육계에서 분리한 Salmonella gallinarum 의 약제내생 및 PFGE 양상 (Antimicrobial resistance and pulsed -field gel electrophoresis patterns of Salmonella gallinarum isolated from broiler)

  • Fowl typhoid (FT) is a septicemic disease caused by Salmonella gallinarum. The purpose of this study was to investigate the antimicrobial resistance and pulsed -field gel electrophoresis (PFGE) patterns of S gallinarum isolated from broiler. During 1999 to 2004, there was isolated a total of 26 strains in liver and spleen. The biochemical characteristics of S gallinarum isolates was nonmotile, no production of $H_2S$, glucose gas, non-fermented rhamnose, indole-negative, fermentation of dulcitol, mannitol, maltose, and ornithine decarboxylase. At antimicrobial susceptibility, all of isolates were susceptible to amoxicillin/clavulanic acid, amikacin, neomycin, kanamycin, and cephalothin. Twenty-six isolates were divided into 19 resistant patterns and 6 strains was 8-multi-drug resistance. PFGE of Xba I restriction fragments of S gallinarum isolates was 22 patterns.

  • During a 10 year periods, 42 patients with well-documented Midline Malignant Reticulosis were treated with local irradiation and followed for extended periods of time. 13 cases with systemic manifestation after irradiation illustrate the protean features of this disease. Although it commonly Presents in upper airway tracts such as nasal cavity and/or septum, soft palate and palatine tonsil, the lesion may be localized as well as diffuse. 2 cases among them showed systemic relapse on lung, pancreas and scrotum probably from either a multifocal or metastatic deposits. Another 3 eases were associated with systemic diseases such as stomach carcinoma, typhoid enteritis and CNI of fat tissue. Remained cases were diagnosed by clinical or radiological examination. Possibility of close relation to lymphoma with Midline Malignant Reticulosis are also suggested in 1 case. In Midline Malignant Reticulosis, the best results of treatment are obtained in localized lesion of the upper airway treated early with irradiation: A poorer outcome is associated with multifocal systemic involvement, which necessitates a systemic therapy.

수인성 전염병의 현황 및 효율적 관리 (Present status and effective control measure of water-borne infectious diseases in Korea)

  • Water- borne infectious diseases can be acquired by contact with contaminated water or by ingestion of contaminated water. There are many water- borne infectious agents such as bacteria, virus, and parasite. Among many of water- borne infectious diseases, health authorities of Korean government has particularly intensified to prevent and control typhoid fever(class I ), shigellosis(class I ), cholera(class I ), paratyphoid fever(class I), amebiasis(class II ) and leptospirosis(euivalent to class II ) under the communicable disease control law. Water- borne disease Prevention and control guideline itself has been also well provided by the health authorities. However, in practical public health point of view, there are still many problems remained to be solved out; no prospective investigation project to survey water borne infectious diseases under the national disease prevention and control programmes, incredible statistic data of annual notifiable disease report frequent appearance and varieties of drug resistance water- borne infectious agents, little cooperation and information- exchange system in between the related government authorities( the health authorities, the environment sanitation authorities and the food hygiene authorities) which should be closely collaborated, lack of health consciousness of the people, necessity of evaluation and Hndification on to the outcomes of performed health activities and programmes, neglect activities for water quality investigation, shortage of expertise and human resources in the related field, and poor investment of the government budget to develope and improve public health and sanitation field. In order to prevent and control water- borne infectious diseases effectively, it is emphasized that all the above indicated should be considered and performed to improve under the national health and sanitation development programmes.

닭에서 분리한 Salmonella gallinarum의 병원성 및 Plasmid Profile (Virulence and Plasmid Profiles of Salmonella gallinarum Isolated from Chickens)

  • The present study was conducted to investigate the antibiotic resistance of 24 Salmonella gallinarum isolated from 48 chicken samples of diagnosed fowl typhoid cases during the period from November 1998 to November 1999. And the isolates of S gallinarum were also tested for their invasion abilities to experimental infection as one of virulence tests, and the presence of virulence-related plasmid in S gallinarum isolates. The results obtained through this study were summarized as follows; 1. All of isolates from 48 cases of 24 farms were identified S gallinarum by biochemical and serological tests.2. Antimicrobial drug resistance test against 24 isolates showed that the isolates were resistant to Colistin(95.8%), and Penicillin(79.2%), Polymyxin B(75.0%), Streptomycin (65.2%), Gentamycin(54.2%), and Tetracycline(33.3%). 3. Mortality in chicken following peroral inoculation of four isolates of S gallinarum during 14-days inoculation pecked at 5 days(40%) after inoculation and all of experimental chickens died within 13 days after inoculation.4. Based on the pattern and number of isolated plasmid from each isolate, plasmid profiles were divided into five groups; group I with 3 plasmids, group II to group IV with 4 plasmids and group V with 5 plasmids.

마행의감탕의 소염.진통.해열작용 (Anti-Inflammatory, Analgesic and Antipyretic Actions of Mahaengeuigam-Tang)

  • The anti-inflammatory activity of water extract of Mahaengeuigam-Tang(MHEGTWE) was examined using the carrageenin and acetic acid induced edema, croton oil induced granuloma pouch, and adjuvant arthritis in rats. In addition, the acute toxicity, analgesic and antipyretic effects of MHEGTWE were investigated by the general experimental methods. In acute toxicity test in mice, MHEGTWE showed 10% mortality at 2400 mg/kg(p.o), but it did not showed at 1200 mg/kg(i.p). It was also showed significant analgesic action on the writhing syndrome induced by 0.7% acetic acid at 600 mg/kg(p.o) and its antipyretic activity was observed in the typhoid vaccine induced fevered rats at 300 mg/kg(p.o). By the oral administration of the MHEGTWE, the significant anti-inflammatory activity was observed on 1% carrageenin induced edema, and it significantly inhibited the granuloma and exudate formation in rats. In the adjuvant arthritis experiment, the MHEGTWE decreased the hind paw edema in rats for 19 days. The results suggest that MHEGTWE has analgesic, anti-inflammatory and antipyretic action.

