Microbial Inactivation in Kimchi Saline Water Using Microwave Plasma Sterilization System (Microwave Plasma Sterilization System을 이용한 배추 절임수의 미생물 저감화)

  • This study was conducted to decrease the microbial hazard in kimchi saline water with microwave plasma sterilization system and to evaluate the inactivation of foodborne pathogens by the microwave plasma sterilization system as a non-thermal treatment. Contamination of coliform, Escherichia coli, and yeasts and molds were detected in the used saline water, and the microbial populations increased as the saline water was reused repeatedly. The $D_{10}$-values of E. coli O157:H7, Salmonella Typhimurium, and Listeria monocytogenes by the microwave plasma sterilization system were 0.48, 0.52, and 0.45 cycle, respectively. In addition, the microbial populations of coliform, E. coli, Salmonella spp., total aerobic bacteria, and yeasts and molds in the used kimchi saline water were significantly decreased by treating the saline water using the microwave plasma sterilization system. Therefore, these results suggest that microwave plasma sterilization system can be useful in improving the microbial safety of the used saline water.

Effects of UV-C and Ethanol Treatment on Quality of Fresh-cut Seedless Watermelon (UV-C, 에탄올 처리가 신선편이(Fresh-cut) 씨 없는 수박의 품질에 미치는 영향)

  • To apply UV-C as a non-heating sterilization method to increase the microbiological safety of fresh seedless watermelon products, reductions in E. coli and quality changes by treatment dose (0, 2, 4, 8, 14, 20 kJ/m2) were investigated. The pH, sugar content, and hardness of watermelon inoculated with E. coli were not significantly different according to the UV-C treatment dose, but the polyphenol content was significantly decreased compared to the controls (425.4 GAE ㎍/g F.W.). When treated with 2 and 4 kJ/m2, the lycopene content was 31.6 and 30.9 ㎍/g F.W., respectively, which was increased compared to the controls (28.5 ㎍/g F.W.). The arginine and citrulline content was also significantly increased compared to the controls. The number of E. coli was significantly decreased compared to the controls following UV-C treatment. Considering the degree of E. coli reduction, lycopene content, arginine content, citrulline content, and UV-C irradiation time, subsequent experiments were conducted by selecting a UV-C treatment dose of 2 kJ/m2. The results of confirming the degree of reduction in the number of E. coli colonies by a single treatment and combined treatment with UV-C 2 kJ/m2 and 70% ethanol showed that the combined treatment was most effective as colonies were decreased by 2.3 log CFU/g compared to the controls. Therefore, it is judged that UV-C 2 kJ/m2 radiation and combined treatment with 70% ethanol could be applied as a non-heating sterilization method for fresh watermelon slices.

  • HEMA, AA, MMA, and EGDMA as crosslinking agent and AIBN as an initiator, and 2,4-dihydroxybenzophenone, 2-ethylhexyl-trans-4-methoxy-cinnamate, 2-hydroxy-4-(methacryloyloxy)benzophenone as additives at 0.1-1.0% ratios were used to manufacture hydrophilic ophthalmic lenses through thermal polymerization before their physical properties were measured. The results showed that the samples containing of 2,4-dihydroxybenzophenone and 2-ethylhexyl-trans-4-methoxy-cinnamate resulted in a decrease of the UV blocking performance after high-pressure sterilization whereas the sample containing 2-hydroxy-4-(methacryloyloxy)benzophenone showed no change in the UV blocking performance. It is judged that this is induced by presence or absence of an acyl functional group in benzophenone.

  • In this study, macroporous titania powders were synthesized utilizing the emulsion-assisted self-assembly to apply the removal of B. subtilis under UV irradiation, and the results were compared with the bactericidal effect of commercial titania nanoparticles. By changing the pore size of the porous titania powder, the reduction of B. subtilis by photocatalytic effect was measured, and the bactericidal capacity of the porous particles according to the pore size was compared in order to derive the optimum condition of the sterilization experiment. It was observed that the sterilization effect increased as the pore size became smaller, and it was confirmed that more than 50% of B. subtilis cold be removed for 1 hour of UV irradiation. Also, in order to promote the generation of active chemical species, a diluted solution of hydrogen peroxide was combined with the photocatalytic sterilization method, resulting in the removal of most of the strain after ultraviolet irradiation for 1 hour.

