  • The optimum conditions and mechanisms for the plasmid-mediated genetic transformation of intact cells of Bacillus brevis Pl76-2, an extracellular protein producing bacterium by electroporation were investigated. It was found that pUB110 Plasmid DNA can be introduced into intact bacterial cells by electroporation. The frequency of transformation by this electroporation system depended upon the initial electric field strength, the capacity of the electric discharge capacitor, growth stage, number of successive pulses and composition of electroporation buffer. It was effective for transformation that cells were harvested, washed and resuspended with HSM [7M HEPES(PH 7.4), 272mM sucrose, 1 mM MgCl2] electroporation buffer when cell growth was attained to 1.2 at OD660. A maximum frequency of transformation of 2.40$\times$104 transformants per$\mu$g plasmid DNA was obtained by two succesive Pulses with an initial electric field strength of 12.5kV/cm and with a capacitance of 7.3uF.

  • In present, advanced countries in the world are seen concentring on Military Transformation to create new military capability as there is a shift in Warfare Paradigm. Rapid development of technology enables military system, operational concept and organization innovated. As a result, Revolution in Military Affairs(RMA) that dramatically increased warfighting capability is pursued. Especially, advanced information technology constructs system of systems with abilities of battle visualization, information sharing in battlefield, long-range strike capability that are interoperable and combined. Corresponding to this, engagement methods and organizations are being progressively developed. The purpose of this thesis is overview of newly developing Warfare Paradigm and analysis of tendency of Military Transformation that advanced countries are pursuing preparation to this. Above all, specific aspects and contents of the Warfare Paradigm which emerged by information age were summarized. Subsequently, Military Transformation which provides the basic concepts and principles to the advanced countries in the world for the creation of new military capability was discussed. In conclusion, the tendency of Military Transformation by U.S.A, Japan, China, and Russia surround the Korean peninsular as the major power in military was carefully observed.

  • The aim of this paper is to establish the well-known and very useful classical Saalsch$\ddot{u}$tz's theorem for the series $_3F_2$(1) by following a different method. In addition to this, two summation formulas closely related to the Saalsch$\ddot{u}$tz's theorem have also been obtained. The results established in this paper are further utilized to show how one can obtain certain known and useful hypergeometric identities for the series $_3F_2$(1) and $_4F_3(1)$ already available in the literature.

  • Using the shuttle vector pECCGl between Escherichia coli and Corynebacterium glutamicum and C. glutamicum strain JS231 grown in the medium supplemented with penicillin-G, which inhibits the formation of cross-links in the peptidoglycan of bacterial cell wall, various parameters involved in electroporation system including resistance, electric field strength, capacitance, DNA concentration, and cell density were investigated independently and optimized for the high efficiency transformation of coryneform bacteria. Using cells grown with 0.3U/ml of penicillin-G and harvested at A600 of 0.7-0.8, transformation efficiencies of 107-l08 transformants/$\mu\textrm{g}$ of DNA with Corynebcctertum glutamicum strain JS231 and wild type ATCC13032 were achieved under conditions of 12.5kV/cm of electric field strength, 400 ohms of resistance, $25\mu$F of capacitance, 3$\times$108 cells per transformation(1.2$\times$1010 cells/ml) and 100ng of plasmid DNA per transformation.

  • Observation of standard stars is of crucial importance in stellar photometry. We have studied the standard transformation relations of the UBV RI CCD photometric system at the Maidanak Astronomical Observatory in Uzbekistan. All observations were made with the AZT-22 1.5m telescope, SITe 2k CCD or Fairchild 486 CCD, and standard Bessell UBV RI filters from 2003 August to 2007 September. We observed many standard stars around the celestial equator observed by SAAO astronomers. The atmospheric extinction coefficients, photometric zero points, and time variation of photometric zero points of each night were determined. Secondary extinction coefficients and photometric zero points were very stable, while primary extinction coefficients showed a distinct seasonal variation. We also determined the transformation coefficients for each filter. For B, V, R, and I filters, the transformation to the SAAO standard system could be achieved with a straight line or a combination of two straight lines. However, in the case of the U filter and Fairchild 486 CCD combination, a significant non-linear correction term - related to the size of Balmer jump or the strength of the Balmer lines - of up to 0:08 mags was required. We found that our data matched well the SAAO photometry in V, B - V, V - I, and R - I. But in U - B, the difference in zero point was about 3.6 mmag and the scatter was about 0.02 mag. We attribute the relatively large scatter in U -B to the larger error in U of the SAAO photometry. We confirm the mostly small differences between the SAAO standard UBV RI system and the Landolt standard system. We also attempted to interpret the seasonal variation of the atmospheric extinction coefficients in the context of scattering sources in the earth's atmosphere.

  • Alpha-SiAlON ceramics having various compositions and modifying cations were investigated with respect to their phase stability, transformation kinetics. and resulting microstructures. Each composition was heat treated at 150$0^{\circ}C$ for 1h and measured the $\alpha$-SiAlON transformation. The phase-boundary composition in the single-phase $\alpha$-SiAlON region showed sluggish transformation from $\alpha$-$Si_3N_4$ to $\alpha$-SiAlON compared to the phase-center composition in the diagram. Using the different rare earth modifying cations, dependence of transformation kinetics on the phase stability in a fixed composition was also explained. By changing size of the stable u-phase region with exchanging cations, systematic change in transformation was observed. Transformation rate of $\alpha$-SiAlON at low temperature has an important role on controlling the final microstructure. Less transformation gives more chances to develop elongated grain in the microstructure.

  • The low-lying resolved resonance structure of U-233 is investigated in terms of the Adler-Adler multilevel formalism. The resonance capture and fission cross sections of U-233 below 60 eV are calculated using Adlers' effective resonance parameters. The infinite dilution resonance integrals of U-233 are computed with the use of the Alders' parameters adjusted to fit the cross section data. It is found that the agreement of calculations with experiments is generally good over most of the energy region covered. A transformation of the Adlers' multilevel parameters into an equivalent set of the single level pseudoparameters is made for the use of the existing computer codes which are useful in the reactor calculations but do not have capability to use the multilevel parameters. The results of this transformation are presented in the form of a table.

  • In this paper, we study the problem of transforming to normality. We propose to estimate the transformation parameter by minimizing a weighted squared distance between the empirical characteristic function of transformed data and the characteristic function of the normal distribution. Our approach also allows for other symmetric target characteristic functions. Asymptotics are established for a random sample selected from an unknown distribution. The proofs show that the weight function $t^{-2}$ needs to be modified to have thinner tails. We also propose the method to compute the influence function for M-equation taking the form of U-statistics. The influence function calculations and a small Monte Carlo simulation show that our estimates are less sensitive to a few outliers than the maximum likelihood estimates.
