Cholesterol-Lowering Effects in Rat Liver Fed Barley and ${\beta}-Glucan-enriched$ Barley Fraction with Cholesterol (보리 및 보리 ${\beta}-glucan$ 농축획분 섭취에 의한 흰쥐의 간 콜레스테롤 저하효과)

  • Hypocholesterolemic effects in rats of two hull-less barleys, waxy and nonwaxy-starch genotype, were evaluated by analyzing liver cholesterol and triglycerides. A group fed cellulose control and fiber-free exhibited significant liver lipid accumulation by morphologically observation. In barley fed group, however, lipid disposition in liver was effectively suppressed. The concentrations of cholesterol and triglyceride in liver were significantly reduced in rat fed 5 and 10% fiber diets containing barley flour and ${\beta}-glucan-enriched$ fraction. ${\beta}-Glucan-enriched$ barley fraction as a fiber source showed more cholesterol-lowering than barley flour with cellulose, and this effect of ${\beta}-glucan$ from two hull-less barleys was similar. Liver cholesterol and triglyceride in cellulose control group were 14.5 mg/g and 46.1 mg/g, respectively, while those of barley and ${\beta}-glucan$ fed groups were $5.1{\sim}8.2\;mg/g$ and $20.1{\sim}31.3\;mg/g$, respectively.

Studies on the Lipid Content and Neutral Lipid Composition of Brown Rice and Milled Rice (현미와 백미의 지질 함량 및 중성 지질의 조성에 관한 연구)

  • The lipid contents and neutral lipid components of brown rice, milled rice and bran were studied for four varieties of rice such as Nampung, Milyang #23, Whasung and Jinhung. Total lipid contents of brown rice, milled rice and bran were 2.65%, 1.09% and 20.24% respectively. The ratios of neutral lipids, glycolipids and phospholipids in total lipids were 82.53:12.39:4.08 in brown rice and 87.72:7.02:5.26 in bran. Neutral lipids were separated on the TLC; among them 6 neutral lipids were identified by comparing the RF value of standards. Triglycerides(TG), free fatty acids(FFA) and steryl esters(SE) were major neutral lipid components in brown rice, milled rice and bran. TG content of the bran, compared to that of brown rice and milled rice, was very low in contrast the FFA content was high. The major fatty acids of total lipid and neutral lipid fractions were palmitic, linoleic and oleic acids, comprising over 95% of these classes.

Effects of Gami-Handayeolso-Tang on Body Fat Reduction in High Fat Diet-Fed Obese Mice (가미한다열소탕(加味寒多熱少湯)이 고지방식이 비만생쥐의 체지방감소에 미치는 영향)

  • Objectives In this study, it was investigated whether Gami-Handayeolso-Tang (HDYST) medication has anti-obesity effects in high fat diet (HFD)-fed obese mice. Methods The experimental animals were divided into five groups-normal diet-fed (ND), high fat diet-fed control (HFD), HFD+HDYST 150, HFD+HDYST 300, and HFD+orlistat as a positive drug. The obese markers such as body weight, diet efficiency ratio, serum levels of total cholesterol, triglyceride, lipid contents, leptin, adiponectin, and GOT/GPT were measured. Also, white adipose tissue, liver weight, abdominal fat mass, hepatic lipid contents, and mRNA expression of obese-associating genes were examined in obese mice. Results In high fat diet-fed mice, HDYST administration significantly decreased body weight, diet efficiency ratio, serum levels of total cholesterol, triglyceride, LDL-cholesterol, as well as leptin and GOT/GPT, compared to the HFD group in a dose-dependent manner. HDYST increased significantly the serum levels of HDL-cholesterol and adiponectin. It also reduced the accumulation of lipids, such as total lipid and triglycerides, in organs such as liver and abdominal adipose tissue. Moreover, HDYST administration significantly decreased the expression levels of fatty acid synthetic genes, such as sterol regulatory element-binding protein-1c (SREBP-1c), FAS and Stearoyl-Coenzyme A desaturase 1 (SCD-1), in the liver tissues, while it increased the messenger RAN (mRNA) levels of fatty acid catalytic genes, such as Peroxisome proliferator activated receptor alpha (PPAR-${\alpha}$), acyl-COA oxidase (ACO), and Carnitine palmitoyltransferase-1a (CPT-1a). Conclusions Based on the results above, HDYST reveals anti-obesity effects declining body fat accumulation through the regulation of fatty acid metabolism and leptin/adiponectin serum levels. It therefore suggests that HDYST can be clinically useful for the treatment of obesity.

