To find the antidiabetic activity of the tripterpenoid-rich fractions of Rubus coreanus (TRF-cor) and R. crataegifolius (TRF-cra) leaves or its main component niga-ichigoside $F_1$ (Niga-$F_1$), anti-hyperglycemic and antihyperlipidemic effects were investigated in the diabetic rat model induced by streptozotocin (STZ). Treatments of rats with 200 mg/kg of the TRF-cor, TRF-cra (each, p.o.) or 20 mg/kg of Niga-$F_1$ significantly inhibited the increase of blood glucose concentration about 44.8%, 28.7% or 20.6%, respectively, in the diabetic rats. In addition, treatments with those fractions inhibited the increase of serum concentrations of triglyceride, total cholesterol or LDL-cholesterol caused by STZ. The inhibitory rate on atherogenic index (AI) values of the TRFcor (200 mg/kg), TRF-cra (200 mg/kg) or Niga-$F_1$ (20 mg/kg)-treated groups were decreased about 55.7%, 36.3% or 22.6%, respectively, comparable to STZ-treated group. In the oral glucose tolerance test, treatment of TRF-cor or TRF-cra inhibited the increase of blood glucose concentration in the STZ-induced rats. Administration of 20 mg/kg of Niga-$F_1$ (p.o.) also exhibited similar effects with the effects of both TRFs at 200 mg/kg dose (p.o.). These results support that the triterpenoids, in particular Niga-$F_1$, are contributed to the antidiabetic effects of R. coreanus or R. crataegifolius.

To produce an intestinal immunomodulatory beverage containing Centella asiatica extract (CAE), three types of CAE-added beverage prototypes were prepared, and their immunomodulatory activities and marker compounds were analyzed. As a result of the cytotoxicity assessment, all the beverages did not show significant toxicity compared to the control group. Next, the immunomodulatory activities of the beverage prototype were evaluated using the inflammatory model of IL-1β-induced intestinal epithelial cell line. All the samples significantly reduced the production of IL-6, IL-8, and MCP-1 in a CAE concentration-dependent manner. In addition, CAE-added beverages inhibited NO, IL-6, and IL-12 production in LPS-induced RAW 264.7 cells. When the major triterpenoids, as marker compounds for the production of CAE-added beverages, were analyzed by HPLC-DAD, only asiaticoside was detected beyond the limit of quantification, while madecassoside, madecassic acid, and asiatic acid were not detected. The amounts of asiaticoside in CAE-added beverage prototypes were confirmed in No. 1 (19.39 ㎍/mL), 2 (19.25 ㎍/mL), and 3 (19.98 ㎍/mL). In conclusion, the results of this study suggested that CAE-added beverage prototypes induced immunomodulatory effects in the intestinal inflammatory cell line models and asiaticoside could be used as a marker compound for CAE-added beverage production.
Achyranthes japonica extract (AJE) is derived from a medicinal plant Achyranthes japonica, known for its anti-inflammatory, antioxidant, and antimicrobial properties. AJE contains multiple bioactive compounds, including saponins, triterpenoids, phytoecdysteroids, 20-hydroxyecdysone, and inokosterone. The aim of this investigation was to examine the impact of AJE as a phytogenic feed additive on growth performance, nutrient digestibility, excreta microbial count, noxious gas emissions, breast meat quality in broilers. About three hundred and sixty, day-old broilers (Ross 308) were assigned into four treatments (five replication cages/treatment, and 18 birds/cage). Dietary treatments: CON, basal diet; 0.02% AJE, basal diet with 0.02%; 0.04% AJE, basal diet with 0.04% AJE, and 0.06% AJE, basal diet with 0.06% of AJE. Body weight gain increased linearly (p < 0.05) through the inclusion of AJE during days 7 to 21, 21 to 35, as well as the entire experimental period. Besides, feed intake increased (p < 0.05) linearly during days 21 to 35 and the entire experiment with the increased AJE doses in broiler diet. Dry matter digestibility was increased (p < 0.05) linearly along with increasing amounts of AJE. With increasing AJE supplementation, nitrogen and energy utilization tended to improve (p < 0.10). In summary, the addition of AJE in the corn-soybean meal diet led to higher body weight gain and increased feed intake as well as enhanced nutrient digestibility, among them the highest improvement was found in 0.06%-AJE indicating the acceptance of AJE as a phytogenic feed additive.
The aerial parts of Sajabalssuk (Artemisia princeps PAMPANINI, Sajabalssuk) was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc fraction, three cycloartane-type triterpnoids and one ursane-type triterpenoid were isolated through the repeated silica gel, ODS and Sephadex LH-20 column chromatographies. From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the triterpenoids were determined as wrightial (1), wrightial acetate (2), 27-norcycloart-20(21)-ene-25-al-3${\beta}$-ol acetate (3) and ursolic acid (4). No report has been found for isolation of compound 3 in the literature so far, and compounds 1, 2 and 3 were the first to be isolated from Sajabalssuk (Artemisia princeps PAMPANINI, Sajabalssuk). Also, compound 1 showed Acyl-CoA:Cholesterol acyltransferase (hACAT-1) and hACAT-2 inhibitory activity with the $IC_{50}$ values of 33.0 and 45.0 ${\mu}g/ml$, respectively. Compounds 2 and 3 inhibited hACAT-1 activity with the $IC_{50}$ values of 12.0 and 16.0 ${\mu}g/ml$, respectively.

