A study on SiC crystal growth by sublimation process using resistance heating method (저항가열 방식을 적용한 승화법에 의한 SiC 결정 성장에 대한 연구)

  • SiC crystals are well known for their true potential as high power devices and their crystal growth activity is actively carried out in domestic as well as in abroad. Until now the process to grow this crystal has been done by sublimation technique using radio frequency induction heating method. However in order to get better quality of SiC crystals, the stability of temperature is needed because SiC crystal tends to transform to other polytypes. So, the possibility of SiC crytals growth was evaluated by different heating method. This study aimed to observe whether the resistant heating method would show stable growth and better quality of SiC single crystal than that of RF induction heating. As a result, polycrystalline SiC crystals were grown by the growth rate of 0.02~0.5 mm/hr under the condition of $2100{\sim}2300^{\circ}C$ at the bottom side of the crucible and 10~760 torr. The polycrystalline SiC crystals with 0.25 and 0.5 mm in thickness were grown successfully without seed and characterized by optical stereo microscopic observation.

Current status of CRISPR/Cas9 base editor technologies and their applications in crop precision breeding

  • Plant biotechnologists have long dreamed of technologies to manipulate genes in plants at will. This dream has come true partly through the clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 technology, which now has been used to edit genes in several important crops. However, there are many restrictions in editing a gene precisely using the CRISPR/Cas9 technology because CRISPR/Cas9 may cause deletions or additions in some regions of the target gene. Several other technologies have been developed for gene targeting and precision editing. Among these, base editors might be the most practically and efficiently used compared to others. Base editors are tools which are able to cause a transition from cytosine into thymine, or from adenine into guanine very precisely on specific sequences. Cytosine base editors basically consist of nCas9, cytosine deaminase, and uracil DNA glycosylase inhibitor (UGI). Adenine base editors consist of nCas9 and adenine deaminase. These were first developed for human cells and have since also been applied successfully to crops. Base editors have been successfully applied for productivity improvement, fortification and herbicide resistance of crops. Thus, base editor technologies start to open a new era for precision gene editing or breeding in crops and might result in revolutionary changes in crop breeding and biotechnology.

Influence of different fatigue loads and coating thicknesses on service performance of RC beam specimens with epoxy-coated reinforcement

  • Epoxy-coated reinforcing bars are widely used to protect the corrosion of the reinforcing bars in the RC elements under their in-service environments and external loads. In most field surveys, it was reported that the corrosion resistance of the epoxy-coated reinforcing bars is typically better than the uncoated bars. However, from the experimental tests conducted in the labs, it was reported that, under the same loads, the RC elements with epoxy-coated reinforcing bars had wider cracks than the elements reinforced with the ordinary bars. Although this conclusion may be true considering the bond reduction of the reinforcing bar due to the epoxy coating, the maximum service loads used in the experimental research may be a main reason. To answer these two phenomena, service performance of 15 RC beam specimens with uncoated and epoxy-coated reinforcements under different fatigue loads was experimentally studied. Influences of different coating thicknesses of the reinforcing bars, the fatigue load range and load upper limit as well as fatigue load cycles on the mechanical performance of RC test specimens are discussed. It is concluded that, for the test specimens subjected to the comparatively lower load range and load upper limit, adverse effect on the service performance of test specimens with thicker epoxy-coated reinforcing bars is negligible. With the increments of the coating thickness and the in-service loading level, i.e., fatigue load range, load upper limit and fatigue cycles, the adverse factor resulting from the thicker coating becomes noticeable.

