Triploidy Production of Nile Tilapia, Oreochromis niloticus (3 배체 나일틸라피아 생산에 관하여)

  • Fertilized eggs of Oreochromis niloticus were subjected to cold treatments at $14^{\circ}C$ for 30, 45 and 60 minutes starting 5 minutes after insemination at $27^{\circ}C$. Ploidy levels were determined by chromosome preparations and the analysis of both cell and nuclear sizes. A temperature shock of $14^{\circ}C$ for 60 minutes yielded the best results ( $83.3\%$). Gonadal development in both sexes was severely retarded in all triploid groups at 6 months of age.

Utility of Selected Non-coding Chloroplast DNA Sequences for Lineage Assessment of Musa Interspecific Hybrids

  • Single-copy chloroplast loci are used widely to infer phylogenetic relationship at different taxonomic levels among various groups of plants. To test the utility of chloroplast loci and to provide additional data applicable to hybrid evolution in Musa, we sequenced two introns, rpl16 and ndhA, and two intergenic spacers, psaA-ycf3 and petA-psbJ-psbL-psbF and combined these data. Using these four regions, Musa acuminata Cola(A)- and M. balbisiana Colla (B)-containing genomes were clearly distinguished. Some triploid interspecific hybrids contain A-type chloroplasts (the AAB/ABB) while others contain B-type chloroplasts (the BBA/BBB). The chloroplasts of all cultivars in 'Namwa' (BBA) group came from the same wild maternal origin, but the specific parents are still unrevealed. Though, average sequence divergences in each region were little (less than 2%), we propose that petA-psbJ intergenic spacer could be developed for diversity assessment within each genome. This segment contains three single nucleotide polymorphisms (SNPs) and two indels which could distinguish diversity within A genome whereas this same region also contains one SNP and an indel which could categorize B genome. However, an inverted repeat region which could form hairpin structure was detected in this spacer and thus was omitted from the analyses due to their incongruence to other regions. Until thoroughly identified in other members of Musaceae and Zingiberales clade, utility of this inverted repeat as phylogenetic marker in these taxa are cautioned.

  • Present study was aimed to summary the recent reports of chromosomal technology such like a polyploidv, sex differentiation, gynogenesis, transgenic fish and gene manipulation. Triploid cells for rainbow trout and channel catfish were induced through thermal shocks of varying temperature levels and produced as a industrial use. A monosex fish with homogametic females of 15 species of high valued fish were produced by exposing to irradiation. It seemed that different irradiation was suitable to inactivate the sperm and block the formation in producing the gynogenetic diploids. Since 1985, transgenic fish have been successfully produced by microinjecting or electroporating desired foreign DNA into unfertilized or newly fertilized eggs using about 40 fish species. More recently, transgenic fish have also been produced by infecting newly fertilized eggs with pantropic, defective retroviral vectors carrying desired foreign DNA. These transgenic fish can serve as excellent experimental models for basic scientific investigations as well as in marine biotechnological applications.

Breeding of a Seedless Table Grape Cultivar 'Heukisul' (Vitis sp.) with High Quality (고품질 무핵 포도 품종 '흑이슬' 육성)

  • 'Heukisul' (3x) a cross made in 2000 between 'Kyoho' (4x, Vitis sp.) and 'Thompson Seedless' (2x, Vitis vinifera L.) in a seedless grape breeding program, was preliminarily selected in 2004. After regional adaptation testing under the name of 'KTS014' at two sites during 2004-2005, it was finally selected in 2006. 'Heukisul' showed low incidence of berry shattering, resulting in a good berry set compared to 'King Dela'. 'Heukisul' had budburst on April 23, flowering on in June 9, and fruit maturation in September 29 (almost the same as 'King Dela' at Chuncheon), and it was considered a midseason cultivar. The mean berry weight was 4.0 g, about 0.7 g heavier than 'King Dela', and mean soluble solids were $21.9\;^{\circ}Brix$, about $2.7\;^{\circ}Brix$ higher than 'King Dela'. The skin color was dark violet with abundant bloom and the flesh was very firm. Although the cluster was compact, it required no cluster thinning. Also the incidence of berry cracking was very low.

Identification of Gene Locus by the Somatic Cell Hybridization in Chicken (체세포 융합에 의한 닭의 유전인자구명에 관한 연구)

  • This experiment was conducted to improve the performance of chickens by the precise separation and analysis of chromosomes which are integrated genetic materials, and by the use of gene manipulation techniques. Following are the main results obtained. 1. When the chromosomes were separated through the leucocyte culture and analyzed by Giemsa banding techniques (especially by the method in which 20 layers of banding patterns could be found in chromosome #1), the normal Patterns of chromosomes #l-9 and sex chromosomes, and the location of constitutive heterochromatin without any gene activities in all chromosomes were discovered. 2. To utilize the primodial germ cells (PGC) as the genetic vector which is one of the most important gene manipulation techniques, PGC's from triploid were transplanted to normal host embryos. Since the donor PGC's(3n) were found in the gonads of growing host embryos gene manipulation in poultry using PGC's, seemed to be possible.

