Dopaminergic neurons of the substantia nigra and ventral tegmentum in the stripped field mouse(apodemus agrarius coreae) (야생등줄쥐 흑색질 및 배쪽피개의 dopamine성 신경세포)

  • The distributions characteristics of neurons displaying immunoreactivity to the catecholamine synthetic enzymes, tyrosine hydroxylase(TH), dopamine-${\beta}$-hydroxylase(DBH), and phenylethanolamine-N-methyltransferase(PNMT) were examined in the adjacent sections of the substantia nigra & ventral tegmentum of the Striped Field Mouse(Apodemus agrarius coreae). None of these cell groups displayed either DBH or PNMT immunoreactivity. Many TH-immunoreactive neurons were present in the substantia nigra & ventral tegmentum. The major dopaminergic cell(TH-positive, DBH- & PNMT-negative) group in the midbrain was present in the pars compacta of substantia nigra and adjacent ventral tegmentum. And smaller dopaminergic cell groups Were found in the pars reticulata of the substantia nigra and central liner nucleus.

Effect of Intracerebroventricular Administration of Ethylcholine Aziridinium (AF64A) on Dopaminergic Nervous Sys-tems

  • Changes in dopaminergic activities were investigated after the intracerebroventricular (icv) administration of ethylcholine aziridium (AF64A) in rats. The levels of dopamine (DA) and metabolites, the activities of tyrosine hydroxylase (TH) and monoamine oxidase (MAO), and the specific binding sites of dopamine receptros in striata, hippocampus, and frontal cortex were assessed 6 days after the AF64A treatment with 3 nmol/each ventrcle. In frontal cortex, the levels of DA and metabolities were significantly decreased without changes in metabolites/DA ratios in the AF64A-treated groups. In contrast, the ratios of metabolites/DA were significantly decreased in striatum and hippocampus in the AF64A treatment. The activity of TH in frontal cortex was significantly decreased. However, that in other areas was not changed. Also the activity of MAO-A was not changed in the studied brain regions. However, the activity of MAO-B in striatum was significantly increased with no change in other areas. The specific binding sites of dopamine D1 and D2 receptors were increased in AF64A-treated frontal cortex. However, those were not changed in striatum and hippocampus except the small decreased specific binding sites of dopamine D-1 receptors in striatum after AF64A treatment. These results indicate that the dopaminergic activity was altered in AF64A treatment. Furthermore, it suggest that the decreased dopaminergic activities in each brain regions might be differently affected by AF64A treatment.

  • The carotid sinus nerve (CSN) is well known as mediating baroreflexes. However, studies of its detailed histological analysis are scant in the literature. Therefore, the current anatomical study sought to better elucidate the microanatomy of the CSN. Ten fresh frozen adult cadavers underwent dissection of the CSN. Then, it was harvested and submitted for histological and immunohistochemical staining. Specimens were all shown to be nerve fibers on histology and immunohistochemistry. We identified tyrosine hydroxylase positive fibers in all CSN specimens. These fibers were always found to be within the CSN and not on its surface i.e., epineurium. Based on our findings, the majority of fibers contained in the CSN are tyrosine positive in nature. Further studies are necessary to understand the true function of this autonomic nerve fibers.

Antistress effects of Lycii Radicis Cortex in the passive avoidance test and the forced swimming test (지골피(地骨皮)가 스트레스로 인한 기억저하와 우울행동에 미치는 영향)

  • Objectives : Investigation of the anitstress effects of Lycii Radicis Cortex. Methods : Behavior was measured in PAT and FST after applying stress to rats. Also, TH expression-degree in the regions of VTA and LC was measured, respectively. Results : The memory was improved in the LH 50mg/kg treated group in the passive avoidance test(P<0.01). The duration of immobility in the forced swimming test was significantly decreased in the LR 50mg/kg and 400mg/kg treated groups(P<0.0l). Stress-induced TH increases were meaningfully suppressed in the LR 100mg/kg and 400mg/kg treated group(P<0.001). Conclusion : Lycii Radicis Cortes has the effect of memory improving and antidepressant.

