Effects of Pueraria radix in Water Extract on the Detoxification in Rat administered with Cadmium (카드뮴을 급여한 흰쥐에서 갈근 열수 추출액의 해독작용효과)

  • This study was designed to investigates the effects of Korean pueraris radix water extract in Cd(cadmium) administered rats. Forty male Sprague-Dawley rats weighing $100{\pm}10g$ were used for this experiment and divided into following 4 groups; control group, 3% pueraria radix in water extract group, 50 ppm Cd group, 50ppm Cd group with 3% pueraria radix in water extract group. The Cd administered rats were given 50 ppm of $CdCl_2\;{\cdot}\;2H_2O$ disolved in the distilled water. The Cd content in the rats tissue of Cd administered group was lower than in the rats tissue of Cd group with 3% pueraria radix in water extract group. Plasma levels of renin activity was increased by Cd administration group, compared with 3% pueraria radix in water extract group and Cd administred group. Glutamate oxaloacetate transaminase(GOT) and Glutamate pyruvate transaminase(GPT) were increased in Cd-administered group and lower in the 3% extracts of pueraria radix in water extract group. Lactate dehydrogenase(LDHase) was lower in the 3% extracts of pueraria radix-Cd group than in the Cd group. This results suggested that pueraria radix in water extract group, has a lowering effects on the accumulation of Cd and it is belived that the pueraria radix in water extract group has some protective effects to Cd administered in rats, but the mechanism of these effects was obscure.

Anti-Obesity Effect of Crataegus Fructus Extract from Chinese Cultivation (중국산 산사자 추출물의 항비만 효과)

  • This study was carried out to evaluate the antiobesity effects of Crataegus fructus in 3T3-L1 adipocytes and mice fed a high fat diet (high fat 45% cal). The inhibitory effect of methanol extract from Crataegus fructus on lipid accumulation in 3T3-L1 adipocytes was quantified using Oil red O staining. Compared with the control, lipid accumulation was significantly decreased by 10-25% with treatment with Crataegus fructus extract at a concentration of 600-2,000 ug/ml. Three-week old ICR mice (n=24) were randomly divided into four groups (T0: normal diet, T1: high fat diet, T2: high fat diet and 50 ug of Crataegus fructus extract, T3: high fat diet and 100 ug of Crataegus fructus extract) and were fed an experimental diet for 5 weeks. At the end of the experiment, body weight gain in the T1 group (3.9${\pm}$0.24 g) was higher than that in the T0 group (2.56${\pm}$0.14 g), while body weight gain in the T2 (3.02${\pm}$0.25 g) and T3 (2.58${\pm}$0.16 g) groups was significantly reduced as compared with that of the T1 group. Moreover, liver weight in the T1 (4.8${\pm}$0.17 g) and T2 (4.8${\pm}$0.16 g) groups was significantly higher than that of the T0 (4.05${\pm}$0.16 g) and T3 (4.57${\pm}$0.10 g) groups, while kidney weight was significantly lower than that of the T0 and T3 groups (p<0.05). The levels of total cholesterol and triglyceride in serum in the T2 and T3 groups were significantly decreased compared to the T1 group. These results suggest that Crataegus fructus can be used as functional materials in food and medicine.

Changes in the Contents of Major Minerals and Trace Elements of Human Milk During the Breast-Feeding (수유기간별 모유의 주요 무기질 및 미량원소 함량 변화)

  • Concentrations of 5 major minerals(Na, K, Ca, P, Mg) and 3 trace elements(Mn, Mo, Ni) were measured in human milk samples collected from 21 highly selected healty lactating women at 2-5 days and at 1, 2, 4, 6 and 12 weeks postpartum. Significant decrease in contents of the major minerals and trace elements were found with time postpartum. Sodium contents decreased from 293.01ug/N at 2-5 days to 194.27ug/N at 12 weeks. Potassium contents also decreased from 436.18ug/N in colostrum to 358.51ug/N in matured milk at 12 weeks. The Ca/P ratios of colostrum transitional and matured milk were 3.39, 281 and 2.45 respecti-vely. Mean manganese levels of colostrum and transitional were 0.024ug/N and those of matured milk were 0.014ug/N. Molybdenum concentrations in the breast-milk collected at 1, 2, 4 weeks were higher than those in the milk samples at 2-5 days and 12 weeks Nickel content of colostrum was 0.062ug/N and those of trnsitional matured milk were 0.22ug/N and 0.017ug/N, These determinations will provide the basic information on the variability of minerals and trace elements as lactation proceed and the comparison of the components between term and preterm milk.

