Experimental life history of Spirometra erinacei (우리 나라에 분포하는 스파르가눔의 실험실 내 생활사)

  • The complete life cycle of Spirometra erinacei has been experimentally maintained in the laboratory. The cyclops were reared as the first intermediate host, and the tadpoles of Rana nigromaculata as the second intermediate host. ICR mice were used as another second host. The experimental definitive hosts were dogs and cats. Maturation and hatching of the eggs took 8 to 14 days by incubation at 29℃. The coracidium measured 43.8×36.9㎛. Mesocyclops leuckarti and Eucyclops serrulatus were susceptible to the coracidial infection. The procercoids older than 5 days in the cyclops had minute spines at the anterior end, calcium corpuscles in the body parenchyme and the cercomer at the posterior end. Procercoids 10 to 20 days old were infective to tadpoles, and 15 or 21 day old worms could infect the mice. The plerocercoids from the tadpoles at 15 days after experimental infection were pear-shaped and shorter than 1 mm in the length and were infective to mice. Fifteen to 18 days after experiMental inoculation of plerocercoids to dogs or cats, the adult worms began to produce eggs. One life cycle from egg to egg needed 48 to 67 days in the laboratory. The morphology of larval or adult worms was compatible with the description of Spirometra erinacei.

Growth and Antioxidant Activity of Gynura procumbens by Natural Media Composition in Hydroponic Cultivation Using Organic Nutrient Solution (유기 수경재배 시 천연배지 조성에 의한 명월초 생육 및 항산화 활성)

  • Gynura procumbens (Family Asteraceae) is a medicinal plant commonly found in tropical Asia countries such as China, Thailand, Indonesia, Malaysia, and Vietnam. Traditionally, it is widely used in many different countries for the treatment of a wide variety of health ailments such as kidney discomfort, rheumatism, diabetes mellitus, constipation, and hypertension. The objective of this study was to select the natural growth media for hydroponic cultivation of Gynura procumbens. The commercially available thirty four different organic nutrient solutions were analyzed for growing Gynura procumbens. After analysis, two organic nutrient solution were selected, mixed (pH 5.5~6.5 / EC 0.5~1.0 ds/m) and then used as organic fertilizers. Thirty day old seedlings were planted and the environmental conditions of the rhizosphere were also examined to select the natural media composition. The minimum temperature of rhizosphere by type of natural media was highest in cocopeat media and lowest in perlite media. Furthermore, plant growth was examined from 50 days old seedlings, and vermiculite was observed to be most effective and perlite was the least effective component as natural media. The total polyphenol content of the studied plant material was also higher in vermiculite in comparison to other growth media. The Gynura procumbens yields were also higher in vermiculite in comparison to other growth media. Thus, vermiculite can be used to improve Gynura procumbens plant growth as a natural growth media.

Ultrastructural observation of Naegleyiu fowleri trophozoite in mouse brain and axonic culture (조직내 및 배야기내 자유생활아메바의 전자현미경적 비교연구)

  • Present study was undertaken to elucidate the changes of the ultrastructure of Naegleria fowleri trophozoite in brain tissue of mice and culture medium. Naegleria fowleri, 0359 strain, which used in this study was cultured in axonic liquid medium, CGVS medium. Each mouse was inoculated with amoebas intranasally under secobarbital anesthesia, and sacrificed on 7th day after the infection. Comparative observation of the ultrastructure of the amoebas in axonic culture and experimentally infected mice brain was done with transmission electron microscope. The results are summarized as follows: 1. The amoebas in mouse brain tissue were round in outline, whereas those of amoebas from axonic culture showed irregular appearance. 2. Mitochondria in the amoebas from axonic culture was oval, round and cylindrical shape and darkly stained, whereas those of the amoebas from mouse brain tissue showed dumbbell shape together with above forms. The stain was not unique, but light and/or dark. 3. Rough endoplasmic reticulum of amoebas in brain tissue was tubular, but from culture it was vesicular or tubular in shape. 4. Emity vacuoles were demonstrated in amoebas from culture, while food vacuoles with myelinated structures were abundant in those from tissue, suggesting a strong phagocytic activity. 5. Mouse brain tissue in ected were extensively destroyed, and Polymorphonuclear leukocytes were infiltrated predominantly with inflammatory lesion. Amoebas were observed in the vicinity of the capillary.