The Correlations of Walking Exercise Program-Induced Abdominal Visceral Fat Loss with Metabolic Syndrome Risk Factors (걷기운동 프로그램에 의한 복부 내장지방의 감량과 대사증후군 위험인자 간의 관련성)

  • The purpose of current study was to investigate the correlations of walking exercise program-induced abdominal visceral fat loss with metabolic syndrome risk factors in middle-aged women. The walking exercise program was provided at a frequency of 3 sessions, respectively, per week for a duration of 12 weeks. Then, the subjects were classified into either those whose abdominal visceral fat loss belonged to low 25 percentile (low 25% group, n=8) or those whose abdominal visceral fat loss belonged to high 25 percentile(high 25% group, n=8) based on the amount of abdominal visceral fat loss induced by the walking exercise. Metabolic syndrome risk factors(waist circumference, triglycerides, HDL-cholesterol, fasting blood glucose, blood pressure) between groups according to the 12 weekly walking exercise program had no difference. In conclusion, the findings of the present study show that the walking exercise-induced total abdominal fat and visceral fat loss were positively relate to waist and blood pressure(SBP, DBP) and that the walking exercise-induced subcutaneous fat loss were positively relate to waist and blood pressure(SBP).

Antidiabetic Activity of a Chloroform Fraction of Vigna nakashimae in db/db mice (db/db 마우스 동물모델에서 Vigna nakashimae 클로로포름층 분획물의 항당뇨 효능)

  • In this study, we evaluated the antidiabetic effect of a chloroform fraction of a methanol extract of Vigna nakashimae (designated VN) and compared it with that of a water fraction. Both fractions were administrated to eight-week old db/db mice for two weeks, after which the plasma glucose, triglyceride, and total cholesterol levels were measured. The chloroform fraction (VN-C) lowered the fasting glucose and blood glycated hemoglobin in the db/db mice more effectively than those of the water fraction (VN-W). VN-C also improved the glucose tolerance and led to a significant decrease in the plasma levels of free fatty acids and triglycerides. VN-C enhanced the phosphorylation of AMP-activated protein kinase (AMPK) and increased the expression of carnitine palmitoyltransferase-1 (CPT-1) in HepG2 and C2C12 cells more significantly than VN-W. Consistent with AMPK activation, VN-C inhibited cAMP/Dex-stimulated expression of gluconeogenic genes and increased glucose uptake in C2C12. Collectively, these results suggest that VN-C has an antidiabetic effect, which is exerted via AMPK activation, and that this effect is stronger than that of VN-W.

Fermented Chaga Mushroom (Inonotus obliquus) Effects on Hypolipidemia and Hepatoprotection in Otsuka Long-Evans Tokushima Fatty (OLETF) Rats