Evaluation of QuEChERS Method for Determination of pesticide Residues Using GC/NPD and GC/ECD (GC/NPD와 GC/ECD를 이용한 잔류농약 정량 분석법으로써의 QuEChERS의 평가)

  • The modified QuEChERS method was evaluated for rapid determination of pesticide residues in spinach by gas chromatography-nitrogen phosphorous detector (NPD) and electron capture detector (ECD). Fifty GC-amenable pesticides which were most frequently detected in monitoring were selected in the current study. Matrix-matched calibration was performed. The detector response for all pesticides was linear with determination coefficients higher than 0.995. LODs for most compounds ranged between 0.001 and 0.1 ug/g, and about 90% of the compounds had LODs of less than 0.05 ug/g. LOQs ranged from 0.001 to 0.32 ug/g, which were well below the MRLs permitted for most of pesticides. In the majority of cases, the recoveries (80-120%) and relative standard deviations (RSDs) (less than 20%) were within acceptable levels except for dichlorvos, propamocarb, chlorothalonil, dichlofluanid, cyhalothrin and fenvalerate. Also, this method which is applied to routine samples showed good results when comparing with traditional multi-residue method.

Estrogen activity of Silkworm (Bombyx mori) Pupa water extract and its fractions

  • This study was conducted to evaluate the estrogen activity of silkworm (Bombyx mori) pupa extracts and their fractions. Powdered samples of freeze-dried silkworm pupa were extracted at room temperature (RT), $40^{\circ}C$, $60^{\circ}C$, $80^{\circ}C$, and $100^{\circ}C$ in water (D.W), chloroform, ethyl acetate, and methanol for 6h and then filtered (0.45 um). The extracts were then freeze-dried. The estrogenic activity of these extracts was then investigated by competition binding assays using estrogen receptor ${\alpha}\;(ER{\alpha})$ and $ER{\beta}$, and by evaluating their effects on the proliferation of the human breast cancer cell line, MCF-7. Among the extracts evaluated, water extracts prepared at RT showed the highest binding affinity to $ER{\alpha}$ ($IC_{50}$, 1.76 ug/ml) and $ER{\beta}$ ($IC_{50}$, 0.07 ug/ml). In addition, MCF-7 cells that were treated with 62.5 ug/ml of the RT extract showed the greatest increase in proliferation (2-fold; 1291.79%) when compared to control cells (659.82%). Next, the water extract that was prepared at RT (sample 1) was dissolved in D.W. and further fractionated using a Dowex 50W - 8X ($H^+$) column. The flow-through and wash were then pooled together and freeze-dried (sample 2). The bound materials were then eluted with 20 mM NaCl, after which they were applied to a Dowex 1X2 - 200 ($Cl^-$) column and washed with D.W. to remove the sodium ions. The eluants were then freeze-dried (sample 3). Of these fractions, sample 2 showed the highest binding affinity to ER{\alpha} ($IC_{50}$, 1.44 ug/ml) and $ER{\beta}$ ($IC_{50}$, 1.18 ug/ml). In addition, MCF-7 cells that were treated with sample 2 (15.6 ug/ml) showed the largest increase in growth (1159.39%) when compared to control cells (525.26%). Taken together, these results suggest that the fraction of the RT water extract of silkworm pupa referred to as sample 2 may be useful as a phytoestrogen.

Effects of Amyda sinensis on Allergic Inflammation Mechanism related Atopy Dermatitis (별갑이 아토피 피부염에서의 알러지성 염증 반응에 미치는 영향)

  • Objectives: This study aimed to investigate the effects of Amyda sinensis (AS) on allergic inflammation mechanism related atopy dermatitis. Methods: To investigate the effects of AS, We study inhibitory effect of AS on the levels of pro-inflammatory cytokines released from Raw264.7 cell stimulated with LPS (lipopolysaccaride), and EoL-1, THP-1, Jutkat cell stimulated with DP (Dermatophagoides pteronyssinus), and LPS indused acute inflammatory BALB/c mouse model. Result: AS reduced the levels of IL-$1{\beta}$ released from Raw264.7 cell stimulated with LPS at 20 ug/ml, 5 ug/ml concentration, and reduced the levels of IL-6 in a dose-dependent. AS significantly reduced the levels of MCP-1 released from EoL-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at all the concentration, and significantly reduced the level of IL-8 at 0.1 ug/ml concentration. AS significantly reduced the levels of MCP-1 released from THP-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at 1 ug/ml concentration, and reduced the level of IL-6 in a dose-dependent. AS significantly reduced the levels of IL-4 released from Jutkat cell stimulated with DP at all the concentration, and significantly reduced the level of IL-5 at 0.1 ug/ml. 1 ug/ml concentration,. and reduced the level of TNF-${\alpha}$ in a dose-dependent. AS significantly reduced the levels of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, in LPS indused acute inflammatory BALB/c mouse model, in a dose-dependent. Conclusion: These result suggested that AS has suppressive effect on pro-inflammatory cytokines in various cell lines through the regulation of immune system. AS could be applied on the medicinal sources for treatment of immune abnormal diseases such as atopy dermatitis afterward.

