The Regulation of LexA on UV-Induced SOS Response in Myxococcus xanthus Based on Transcriptome Analysis

  • SOS response is a conserved response to DNA damage in prokaryotes and is negatively regulated by LexA protein, which recognizes specifically an "SOS-box" motif present in the promoter region of SOS genes. Myxococcus xanthus DK1622 possesses a lexA gene, and while the deletion of lexA had no significant effect on either bacterial morphology, UV-C resistance, or sporulation, it did delay growth. UV-C radiation resulted in 651 upregulated genes in M. xanthus, including the typical SOS genes lexA, recA, uvrA, recN and so on, mostly enriched in the pathways of DNA replication and repair, secondary metabolism, and signal transduction. The UV-irradiated lexA mutant also showed the induced expression of SOS genes and these SOS genes enriched into a similar pathway profile to that of wild-type strain. Without irradiation treatment, the absence of LexA enhanced the expression of 122 genes that were not enriched in any pathway. Further analysis of the promoter sequence revealed that in the 122 genes, only the promoters of recA2, lexA and an operon composed of three genes (pafB, pafC and cyaA) had SOS box sequence to which the LexA protein is bound directly. These results update our current understanding of SOS response in M. xanthus and show that UV induces more genes involved in secondary metabolism and signal transduction in addition to DNA replication and repair; and while the canonical LexA-dependent regulation on SOS response has shrunk, only 5 SOS genes are directly repressed by LexA.

Degradation of Dye Wastewater by Advanced Oxidation Process: A Comparative Study (고급산화공정에 의한 안료폐수 처리: 비교 연구)

  • The degradation of Rhodamine B (RhB) in water was investigated in laboratory-scale experiments, using five advanced oxidation Processes (AOPs) $UV/H_2O_2$, lenten, photo-lenten, $UV/TiO_2,\;UV/TiO_2/H_2O_2$. The photodegradation experiments were carried out in a fluidized bed photoreactor equipped with an immersed 32 W UV-C lamp as light source. initial decolorization rate and COD removal efficiency were evaluated and compared. The results obtained showed that the initial decolorization rate constant was quite different for each oxidation process. The relative order of decolorization was: photo-fenton > $UV/TiO_2/H_2O_2$ > fenton > $UV/H_2O_2$ > $UV/TiO_2$ > UV > $H_2O_2$. The relative order of COD removal was different from decolorization: photo-fenten ${\fallingdotseq}$ $UV/TiO_2/H_2O_2\;>\;UV/TiO_2\;>\;fenton\;>\;UV/H_2O_2$. The Photo-lenten and $UV/TiO_2/H_2O_2$ processes seem to be appropriate for decolorization and COD removal of dye wastewater.

Innate immune responses of common carp, Cyprinus carpio L. against antiviral activity inducers (항바이러스 활성 유도 물질에 대한 잉어의 선천성 면역 반응)

  • To investigate the innate immune response involved in early stage of anti-viral defence, carps were injected with UV-inactivated spring viraemia of carp virus (SVCV), poly inosinic:cytidylic acid (Poly I:C) and concanavalin A (Con A), respectively and examined lysozyme activity, serum complement activity and chemiluminescent (CL) response of leucocytes isolated from head kidney at 3 days post-injection. There was no significant difference in plasma lysozyme activities among all experimental groups. However, lysozyme activities of head kidney in the groups injected with antiviral activity inducers were significantly higher than those of the control injected with physiological saline. Bactericidal activities of serum of the groups injected with antiviral activity inducers were not significantly different from control group. However, the CL responses were significantly higher at lower dose of Poly I:C and Con A, whilst dose-dependent increase was shown in UV-inactivated SVCV-injected group. In the challenge test with 1×104 TCID50/fish of SVCV at 4 days post-injection, UV-inactivated SVCV- and Poly I:C-injected groups showed higher relative percent survival (RPS) than Con A-injected group. Furthermore, strong protection was observed in the group injected higher dose of Poly I:C although showed lower activities in lysozyme and CL response. These results suggested that Poly I:C might stimulate other factors belonging to non-specific immune system have induced protective immunity against the SVCV challenged.