Migration and Distribution of Spargana in Body of Experimentally Infected Mice (실험감염 스파르가눔의 마우스체내 이행경로 및 분포)

  • The migration and distribution pattern of spargana in mouse body was observed after experimental infection through mouth. The spargana were obtained from the snake, Natris tigrina lateralis, caught in Hoengseong-gun, Kangwon-do. A total of 28 male mice (ICR strain), 21∼259 in body weight, were fed each with 5 scolices (and necks) of spargana and killed after 10 minutes to 14 days. Systemic autopsy was performed on each mouse to recover the spargana. The results are as follows: 1. The spargana were found to penetrate into the stomach or duodenal wall of mice as early as 10 minutes after infection. They completed the penetration within 30 minutes and appeared in abdominal cavity. It was observed that spargana did not migrate tangentially along the gut wall but directly perforated the wall. 2. After 1 hour to 1 day the majority of spargana distributed in abdominal cavity of mice except a few which migrated to muscles or subcutaneous tissues. 3. It was within 7 days that nearly all of the spargana migrated to subcutaneous tissues. Out of total 28 in number found from subcutaneous tissues, 13 distributed around neck region, 12 around trunk and other 3 on head of mice and the most common sites were submandibular and subscapular areas. There was nearly no host tissue reaction to migrating spargana. 4. The initial length of spargana given was 4 mm in average but it increased to 12 mm after 7 days and to 35 mm after 14 days. The results suggest that spargana orally given to mice penetrate the gut wall within 30 minutes followed by escaping into abdominal cavity, and after passing through thoracic cavity or abdominal wall they anally Localize in subcutaneous tissues chieay around neck region within 7 days.

Studies on Intestinal Trematodes in Kores XIV. Infection Status of Loaches with Metacercariae of Echinostoma cinetoychis and Their Development in Albino Rats (한국의 간흡충에 관한 연구 XIV. 미꾸리의 이전고환극구흡충(Echinostoma cinertorchis) 피낭유충 감염상황 흰쥐내에서의 성장발육)

  • The metacercariae of Echinostoma cineterchis (Trematoda: Echinostomatidae) were found infected in the loaches (Misgurnus anguillicaudatus) purchased from two local markets in Korea. Identification of the metacercariae was done after obtaining adult worms from experimentally infected albino rats. Brief course of worm development in rats was also observed up to 28th post-infection day. The results are as follows: 1. The metacercariae of E. cinetorchis were detected from 5 (10.9%) out of 46 loaches examined and the average metacercarial burden per infected loach was 34.4. They were always found from the posterior abdominal wall. 2. The worm recovery rate from 8 rats was 24.6% in average. It was observed that the rate decreased according to age of infection; from 40.7% after 6 days to 15.0% after 28 days. 3. The adult worms were morphologically characterized by the presence of head crown with derfally uninterrupted collar spines, 37-38 in number, and the tendency of migration or disappearance of testes. So that they were identified as Echinostoma cinetorchis Ando et Ozaki, 1923. 4. The development of worm was very rapid during 6-10 days after infection and became much slower thereafter. During the rapid worm growth the development of genital organs was more prominent than that of non-genital organs. From the results it is concluded that the loach is involved in the life cycle of E. cinetorchis in this country and may take a role for infection source to animals and man. It is also confirmed that migration or loss of testes is the distinct specific character of this echinostomatid fluke.

Expression patterns of proliferating cell nuclear antigen in the small intestine of mice infected with Metagonimus yokogawai and Metagonimus Miyata type (요코가와흡충 및 미야타흡충 감염 마우스 소장의 PCNA발현 양상)

  • Genus Metasonimus has been a subject of taxonomic debates for several years. In morphological aspects, M. vokogawci has been thought to have three subtypes, namely Yokogawa type (M. yokogawai in strict sense) , Miyata type and Koga type. But dif- ferences in the intestinal pathology induced by these subtypes have not been studied yet. In this study we compared the pathological reactions inducted by M. yokoBnwai and Metcgonimus Miyata type using proliferating cell nuclear antigen (PCNA) index. Metacercariae (Mc) of M. yokogcuani were collected by artificial digestion of Plecoslossw oLtiveLis and Mc of Metngonimus Miyata type were collected from Zacco pEntvpus. Three hundreds Mc of each species were infected orally to ICR mice. The mice were sacrificed at 3, 6. 10. 16 and 23 days after infection and the small intestines were resected into three portions (proximal. middle, and distal) . Immunohistochemical staining for PCNA was done using PC-10 (DAKO-PCNA, CA, USA) The PCNA indices in M. Wokosnwci infected group on the 6th and 23rd day after infection were lower than in the control and Miyata type infect ed groups (p < 0.05) from all of the three intestinal regions. On the other hand, the control group and Ifetngonimus Miyata type infected group did not make any differences in PCNA indices. The villus/crypt WIC) ratio was also decreased significantly in M. Wokogcwni infected mice but not in Metagonimus Miyata type infected ones. It is suggested that M. wokogawci induce villous atrophy through a decrease in the cell proliferation at the crypt. The results of this study suggested that M. WokoBnwai induce more serious intestinal pathology than Metcsonimuf Miyata type.