원지(遠志) Saponin의 이뇨효과(利尿效果) 및 중추억제작용(中樞抑制作用)에 관한 연구(硏究) (Effects of the Saponin of Polygalae Radix on the Renal Function and CNS-Depression)

  • Polygalae Radix was used as diuretics, analgesics and expertorants in oriental medicine. The root of Polygala tenuifolia Willd. (Polygalaceae) is comprised saponin (Onjisaponin A,B,C,D,E,F and G) polygalitol, onsitin and sugars. The pharmacological action of crude Polygala-saponin (PS) obtained from the roots are studied. The following results were obtained; 1) The median lethal dose $(LD_{50})$ of PS in mice is presented 71.1mg/kg s.c. and 694. 5mg/kg p.o.. 2) PS demonstrated diuretic action of relatively long acting duration in mice. 3) The diuretic mechanism of PS was found due to inhibitory effect of renal tubular reabsorption of electrolytes and glomerular vascular dilatation. 4) The group, administered simultaneously PS and cefadroxil monobydrate was significantly increased with PS alone group on diuretic action. Synergistic effect cefadroxil monohydrate on the diuretic action of PS seems due to competitive inhibition of plasma protein binding with PS. 5) PS demonstrated analgesic action by the acetic acid stimulating method and Randall-Selitto test in mice. 6) PS presented antipyretic action against febrile treated with the typhoid vaccine. 7) PS was significantly prolonged against the hypnotic duration of pentobarbital in mice. 8) Onset time convulsion and death induced by picrotoxin and strychnine in mice were not delayed. According to the above results, the PS was identified as a pharmacological active component obtained from roots of Polygala tenuifolia Willd.

Gallinarum isolates was nonmotile, no production of H$_2$S, glucose gas, non-fermented rhamnose, indole-negative, fermentation of dulcitol, mannitol, maltose, and ornithine decarboxylase. At antimicrobial susceptibility, all of isolates were susceptible to amoxicillin/clavulanic acid, amikacin, neomycin, kanamycin, norfloxacin and enrofloxacin. One hundred isolates were divided into 54 resistant patterns and 37 strains was 6-multi drug resistance. PFGE of Xba I restriction fragments of S. Gallinarum isolates was 20 patterns.

사간탕(瀉肝湯)의 효능(效能)에 관(關)한 실험적(實驗的) 연구(硏究) (Experimental Studies on the Effiency of Sagantang)

  • The present investigation was undertaken to confirm of clinical effects of Sagnating. So, experimental studies were done, especially, to ascertain whether Sagantang had pharmacological effect of analgesia, antipyresis, diuresis and decreasing blood pressure and so on. The results of the studies were obtained as follows: 1. The analgesic effects of Sagantang were obtained in mice. 2. Sagantang prolonged the duration of hypnosis which is induced thiopental-Na in mice, but it was not effective in rotor rod method. 3. Antipyretic effect of Sagantang was known on the typhoid vaccine induced fever in rats. 4. Sagantang inhibited automatic movement on the isolated ileum of mice, and antiacetylcholine effect and antibarium chloride effect of Sangantang were known on that. 5. Sangantang decreased blood pressure due to vasodilatation in anesthetized mice. 6. The effect of diuresis were noted in mice. According to the above results, clinical effects of Sagantang on oriental medical references were similar to the actual experimental results.

Identification of Genes Differentially Expressed in RAW264.7 Cells Infected by Salmonella typhimurium Using PCR Method

  • Salmonella typhimurium, causing mouse typhoid, infects hosts such as macrophage cells, and proliferates in intracellular vacuoles causing infected cells to trigger numerous genes to respond against the infection. In this study, we tried to identify such genes in RAW264.7 cells by using the PCR screening method with degenerate primers. Fourteen genes were found to be differentially expressed after a 4 h infection in which the expression of 8 genes increased while expression of the others decreased. Most of the genes were involved in proinflammatory responses such as cytokines production and cell death. The mutation in msbB gene encoding the myristoyl transferase in lipid A of lipopolysaccharide (LPS) resulted in much lower toxicity to the inoculated animals. We compared the expression of the identified genes in wild-type and msbB-mutated S. typhimurium infections and found that Lyzs encoding lysozyme type M was differentially expressed. This gene is quite likely to be related to bacterial survival in the host cells.

  • Salmonella enterica serovar Gallinarum is the causative agent of fowl typhoid (FT) and Salmonella enterica serovar Pullorum is pullorum disease (PD), a severe systemic disease of chick and it has the same antigenic fomula, the close relation but distinct pathogen. The traditional bacteriologic and serologic methods routinely used but tedious, time consuming. some of biochemical differences are helpful in differentiating the two organisms, however variation in the characteristics of some strains can be observed. During 2006 to 2008, there was isolated 30 strains. The biochemical characteristics of S. Gallinarum was nonmotile, fermentation of dulcitol, maltose but positive arginine (6.6%), lysine (83.3%) and arabinose (20.0%). The antimicrobial susceptibility test showed 100% sensitive to amikacin, ampicillin, amoxicillin/clavulanic acid and florfenicol, but resistant to penicillin (100%) and erythromycin (60.0%). This PCR method can be applied in the diagnosis between S. Gallinarum and S. Pullorum.
