미생물 효소에 의한 핵산 및 그의 관련물질의 분해에 관한 연구 (Studies on Degradation of Nucleic acid and Related Compounds by Microbial Enzymes)

  • 핵산 및 그 관련물질 연구의 일환으로서 미생물 효소에 의하여 핵산을 분해하여 5'-mononucleotides 생성을 목적으로 미생물에서 5'-phosphodiesterase 생성균주를 얻기위하여 전국 각지 76개 지역에서 논, 밭, 산, 하천의 토양 그리고 퇴비, 누룩, 메주 등 시료 210종을 수집하였다. 이 수집 시료로 부터 dilution pour plate method 로서 Aspergillus속 240주, Penicillium속 232주, Neurospora속, 19주, Monascus속 16주, 그리고 Streptomyces속 265주로 총 758주를 순수 분리하였다. 분리된 모든 균주에 대하여 RNA-depolymerase 균의 crude enzyme 중에는 5'-AMP deaminase가 공존하고 있으므로 효소반응 중에 RNA 가 5'-mononucleotide로 분해 축적하는 동안에 5'-AMP의 adenine의 6위치에 붙은 $NH_2$기를 탈아미노하여 hypoxanthine으로 하기 때문에 5'-IMP 로 되는 것이라 생각된다. 분리 동정 된 Penicillium citreo-viride PO 2-11 strain과 Streptomyces aureus SOA 4-21 strain 이 생성한 5'-phosphodiesterase 로서 RNA를 효소 분해하여 5'-mononucleotide 의 정량한 결과는 Table 10과 같다. productivities를 1차 screening 하고 5'-phosphodiesterase productivities로서 2차 screening 하여 우수균주를 얻고 동정하였다. 우수균주의 5'-phosphodiesterase productivity에 대하여 배양상의 optimum condition을 검토하고 5'-phosphodiesterse activity에 미치는 여러 화합물의 영향과 효소반응의 최적 조건을 구명하였다. 우수균주가 생성한 5'-phosphodiesterase에 의하여 RNA 분해로 반응 최종 산물을 ion exchange column chromatography법으로 정량하고 최종 분해 산물엔 5'-mononucleotide를 paper chromatography, thinlayer chromatography, UV-absorption spectra, carbazole reaction 및 Schiff's reaction 등으로 동정한 결과는 다음과 같다. [1] 5'-phosphodiesterase productivity가 가장 우수한 두 균주를 선정하였고 이들은 토양에서 분리 되었으며 선정된 푸른 곰팡이는 Penicillium citreoviride PO 2-11로 동정되었고 방사선균은 Streptomyces aureus SOA 4-21로 동정되었다. [2] 분리 선정 된 Penicillium citreo-viride PO 2-11 strain은 배양상의 optimum condition 이 pH 5.0이고 temperature는 $30^{\circ}C$이었고 이 균이 생성한 5'-phosphodiesterase의 효소 반응상의 optimum condition 이 pH 4.2이고 temperature는 $60^{\circ}C$이었다. 그리고 5'-phosphodiesterase 생성에서 최적 탄소원은 sucrose이고 질소원은 $NH_4NO_3$이고 corn steep liquor나 혹은 yeast extract를 각각 0.01%씩 첨가한 구는 첨가하지 않은 control 구보다 20%의 5'-phosphodiesterase 생성 증가를 나타 내었다. 이 균이 생성한 5'-phosphodiestrase는 $Mg^{++},\;Ca^{++},\;Zn^{++},\;Mn^{++}$ 등 금속이온은 activator이고 EDTA, citrate, $Cu^{++},\;Co^{++}$ 등은 inhibitor 임을 알았다. 이 균이 생성한 5'-phosphodiesterase는 RNA를 분해하여 분해율 65.81%이었고 5'-AMP, 5'-GMP, 5'-UMP 및 5'-CMP를 생성하며 이때 축적되는 5'-mononucleotides중 5'-GMP 만이 정미성이 있음을 알았고 이균은 5'-AMP deamaminase가 없음을 확인하였다. 이 균의 효소에 의하여 RNA에서 정미성 5'-GMP 186.7 mg/RNA(g)를 생산할수 있음을 알았다. [3] 분리 선정된 Streptomyces aureus SOA 4-21 strain은 배양상의 optimum condition이 pH 7.0이고 temperature는 $28^{\circ}C$이었고 이 균이 생성한 5'-phosphodiesterase의 효소반응상의 optimum condition이 pH 7.3이고 temperature는 $50^{\circ}C$이었다. 그리고 5'-phosphodiesterase 생성에서 최적탄소원은 glucose이고 질소원은 asparagine이고 yeast extract 0.01%첨가구가 control 구보다 40%의 5'-phosphodiesterase 생성증가를 나타내었다. 이 균이 생성한 5'-phosphodiesterase는 $Ca^{++},\;Zn^{++},\;Mn^{++}$ 등 금속이온은 activator 이고 citrate, EDTA, $Cu^{++}$ 등은 inhibitor 임을 알았다. 또한 이 균은 5'-phosphodiesterase 뿐만 아니라 5'-AMP deaminase도 생성함을 확인하였다. 그러므로 RNA 분해율은 63.58% 이었고 RNA를 분해하여 5'-AMP, 5'-CMP, 5'-GMP 및 5'-UMP로 축적시키고 RNA가 효소 분해됨과 동시에 5'-AMP deaminase도 작용하며 생성된 5'-AMP 의 60% 상당을 5'-IMP로 전환시키는 특성이 있어서 정미성 5'-mononucleotide 생성이 전자의 균보다 두드러지게 증가함을 밝혔다. 이 균에 의하여 RNA에서 정미성 5'-IMP 171.8 mg/RNA(g) 및 5'-GMP 148.2 mg/RNA(g)생산할 수 있어 정미성 5'-mononucleotide 320mg/RNA(g)를 생산할 수 있음을 알았다.

