Effect of Triton X-100 on the Growth and Morphology of Trichoderma koningii (Triton X-100이 Trichoderma koningii의 성장 및 형태에 미치는 영향)

  • We investigated the effect of Triton X-100 on the growth and morphology of Trichoderma koningii by comparing various parameters representing the frowth of mold in the presence or absence of Triton X-100. The specific growth rate and doubling time of T. koningii were not affected by the addition of 0.05% Triton X-100 in batch culture. However, in the presence of Triton X-100, cultures reached its stationary phase earlier and showed reduced level in total yield of biomass. The addition of Triton X-100 into solid medium also resulted in decrease in the colony radial growth rate and this response was correlated with the formation of mycelia which showed increase in branching and septation in the presence of Triton X-100.

Investigation on Minimum Film Boiling Point of Highly Heated Vertical Metal Rod in Aqueous Surfactant Solution (계면활성제 수용액 내 고온 수직 금속봉의 최소막비등점에 대한 연구)

  • In this study, experiments were conducted on the MFB(minimum film boiling) point of highly heated vertical metal rod quenched in aqueous surfactant solution at various temperature conditions. The aqueous Triton X-100 solution(100 wppm) and pure water were used as the liquid pool. Their temperatures ranged from $77^{\circ}C$ to $100^{\circ}C$. A stainless steel vertical rod of initial center temperature of $500^{\circ}C$ was used as a test specimen. In both liquid pools, as the liquid temperature decreased, the time to reach the MFB point decreased with a parallel increase in the temperature and heat flux of the MFB point. However, over the whole present temperature range, in the aqueous Triton X-100 solution, the time to reach the MFB point was longer, while the temperature and heat flux of the MFB point were reduced when compared with pure water. Based on the present experimental data, this study proposed the empirical correlations to predict the MFB temperature of a high temperature vertical metal rod in pure water and in aqueous Triton X-100 solution.

Effect of Triton X-100 on Compactin Production from Penicillium citrinum

  • Glucose alone was found to be the most effective carbon source for producing compactin. An initial glucose concentration of 40 g/L gave the highest compactin concentration of 250mg/L. Among the various nitrogen sources, when 5g/L of pharmamedia and soybean meal as the sole nitrogen source were used, respectively, the compactin concentration was higher than 250mg/L. Especially, in the case of the mixture of 6 g/L of pharmamedia and 8 g/L of soybean meal, the compactin concentration was 400mg/L. To select the best surfactant for effective compactin production, various surfactants were investigated. When Triton X-100 was used, the maximum compactin concentration was 445mg/L. With the initial concentration ranging from 1.5 to 2.0 g/L, the compactin concentration was the highest at 465-450mg/L. The cell concentration was similar to that of the control without the addition of Triton X-100. On the other hand, when the above 4.0 g/L of Triton X-100 were used, the cell concentration decreased. Using the based results, the continuous fed-batch cultures by adding the Triton X-100 were carried out for 10 days in an air-lift bioreactor. When 1.5 g/L of Triton X-100 was added to the culture broth at 0, 4, and 8 days of culture, respectively, the compactin production was increased with the increase of culture time. The maximum compactin concentration after 10days of culture was 1,200mg/L, which was about 2.0-fold higher than that of the control without the addition of Triton X-100.

  • In this study, we reported the effect of porcine pancreas lipase treatment in the presence of a calcium chloride and Triton X-100 on moisture regain and wettability of PET fabrics. The moisture regain of PET fabrics in the presence of 0.5% surfactant showed a 1.5-fold decrease, compared to the absence of it. Triton X-100 acted as an inhibitor to porcine pancreas lipase hydrolytic activity. The moisture regain and wettability of porcine pancreas lipase treated PET fabrics improved when more than 10mM of calcium chloride was added to the treatment solution. Porcine pancreas lipase treatment caused voids and cracks on PET fabrics.

  • Calcification is a major problem in glutaraldehyde-preserved bioprosthetic valves. We have used bovine pericardium processed in a solution containing 0.625% glutaraldehyde, 0.05M HEPES buffer and 0.26% magnesium chloride in saline. And, we also treated the glutaraldehyde-preserved bovine pericardium with a surfactant, Triton X - 100 to reduce calcification. To evaluate the degree of calcification. 4 kinds of pericardial xenografts, group I [Xenomedica, equine pericardial xenografts], group II [0.625% glutaraldehyde-preserved bovine pericardiums], group III [0.5% Triton X - 100 treated bovine pericardiums], and group IV [1.2% Triton X - 100 treated bovine pericardiums] were implanted in subcutaneous layer of growing rabbits, and they were explanted about 3 months later. The mean calcium contents[%/mg of dry tissue] of 0.5% and 1.2% Triton X - 100 treated bovine pericardiums [80.0$\pm$27.1%: 78.6$\pm$47.0% respectively] were lower than those of glutaraldehyde-preserved bovine pericardiums[126.2$\pm$29.8] [p=0.05]. Thus, under the conditions of subcutaneous implantation in rabbits, Triton X - 100 was efficient in calcification mitigation.

