양의 세포만이 임상에 적용되고 있는 실정이며 체외에서의 세포 배양이 세포의 특성에 미치는 영향에 대한 연구와 함께 세포의 특성 변화 없이 체외증식이 가능하도록 하는 연구들이 골수 및 지방유래 중간엽 줄기세포에서 보고되고 있다. 그러나 현재 탯줄유래 줄기세포의 체외 배양에 따른 특성 변화 분석 연구는 아직 잘 이루어지지 않고 있다. 본 연구의 목적은 탯줄유래 줄기세포의 체외 배양 시 계대배양 증가에 따른 줄기세포의 특성 변화를 분석하고자 하였다. 연구방법: 사람의 탯줄유래 줄기세포 (human umbilical cord-derived stem cells, HUC)를 분리하여 in vitro에서 계대배양하였다. 계대배양에 따른 세포의 형태와 population doubling time (PDT)을 조사하고 RT-PCR 방법을 이용하여 mRNA 분석을 하였으며 면역세포화학 염색법을 이용하여 단백질 발현을 분석하였다. 결 과: 탯줄유래 줄기세포는 평균 10번의 계대배양 후 senescence를 나타냈다. 세포의 형태는 7번째 계대배양 이후 세포질이 넓어지고 세포의 크기가 커지는 변화를 나타냈으며 PDT가 증가하기 시작하였다. 계대배양 4, 8, 10번째 시기의 세포의 mRNA 변화를 분석한 결과 Oct-4, HNF-4${\alpha}$, mRNA는 10번째 계대배양까지 지속적으로 발현하였으나 nestin, vimentin mRNA는 지속적으로 발현이 감소하였고 SCF mRNA는 지속적으로 발현이 감소하였다. 이에 반해 HLA-DR${\alpha}$, Pax-6, BMP-2 mRNA는 모든 계대배양 시기의 세포에서 발현되지 않았다. 면역세포화학 분석법을 통한 3, 6, 9번째 계대배양 세포의 단백질 발현 분석 결과 SSEA-3와 SSEA-4는 3, 6, 9번째 계대배양 세포 모두에서 발현하였으나 ICAM-1과 HLA-ABC는 계대배양이 증가함에 따라 발현이 증가되었다. Thy-1 단백질은 p9에서 발현이 증가되었으며 이와 반대로 TRA-1-60와 VCAM-1 단백질은 p6과 p9 시기에 발현이 감소되었다. HLA-DR 단백질은 모든 계대배양 시기에 발현되지 않았다. 결 론: 본 연구결과 탯줄유래 줄기세포는 체외 배양 시 줄기세포 특성이 일부 변하는 것을 관찰하였다. 앞으로 줄기세포의 특성을 유지할 수 있는 체외 배양법의 발달을 위한 연구들이 수행 되야 할 것으로 사료된다.

Nanosphere Form of Curcumin Stimulates the Migration of Human Umbilical Cord Blood Derived Mesenchymal Stem Cells

  • Curcumin, a hydrophobic polyphenol derived from turmeric, has been used a food additive and as a herbal medicine for the treatment of various diseases. In the present study, we found the functional role of a nanosphere loaded with curcumin (CN) in the promotion of the motility of human umbilical cord blood derived mesenchymal stem cells (hUCB-MSCs) during the wound closure. We found that the efficacy of hUCB-MSCs migration induced by CN was 1000-fold higher than that of curcumin powder. CN significantly increased the motility of hUCB-MSCs by activating c-Src, which is responsible for the phosphorylation of protein kinase C (PKC) and extracellular signal-regulated kinase (ERK). CN induced the expression levels of α-actinin-1, profilin-1 and filamentous-actin, as regulated by the phosphorylation of nuclear factor-kappa B during its promotion of cell migration. In a mouse skin excisional wound model, we found that transplantation of UCB-MSCs pre-treated with CN enhances wound closure, granulation, and re-epithelialization at mouse skin wound sites. These results indicate that CN is a functional agent that promotes the mobilization of UCB-MSCs for cutaneous wound repair.

사람의 골수와 제대정맥에서 유래된 중간엽 줄기세포에서 TREK1 통로의 기능적 발현 (Functional expression of TREK1 channel in human bone marrow and human umbilical cord vein-derived mesenchymal stem cells)

  • 사람의 골수 또는 제대정맥에서 유래된 중간엽 줄기 세포 (hBM-MSC 또는 hUC-MSC)는 임상적 치료 적용에 매우 유용한 세포유형으로 알려져 왔다. 우리는 이러한 세포에서 two-pore 도메인 포타슘 (K2P)채널을 조사하였다. K2P 채널은 다양한 세포유형들에서 안정막 전위를 형성하는데 중요한 역할을 한다. 그들 중 TREK1은 수소, 저산소증, 다불포화 지방산, 항우울제 및 신경전달물질들의 표적이다. 우리는 RT-PCR 분석과 팻취고정기법을 이용하여 hBM-MSCs와 hUC-MSC가 기능적인 TREK1 채널을 발현하는지 조사했다. hBM-MSCs와 hUC-MSCs에서 100 pS 단일 채널 전도도를 가진 포타슘채널이 발견되었고, 그 채널은 세포막 신전 (-5 mmHg ~ -15 mmHg), 아라키도닉산 ($10{\mu}M$), 세포내 산성화 (pH 6.0)에 의해 활성화 되었다. 이러한 전기생리학적 성질은 TREK1과 유사하였다. 우리의 결과는 안정막 전위에 기여하는 TREK1 채널이 hBM-MSC와 hUC-MSC에 기능적으로 존재하고 있음을 제시한다.

In Vitro Differentiation of Mesenchymal Progenitor Cells Derived from Porcine Umbilical Cord Blood

  • Mesenchymal stem/progenitor cells (MPCs) were isolated from porcine umbilical cord blood (UCB) and their morphology, proliferation, cell cycle status, cell-surface antigen profile and expression of hematopoietic cytokines were characterized. Their capacity to differentiate in vitro into osteocytes, adipocytes and chondrocytes was also evaluated. Primary cultures of adherent porcine MPCs (pMPCs) exhibited a typical fibroblast-like morphology with significant renewal capacity and proliferative ability. Subsequent robust cell growth was indicated by the high percentage of quiescent (G0/G1) cells. The cells expressed the mesenchymal surface markers, CD29, CD49b and CD105, but not the hematopoietic markers, CD45 and CD133 and synthesized hematopoietic cytokines. Over 21 days of induction, the cells differentiated into osteocytes adipocytes and chondrocytes. The expression of lineage specific genes was gradually upregulated during osteogenesis, adipogenesis and chondrogenesis. We conclude that porcine umbilical cord blood contains a population of MPCs capable of self-renewal and of differentiating in vitro into three classical mesenchymal lineages.
