갯방풍 지상부 정유 및 그 주성분의 항상제 감수성 및 내성 Streptococcus pneumoniae 균주에 대한 억제효과 (In Vitro Inhibitory Activities of Essential Oils from the Aerial Parts of Glehnia littoralis and its Main Components Against Antibiotic-Susceptible and -Resistant Strains of Streptococcus pneumoniae)

  • To develop a new natural antibiotics from Koran plant resources for dealing with the current situation regarding the rapid increase in antibiotic-resistant pathogen, the in vitro inhibitory activities of essential oils from the young leaves of Glehnia littoralis (Umbelliferae) as well as its main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae. The essential oil fraction of G. littoralis and its main components, $\alpha-and\;\beta-pinene$, exhibited significant inhibitory activities against the antibiotic-susceptible and resistant strains of S. pneumoniae, with MICs (minimum inhibiting concentrations) ranging from 4.0mg/ml to 16mg/ml. No remarkable differences were shown between the susceptible and resistant strains. Moreover, the disk diffusion test disclosed that these inhibitory activities were dose­dependent. Furthermore, data from the checkerboard titer test with FICIs (fractional inhibiting concentration indices) from 0.15 to 0.28 indicated synergisms between norfloxacin and $\alpha-or\;{\beta}-pinene$ in activity against S. pneumoniae KCCM49629 and S. pneumoniae CCARM4059.

기후대별 산림환경에 따른 참당귀의 생육 및 지표성분 특성 (Characteristic of Growth and Active Ingredient in Angelica gigas Nakai according to Forest Environment by Climate Zone)

  • Background: Angelica gigas Nakai, that belong to the Umbelliferae family, is one of the traditional medicinal plants in Korea. Its roots have been used to treat gynecological diseases. In this study, growth characteristics and index components were compared with the forest microclimate at several forest sites. Methods and Results: A. gigas was planted in three climatic zones according to the temperature (southern temperature zone - Hamyang, central temperature zone - Bonghwa, and northern temperature zone - Jeongseon) and growth characteristics were investigated in comparison with the forest microclimate. Our results indicated that the root diameter and length, and fresh and dry weight were the highest in Jeongseon. The total content of decursin was the highest in Jeongseon (9.52%), followed by those in Hamyang (8.07%) and Bonghwa (7.48%), respectively. Additionally, the yield of decursin (1.39 g) was the highest in Jeongseon. Conclusions: The yield and index components were influenced by the microclimate in the forests, and it was assumed that high altitude and low temperature affected the increase in growth and index components. These results will be useful as basic data to study the correlation among environmental conditions, growth, and index components.

The Effects of Glehnia littoralis on the Inflammatory mediators in Mouse Macrophage Cells

  • Glehnia littoralis (Umbelliferae) is the medicinal plant used traditionally for treatment of immune-related diseases. Prostaglandins and nitric oxide (NO) have been implicated as important mediators in the processes of inflammation and carcinogenesis. For understanding the mechanisms for pharmacological activities of Glehnia littoralis, we evaluated the inhibitory activity of Glehnia littoralis on lipopolysaccharide (LPS)-induced prostaglandin $E_2$ ($PGE_2$) and NO production in mouse macrophage RAW264.7 cells. The results showed that the extract of Glehnia littoralis inhibited LPS- induced $PGE_2$ production effectively, but not NO. Additional study revealed that the extract of Glehnia littoralis suppressed cyclooxygenase-2 (COX-2) expression in a dose-dependent manner. Present study suggests that Glehnia littoralis may have anti-inflammatory and/or cancer chemopreventive activity through the inhibition of $PGE_2$ production by the suppression of COX-2 activity.

Effects of Furanocoumarins from Angelica Dahurica on Aldose Reductase and Galactosemic Cataract Formation in Rats

  • The ether extract from the roots of angelica clahurica was found to inhibit bovine lens aldose reductase (BLAR0 activity in vitro by 100% at 100.mu.g/ml. Systematic fractionation of the ether soluble fraction and subsequent active frctions monitorede by bioassy led to isolation of four furanocoumarins, isoimperatorin (I0, imperatorin (II), ter-O-methyl byakangelicin (III) and byakangelicin (IV), among which compound III and IV were identified as potential AR inhibitors, their $IC_{50}$ values being $2.8{\mu}M{\;}and{\;}6.2{\mu}M$, respectively. Galactosemic cataract formation tors, in rats treated with 40 g/kg/day of galactose was blocked almost completely throughout the exeprimental periods up to 44 days by i.p. administrations of byakangelicin (IV) at 50 mg/kg/day. In coincidence with the inhibitory action on cataract formation, the galactitol accumulation in rats treated with byakangelicin (IV) was found to be markedly prevented by approximately 80.5% compared to those of the contro. These results indicate that byakangelicin (IV), as a main principle of this plant, possesses high potential for a clinically useful drug of the future which prevents and/or improves sugar cataract as well as diabetic complications.

Antibacterial activity of methanol extract of roots of Heracleum nepalense D Don. on bacteria causing diarrhoea

  • Heracleum nepalense D Don. (Umbelliferae) is a small shrub having high glabrescent stem found in stream banks in Sikkim. Various medicinal properties which include antidiarrhoeal, antiseptic, anti-influenzal etc. have been attributed for this plant in the traditional system of medicine in Sikkim. In present investigation the methanol extract of roots of Heracleum nepalense was subjected for its effectiveness against both Gram-positive and Gram-negative bacteria causing diarrhoea. The roots extract was tested for its minimum inhibitory concentration (MIC) against both Gram-positive and Gram-negative organisms causing diarrhoea. Further, the zones of inhibition produced by the crude extract against few sensitive strains was measured and compared with those of standard antibiotic ciprofloxacin. It is evident that the methanol extract is very active against the bacteria causing diarrhoea at low concentrations. The antibacterial efficacy of the root extract was found to decrease in the following order against different tested bacterial strains like Shigella dysenteriae, Escherichia coli, Shigella boydii, Vibrio cholerae, Salmonella typhimurium.