영아자 (Phyteuma japonicum Miq)의 성분 조성 (Chemical Components of Wid and Cultivated Horned Rampion, Phyteuma japonicum Miq.)

  • This research is to establish the basic data of the nutritive value and improve our diet. In the part of the leaf and stem of the wild and cultivated horned rampion (Phyteuma japonicum Mig), the components such as chemical compositon, vitamin C, free sugar, mineral, nucleotide and its related compounds, composition and free amino acid were analyzed one after another. Content of the crude lipids and proteins was determined much higher in its wildness than in its cultivated horned rampion; while, that of carbohydrates was higher in the former than in the latter. The content of vitamin C was retained higher in the leaf than in the stem horned rampion. And the content of calcium among the detected minerals was outstanding in all of the samples collected, and potassium and magnesium was the next ones in its order. The main components of free sugars in both the wild and cultivated horned rampion were glucose and fructose, and their content was higher in the stem than in the leaf. Nucleotide and its related compounds were identified with 5 kinds of nucieotides such as CMP, UMP, IMP, AMP and hypoxanthine (Hx), and the content of Hx and AMP was the highest I the wild and cultivated samples, respectively. In the composition amino acid of the wild horned rampion, glutamic acid, aspartic acid and phenylalanine was outstandingly abundant; while, such amino acid as methinone and proline was small and besides cysteine couldn't be detected in the stem. Total amounts of composition amino acid in the leaf was 2118.0 and 1120.1mg% in the wild and cultivated sample, respectively. In the free amino acid of horned rampion, the total amount ranged from 8.5 to 50.1mg.%, which were lower level than that of composition amino acid. But the number of free amino acid was 29 kinds, which was bigger in its number than that of composition amino acid detected 17 kinds.

수치모델을 이용한 와편모조류 Alexandrium tamarense, Gymnodinium catenatum 및 규조류 Skeletonema costatum의 종간 경쟁에 미치는 용존태 유기인의 영향 (Use of n Mathematical Model to Assess the Effects of Dissolved Organic Phosphorus on Species Competition Among the Dinoflagellates Alexandrium tamarense and Gymnodinium catenatum and the Diatom Skeletonema costatum)

  • Species competition among the toxic dinoflagellates Alexandrium tamarense and Gymnodinium catenatum and the diatom Skeletonema costatum was simulated using a mathematical model. Prior to the model simulation competition experiments using the three species were conducted to obtain data for validation by the simulation model. S. costatum dominated at a density of ${\sim}10^{4}\;cells/mL$ compared to the other species in the medium with dissolved inorganic phosphorus (DIP). The growth of S. costatum was also stimulated by the addition of dissolved organic phosphorus (DOP), such as uridine-5-monophosphate (UMP) or glycerophosphate (Glycero-P), although this species is unable to take up DOP. This implies that the growth of S. costatum may be supported by DIP, which is hydrolyzed by alkaline phosphatase produced from A. tamarense and G. catenatum. The species competition model was run assuming the environmental conditions of northern Hiroshima Bay, Japan, during spring and summer. G. catenatum increased in cell density and neared the level of S. costatum at the end of the calculation. In the sensitivity analyses by means of doubling and halving parameters, depleted DIP had little effect on the cell density of G. catenatum. However the growth of A. tamarense and S. costatum was significantly affected by changes in the parameter values. These results indicate that if DIP depletion is ongoing, species that have a large phosphate pool in their cells, such as G. catenatum, will predominate in the community.

