Excessive production of reactive oxygen species (ROS) is a key phenomenon in tumor necrosis factor (TNF)-α-induced cell death. However, the role of ROS in necroptosis remains mostly elusive. In this study, we show that TNF-α induces the mitochondrial accumulation of superoxide anions, not H2O2, in cancer cells undergoing necroptosis. TNF-α-induced mitochondrial superoxide anions production is strictly RIP3 expression-dependent. Unexpectedly, TNF-α stimulates NADPH oxidase (NOX), not mitochondrial energy metabolism, to activate superoxide production in the RIP3-positive cancer cells. In parallel, mitochondrial superoxide-metabolizing enzymes, such as manganese-superoxide dismutase (SOD2) and peroxiredoxin III, are not involved in the superoxide accumulation. Mitochondrial-targeted superoxide scavengers and a NOX inhibitor eliminate the accumulated superoxide without affecting TNF-α-induced necroptosis. Therefore, our study provides the first evidence that mitochondrial superoxide accumulation is a consequence of necroptosis.
차(Camellia sinensis)는 세계적으로 애용되는 음료이다. 그 중, 아시아권에서 주로 소비되는 녹차는 다양한 생리활성을 가지고 있어 화장품, 기능성식품을 위한 기능성 소재로서 선호되고 있다. 녹차의 피부에 대한 활성은 세포보호에서부터 피부 기질 단백질의 합성까지 다양하게 나타난다. 녹차 폴리페놀(green tea polyphenols; GTPs)은 활성을 나타내는 주성분으로서 항산화 활성 이외에 항암, 항염증, 피부면역능 저하방지활성 등을 보이며, 세포내 신호전달 경로에도 관여한다 GTPs는 표피각질형성세포에서 자외선 조사에 의한 산화 스트레스를 감소시키고, 그에 따르는 mitogen-activated protein kinase (MAPK) 신호전달과 세포사멸을 억제한다. 또한, 같은 세포에서 tumor necrosis factor alpha $(TNF{\alpha})$나 다른 화학물질에 의해 cyclooxygenase-2(COX-2), interleukin-8 (IL-8) 및 vascular endothelial growth factor (VEGF) 같은 염증매개물질이 유발되는 것을 막아준다. 또한, GTPs는 등물실험에서 화학물질이나 자외선에 의한 피부암의 발생도 억제하는데, 경구투여 외에 경피투여로도 효과가 있었다. 피부보호작용 이외에도 GTPs는 각질형성세포의 분화촉진, 노화된 피부세포의 증식능 회복, 피부기질단백질의 분해억제 등의 기능이 있으며, 피부세포에서의 기질단백질 생합성을 직접적으로 촉진하기도 한다. 녹차성분이 보이는 이러한 피부에 대한 활성은, 제형 측면에서의 연구가 진행되어 감에 따라 보다 효과적인 피부를 위한 소재로서의 사용 가능성을 더욱 높여주고 있다.
연구배경 : 종양괴사인자(tumor necrosis factor ; TNF)는 다양한 생물학적 기능을 가지고 있는바, 그 중 생체외에서 증명된 뚜렷한 항암효과로 말미암아 최근 항암유전자요법의 중요한 대상으로 관심을 모으고 있다. 그러나 유전자 이입의 기술적 문제로 생체외에서 암세포에 유전자 이입을 시행한 후 이를 다시 환자의 생체내로 이식하는 방법이 연구의 주종을 이루고 있다. 그러나 저지들의 과거의 연구를 포함한 여러 연구에서 TNF가 이입된 암세포는 TNF에 대해 내성을 보이는 것으로 증명되었다. 이 획득내성의 기전을 밝히는 것이 종양생물학의 이해를 넓히고 보다 효과적인 항암유전자 요법을 개발하기 위한 매우 중요한 과제로 생각된다. 저자들은 TNF 유전자 이입에 따른 암세포의 TNF에 대한 획득 내성에 새로운 방어단백질의 합성이 관여하는 지를 규명하고자 본 실험을 수행하였다. 방 법 : TNF에 감수성을 보이는 생쥐 섬유육종 세포주인 WEHI164에 TNF-$\alpha$ 유전자를 retroviral vector를 이용하여 이입하고 TNF의 발현을 시도하여 PCR, ELISA, MTT assay로 확인하였고, TNF 유전자가 이입된 세포(WEHI164-TNF)는 TNF에 내성을 보이는지 역시 MTT assay로 검증하였다. WEHI164-TNF세포를 transcription 억제제인 actinomycin D와 translation 억제제인 cycloheximide로 처리한 후 역시 MTT assay로 TNF에 대한 감수성에 변화를 보이는지 확인하였다. 