  • As a pary of investgation on effective accumulation of cadmium in yeast cells, effect of surfactant was studied on intracellular accumulation of cadmium in extremely cadmium-tolerant yeast, Hansenula anomala B-7. Cadmium accumulation was enhanced up to approximately 40% by the addition of non-ionic surfactant, Triton X-100 and its optimal concentration was found to be 0.1-0.2%. Furthermore, optimum conditions for intracellular accumulation of cadmium were at $40^{\circ}C$ and initial pH 6.0 for 48 hours under shaking culture.

  • Triton X-100 enhances to a marked extent the analytical sensitivity of the nitrobluetetrazolium (NBT) reduction method for the assay of superoxide$(O^-_{\.{^.2}})$ production. In the present work, it was attempted to elucidate the physicochemical nature of this Triton X-100 effect, focusing on not only the surfactant-caused stabilization of the water-insoluble formazan colloid but also the kinetic competition between the $NBT-O^-_{\.{^.2}}$ reaction and the autodisproportionation of concommitantly occuring in aqueous media. The measurements of formazan and $H_2O_2$ formed in a number of reaction systems, as prepared by vortex-mixing potassium superoxide dissolved in an aprotic solvent with aqueous solutions of NBT, revealed that Triton X-100 exerts its effect both through preventing formazan colloid from aggregation and thereby increasing the formazan absorbance and through suppressing the autodisproportionation reaction of $O^-_{\.{^.2}}$. It also turned out that the relative shares of the colloid stabilization effect and the kinetic effect in the contribution to the sensitivity amplification of the NBT method are dependent upon the reaction conditions, particulary the molar concentration ratio of NBT to $O^-_{\.{^.2}}$ in the reaction systems.

Surfactant Selection for the Enhanced Biological Degradation of Toluene (Toluene의 생물학적 분해능 향상을 위한 계면활성제의 선정)

  • Surfactants can be used to enhance the mass transfer rate of hydrophobic compounds into the biologically active liquid phase, resulting in an increase in biodegradation rate of toluene. In this study, the mass transfer rate and the biocompatibility of toluene in the presence of various surfactants were evaluated. Four anionic and non ionic surfactants were tested: sodium dodecyl sulfate (SOS), TritonX-100, Tween 80, and BYK-345 (silicone surfactant). Experimental results showed that BYK-345 at the critical micelle concentration (CMC) enhanced the solubility of toluene. However, there was no increase in the solubility of toluene by SOS and TritonX-100 at their CMCs. With the addition of each surfactant into deionized water the mass transfer rate became faster than that of the case with no surfactant. A bottle study using toluene-degrading microorganisms showed that SOS seriously reduced toluene removal presumably due to the toxicity of the anionic surfactant and/or the substrate competition between the surfactant and toluene. In addition, the degradation rate of toluene was decreased in the presence of BYK-345, indicating that BYK-345 adversely affects the activity of microorganisms. However, TritonX-100 and Tween 80 did not decrease the degradation rate of toluene significantly. Rather, at the low concentration of TritonX-100 toluene degradation rate was even increased. Overall the experimental results suggest that TritonX-100 be the appropriate surfactant for enhanced biological degradation of toluene.

Composition of Lipids Associated with Dense Coat-enriched Fractions of Bovine Milk Fat Globule Membrane (우유 지방구막의 고밀도 표피에 결합된 지질의 조성)

  • To analyze the components of dense coat fractions associated with fat globule membrane, The membrane was treated with various concentrations of Triton X-100, a non-ionic detergent, and the composition of lipids associated to the detergent insoluble material was analyzed. The amount of protein, phospholipid, cholesterol and ganglioside in milk fat globule membrane was reduced consistently with increasing concentrations of Triton X-100. Butyrophilin (band 12), xanthine oxidase (band 3) and band 16 as constituents of insoluble coat materials was revealed after electrophorisis on SDS-polyacrylamide gels. Phosphatidyl ethanolamine, phosphatidyl choline, phosphatidyl serine, phosphatidyl inositol and sphingomyelin were identified as the major phospholipids of the coat materials without selective concentration relative to the original membranes. Percentages of total phospholipid were not changed by any of the treatments. Fatty acids of total lipid were myristate, palmitate, stearate (major saturated acids), oleate and linoleate (major unsaturated acids). Cholesterol contents on a protein basis were slightly reduced with increasing concentrations of Triton X-100. Cholesterol adhered to protein more tightly than other constituents The contents of gangliosides was proportionally refuted with increasing concentration of Triton X-100.
