The melanin pigment in human skin is a major defense mechanism against ultraviolet light to the skin, but darken skin color. Tyrosinase is mainly responsible for melanin biosynthesis (melanogenesis) in animals and enzymatic browning (melanosis) in plants. The purpose of this study was to optimize the fermented milk process for the melanin formation inhibition by using Lactobacillus plantarum M23 with tyrosinase inhibitory activity. We used 4-factor-3-level central composite design combining with response surface methodology. Yeast extract concentration (%, $X_1$), addition of grape (%, $X_2$), incubation temperature ($^{\circ}C$, $X_3$) and incubation time (h, $X_4$) was used as an independent factor, on the other hand, pH (pH, $Y_1$), overall palatability (score, $Y_2$) and tyrosinase inhibitory activity (%, $Y_3$) was used as a dependant factor. Based on the optimization for the highest tyrosinase inhibitory activity with pH 4.4, the expected data of pH, palatability and tyrosinase inhibitory activity with 14.8 h incubation at $37.1^{\circ}C$ by the addition of 0.127% of yeast extract, 2.95% of grape was 4.42, 7.06 and 86.65%, but the real data was 4.35, 6.86 and 84.05%, respectively. Based on the previous results, fermented milk using Lactobacillus plantarum M23 with the tyrosinase inhibitory activity could contribute for the whitening and antiaging of human skin.
To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.
This study was performed to investigate the effects of acteoside and isoacteoside isolated from Clerodendron trichotomum Thunberg on melanin production in B16 melanoma cells. In DPPH radical scavenging activity, acteoside and isoacteoside had a potent anti-oxidant activity in a dose-dependent manner. Both acteoside and isoacteoside dose-dependently inhibited silica-induced ROS (reactive oxygen species) generation in B16 melanoma cells. They significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. The inhibitory effect of acteoside was more potent than that of isoacteosidee. In Western blot of tyrosinase, acteoside inhibited MSH-induced tyrosinase expression in B16 melanoma cells, which is related to the inhibitory action of acteoside on tyrosinase activity and melanin production. These results show that acteoside and isoacteoside from Clerodendron trichotomum Thunberg has a potent antioxidant activity and whitening activity. The underlying mechanism of acteoside on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase expression.

This study investigated the water extracts of fallen pear (FPWE) on tyrosinase activity and melanogenesis. In the present study, we examined the effects of FPWE on mushroom tyrosinase activity in vitro, B16F10 melanoma cell tyrosinase activity, melanin contents, and expression of melanogenic enzyme proteins such as tyrosinase. An apparent down-regulatory effect on tyrosinase activity was observed when B16F10 cells were incubated with FPWE. Results of melanin assay using B16F10 cells treated with different concentrations (50, 125, and $250{\mu}g/mL$) of FPWE showed a dose-dependent decrease in melanin content. To determine whether or not FPWE indirectly affects tyrosinase activity, we assessed mushroom tyrosinase activity upon treatment with various concentrations (125, 250, 500, and $1,000{\mu}g/mL$) of FPWE. In addition, we investigated changes in the protein level of tyrosinase by using Western blotting. Tyrosinase and microphthalmia-associated transcription factor expression levels in B16F10 melanoma cells were reduced in a dose-dependent manner by FPWE. These results suggest that FPWE reduced melanin formation by inhibition of tyrosinase activity. Therefore, we suggest that FPWE could be used an effective whitening agent for skin.
To investigate the possibility of development as a whitening agent using arctigenin, we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B16 melanoma cells. Arctigenin dose-dependently had anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Although arctigenin did not inhibit purified tyrosinase activity, it dose-dependently inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M$${\alpha}$-MSH. In particular, arctigenin at a concentration $100{\mu}M$ inhibited ${\alpha}$-MSH-stimulated tyrosinase activity and melanin production by $50.9{\pm}2.9%$ and $69.0{\pm}6.5%$ respectively. And typical tyrosinase inhibitor, arbutin, inhibited $57.7{\pm}2.9%$ and $65.1{\pm}5.0%$ respectively. Such an similar inhibitory effect of arctigenin and arbutin in B16 melanoma cells may be due to the inhibition of MSH signal pathway rather than the direct inhibition of tyrosinase. Therefore, these results suggest that arctigenin may be useful for the development as whitening agents.