천궁 엑스 및 분획의 소염.진통작용 (Anti-inflammatory and Analgesic Activities of the Extracts and Fractions of Cnidii Rhizoma)

  • Cnidii rhizoma is one of the most important crude drugs used particularly for the treatment of female genital inflammatory diseases in traditional oriental medicine. In this study, its anti-inflammatory and analgesic activities were examined employing animal models. It was found that $H_2O$ extract and n-BuOH fraction inhibited significantly the edema formation after the subplantar injection of carrageenin at oral doses of 100 and 200 mg/kg in a dose-dependent fashion, where MeOH extract showed significant anti-inflammatory activity at a oral dose of 200 mg/kg. In Freund's complete adjuvant-induced arthritis, H2O extract and n-BuOH fraction exerted their significant inhibitory activity on the edema formation at oral doses of 100 and 200 mg/kg. The $H_2O$ extract and n-BuOH fraction also showed significant analgesic activity in a dose-dependent manner in acetic acid-induced writhing test.

사상자 미백성분 Torilin의 추출조건 최적화 (Optimization of Extraction Conditions of Torilin, a Melanogenesis Inhibitor from Torilis japonica Fruits)

  • Torilin is a major sesquiterpene of Torilis japonica (Umbelliferae) fruits and known to be a melanogenesis inhibitor. Extraction conditions are important factor for the efficient preparation to save cost and time in economic aspects. For this reason, this study was conducted to optimize the extraction condition for maximal yield of torilin. For optimization, extraction factors such as extraction solvent, extraction temperature and sample/solvent ratio were tested and optimized for maximum yield of torilin using response surface methodology with Box-Behnken design (BBD). The optimal condition was obtained as a EtOAc concentration in MeOH of 31.8%, an extraction temperature at $30.3^{\circ}C$ and a sample/solvent ratio, 1000 mg/2 ml. The torilin yield under optimal conditions was found to be 9.9 mg/g dried samples, which were well-matched with the predicted value of 10.4 mg/g dried samples. These results will provide useful information about optimized extraction conditions for the development of torilin as cosmetic therapeutics to reduce skin hyperpigmentation.

  • Aldose reductase(AR), a rate-limiting enzyme in the polyol pathway, has been demonstrated to cause the intracellular accumulation of sorbitol or galactitol and hence to play key roles not only in the cataract formation in the lens but also in the pathogenesis of diabetic complications such as neuropathy, retinopathy and nephropathy, etc. In a series of investigations to evaluate potential AR inhibitors from medicinal plants, we have shown that some hot water extracts exhibited a significant inhibition of a significant inhibition of bovine lens AR in vitro. Among active plants, the roots of Angelica dahuria (Umbelliferae) were shown to have relatively potent AR inhibitory activity. Systematic fractionation of the ether soluble fraction monitored by bioassay led to isolation of two furanocoumarins, byakangelicin(I) and ter-O-methyl byakangelicin( II), were identified as potential AR inhibitors, their $IC_{50}$ values being 6.2 M and 2.8 M, respectively.

천궁의 향신료로서의 이용 연구 (Studies on Cnidium officinale As Natural Spices)

  • 신선, 열풍건조, 동결건조 천궁(Cnidium officinale)을 대상으로 향신료로의 이용가능성을 검토하기 위한 기초 실험으로 건조방법을 달리한 천궁 자체에 대한 관능평가 및 이들이 육류의 누린내 및 생선의 비린내에 미치는 영향을 관능검사 하였다. 관능검사 결과 천궁자체에 대한 관능검사에서는 신선, 동결건조, 열풍건조 시료 순으로 전체적인 선호도가 좋게 나타났으며, 천궁의 산뜻한 향과 노르스름한 색에 대한 선호도가 높았다. 신선, 열풍건조 및 동결건조 천궁 0.1% 첨가는 돼지고기의 누린내 및 고등어의 비린내를 제거하였으며 전체적인 선호도도 무첨가군보다 유의적으로 높았다. 쇠고기 및 닭고기 stock에서도 건조 천궁을 첨가함으로써 stock의 누린내를 제거하면서 색이나 맛에 나쁜 영향을 미치지 않았다. 따라서 천궁은 강한 향을 선호하는 소비자에게 알맞은 향신료로 사용될 수 있다고 판단된다.

강활에서 종양괴사인자 생성을 억제하는 활성성분의 분리 (Isolation of Inhibitory Components on Tumor Necrosis Factor-${\alpha}$ Production from Angelica koreana)

  • The total ethanol extract of Angelica koreana radix (UmbeUiferae) showed potent inhibitory effect on tumor necrosis factors (TNF-${\alpha}$) production from LPS-stimulated RAW 264.7 cells in screening studies on 118 Korean medicinal plants. The activity-guided fractionation of the total ethanol extract resulted in the isolation of 4 compounds, including active substances. The chemical structures of the compounds isolated were established by chemical and spectrometric analyses as imperatorin, isoimperatorin, osthol and oxypeucedanin. Among them, osthol, oxypeucedariin and isoimperatorin showed significant inhibitory activities on TNF-${\alpha}$ production as 54.3${\pm}$2.1%, 33.6${\pm}$1.0% and 17.7${\pm}$3.0%, respectively, at 12.5${\mu}$g/ml.