  • The effects of fermented chaga mushroom (Inonotus obliquus) powder on the lipid concentrations and the activities of liver marker enzymes of serum in genetically diabetic Otsuka Long-Evans Tokushima fatty (OLETF) rats were investigated. Rats were fed a semisynthetic diet supplemented with 50 g/kg chaga mushroom powder (CM) or 50 g/kg fermented chaga mushroom powder (FCM) for 8 weeks (26 to 34 weeks of age). Nondiabetic Long-Evans Toknshima Otsuka (LETO) rats were used as age-matched nondiabetic control animals. Water consumption was significantly higher in the OLETF control than the LETO rats. Water consumption in the FCM-fed OLETF rats tended to be less than in both the OLETF control and CM-fed OLETF rats. Serum concentrations of triglycerides and total cholesterol were significantly higher in the OLETF control rats than in the LETO rats while within the OLETF rat groups, the consumption of FCM resulted in a significantly lower serum triglyceride concentration and slightly lowered serum total cholesterol concentration when compared to the OLETF control and CM-fed rats. The alanine aminotransferase (ALT) activity was significantly higher in the OLETF control than in the LETO rats, but this difference was significantly reduced compared to the CM-fed rats and essentially no difference in the ALT levels was observed between the LETO and OLETF-FCM rats. This observation suggests an adaptive effect of the fermented chaga mushroom in liver function. Livers of the LETO rats showed no histopathological changes, whereas those of the OLETF control rats were characterized by many fat depositions in the central zone of the hepatocytes. The livers of the OLETF CM-fed rats showed less fatty changes compared to the OLETF control rats and fat deposition in the hepatocytes was nearly absent. These results suggest that orally ingested fermented chaga mushroom has a potential beneficial effect on the complications known to occur in the obesity-related non-insulin dependent diabetes mellitus (NlDDM) OLETF rat.

Effect of Dietary Protein Levels, Caffeine and Green Tea on Body Fat Deposition in Wistar Rats (Wistar Rat에 있어서 체지방 축적에 미치는 식이단백질 수준, 카페인 및 녹차의 영향에 관한 연구)

  • This study was performed to investigate the effects of dietary protein levels(5% and 15% PEP), caffeine or green tea powder on body fat deposition in rats. Male weanling Wistar rats weighting the average of 90g were allotted into 6 experimental groups, each of which was fed two different levels of dietary protein with or without caffeine or green tea powder(5p+0.15% caffeine ; 5p+6.1% green tea powder ; 15p+0.15% caffeine ; 15p+6.1% green tea powder) during 8 weeks of the experimental period. Caffeine and green tea powder were supplemented at the levels of 0.15% and 6.1% of experimental diets. The rats fed 5% PEP diet which had received caffeine or green tea powder showed significantly(p<0.01) reduced gain in body weight. The food efficiency of which rats fed both 5% and 15% PEP diet supplemented with 6.1% green tea powder was significantly low(p<0.05, respectively) compared with the control group. Rats fed diets containing 0.15% caffeine and 6.1% green tea powder showed the significant reduction(p<0.01) of hite adipose tissue weight, triglycerides levels of liver and plasma. The addition of 0.15% caffeine or 6.1% green tea powder to 5% PEP diet resulted in significantly(p<0.01) higher levels of plasma total cholesterol, free cholesterol, cholesterol ester, but HDL-cholesterol levels were significantly(p<0.01, in 5% PEP group) high. The atherogenic index(Tchol-HDLchol/HDLchol) in rats fed 6.1% green tea powder diets decreased especially compared with the control group.

Studies on the Composition of Lipid Class and Fatty Acid of Korean Black Soybean (한국 재래 검점콩의 지방질 조성에 관한 연구)

  • Total lipids of Korean black Soybean were extracted, purified and fractionated into three lipid classes (Neutral lipid, NL; glycolipid, GL;phospholipid, PL) and the composition of lipid class and fatty acid were studied. Total lipids of black soybean consisted of 88.5% NL, 2.5% GL and-9.0% PL. In the NL, triglycerides were predominant(80.4%) and also sterol esters, hydrocarbons, diglycerides, free fatty acids and sterols were identified. The major component of GL were monogalactosyl diglycerides and esterified steryl glycosides, and then cerebrosides, steryl glycosides, digalactosyl diglycerides were also found. As major component of the PL, phosphatidyl ethanolamine and phosphatidyl choline were observed, other compounds such as phosphatidyl inositol, phosphatidyl serine and lysophosphatidyl choline were also determined. The major fatty acids in the NL and GL were linoleic acid, oleic acid and palmitic acid, however, PL contained higher relative content of palmitic acid and lower level of oleic acid compared with those of NL and GL.