A Novel Approach to Cloning and Expression of Human Thymidylate Synthase

  • Thymidylate synthase (TS) catalyzes the transfer of a methyl group from methylenetetrahydrofolate to dUMP to form dTMP. It is a primary target in the chemotherapy of colorectal cancers and some other neoplasms. In order to obtain pure protein for analysis of structure and biological function, an expression vector TS-pET28b (+) was constructed by inserting wild-type human thymidylate synthase (hTS) cDNA into pET28b (+). Then an expression strain was selected after transformation of the recombined plasmid into Rosetta (DE3). Fusion protein with His-tag was efficiently expressed in the form of inclusion bodies after IPTG induction and the content was approximately 40.0% of total bacteria proteins after optimizing expression conditions. When inclusion bodies were washed, dissolved and purified by Ni-NTA under denatured conditions, the purity was up to 90%. On SDS-PAGE and West-blotting, the protein band was found to match well with the predicted relative molecular mass-36kDa. Bioactivity was 0.1 U/mg. The results indicated that high-level expression of wild-type hTS cDNA can be achieved in prokaryotes with our novel method, facilitating research into related chemotherapy.

담수어(淡水魚)의 식품학적(食品學的) 연구(硏究)(II)-붕어, 메기, 가물치 및 미꾸리의 맛성분(成分)- (Studios on the Food from Fresh Water Fish(II)-The Taste Compounds in Meat of Crucian Carp, Skate$\cdot$Fish, Snake Head and Loaches-)

  • Crucian carp Carassius carassius L., Skate-fish Parasilurus asolus, snake head Ophicephalus argus and loaches Misgrunus anguillicaudatus have been esteemed as the tasty fresh water fish in Korea. However, a little has been studied on their taste compounds. Amino acids, nucleotides and their related compounds as the taste giving substance in the dorsal muscle of crucian carp, skate-fish, snake head and loaches were analyzed. Hypoxanthine (4.6-30.3 ${\mu}mole/g$, on dry base) was dominant among the nucleotide materials in the dorsal muscle of crucian carp, skate-fish, and snake head while IMP (12.8 ${\mu}mole/g$) was detected with the highest amount in loaches meat. IMP was also high in the muscle of skate-fish (13.5 ${\mu}mole/g$) and snake head (5.6 ${\mu}mole/g$). The other nucleotide materials, CMP, UMP, GMP, AMP, and inosine were present less than 2.0 ${\mu}mole/g$ in all the fish meat. The amino acid composition showed that glutamic acid and aspartic acid were the most abundant in the dorsal muscle of crucian carp, skate-fish, snake head and loaches. The analysis of free amino acids showed that histidine, lysine, glycine, and serine were the leading amino acids in the dorsal muscle of crucian carp, alanine, serine, and lysine were of skatefish, glycine, lysine, alanine, and serine were of snake head, while alanine, serine, and glycine were of loaches. Isoleucine and leucine were detected in trace amount in the muscle of skate-fish, snake head, and loaches. It is believed that alanine, Iysine, serine, glycine and histidine along with IMP and hypoxanthine may play an important role as the taste compounds in fresh water fish.

Simvastatin이 UMR-106 세포의 조골세포 형성에 미치는 영향 (Effects of Simvastatin on osteogenesis of rat osteoblast-like cells, UMP-106)

  • Purpose : The purpose of this study is to investigate the effects of Simvastain, which is HMG-CoA reductase inhibitor, on proliferation and differentiation of osteoblast. Materials & Methods : Twenty-four cell culture plates containing essential medium were seeded with UMR-106 cell lines, at density of 5 x $10^4$ cells per plate. Each plates were incubated with 5% $CO^2$incubator $37^{\circ}C$. Starting from 2 days after incubation, cell culture medias were replaced with Osteogenesis induction media every 2 days, for 12 days. In some plates, 0.01, 0.1, 1, 10, $100\muM$ of Simvastatin were added with Osteogenesis induction media, and classified as "test group". Those not added with Simvastatin were classified as "control group". Results : 1. When Alrizarin Red staining was observed with naked eye, control group showed normal deep red color, but test group show rapid decrease of red color as Simvastatin concentration increased more than $0.1\muM$. 2, When observed with microscope, compared to control group, amount of osteo matrix stained with Alrizarin Red decreased rapidly in Simvastatin concentration more than $0.1\muM$. 3. In optical density analysis, regarding control group as a basis, mineral deposition decreased rapidly when Simvastatin concentration increased more than $0.1\muM$. 4. In flow cytometry analysis, survival rate of UMR-106 cell showed no changes in both control group and test group. Conclusion : From the above results, we were able to identify that Simvastatin inhibited osteogenesis without effecting survival or cell number of osteoblasts.