결 과 : 1) TNF-$\alpha$ 유전자 이업 및 발현 확인 PCR을 시행한 결과 TNF 유전자가 이입된 WEHI164-TNF 세포주는 790 base pair 크기의 진한 DNA band를 보인 반면 모세포주는 보이지 않아서 retroviral vector를 이용한 유전자 이입이 DNA 수준에서 이루어졌음을 확인할 수 있었다. 그리고 WEHI164-TNF의 배양상층액에서 TNF양을 ELISA와 MTT assay로 측정한 결과, 생물학적 활성을 지닌 TNF를 $10.9{\pm}1.47ng/24hr/10^6cells$ 생산함을 알 수 있었다. 2) TNF 유전자 이입 전후, 암세포의 TNF에 대한 감수성 비교 TNF 농도 100ng/ml 에서 모세포는 $73{\pm}5%$의 세포독성을 보인 반면 WEHI164-TNF 세포는 $3{\pm}2%$의 세포독성을 보여 통계적으로 유의하게 (p < 0.00) TNF에 대한 내성을 획득함을 알 수 있었다. 3) TNF 유전자 이입 후 획득된 TNF에 대한 내성의 기전 WEHI164-TNF 세포를 actinomycin D로 처리한 경우 TNF 농도 10ng/ml과 100ng/ml에서 각각 $24{\pm}7%$, $44{\pm}6%$의 세포독성을 보여 control의 $6{\pm}2%$, $17{\pm}2%$보다 통계적으로 유의하게(p < 0.01) TNF에 대한 감수성이 부분적으로 회복됨을 관찰할 수 있었다. 그러나 cycloheximide로 처리한 경우에서는 TNF에 대한 감수성에 변화를 관찰할 수 없었다. 결 론 : TNF에 감수성을 보이는 암세포주인 WEHI164에 TNF 유전자를 이입하여 TNF를 발현하게 하였을 때 그 세포 자신은 TNF에 대해 내성을 획득하게되며 이에는 미지의 방어단백질의 합성이 일부 관여할 것으로 판단된다.
Obesity may cause metabolic syndrome and adult diseases. This study was undertaken to investigate the ameliorative or useful effects of beanleaves on inflammation and liver damage in obese rat models. Rats were divided into three groups: a control group (normal diet, n=6), a fat diet group (45%-fat diet, n=7), and a bean leaf group (45%-fat+Korean bean leaves diet, n=7). Body weights in the bean leaf group were lower than those of the fat group (P<0.05). Serum tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and prostaglandin $E_2$ ($PGE_2$) concentrations were lower in both the control and bean leaf groups than in the fat group (P<0.001). TNF-${\alpha}$ concentrations in the bean leaf group were slightly higher than in the control group but statistically significant (P<0.05). The bean leaf group histologically exhibited lower fatty degeneration, spotty necrosis, and leukocyte infiltrations in hepatic tissues than those of the fat group. In the homogenized liver tissues, the cyclooxygenase-2 (COX-2) gene was only expressed in the fat group. The gene expression levels of hepatic TNF-${\alpha}$, inducible nitric-oxide synthase, peroxiome proliferator-activated receptor-${\alpha}$ (PPAR-${\alpha}$), poly (ADP-ribose) polymerase (PARP), and transforming growth factor-${\beta}1$ (TGF-${\beta}1$) were weaker in the bean leaf group than in the fat group. These results suggest that adding bean-leaves to the diet may ameliorate obesity-induced systemic inflammation and liver damage and that bean leaves may be a useful food for preventing obesity and thereby metabolic syndrome and adult diseases.