The leaves and stems of Centella asiatica have a long history of their usage as a medicine for the treatment of skin diseases such as ulcers and psoriasis, especially in Asia. Triterpenoids, the active components of Centella asiatica including asiaticoside, madecasosside, asiatic acid and madecassic acid, have shown to inhibit skin inflammation as well as improve skin photoaging. The main objective of this study is to investigate whether the Centella asiatica ripened with lava seawater which is rich in minerals known to be beneficial to human body can provide anti-inflammatory and moisturizing effects to skin. HPLC analysis showed that the concentration of triterpenoids increased further after ripening Centella asiatica with lava seawater. In order to confirm the inflammatory efficacy of the extract of the extract of the ripened Centella asiatica, the production of NO in LPS-activated RAW 264.7 cells and the expression of inflammatory cytokines in PM10 or UVB-induced HaCaT cells were observed. We found that the extract of the ripened Centella asiatica inhibited the expression of NO, IL-6, IL-8, and TNF-a and had higher inhibitory effect compared to the extract of the non-ripened Centella asiatica. In order to confirm the skin moisturizing effect, we investigated the synthesis of HA in HaCaT cells. The result showed HA production was enhanced in a concentration-dependent manner from the ripened group, while there was no efficacy from the non-ripened group. Taken together, it is concluded that the extract of the Centella asiatica ripened with lava seawater was effective in anti-inflammation and moisturization.

Skin aging is a complex biological process due to intrinsic and extrinsic factors. Free radical oxidative is one of extrinsic factors that induce activation of collagenase, elastase and hyaluronidase. Natural product from plants has been used as antioxidant and antiaging. This study aimed to evaluate antioxidant and antiaging properties of Hibiscus sabdariffa extract (HSE) and its compounds including myricetin, ascorbic acid, and ${\beta}$ carotene. The phytochemical of H. sabdariffa was determined using modified Farnsworth method and presence of phenols, flavonoids and tannins were in moderate content, whereas triterpenoids and alkaloids were in low content. Total phenolic content performed using Folin-Ciocalteu method, was $23.85{\mu}gGAE/mg$. Quantitative analysis of myricetin, ${\beta}-carotene$, and ascorbic acid of HSE was performed with Ultra-High Performance Liquid Chromatography (UHPLC) that shows $78.23{\mu}g/mg$ myricetin, $0.034{\mu}g/mg$${\beta}-carotene$, whilst ascorbic acid was not detected. HSE has lower activity on DPPH ($IC_{50}=195.73{\mu}g/mL$) compared to ${\beta}-carotene$, the lowest in ABTS assay ($IC_{50}=74.58{\mu}g/mL$) and low activity in FRAP assay ($46.24{\mu}MFe(II)/{\mu}g\;$) compared to myricetin, ${\beta}-carotene$. Antiaging was measured through inhibitory activity of collagenase, elastase, and hyaluronidase. HSE had weakest collagenase inhibitory activity ($IC_{50}=750.33{\mu}g/mL$), elastase inhibitory activity ($103.83{\mu}g/mL$), hyaluronidase inhibitory activity ($IC_{50}=619.43{\mu}g/mL$) compared to myricetin, ${\beta}-carotene$, and ascorbic acid. HSE contain higher myricetin compared to ${\beta}-carotene$. HSE has moderate antioxidants and lowest antiaging activities. Myricetin is the most active both antioxidant and antiaging activities.
Tiarella polyphylla D. Don(Saxifragaceae) is a perennial herb and distributed in China, Japan, Taiwan and Korea. Especially, it only grows in Ulleung island of Korea. It has been using for asthma, bruise and audition troubles with main components of some Triterpenoids and seven oleanolic Saponins. There is only known its chromosomal number rarely and cytogenetic study was not done. From this study, karyotype analysis and chromosomal localization of 5S and 45S rDNAs using bicolor-FISH(fluorescence in situ hybridization) were carried out. The somatic metaphase chromosome number was 2n=2x=14 and the size of chromosomes ranged $1.66{\sim}3.50{\mu}m$. The chromosome complement consisted of four pairs of submetacentrics(chromosomes 1, 2, 3 and 6), two pairs of subtelocentrics(chromosomes 5 and 7) and one pair of telocentrics(chromosome 4). We also observed NOR(nucleolus organizer region) on the chromosome 4. In bicolor-FISH, one pair of 55 and 45S rDNA sites was detected on the centromeric region of chromosome 3 and short arm of chromosome 4, respectively. Bicolor FISH was very useful tool for the localization and identification of rDNAs on the chromosomes in Tiarella polyphylla.