Exofocal Damage to the Substantia Nigra by Transient Middle Cerebral Artery Occlusion in Rats

  • The present study examined chronic effects of transient focal cerebral ischemia on the substantia nigra, a remote exofocal area, using immunohistochenmical and receptor autoradiographic techniques. Transient focal cerebral ischemia was induced by middle cerebral artery (MCA) occlusion for 60 or 90 min followed by reperfusion using silicone-coated 4-0 nylon monofilament in male Wistar rats. After 1- or 2-week reperfusion following transient MCA occlusion, there were partial losses of tyrosine hydroxylase-immunoreactive dopaminergic neurons, incieases in glial fibrillary acidic protein-immunoreactive cells (gliosis), decreases in [$^3$H]YM-09151-2 binding for dopamine D$_2$ receptors, and marked atrophy in the ipsilateral substantia nigra. The precise mechanism(s) of exofocal damage to the substantia nigra is remained to be elucidated.

Genetically Modified Human Embryonic Stem Cells Relieve Symptomatic Motor Behavior in a Rat Model of Parkinson′s Disease

  • Embryonic stem cells have several characteristics suitable for cell replacement therapy. To investigate a possibility of using human embryonic stem cell (hESC) as a carrier of therapeutic gene(s), hESC (MB03) was co-transfected with cDNAS coding for tyrosine hydroxylase (TH) and GTP cyclohydrolase Ⅰ (GTPCH Ⅰ) and bulk-selected using neomycin and hygromycin-B. Successful transfection was confirmed by western immunoblotting and RT-PCR. The genetically modified hESC (bk-THGC) relieved apomorphine-induced asymmetric motor behavior by approximately 54% when grafted into striatum of 6-OHDA-denervated rat brain. The number of rotation, however, increased up to 176+18% in 6 weeks when sham-grafted compared with number of rotation before graft. Immunohistochemical staining revealed that the grafted hESC survived and expressed TH for at least 6 weeks while the experiment was continued.

  • The vocal fold has three major function-phonation, respiration and protection, and is richly innervated. The vocal 1314 its autonomic innervation-adrenergic and cholinergic from superior cervical ganglion and the vagus nerve, respectively. The action of both system account for vasoregulation and glandular activity. In e vocal fold several kin of neuropeptides, including SP, CGRP, VIP, TH, NPY, ENK have been reported at the animal including cat or dog. But information regarding the distribution of autonomic nerve fibers containing neuropeptides in the human vocal fold is lacking. Two neuropeptides are of special interest : 1) vasoactive intestinal polypeptide(VIP)that is known to be contained in the parasympathetic(cholinergic) neuron. 2) tyrosine hydroxylase(TH)is located in the cytoplasm of noradrenergic neuron and is the rate-limiting enzyme in noradrenaline synthesis. To understand specific autonomic function of vocal fold we did immunohistochemical examination of VIP and TH in the human vocal fold.

Tricyclic Antidepressants Amitriptyline and Desipramine Induced Neurotoxicity Associated with Parkinson's Disease

  • Recent studies report that a history of antidepressant use is strongly correlated with the occurrence of Parkinson' disease (PD). However, it remains unclear whether antidepressant use can be a causative factor for PD. In the present study, we examined whether tricyclic antidepressants amitriptyline and desipramine can induce dopaminergic cell damage, both in vitro and in vivo. We found that amitriptyline and desipramine induced mitochondria-mediated neurotoxicity and oxidative stress in SH-SY5Y cells. When injected into mice on a subchronic schedule, amitriptyline induced movement deficits in the pole test, which is known to detect nigrostriatal dysfunction. In addition, the number of tyrosine hydroxylase-positive neurons in the substantia nigra pars compacta was reduced in amitriptyline-injected mice. Our results suggest that amitriptyline and desipramine may induce PD-associated neurotoxicity.