Taxonomic Identity of Leaf Fragments Found in the Annals of the Joseon Dynasty and Botanical Origin of a Herbal Medicine 'Cheongung' (조선왕조실록 갈피에서 발견된 잎 조각의 실체 및 천궁의 식물학적 기원)

  • Tiny leaf fragments were found in the Annals of the Joseon Dynasty, which were compiled about 500 years ago. The records describing the detailed process of compiling the Annals indicate that silk bags packed with the powders of 'Cheongung' and 'Changpo', which have been used as traditional herbal medicines in the northeast Asian countries such as China and Japan as well as Korea, were put in the wooden storage boxes together with the volumes of the Annals. However, there is no record that parts of plants were used in the process of compiling the Annals. The botanical origin of leaf fragments was identified as Ligusticum sinense 'Chuanxiong' by the analysis of trnK of chloroplast DNA as well as the examination of leaf surface with SEM. The comparative analysis of trnK sequences showed that the chloroplast DNA haplotype of 'Tocheongung', a triploid species cultivated in Korea, was identical with Cnidium officinale, but different from L. sinense 'Chuanxiong'. The molecular results provide a new suggestion on the botanical origin of crude drugs used as 'Cheongung', which has been disputed in Korea.

Breeding of Good Quality Leaves High Yielding Mulberry Cultivar "Suilppong" (Morus alba L.) (양질다수성 뽕품종 "수일뽕" 육성)

  • New triploid mulberry cultivar, Suilppong was bred among selected lines from F$_1$ seedlings obtained by crossing the female, artificial tetraploid No. 411 which obtained from Chungilppong seedlings, and the male, Kugsang 21. Local adaptability test has been carried out at four places (Suwon, Kongju, Jeonju and Jangsung) for five years since 1997. Leaf quality was tested by silkworm rearing for two silkworm rearing seasons (spring and autumn) at Suwon in 2000. Suilppong was recognized to be superior in productivity to control cultivar, Kaeryangppong. The new cultivar, Suilppong, has following agronomic characteristics as compared with Kaeryangnppong, which is leading cultivar widely cultivating in Korea. It is belonging to Morus alba L. with mixed shapes from the ovate to 5 lobated leaf shape, sprouting in spring nearly the same to the Kaeryangppong (Morus alba L.), high leaf yielding of 20% compared to Kaeryangppong (Morus alba L.). It is adaptable to everywhere except the places where cold damage occurs frequently in Korea. Suilppong is recommended to rear silkworms not only in spring but also in autumn besides for young silkworms.

Current status on Miscanthus for biomass (바이오매스로서의 억새에 대한 연구 동향)

  • The carbon dioxide concentration of the atmosphere is projected to increase by almost 50% over the first 50 years of this century. The major cause of this increase is continued combustion of fossil fuels. As a result, the significant changes in climate that have already occurred will be amplified, in particular a global temperature increase. Renewable energy production has a central role to play in abating net $CO_2$ emissions to a level that will arrest the development of global warming. Especially, biomass crops are becoming increasingly important as concerns grow about climate change and the need to replace carbon dioxideproducing fossil fuels with carbon-neutral renewable sources of energy. To succeed in this role, biomass crop has to grow rapidly and yield a reliable, regular harvest. A prime candidate is Miscanthus, or Asian elephant grass, a perennial species that produces over 3 metres of bamboo-like stems in a year. Miscanthus species are typically diploid or tetraploid. Hybrids between species with different ploidy levels result in the highly productive triploid hybrids, M. ${\times}$ giganteus. Here we will detail the Miscanthus characteristics desired of a biomass fuel crop.

Relationship of diploid East Aisan Taraxacum Wiggers using the capitulum morphological character (형태형질에 의한 동아시아산 민들레속 2배체 식물의 유연관계)

  • Genus Taraxacum propagated through diploid sexual reproduction and polyploid agamospermy. The cluster analysis of Korean(2 species, 4 population), Japanese (2 species 4 taxa 6 population) and Taiwanese (1 species, 3 population) Taraxacum species using 15 measured capitulum morphological characters was conducted to study the speciation of diploid Taraxacum in East Asia. We measured 15 capitulum morphological characters including length of capitulum, length and shape of outer-involucre, corniculate appandage. Within one population, these characters were very various and were overlapped. The result of cluster analysis using morphological character showed that all species were clustered into four groups (Korean species & T. japonicum group, T. platycarpum subspecies group, T. platycarpum subsp. hodense group, T. formosanum group). Korean species, T. hallaisanense (diploid) and T. ohiwanum (diploid, triploid) were clustered into T. japonicum that was occurred in Kansai provinces of Japan. Therefore, we could infer that Korean species was closely related to Japanese T. japonicum of Japanese species.

Production of Hybrid and Alotriploid between Rainbow Trout, Oncorhynchus mykiss and Cherry Salmon, O. masou I. Cytogenetic study (무지개송어, Oncorhynhus mykiss와 산천어, O. masou간의 잡종 및 잡종 3배체 생산 I. 세포유전학적 연구)

  • Induction of hybrid and allotriploid between female rainbow trout, oncorhynchus mykiss and male cherry salmon, O. masou were performed. Triploid rate in allotriploid was 92% and the survival rates of allotriploid exceeded that of their hybrid counterparts. Karyological studies revealed that the hybrid had exactly the same intermediate diploid karyotypers as that of two parental species, while allotriploid had two sets of the maternal haploid complement and one set of the paternal haploid complement. Hybrid revealed an intermediate size in celluar and nuclear size of erythrocyte, while allotriploid revealed larger cellular and nuclear size of erythrocytes than hybrid. The number of nucleolar organizer regions of hybrid were 1 or 2, while those of allotriploids were 1, 2 or 3. These results proved that allotriploidization can lead to increase cytogenetical characters compared to counterpart hybridization.