A study of dry cleaning for metallic contaminants on a silicon wafer using UV-excited chlorine radical (UV-excited chlorine radical을 이용한 실리콘 웨이퍼상의 금속 오염물의 건식세정에 관한 연구)

  • The reaction mechanisms of dry cleaning with UV-excited chlorine radical for Zn, Fe and Ti trace contaminants on the Si wafer have been studied by SEM, AFM and XPS analyses in this work. The patterned Zn, Fe and Ti films were deposited on the Si wafer surface by thermal evaporation and changes in the surface morphology after dry cleaning with $Cl_2$and UV/$Cl_2$at $200^{\circ}C$ were studied by optical microscopy and SEM. In addition, changes in the surface roughness of Si wafer with the cleaning was observed by AFM. The chemical bonding states of the Zn, Fe and Ti deposited silicon surface were observed with in-line XPS analysis. Zn and Fe were easily cleaned in the form of volatile zinc-chloride and iron-chloride as verified by the surface morphology changes. Ti which forms involatile oxides was not easily removed at room temperature but was slightly removed by UV/$Cl_2$at elevated temperature of $200^{\circ}C$. It was also found that the surface roughness of the Si wafer increased after $Cl_2$and UV/$Cl_2$cleaning. Therefore, the metallic contaminants on the Si wafer can be easily removed at lower temperature without surface damage by a continuous process using wet cleaning followed by UV/$Cl_2$dry cleaning.

Degradation of Natural Dyed Silk Fabrics under Ultraviolet Light(UV) -Focused on Gardenia and Sappanwood- (자외선에 의한 천연 염색 견직물의 취화 연구 -치자, 소목 염색을 중심으로-)

  • The purpose of this study was to investigate the degradation of silk fabrics dyed with gardenia and sappanwood by Ultraviolet Light(UV). To asses the effect of uv on dyed silk fabrics, physical and chemical properties of samples were explored. K/S value rapidly decreased with increasing exposure time. Mordanting improved ultraviolet-cut ability and the sappanwood dyed samples were superior to those of gardenia dyed in ultraviolet-cut ability. Color progressively faded away as uv exposure time increased, accordingly, $L^*,\;a^*,\;b^*$, H/VC, ${\Delta}E$ were changed. Morphological change observed with SEM represented degradation of silk fabrics from the outer fibril to the inner fibril. Tensile Strength abruptly decreased as uv exposure time increased and the mordanted samples showed higher tensile strength than the unmordanted. FT-IR analysis confirmed that main peaks at 3297 and $1704cm^{-1}$ band for silk fabric were due to N-H and C=O stretching, gardenia peaks at 1654 and $668cm^{-1}$ band representing C=O(ester), C=C(alken) and O-C=O(carboxylic acids) of crocin and sappanwood peaks at $1715cm^{-1}$ band representing C=O(cyclic keton) of brazilin appeared on the samples exposed for 14 days, but these peaks indicating colorants after 28 days of uv exposure faded away due to prolonged exposure of uv.

Inactivation of Infectious Cryptosporidium parvum by Various Disinfectants (다양한 소독제에 의한 감염성 크립토스포리디움 불활성화율 평가)

  • Cell culture infectivity assay using HCT-8 cell was combined with most-probable-number technique to evaluate the inactivation of Cryptosporidium parvum by various disinfectants, including chlorine, ozone, and UV light. The assay was demonstrated to be as sensitive as animal infectivity assay, which has been considered the "gold standard" for assessing Cryptosporidium oocyst infectivity, and a valuable tool to evaluate inactivation of C. parvum by disinfectants. Bench-scale inactivation study showed that at the condition of $5^{\circ}C$ and pH 7.0, CT value of $1,250mg{\cdot}min/L$ by chlorine and $16mg{\cdot}min/L$ by ozone were required to achieve approximately 1.0 log inactivation of C. parvum, suggesting that even ozone could not be sufficient to inactivate C, parvum at low. temperature. Unlike chlorine and ozone, UV light is very effective to inactivate C. parvum, regardless of temperature. A UV light dose of 2 $mJ/cm^2$ provided at least 3 log inactivation of C. parvum.