협력적 스펙트럼 공유의 자동 반복 프로토콜 (An Automatic Repeating Protocol in Cooperative Spectrum Sharing)

  • 본 논문에서는, NACK 메시지가 스펙트럼 공유와 협력을 위한 명령으로 사용되어지는 방법을 제안하였다. 1차 사용자의 직접 연결이 중단될 때, 협력과 공유의 자동 반복을 허가하고, 패러다임 기반의 협력 스펙트럼 공유의 조정 메시지의 수를 절약하기 위해 허가한다. 공유 시, ${\alpha}$의 남은 전력 부분이 1차 재전송 신호인 동안, 2차 전송된 신호를 위한 $1-{\alpha}$ 전력 부분은 중계기 공유로 선택된다. 중계기를 사용하지 않는 경우, 1차 송신단은 전체 전력($1-{\alpha}$)를 사용하여 신호를 재전송 하기 위해 NACK를 사용한다. 두 시스템은 BPSK 신호를 적용한 것으로 가정한다. 이 기법에서, 2차 사용자는 공동 최적화 복호를 하는 것으로 제안한다. 프레임 오류율(FER) 성능은 양 시스템에서 분석된다. 이론과 시뮬레이션 결과는 본 프로토콜의 유효성을 분석하고 효율적임을 확인하였다.

Possible Roles of the Xenobiotic Transporter P-glycoproteins Encoded by the MDR1 3435 C>T Gene Polymorphism in Differentiated Thyroid Cancers

  • Background: P-glycoprotein (Pgp), encoded by the multidrug resistance 1 (MDR1) gene, is an efflux transporter which plays an important role in pharmacokinetics. The current preliminary study was designed to determine associations between a germ-line polymorphism in the MDR1 gene with differentiated thyroid carcinoma (DTC). Materials and Methods: In the current case-control study, 60 differentiated thyroid cancers (DTC)- 45 papillary TC (PTC), 9 follicular TC(FTC) and 6 well-differentiated tumors of uncertain malignant potential (WDT-UMP) were examined. Results were compared to a healthy control group (n=58) from the same population. Genomic DNA was extracted from peripheral blood with EDTA and the target gene was genotyped by real-time PCR. Results: Carriers of the variant allele of MDR1 exon 26 polymorphism were at 2.8-fold higher risk of DTC than the control group (odds ratio [OR]: 0.3805, 95% confidence interval [Cl]: 0.1597-0.9065 (p> 0.046). Conclusions: Presented results suggest that the MDR1 3435TT genotype might influence risk of development of DTC and that the CC genotype might be linked to a poor prognosis. Large-scale studies are now needed to validate this association.

진주담치 건조중의 Nucleotides의 변화 (DEGRADATION OF NUCLEOTIDES IN THE MUSCLE OF SEA MUSSEL DURING DRYING)

  • 진주담치의 건조중의 nucleotides의 변화를 알기 위하여 생체시료, 열풍건조시료 및 자숙열풍건조시료에 대하여 분석 검토하였다. 1. ATP 함양은 $8.2{\mu}moles/g(dry basis)$ 로서 높았고, 건조후외 잔존율도 $30\%$ 이상으로 많았다. 2. ADP 및 AMP는 건조중에 축적되는 결과를 보여 nucleotides 총량(ATP+ADP+AMP)으로 볼때는 건조중에 크게 감소하지 않았다. 3. 분획물중 UMP 및 adenosine이라고 추정되는 성분이 있었으나 동정하지 못하였다. 4. IMP의 생성은 볼 수 없어 AMP deaminase 활성은 없다고 보며, inosine 및 hypoxanthine 함양은 건조중에 증가하므로 adenosine deaminase 활성은 있다고 볼 수 있다. 5. 진주담치의 ATP 주분해경로는 그 분해생성물로부터 볼때 $ATP{\to}ADP{\to}AMP{\to}adenosine{\to}inosine{\to}hypoxanthine$이라고 추정된다.

수동 소나 쌍을 이용한 에너지 인식 분산탐지 체계의 설계 및 성능 분석 (Design and Performance Analysis of Energy-Aware Distributed Detection Systems with Two Passive Sonar Sensors)

  • 이 논문에서는 한 쌍의 수동 광대역 소나로 구성된 병렬구조 분산센서 망에 적용할 수 있는 에너지 인식 분산탐지 체계의 설계 및 성능분석에 대해 검토한다. 이 분산탐지기는 융합규칙으로 AND 규칙과 OR 규칙을 사용하였다. 이 경우 각 센서노드의 문턱값 규칙은 전체 분산센서망의 탐지확률을 최대로 하는 UMP(uniformly most powerful)임을 증명하였다. 그리고 소나 체계에서 오경보에 의해 발생하는 에너지 비용을 제한 조건으로 주었을 때, 탐지확률을 최대화하는 센서노드의 최적 문턱값이 갖는 특성을 이론적으로 검토하였다. 나아가 신호 강도, 에너지 비용에 대한 제한조건, 센서노드 사이의 간격등이 탐지성능에 미치는 영향을 수치실험을 통해 분석하였다.