Determination of ethyl carbamate in maesil wine by alcohol content and ratio of maesil (Prunus mume) during ripening period (알코올 농도 및 담금비에 따른 숙성 기간별 매실주의 에틸카바메이트 함량조사)

  • This study was designed to investigate the formation of ethyl carbamate (EC) during the ripening of Maesil with sugar and Soju (19.5~35% alcohol contents) using a homemade method. Maesil, sugar and Soju were purchased at ordinary market in June of 2012. The preparation of sample for analysis was conducted by method of Henry et al. The analysis of GC/MS was used SIM mode (m/z 89, 74, 62). Quantification was performed in terms of the 62 ion and was based on an internal standard procedure. Good linearity was obtained with a regression coefficient ($r^2$ = 0.993). Low detection limits (LOD) was achieved 4.31 ug/kg and recovery for alcohol was 74.8%. During 90 days, fermentation with sugar was not detected EC (under LOQ). 15 days ripened Maesil wine contained EC between non detected~32.7 ug/kg and 90 days ripened Maesil wine was 19.7~87.4 ug/kg. Higher proportion of Maesil and Soju increased EC contents. EC levels were increased 32.7 ug/kg to 87.4 ug/kg in the ratio of Maesil to 35% alcohol-Soju (1:1). In the ratio of Maesil to 35% alcohol-Soju (1:3) was increased non detected to 69.7 ug/kg. After 90 days, Maesil wine was filtered Maesil through a seive and ripened by 180 days to investigate the formation of EC compared with non filtered. Treatment of filtered, EC contents was much higher level compared with non filtered. Therefore, this result showed that alcohol contents contribute to increase EC formation more than Maesil.

  • A biological active substance was isolated from the cultured medium of Streptomyces sp. and its biochemical characteristics were investigated. Isolation process of the substance was as follows; the pH of filterate of the cultured medium was adjusted to 3.0 with N-hydrochloric acid and saturated with sodium chloride, then chloroform was added to this filterate in one fifth portions and stirred vigorously. After extracting the active substance with chloroform in 3 stages, the chloroform layer combined and evaporatea after dehydrating with sodium sulfate. The substance was found to be to be toxic to various fresh water fishes; the lethal dose for an average size Pseudorasbora parva T. et. S. was 50ug per ml. In the acidic condition, the toxicity of the substance remained fora long time, while in the alkaline state, the toxicity was decreased very fast. This substance was found to be stable to organic solvents, but labile to heat treatment. The maximal revival time of Pseudorasbora parva T. et. S. was about 20 minutes in 25 ug/ml of the substance solution.

The Formaion of N-nitrosamine in Soy Sauce, Soybean Paste and Beer under Simulated Gastric Digestion (간장, 된장 및 맥주의 인공소화시 N-nitrosamine의 생성)

  • The aim of the study was to analyze N-nitrosamine (NA) and its precursors in serveral fermented foods which were treated with nitrite, thiocyanate and ascorbic acid under simulated gastric digestion. Every analyzed sample contained nitrate, with levels ranging from 0.3 to 1.3 mg/kg, but nitrite was present at very low levels of less than 0.3 mg/kg. And other precursors of amines such as dimethylamine and trimethylamine were detected less than 0.5 mg/kg in every samples. N-nitrosodimethylamine (NDMA) was detected in the levels of <0.5 ∼ 2.7 ug/kg in soy sauce,1.5∼3.1 ug/kg in soybean paste and <0.5∼1.8 ug/kg in beer, while NDMA levels increased by 1.1∼4.5 times in the fermented foods which were digested under simulated gastric conditions.

Annual Change and C:N:P ratio in Particulate Organic Matter in Chinhae Bay, Korea (한국진해만 입자유기물 함량과 C:N:P 비의 연변화)

  • An investigation of the annual change and C:N:P ratio in particulate organic matter (POM) in Chinhae Bay, a semi-enclosed bay of the southern coast of Korean Peninsula, was carried out for a period of 12 months between January and December, 1993. The concentrations of POM have a broad range: 198∼4,416 ugC/l, 24∼792 ugN/l and 4.5∼69.0 ugP/l, Marked seasonal changes of POM, particularly particulate organic carbon (POC) and nitrogen (PON), were observed in the surface water. Generally, the concentration of POM peaks in summer. The C:N:P composition ratio of particulate organic matter, which is high in summer, also shows a seasonal change. The C:N assimilation ratio is constant at 6.53, which is consistent with the Redfield ratio. The significant linear relationship between POM and chlorophyll-a in the surface water during the survey period (except for January and February) and the C:N ratio suggest that the concentration of POM is controlled by phytoplankton biomass. POM peaks in summer, a period characterized by high freshwater input and the strong stratification, as a result of the intense proliferation of phytoplankton by a large amount of nutrient loading from the tributaries. On the other hand, the high C:P and N:P ratios in summer indicate that P is limited for phytoplankton growth owing to N-enrichment from a high input of freshwater with a high dissolved inorganic N:P ratio.