  • Investigation of improving the properties of UV detector which uses the wide bandgap of ZnO are under active progress. The present study focused on the design and fabrication of i-ZnO/p-inversion $layer/n^--Si$ Epi. which is characterized with very thin p-type inversion layer for UV detectors. The i-ZnO thin film for achieving p-inversion layer which was grown by RF sputtering at $450^{\circ}C$ and then annealed at $400^{\circ}C$ in $O_2$ gas for 20 min shows good intrinsic properties. High (0002) peak intensity of the i-ZnO film is shown on XRD spectrum and it is confirmed by XPS analysis that the ratio of Zn : O of the i-ZnO film is nearly 1 : 1. Measurement shows high transmission of 79.5 % in UV range (< 400 nm) for the i-ZnO film. Measurement of $V_r-I_{ph}$ shows high UV photo-current of 1.2 mA under the reverse bias of 30 V.

  • Vinylbenzyloxystyrylpyridine (VSP) as a photosensitive monomer was synthesized by the reaction of 4-hydroxystyrylpyrydine with 4-vinylbenzyl chloride. Photocrosslinkable polymers containing rosin moiety were then prepared by radically copolymerizing VSP with a methacrylic monomer having rosin moiety. In these copolymerizations, the VSP feed ratios of 5 to 20 mol% were used. The contents of VSP units in the copolymers were determined by UV spectroscopy to be 5.3 to 17.3 mol%. The numberaverage molecular weights of these polymers were in the range of 18000 to 28000 and the polydispersity indexes were about 1.8. The glass transition temperatures were about 15$0^{\circ}C$ and the initial decomposition temperatures were about 34$0^{\circ}C$. The polymers were relatively fast photocrosslinked and the photocrosslinking reaction could be traced by the UV spectroscopy and the residual yield method.

Changes of Rutin Content and Photosynthesis Rate of Korean Buckwheat Cultivars under Various Environmental Stresses

  • This study was conducted to classify photosynthesis rate and changes of rutin content of Korean buckwheat (cv. Chunchon-jaerae and Yangjeul-memil) treated with salinity, UV-C and low temperature. In case of cv. Chunchon-jaerae and Yangjeul-memil, according to the salt stress, transpiration rate, stomatal conductance and photosynthesis rate were decreased. Both cultivars also showed decrease of transpiration rate and photosynthesis rate under the UV-C. Rutin contents within leaf and stem of cv. Yangjeul-memil were decreased when NaCl concentration was high. Rutin contents within leaf and stem of cv. Yangjeul-memil were generally decreased when the time laps under the UV-C stress. Rutin contents within leaf and stem of cv. Chunchon-jaerae was also generally decreased when the time laps under the low temperature stress.

Quantitation and Validation of Atorvastatin using HPLC-UV

  • A reversed phase HPLC analysis of atorvastatin (AS) standard solution was performed using diclofenac (DF) as internal standard. Column oven temperature, flow rate and the composition of the mobile phase were varied in order to determine a practical system setup using a C18 column and UV detector. Two C18 columns of different length were compared regarding their influence on the AS peak shape. Based on these preliminary experiments a validation study was performed utilizing a C18 column at $62^{\circ}C$ with a mobile phase consisting of sodium phosphate buffer (0.05 M, pH 4.0), methanol and acetonitrile (40:50:10, v/v/v). The detection limit for AS was $0.1{\mu}g/ml$ and inter- and intra-day calibration curves were linear over a concentration range of $0.2-50{\mu}g/ml$. Accuracy and precision were satisfactory in the AS concentration range of $0.5-50{\mu}g/ml$.