Effects of Dietary Additives and Early Feeding on Performance, Gut Development and Immune Status of Broiler Chickens Challenged with Clostridium perfringens

  • The effects of dietary additives and holding time on resistance and resilience of broiler chickens to Clostridium perfringens challenge were investigated by offering four dietary treatments. These were a negative control (basal), a positive control (Zn-bacitracin) and two dietary additives, mannanoligosaccharides (MOS), and acidifier. Two holding times included (a) immediate access to feed and water post hatch (FED) and (b) access to both feed and water 48 h post hatch (HELD). Chicks fed Zn-bacitracin had no intestinal lesions attributed to necrotic enteritis (NE), whereas chicks fed both MOS or acidifier showed signs of NE related lesions. All dietary treatments were effective in reducing the numbers of C. perfringens in the ileum post challenge. The FED chicks had heavier body weight and numerically lower mortality. The FED chicks also showed stronger immune responses to NE challenge, showing enhanced (p<0.05) proliferation of T-cells. Early feeding of the MOS supplemented diet increased (p<0.05) IL-6 production. The relative bursa weight of the FED chicks was heavier at d 21 (p<0.05). All the additives increased the relative spleen weight of the HELD chicks at d 14 (p<0.05). The FED chicks had increased villus height and reduced crypt depth, and hence an increased villus/crypt ratio, especially in the jejunum at d 14 (p<0.05). The same was true for the HELD chicks given dietary additives (p<0.05). It may be concluded that the chicks with early access to dietary additives showed enhanced immune response and gut development, under C. perfringens challenge. The findings of this study shed light on managerial and nutritional strategies that could be used to prevent NE in the broiler industry without the use of in-feed antibiotics.

Kinetic Study on the Immobilized Penicillin Amidase in a Differential Column Reactor (Differential column reactor에 있어서 고정화페니실린 아미다제의 반응속도론에 관한 연구)

  • The penicillin amidase from Escherichia coli (ATCC 9637) was immobilized by entrappment in gelatin and DEAE-cellulose mixture cross-linked with glutaraldehyde, and the kinetics in a differential column reactor was studied. The optimal operating condition of a differential reactor was reasonably met when the enzyme loading was 1g, and 30 mM substrate solution in 0.1 M phosphate buffer (pH 8.0) was fed at flow rate 4$m\ell$/min and 4$0^{\circ}C$. The optimal pH and temperature were found to be 8.0 and 55$^{\circ}C$, respectively. The Michaelis-Menten constant was 4.8 mM while the maximum velocity was 308 units/g of the immobilized enzyme under the condition of the differential reactor. The effect of substrate inhibition disappeared in the immobilized enzyme preparation. The differential reactor was proved to be good for studying the true kinetics since the pH drop and the external diffusional resistance could be eliminated.

Distance Learning for Higher Education Applicants in War: Information Competence

  • Modern challenges in the educational environment force scientists and practitioners to search for an adequate answer. In particular, the war in Ukraine demonstrated the importance of developing information competence as one of the main means of distinguishing true information from a whole stream of fake news. This is especially relevant in connection with the introduction of distance learning when students must find and process a large amount of information on their own. Therefore, the purpose of the article is to analyze the training of higher education students through the prism of acquiring the necessary level of informational competence in war conditions. For this, general scientific and special research methods, as well as the experimental method, were used. In the results, the peculiarities of the interpretation of information competence in the distance form of education among modern researchers are determined, the psychological components of resistance to fakes are analyzed. Based on the conducted empirical measurements, it was established that thorough work on student education gives positive skills when working independently with Internet materials, strengthens the ability to distinguish false information and propaganda from the real state of affairs. The conclusions summarize the results of the empirical research and suggest ways to improve the situation with the formation of information competence.

A Study on the Performance Predictions of Twin Sail Drone (트윈 세일 드론의 성능추정에 관한 연구)

  • Recently, marine surveys using unmanned ships are attracting attention, and research on small unmanned ships using sails is on the rise. Sail drones can be used for marine surveys, monitoring, and pollution management. Therefore, in this study, using the method of estimating the ship speed for twin sail drones, the optimal conditions for sailing are checked, and the performance to be considered in the initial design stage, such as the motion performance and resistance of the sail drone. Consequently, the twin sail drone had a speed lower than 2.0 m/s, and the stability satisfied the rule by DNV. In addition, the maximum speed at an angle of attack of 20° at TWA 100° was 1.69 m/s and that at an angle of attack of 25° at TWA 100° was 1.74 m/s.