Centella asiatica (L.) Urban is a medicinal plant containing madecassic acid, triterpenoids and asiaticoside, and has a wound healing effect. However, since it grows only in some regions of Korea, it is necessary to breed stable varieties. In this study, it were analyzed for the growth characteristics, asiaticoside, anxiodant components and 2,2-diphenyl-1picryl hydrazyl (DPPH) radical scavening activity of C. asiatica collected from domestic natural habitat (Hapcheon, Gyeongnam). In the growth characteristics of each resource, CA9 and CA10 were superior in characteristics of aerial part and in dry weight. In asiaticoside content analysis, CA5 and CA3 were high with 53.96 mg/g and 52.40 mg/g, respectively. CA12 showed the possibility of development because the resource showed high in total flavonoid (7.3 mg/g), total polyphenol (10.64 mg/g) and DPPH activity (11.96 mg AAE/g) among other resources. However, it is necessary to select a resource suitable for each characteristic of C. asiatica and to study charateristics through additional resources collection since the excellent growth characteristics of the collected resources do not necessarily coincide with the content and antioxidant activities.

Purpose: The Rubus coreanus fruit (RF) is an important traditional medicinal herb having antioxidant, anti-inflammatory, and immunoregulatory properties. These activities are known to change dramatically, depending on maturity of the RF. It is presumed that change of functional components, such as flavonoids, tannins, phenolic acids, triterpenoids and organic acids in RF, affect the various bioactivities. This study aimed to confirm changes in the anti-melanogenic effects of RF based on maturity, and to identify the bioactive compounds responsible. Methods: The cell viability of mature RF (MRF) and immature RF (IRF) extracts was investigated using B16F10 cells. To compare the anti-melanogenic effect of MRF and IRF extracts, we first assessed the melanin content. High-performance liquid chromatography analysis was performed to evaluate changes in the level of ellagic acid according to maturity of the RF. In addition, tyrosinase inhibitory activity of both extracts was examined. Results: MRF and IRF extracts (50-200 ㎍/mL) do not affect the cell viability of B16F10 melanoma cells. IRF extract more effectively inhibited melanin synthesis than MRF extract. The content of ellagic acid in IRF extract was higher than that obtained in MRF extract. Furthermore, greater inhibition of tyrosinase activity was observed after exposure to IRF extract than MRF extract. A positive correlation was determined between ellagic acid content and tyrosinase inhibitory activity, and a negative correlation was obtained between ellagic acid content and melanin content. Taken together, our results indicate that ellagic acid is one of the major bioactive compounds of RF that imparts a whitening effect. Conclusion: Our results indicate that ellagic acid in MRF and IRF extracts affect the anti-melanogenesis effect through inhibition of tyrosinase activity. Therefore, the ellagic acid rich IRF has greater potential for application as a natural and functional cosmetic material.
Sorbus commixta Hedl. has traditionally been used as a remedy for cough, asthma, and other bronchial disorders. In this study, three major triterpenoids-lupeol, β-sitosterol, and ursolic acid and a coumarin, scopoletin, were isolated from a CHCl3-soluble fragment of the bark of S. commixta. Their structures were identified by spectroscopic analyses, including mass spectrometry (MS), 1D-, and 2D- nuclear magnetic resonance spectroscopy (NMR), as well as by comparing the data with data reported in the literature. Scopoletin was isolated from this plant for the first time. It is a nutraceutical compound contained in many plants that has been reported to exert diverse biological activities, including anti-inflammatory effects. This study examined the inhibitory effect of scopoletin on TNF-α-induced vascular endothelial inflammation. Unlike the marginal impact of other compounds against low-density lipoprotein (LDL) oxidation and vascular endothelial inflammation, scopoletin showed remarkable activity on LDL oxidation (IC50 = 10.2 μM) and exerted vascular anti-inflammatory effects in EA.hy926 human endothelial cells activated by TNF-α. It suppressed the expression of adhesion molecules, such as ICAM-1, VCAM-1, and E-selectin, and blocked the adhesion between THP-1 monocytes and EA. hy926 endothelial cells. It also inhibited TNF-α-induced NF-κB translocation from the cytosol to the nucleus. Moreover, IκBα phosphorylation, which was increased by TNF-α treatment, was reduced after treatment with scopoletin. Thus, scopoletin inhibited TNF-α-induced vascular inflammation in endothelial cells by suppressing the NF-κB signaling pathway. These results demonstrate that owing to its anti-inflammatory activity in the vascular endothelium, scopoletin has the potential to inhibit atherosclerosis development.
