Electrochemical degradation of phenol was evaluated at DSA (dimensionally stable anode), JP202 (Ru, 25%; Ir, 25%; other, 50%) electrode for being a treatment method in non-biodegradable organic compounds such as phenol. Experiments were conducted to examine the effects of applied current (1.0~4.0 A), electrolyte type (NaCl, KCl, $Na_2SO_4$, $H_2SO_4$) and concentration (0.5~3.0 g/L), initial phenol concentration (12.5~100.0 mg/L) on phenol degradation and $UV_{254}$ absorbance as indirect indicator of by-product degraded phenol. It was found that phenol concentration decreased from around 50 mg/L to zero after 10 min of electrolysis with 2.5 g/L NaCl as supporting electrolyte at the current of 3.5 A. Although phenol could be completely electrochemical degraded by JP202 anode, the degradation of phenol COD was required oxidation time over 60 min due to the generation of by-products. $UV_{254}$ absorbance can see the impact of as an indirect indicator of the creation and destruction of by-product. The initial removal rate of phenol is 5.63 times faster than the initial COD removal rate.
Photoreactivation of microorganism following UV-disinfection is one of the research topics of interest in assessing the UV-disinfection performance. Apparent photoreactivation was examined under fluorescent lamp and solar radiation as well as in darkness. Total coliform, fecal coliform, and Escherichia coli were used as indicator microorganisms, and their concentration was monitored with time after UV-disinfection. Under the darkness, their initial concentration of 10∼30 MPN/100 mL increased to the level of 100 MPN/100 mL after 24 hours, which implied that part of damaged microorganisms by UV-disinfection might be repairable with time. Under the fluorescent lamp, photoreactivation was more apparent that their concentration increased up to 1,000 MPN/100 mL which might significantly impair the water uses specially in reuse of reclaimed wastewater. However, their concentration further decreased down to below 2 MPN/100 mL under the solar radiation primarily due to additional disinfection by solar radiation rather than photoreactivation. Samples not disinfected by UV-disinfection also demonstrated substantial decrease of their concentration under solar radiation from about 5,000 MPN/100 mL to less than 30 MPN/100 mL in 24 hours. But direct reuse of effluent without disinfection is not recommended because natural decay by solar radiation may take time and be affected by climatic conditions. The result suggests that photoreactivation of pathogenic microorganisms may not be concerned in agricultural reuse of reclaimed wastewater because solar radiation may provide further disinfection after UV-disinfection.
The aim of this research was to evaluate the effect of combination of disinfection process (electrolysis, UV process) on Escherichia coli (E. coli) disinfection and oxidants (OH radical, $ClO_2$, HOCl, $H_2O_2$ and $O_3$) generation. The effect of electrolyte type (NaCl, KCl and $Na_2SO_4$) on the E. coli disinfection and oxidants generation were evaluated. The experimental results showed that performance of E. coli disinfection of electrolysis and UV single process was similar. Combination of electrolysis and UV process enhanced the E. coli disinfection and 4-carboxybenzaldehyde (4-CBA, indicator of the generation of OH radical) degradation. It is clearly showed synergy effect on disinfection and OH radical formation. However chlorine ($ClO_2$, HOCl) and oxygen type ($H_2O_2$, $O_3$) oxidants were decreased with the combination of two process. In electrolysis + UV complex process, electro-generated $H_2O_2$ and $O_3$ were reacted with UV light of UV-C lamp and increased 4-CBA degradation(increase OH radical). Disinfection of electrolyte of chlorine type was higher than that of the sulfate type electrolyte due to the higher generation of OH radical and oxidants.
The aim of this study was to evaluate the difference of analytical methods for the pharmacokinetic study of bromosulfophthalein (BSP), an indicator of hepatobiliary function. The plasma and bile concentrations of BSP after intravenous administration were measured according to custom UV spectroscopy and HPLC, respectively. Plasma concentration of BSP measured by UV spectroscopy was similar to that measured by HPLC. There was no significant difference in the distribution volume, total body clearance, area under the curve and mean residence time of BSP between different analytical method groups. However, bile concentration of BSP measured by UV spectroscopy was overestimated compared with concentration measured by HPLC method. Biliary clearance of BSP obtained from UV spectroscopy method was almost 3 times higher than that obtained from HPLC method. Thus, a feasibility of UV spectroscopy method for high throughput pharmacokinetic evaluation of BSP was limited to the study based on the plasma concentration of BSP, not bile concentration.
The monthly-mean irradiance of ultra violet (UV)-B and UV-A observed from 2005 to 2014 and 2012 to 2014, respectively, at noon in Gosan, Jeju, South Korea are analyzed. We compare cloudiness, total ozone, visibility, and relative humidity with an emphasis on the four months from May to August (MJJA), which shows the largest UV radiation. While the incoming UV-B radiation at the top of the atmosphere in Gosan is the largest in June due to the small solar zenith angle, the observed surface UV-B shows an unexpected smaller value in June than those in May, July or August. In June, the meteorological conditions affecting Gosan are completely dominated by cloudiness and thus, frequent overcast seems to determine the minimum UV-B. Another important UV-determining factor is the total ozone, which exhibits a monotonic decrease during MJJA without agreeing to the characteristic feature of UV. The ratio of UV-B to UV-A is not generally influenced by cloudiness. Thus, the ratio is a useful indicator of atmospheric turbidity showing larger values for increasing visibility, except in June. A simple model has been used to estimate surface UV by using the observed ozone and visibility in the cloudless condition. The result shows that UV has the lowest value in June with small variation during MJJA. Model estimation also shows that the different characteristic features observed in July between surface UV-B and UV-A is the result of less absorption of UV-B by ozone than that of UV-A by a smaller amount of total ozone.
The effects of UV-B radiation on the seedling growth, carbohydrate metabolism and antioxidants activities of rice (Oryza sativa L.) were investigated under environmentally controlled chamber. Supplementary UVB radiation reduced dry matter as well as leaf area, therefore, relative growth rates (RGR) of seedlings were decreased by up to half compared to control. Photosynthetic products such as soluble sugars and starch were rapidly and significantly reduced by within 1 day of enhanced UV-B radiation due to the inhibition and degradation of photosynthetic processes and thylakoid membrane integrity. In our study, nonstructural carbohydrate levels were proved to be a main indicator on UV-Binduced stress. The behavior of SOD, CAT, APX and POD activities was monitored in the leaves of rice seedlings subjected to UV-B radiation. Under UV-B treatments, SOD activity was initially increased, whereas CAT and POD activities were slowly and slightly increased. However, APX activity showed no presumable results with an increase of UV-B dose. In leaves of rice seedlings, supplementary UV-B radiation caused an increase in free putrescine and spermidine, however spermine remained unaltered, although 24-hrs UV-B treatment slightly increased. This result presumes that an excess UV-B dose may induce ethylene biosynthesis (senescence) rather than polyamine biosynthesis (defense).
The simple and convenient measurement of the dissociation constant of an indicator, thymol blue, was achieved by using a portable diode-laser/fiber-optic thermal lensing spectroscopy, which consisted of a visible diode laser, a photodiode, and an optical fiber. It gives comparable results to the cited value obtained from a conventional UV/VIS spectroscopy.