Fatigue Behavior of Reinforced Concrete Beams Externally Strengthened using FRP Tendons (FRP 긴장재로 외부 보강된 철근콘크리트 보의 피로거동)

  • Recently, the external prestressing method is being much frequently used in strengthening reinforced concrete structures because of it's excellent load resistance and serviceability increases comparing to other strengthening methods. However, it is true that the research on fatigue performance of concrete structures strengthened by the external prestressing using FRP tendons is rare. Therefore, the purpose of this study is to evaluate the safety and feasibility of the external prestressing method by analyzing the characteristics of the reinforced concrete beam strengthening using FRP tendons under repeated loads. Test variables adopted in this experimental study are the types of external prestressing material (steel or FRP tendon) and the repeated load ranges. The repeated load range have the minimum 50% of yield load of reinforced concrete beam and the maximum 70-85%. The test beams are loaded by 4 point loadings with 3 Hz sine wave. From this experimental study, it is confirmed that the reinforced concrete beams strengthened using FRP tendons have sufficient safety against fatigue, especially in FRP tendon itself, tendon at deviators and tendon at anchorages.

Screening for Resistance to Downy Mildew among Major Commercial Cucumber Varieties (주요 오이 품종의 노균병에 대한 저항성 검정)

  • This study was carried out for the downy mildew resistant test between 2010 and 2012. A set of 22 accessions belonging to 2 wild species and 20 varieties of the genus Cucumis, originating mainly from the Asian Vegetable Research and Development Center (AVRDC) Gene Centre, was evaluated for resistance to Pseudoperonospora cubensis, causal agent of cucumber downy mildew. The youngest fully expanded true leaves were found suitable for in vitro screening. Both leaf discs and full leaves could be kept fresh longer when applying 0.2 ${\mu}g/ml$ of gibberellin acid (GA). The incubation temperature of $20^{\circ}C$ was found to be the most suitable temperature for symptom development comparing with 15 and $25^{\circ}C$. Symptom development was faster when contact diseased leaf discs (2 weeks after inoculation) on to fresh leaf samples comparing with using conidia suspension ($10^5$ spores/ml). The numbers of spots in 'C-19' were lower than other varieties. 'C-19' variety was also showed the highest level of downy mildew resistant at $20^{\circ}C$ chamber in 6 days after inoculating with pathogen and displayed 0.90 (under 10%) of the infected rate. However, other varieties displayed susceptible in the pathogen sprayed plots. 'C-19' was the most resistant variety and no lesion was observed. Based on all data, 'C-19' can be a useful variety for the prevention of downy mildew.

'Duo', White Gladiolus with Pink Core (백색의 분홍 화심 글라디올러스 '듀오' 육성)

  • 'Duo', a late-flowering white gladiolus with pink core, was released by the National Institute of Horticultural & Herbal Science (NIHHS) in 2000. The selection was derived from the cross between the progenies of the early flowering dark pink 'True Love' and bright pink domestic cultivars 'Madame Valdiek'. The breeding was conducted in 1996 and selected finally in 2000 after four years (1997 to 2000) of experimentation. 'Duo' is a cut flower with bright white color petals and Pink core which exhibited late flowering and vigor without stem bending. It can be cultivated in areas or regions with strong winds during summer. 'Duo' has a very firm growth form and sturdy branches which do not bend when planted in the open fields. Likewise, the plant showed high tolerance to flooding and resistance, hence, could grow vigorously in open areas. It is a fast- growing plant which exhibits high corm production rate. 'Duo' had a 12.2 cm wide flower and 150 cm plant height. During summer, the average number of days to flowering was recorded at 75 days of 'Duo'. Top dressing must be conducted once or twice during corm production to enhance corm quality and quantity.