A Study on the Lipid Components in Oyster Mushroom (느타리버섯의 지방성분(脂肪成分)에 관한 연구(硏究))

  • Lipids in oyster mushroom (Pleurotus florida) were extracted by the mixture of chloroform-methanol (2: 1, v/v) and fractionated into neutral lipids, glycolipids and phospholipids by silicic acid column chromatography. Components and fatty acid composition of each fraction were deter- mined by thin-layer and gas-liquid chromatographies. Fresh oyster mushroom contained 0.5% total lipid in which the contents if neutral lipids, glycolipids and phospholipids were 33.8%, 19.7% and 45. 6%, respectively, Triglycerides(38.2%), free fatty acids (20%) and free sterol (10%) were the major components among the neutral lipids. Diglycerides, monoglycerides, sterol esters and three unidentified neutral lipids were the minor components. Major components of glycolipids were steryl glycosides(35.9%) and esterified steryl glycosides (23.7%). Digalactosyl diglycerides, mono-galactosyl diglycerides and two unknown components were also present. Of the phospholipids, phosphatidyl cholines and serines (48.2%), and phosphatidyl ethanolamines(44.4%) were the major components. On the other hand, the major fatty acids of neutral lipids we.e linoleic, palmitoleic, oleic and palmitic acid. Linoleic and palmitic acid were the predominant fatty acids of both glycolipids and phospholipids.

Effects of Aloe on Liver Function and Lipid Metabolism in Alcohol-Consuming Rats (Aloe가 알코올을 섭취한 흰쥐의 간 기능 및 지질대사에 미치는 영향)

  • The effects of aloe on liver function and lipid metabolic disorders induced by alcohol consumption were studied in rats using aloe power (0.25%, 0.5%, 1%) and 10% ethanol. 35 Sprague-Dawley (male, 4 weeks old) rats were divided into five groups and fed experimental diets for six weeks. Body weights of rats tended to be lower in all alcohol supplemented groups than in the control. Food intakes and dry feces per day were significantly lower in all alcohol supplemented groups than in the control. Atherogenic indices (AI) were highest in the alcohol group and decreased in proportion with aloe amount. Serum triglyceride level was significantly higher in the alcohol group than in the control, but tended to be lower in the aloe supplemented groups. In relation to liver function, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), and alkaline phosphatase (ALP) activities tended to be higher in the alcohol groups than in the control, but lower in the aloe groups, especially in the alcohol+0.5% AO group. The levels of liver cholesterol were significantly lower in the alcohol group than in the control and aloe supplemented groups. In the histochemical evaluation, fat droplets appeared extensively on the liver-lobule in the alcohol group, whereas they decreased slightly in the alcohol+0.25% AO group and apparently disappeared in the alcohol +0.5% AO. On the other hand, fat droplets appeared again on the liver-lobule in the alcohol+1% AO group, but were reduced compared with the alcohol group. Regarding the fatty acid composition of adipose tissue triglycerides, the level of linoleic acid (18:2) was significantly higher in the aloe supplemented group. Regarding the fatty acid composition of liver phosphatidylcholine (PC), the level of linoleic acid was higher in the alcohol group and alcohol+1% AO group than the other groups. In contrast, the level of arachidonic acid was significantly lower in the alcohol group. As a result, arachidonic / linoleic acid ratios were significantly lower in the alcohol group compared to the control group, whereas the ratios of the aloe supplemented groups were similar to that of the control group. Therefore, aloe had some beneficial effects on lipid metabolic disorders induced by alcohol and affected desaturation of fatty acids.