The purpose of this study was to investigate the age-related changes of cardiovascular disease risk factors and inflammatory markers in non-obese Korean women. Subjects were 112 women over 20 years old with body mass index (BMI) less than $30 kg/m^2$ and were divided into 3 groups (< 40 years, $40{\sim}59$ years, ${\ge}60$ years). Mean weight and BMI in the oldest group were significantly higher than those in the other 2 younger groups (p < 0.05). Mean total cholesterol, triglyceride, LDL-cholesterol and apolipoprotein B/apolipoprotein A1 ratio (BAR) in the oldest group were significantly higher than those in the youngest group (p < 0.05), and mean HDL-cholesterol of the oldest group was significantly lower than that of the youngest group (p < 0.05). The older-aged group showed significantly higher mean values of atherogenic index (AI) and LDL/HDL ratio (p < 0.05) than the respective younger-aged group, and AI was significantly correlated with age, nitric oxide and thiobarbituric acid reactive substances (p < 0.01). In addition, mean vascular cell adhesion molecule-l (VCAM-1) tended to be higher in the older-aged group than the younger group. Tumor necrosis factor-${\alpha}$, a proinflammatory maker, was significantly positively correlated with serum homocysteine, a cardiovascular disease risk factor (p < 0.01). In addition, a significantly positive correlation was observed between C-reactive protein and BAR (p < 0.01). Overall results suggested that the aging might affect the increase of cardiovascular disease risk factors including the serum lipid profiles, weight and BMI, and age-related increases of weight and BMI might play a role in changes in certain biomarkers of inflammation. Objectives This study was designed to evaluate the effects of Danggwisu-san (Dangguixu-san, DG) on bone repair from femur fracture in mice. Methods Mice were randomly divided into 4 groups (normal, control, positive control and DG 300 mg/kg-treated group). In order to investigate the effects of DG on gene expressions in experimental animals with fracture, we measured the levels of bone morphogenetic protein-2 (BMP2), cyclooxygenase-2 (COX2), Sox9, collagen type II alpha 1 chain (Col2a1), runt-related transcription factor 2 (Runx2), osterix genes. After the cytotoxicity test, we analyzed the levels of expression of osteocalcin and Runx2, and tumor necrosis factor-α (TNF-α), a pro-inflammatory cytokine. The process of fusion in the fracture was also investigated by gross examination. Results Through in vivo BMP2, COX2 gene expression significantly decreased. Sox9 significantly increased. Col2a1, Runx2, osterix gene expression also increased as well, but there was no statistical significance. The degree of unilateral fracture fusion investigated by gross examination was significantly faster than those of the other groups. Through in vitro the level of TNF-α in macrophages was increased by DG in a dose-dependent mannerand and 250 and 500 ㎍/mL showed statistical significance. Osteocalcin and Runx2 genes expressions increased when DG was treated in osteoblasts. Conclusions DG promotes the healing of the fracture through the expression of bone repair-related genes and TNF-α production. This study may set the foundation for the clinical application of DG to the patients with bone fractures.
Objectives : The purpose of this study was to investigate the toll-like receptor (TLR)-4 mediated anti-inflammatory effects of extract from Shigyungbanha-tang (SBT) on the peritoneal macrophage. Methods : To evaluate of TLR-4 mediated inflammatory of SBT. we examined NO and cytokine production in TRL-4 ligand (LPS : lipopolysaccharide) induced macrophages. Furthermore, we examined its molecular mechanism using western blot. Results : Extract from SBT itself does not have any cytotoxic effect in the peritoneal macrophages. Extract from SBT reduced LPS-induced nitric oxide (NO). tumor necrosis factor-alpha ($TNF-{\alpha}$), interleukin (IL)-6 and IL-12 production in peritoneal macrophages. SBT inhibited degradation of inhibitor kappa B-alpha ($I{\kappa}B-{\alpha}$) in the TLR-4 mediated peritoneal macrophages. Conclusions : These results suggest that SBT inhibits NO and cytokines production through inhibiting nuclear factor-kappaB (NF-${\kappa}$B) activation in peritoneal macrophage and that SBT may be beneficial oriental medicine for inflammation.
개 말초혈액 다형핵 백혈구(PMNs)의 탐식능력과 순간산소과소비력 (OBA) 에 대한 fucoidan의 효과를 검토하였다. Fucoidan 그 자체는 PMNs의 탐식능력과 OBA에 직접적인 효과를 보이지 않았다. 그러나 fucoidan으로 배양한 PBMCs의 배양상층액은 PMNs의 탐식능력과 OBA를 농도의존적으로 증가시켰다. 또한, TNF-${\alpha}$에 의한 PMNs의 탐식능력과 OBA의 측정 결과도 fucoidan으로 배양한 PBMCs 배양상층액의 그것과 유사하였다. 이러한 탐식능력과 OBA의 활성은 anti-TNF-${\alpha}$ pAb를 처리했을 때 억제되었다. PBMCs 배양 상층액 속의 TNF-${\alpha}$의 양을 정량한 결과 대조군에 비해 증가되었으며, PBMCs의 TNF-${\alpha}$ mRNA 발현 정도도 fucoidan을 첨가한 경우 증가되었다. 이상의 결과로부터, fucoidan은 개 말초혈액 PMNs의 탐식능력과 OBA에 대하여 면역자극 작용을 가지고 있으며, 이것은 fucoidan의 자극에 의해 PBMCs에서 생산되어 분비되는 가용성 물질인 TNF-${\alpha}$에 의해 나타나는 것으로 사료되었다.