Effect of different soybean meal type on ileal digestibility of amino acid in weaning pigs

  • An experiment was conducted to evaluate apparent (AID) and standardized (SID) ileal digestibilities of crude protein (CP) and amino acids (AA) with 6 soybean products in weaning pigs. A total of 14 weaning barrows with an initial body weight of $6.54{\pm}0.34kg$ were fitted with T-cannula at the distal ileum and allotted to 7 diets containing various soybean products. The soybean products used in the experiment were conventional soybean meal (CSBM), SBM fermented by Aspergillus oryzae GB-107 (FSBMA), SBM fermented by Bacillus subtilis PP6 (FSBMB), UV sterilized SBM fermented by Bacillus subtilis PP6 (UVFSBMB), SBM containing Bacillus subtilis PP6 (PSBM), and soy protein concentrate (SPC). Six corn-based diets were used and each of soybean products was added. All diets contained 5.0 g/kg of chromic oxide as an indigestible indicator and an N-free diet was used to measure basal endogenous losses of CP and AAs. Ileal CP digestibility did not differ by different soybean products. However, SIDs of Ile, Phe and Val were improved in pigs fed the FSBMB, UVFSBMB and SPC diets and the pigs fed the FSBMA diet showed higher SIDs of Phe and Val compared with those fed the CSBM diet (P < 0.05). The FSBMB diet had higher SIDs in most AAs compared with the FSBMA diet (P < 0.05), and higher SIDs of Lys, Ala, Pro, Ser, and Tyr compared with PSBM diet (P < 0.05). However, there was no response of UV-sterilization on the FSBMB in the SIDs of AAs. These results suggest that SIDs of AAs could be improved by the supplementation of fermented soybean products in the diet for weaning pigs but fermentation with Bacillus subtilis is more efficient in improving ileal AA digestibility than that with Aspergillus oryzae. Furthermore, probiotics supplementation in the CSBM and UV-sterilization of the FSBMB had no effects on chemical composition and ileal AA digestibility.

Microbial reduction effect of steam heating, UV irradiation, and gamma irradiation on red pepper powder (스팀 가열 및 자외선, 감마선 조사 처리에 따른 고춧가루의 미생물 저감화 효과)

  • This study evaluated the effect of steam heating, gamma irradiation, and ultraviolet (UV) irradiation on microorganism reduction in order to determine an effective sterilization method for red pepper powder. The effect of each treatment on the reduction of thermoduric bacteria and total aerobic bacteria in red pepper powder were as follows: 10 kGy gamma irradiation, reduction of 4 log and 6 log CFU/g, respectively; 12 mW/㎠ UV irradiation (264 nm UV-C), reduction of less than 1 log CFU/g; steam heating at 120℃ for 40 s, reduction of approximately 2 log CFU/g. High-temperature short-time processing at 110℃ for 30 s reduced the total bacterial count in Gochujang solution from 5.70 log CFU/g to 2.26 log CFU/g; at 121℃, the solution was commercially sterile. Steam heating resulted in 1, 2, and 4 log microbial inactivation in garlic, onion, and pepper powder, respectively. Steam sterilization, which consumers prefer over other methods, may be an effective method for reducing microorganisms in spice powders, including those in red pepper powder.

  • The aim of this research was microbiological hazard analysis which is required to application of HACCP(Hazard Analysis Critical Control Point) system to chili powder. A processing had been conducted in Yong-Geum Agricultural Production located in Dongnyang-myeon, Chungju-si between January 10, 2014 to June 13, 2014. A manufacturing process chart was prepared by entire level of processing. A changes of microorganism was tested by changing sterilization time in ultraviolet ray sterilization processing which can control microbiological Hazard. As a result, critical Limits is decided as ultraviolet rays lamp $20W{\times}12EA$, sterilization $63{\pm}3$ min of each. The result of the microorganism test after ultraviolet ray sterilization was safe. On the other hand, a microorganism test of manufacturing environment and workers suggested that the microbiological hazard should be reduced through systematic cleaning and disinfection accompanied by personal hygiene based on hygiene education on workers.

Studies on the improvement of UV sterilizer for toothbrush (가정용 칫솔 UV 살균기의 성능 개선법에 관한 연구)

  • For the purpose of improving the UV sterilization for toothbrush, the sterilizing effects by the UV light were studied. When toothbrushes were treated with the UV light for 5, 15, 30, 60 minutes, the sufficient antibacterial effect were shown after 15 minutes with 80% of bactericidal effects. To prevent the brownish stain on the sterilizer which was caused by the long exposure of UV light, no UV treatment for two hours after 15 minutes exposure were scheduled and then 3 minutes re-exposure were performed, which showed 97% of bactericidal effect. Also, the antibacterial effects of the sterilizer showed different results depending on the position of the toothbrush, it was recommended to adjust toothbrush to be exposed by the UV light directly.