In this study, we investigated on antioxidative activity, antibacterial activity against pathogens of acne and tyrosinase inhibitory activity in 75% EtOH extract and its fractions from bark of Prunus sargen tii. The total polyphenol and flavonoidcontent of the EtOAc fraction were found to be 378.0 mg/g and 67.5 mg/g as the highest content. In the measurement of DPPH radical scavenging ability, EtOAc and BuOH fraction were exhibited stronger scavenging ability than the other fractions and 75% EtOH extract. In antibacterial activity by disc diffusion assay against pathogen of acne, antibacterial activity of the EtOAc fraction and 75% EtOH extract was stronger than the other fractions. Especially, the EtOAc fraction was the highest effective fraction in the antibacterial activity. Moreover, tyrosinase inhibitory activity of EtOAc fraction, BuOH fraction and 75% EtOH extract was higher than the other fractions. In particular, tyrosinase inhibitory activity of the EtOAc fraction showed higher activity than ascorbic acid used as positive control.
Objectives : Draconis Resina (DR), the resin of Daemonorops draco Bl., is used to circulate the blood and to stop bleeding. It also has been used to generate flesh including ulceration. The present study investigated the effects of DR extract on collagen metabolism in human fibroblasts and tyrosinase activity in mushroom tyrosinase.Methods : The effect of DR extract on type I procollagen production (collagen type I synthesis) and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts cell line. Hs68 cells after ultraviolet B (UVB, 312 nm) irradiation was measured using the enzyme - linked immunosorbent assay (ELISA). The tyrosinase activity was also measured to find out the whitening effects in mushroom tyrosinase by ELISA method.Results : There was no cytotoxicity at DR extract at concentrations of 10 μg/ml, 30 μg/ml, and 100 μg/ml. DR extract significantly inhibited the increase of collagenase activity, whereas it did not show on the reduction of type I procollagen in UVB damaged Hs68 cells. DR extract did not reduce the L - DOPA oxidation. However, it significantly reduced the tyrosinase activity by DR extract at concentraions of 0.1 mg/ml, 1 mg/ml and 10 mg/ml.Conclusions : In conclusion, DR showed the anti-wrinkle and whitening effects via the inhibition of collagenase production and the tyrosinase activity. These results suggest that DR may have potential as an anti-aging ingredient in cosmetic herb markets.
This study was conducted to investigate the antioxidative activity and tyrosinase inhibitory activity of 50% ethanol extract and its fractions from the branch of Rhododendron schlippenbachii. In DPPH radical scavenging ability, butanol and ethyl acetate fractions showed 59.98% and 55.17% of relative activity compared with positive control (ascorbic acid), but the 50% ethanol extract showed relatively low activity. In nitric oxide (NO) scavenging ability, the ethyl acetate and butanol fractions showed 141.80% and 131.55% relative activity compared with ascorbic acid as used for positive control. On the other hand, tyrosinase inhibitory activity of the ethyl acetate and butanol fractions showed about twice higher activity than positive control (arbutin). It means that the ethyl acetate and butanol fractions from the extract of R. schlippenbachii branch has ability for used as effective radical scavenger and tyrosinase inhibitor.
For the use of tannins in the whitening-effect cosmetics, inhibitory effect against tyrosinase activity was determined. Three condensed tannins including gallocatechin, gallocatechin 3',4'-di-O-gallate and epicatechin 3-O-gallate and three hydrolyzable tannins, 1,2,6-tri-O-galloyl-${\beta}$-D-glucose, 2,3-(S)-HHDP-D-glucose and pedunculagin showed 15-29% mild inhibitory effects against tyrosinase activity.
This study was conducted to investigate tyrosinase inhibitory effect, hyaluronidase inhibitory effect and antioxidant activity by DPPH radical scavenging method on the MeOH extract of 50 species medicinal plant for screening of functional properties. As a result, Chaenomeles sinensis Koehne extract among 50 species medicinal plant turned out to be having tyrosinase, hyaluronidase inhibitory effect and antioxidant activity. The major component of tyrosinase and hyaluronidase inhibitory effect was isolated from EtOAc extract of Chaenomeles sinensis Koehne. And the component of antioxidant activity was isolated from n-BuOH extract of Chaenomeles sinensis Koehne. Their structure of compounds were identified as oleanolic acid and (-)-epicatechin by spectroscopic evidence, respectively.