Sensitivity of Plasmodium falciparum to Antimalarial Drugs in Hainan Island, China

  • Pyronaridine and artesunate have been shown to be effective in falciparum malaria treatment. However, pyronaridine is rarely used in Hainan Island clinically, and artesunate is not widely used as a therapeutic agent. Instead, conventional antimalarial drugs, chloroquine and piperaquine, are used, explaining the emergence of chloroquine-resistant Plasmodium falciparum. In this article, we investigated the sensitivity of P. falciparum to antimalarial drugs used in Hainan Island for rational drug therapy. We performed in vivo (28 days) and in vitro tests to determine the sensitivity of P. falciparum to antimalarial drugs. Total 46 patients with falciparum malaria were treated with dihydroartemisinin/piperaquine phosphate (DUO-COTECXIN) and followed up for 28 day. The cure rate was 97.8%. The mean fever clearance time ($22.5{\pm}10.6hr$) and the mean parasite clearance time ($27.3{\pm}12.2hr$) showed no statistical significance with different genders, ages, temperatures, or parasite density (P>0.05). The resistance rates of chloroquine, piperaquine, pyronarididine, and artesunate detected in vitro were 71.9%, 40.6%, 12.5%, and 0%, respectively (P<0.0001). The resistance intensities decreased as follows: chloroquine>piperaquine>pyronarididine>artesunate. The inhibitory dose 50 ($IC_{50}$) was $3.77{\times}10^{-6}mol/L$, $2.09{\times}10^{-6}mol/L$, $0.09{\times}10^{-6}mol/L$, and $0.05{\times}10^{-6}mol/L$, and the mean concentrations for complete inhibition (CIMC) of schizont formation were $5.60{\times}10^{-6}mol/L$, $9.26{\times}10^{-6}mol/L$, $0.55{\times}10^{-6}mol/L$, and $0.07{\times}10^{-6}mol/L$, respectively. Dihydroartemisinin showed a strong therapeutic effect against falciparum malaria with a low toxicity.

Effects of energy-protein supplementation frequency on performance of primiparous grazing beef cows during pre and postpartum

  • Objective: Twenty-four pregnant Nellore primiparous grazing cows were used to evaluate the effects of energy-protein supplementation and supplementation frequency during pre (105 d before calving) and postpartum (105 d after calving) on performance and metabolic characteristics. Methods: Experimental treatments consisted of a control (no supplementation), daily supplementation (1.5 kg/d of concentrate/animal) and infrequent supplementation (4.5 kg of concentrate/animal every three days). During the pre and postpartum periods, concentrations of blood metabolites and animal performance were evaluated. Ureagenesis and energy metabolism markers were evaluated at prepartum period. Results: Supplementation frequency did not alter (p>0.10) body weight (BW), average daily gain (ADG), and carcass traits during pre and postpartum. The BW (p = 0.079), adjusted BW at day of parturition (p = 0.078), and ADG (p = 0.074) were greater for supplemented cows during the prepartum. The body condition score (BCS; p = 0.251), and carcass traits (p>0.10) were not affected by supplementation during prepartum. On postpartum, supplementation did not affect animal performance and carcass traits (p>0.10). The dry mater intake was not affected (p>0.10) by supplementation and supplementation frequency throughout the experimental period. Daily supplemented animals had greater (p<0.001) glucose levels than animals supplemented every three days. Supplementation and supplementation frequency did not alter (p>0.10) the levels of blood metabolites, neither the abundance of ureagenesis nor energy metabolism markers. Conclusion: In summary, our data show that the reduction of supplementation frequency does not cause negative impacts on performance and metabolic characteristics of primiparous grazing cows during the prepartum.

Effect of Varying Levels of Dietary Minerals on Growth and Nutrient Utilization in Lambs

  • Hisardale male lambs (n=24, 4-5 month of age) were maintained on a conventional ration for a month, to equilibrate the body mineral status. Six lambs were sacrificed after one month of feeding; the samples of organs were analyzed to ascertain mineral status. The remaining 18 lambs were divided into 3 groups of 6 each on body weight basis. Three dietary treatments containing 100 ($T_1$), 110 ($T_2$) and 120% ($T_3$) of minerals (Ca, P, S, Zn and Mn) as specified by NRC (1985) were formulated and each treatment was alloted ad libitum to a group for 120 days. Blood and wool samples were collected at monthly intervals. At the end of the experiment a balance trial of 5 days duration was conducted to study the balance of mineral elements. The effective intake of minerals was Ca, 111 and 120, P, 110 and 122; S, 112 and 129; Zn, 112 and 126 and Mn, 109 and 123 percent in $T_2$ and $T_3$, respectively, in comparison to $T_1$ (100). The additional mineral supplementation had no significant effect on dry matter intake. The average daily weight gain was higher (p<0.01) in $T_2$ and $T_3$ than the control ($T_1$). The lambs under treatments $T_2$ (8.72) and $T_3$ (8.47 kg) consumed apparently lesser amount of dry matter per unit gain as compared to $T_1$ (10.81 kg). Significantly higher (p<0.05) dry matter and crude protein digestibility (%) were observed in $T_2$ and $T_3$ than in $T_1$. The mean balances for different elements were Ca, 1.14, 1.68 and 1.67 g; P, 1.70, 1.95 and 2.18 g; S, 0.54, 0.92 and 1.11 g; Zn, 22.56, 25.30 and 28.71 mg; Cu, 7.94, 5.71 and 5.53 mg; Fe, 33.19, 32.94 and 31.03 mg and Mn, 8.24, 14.40 and 16.07 mg/lamb/day. The retention of supplemental minerals increased (p<0.01) while that of Cu decreased (p<0.01) due to supplementation of minerals (Ca, P, S, Zn and Mn). Retention as per cent of intake increased statistically for S and Mn while that of Cu decreased. It can be concluded that supplementation of minerals (Ca, P, S, Zn and Mn) higher than the recommended level improved body weight gain and feed to gain ratio. The retention of minerals increased due to supplementation. Therefore, an additional supplementation of deficient minerals (Ca, P, S, Zn and Mn) by 10% was beneficial for Hisardale male lambs under tropical condition in India.

Degummed crude canola oil, sire breed and gender effects on intramuscular long-chain omega-3 fatty acid properties of raw and cooked lamb meat

  • Background: Omega-3 long-chain (${\geq}C_{20}$) polyunsaturated fatty acids (${\omega}3$ LC-PUFA) confer important attributes to health-conscious meat consumers due to the significant role they play in brain development, prevention of coronary heart disease, obesity and hypertension. In this study, the ${\omega}3$ LC-PUFA content of raw and cooked Longissimus thoracis et lumborum (LTL) muscle from genetically divergent Australian prime lambs supplemented with dietary degummed crude canola oil (DCCO) was evaluated. Methods: Samples of LTL muscle were sourced from 24 first cross ewe and wether lambs sired by Dorset, White Suffolk and Merino rams joined to Merino dams that were assigned to supplemental regimes of degummed crude canola oil (DCCO): a control diet at 0 mL/kg DM of DCCO (DCCOC); 25 mL/kg DM of DCCO (DCCOM) and 50 mL/kg DCCO (DCCOH). Lambs were individually housed and offered 1 kg/day/head for 42 days before being slaughtered. Samples for cooked analysis were prepared to a core temperature of $70^{\circ}C$ using conductive dry-heat. Results: Within raw meats: DCCOH supplemented lambs had significantly (P < 0.05) higher concentrations of eicosapentaenoic (EPA, $20:5{\omega}3$) and EPA + docosahexaenoic (DHA, $22:6{\omega}3$) acids than those supplemented with DCCOM or DCCOC; Dorset sired lambs contained significantly (P < 0.05) more EPA and EPA + DHA than other sire breeds; diet and sire breed interactions were significant (P < 0.05) in affecting EPA and EPA + DHA concentrations. In cooked meat, ${\omega}3$ LC-PUFA concentrations in DCCOM (32 mg/100 g), DCCOH (38 mg/100 g), Dorset (36 mg/100 g), White Suffolk (32 mg/100 g), ewes (32 mg/100 g) and wethers (33 mg/100 g), all exceeded the minimum content of 30 mg/100 g of edible cooked portion of EPA + DHA for Australian defined 'source' level ${\omega}3$ LC-PUFA classification. Conclusion: These results present that combinations of dietary degummed crude canola oil, sheep genetics and culinary preparation method can be used as effective management tools to deliver nutritionally improved ${\omega}3$ LC-PUFA lamb to meat consumers.
